Serial changes of cerebral blood flow (CBF) in nine patients with thalamic hemorrhage are studied. Measurement of CBF was performed in 3 times (acute, subacute and chronic stage) by the 133 Xe inhalation method using rapidly rotating single photon emission CT (SPECT, Tomomatic 64). The outcome was judged at 16 weeks from onset according to the activities of daily life. All patients divided into two groups (good group and poor group) [Result] 1. Serial changes in mean hemispheric CBF (mCBF) of both hematoma and nonhematoma side between good group and poor group. Between two groups, the significant difference was observed only in hematoma side was 77% CBF in acute stage and it was gradually improved in subacute stage. mCBF of nonhematoma side was normal value in all stage. In poor group, mCBF of hematoma side was 64% CBF in acute stage and it was slightly improved in chronic stage (70% CBF). Slice 2 mCBF of nonhematoma side was 69% CBF in acute stage and it was improved in subacute (74% CBF) and chronic (78% CBF) stage. 2. Serial changes in regional CBF (rCBF)--H (hematoma area).A (anterior area of hematoma).L (lateral area of hematoma).P (posterior area of hematoma) of hematoma side between good group and poor group. Between two groups, the significant differences were observed in rCBF-H.L.P on subacute stage and rCBF-P on chronic stage. Serial changes in rCBF were similar to those in slice 2 mCBF of hematoma side. 3. Serial changes in rCBF-H.A.L.P of nonhematoma side between good group and poor group.(ABSTRACT TRUNCATED AT 250 WORDS)

We investigated the relationships between antitumor activity and the inhibition of Thymidylate Synthase (TS) activity after oral administration of 5-fluorouracil (FUra) and its derivatives, FT, UFT, HC-FU, PH-FU and 5'-DFUR, using mainly Sarcoma 180 as an experimental tumor model. The inhibition of TS activity in the tumor, after oral administration of these drugs to Sarcoma 180 bearing mice, reached the plateau phase shortly after drug administration. The inhibition of TS activity remained at the same level for over 24 hours and was dose dependent. UFT and FT showed a very high correlation between the inhibition of TS activity and their antitumor activity. However, such a correlation was not found for other FUra derivatives despite their high TS inhibiting values. The relationship between the tumor growth inhibition (TGI) and the TS inhibition (TSI) of these drugs, presented as a ratio of ED50 of TGI to ED50 of TSI in case of UFT and FT, was shown to be 0.81 and 1.18, respectively (i.e., nearly 1.00). However, the above ratio for FUra and HCFU was shown to be 2.49 and 3.88, respectively. These findings demonstrate that it is necessary to take into account other mechanisms besides TS inhibition of some fluorinated pyrimidines to explain their total antitumor activity.

Orcein-positive hepatocellular material (OPHM), found in 18 of 44 hepatocellular carcinomas (HCC), was compared histochemically and ultrastructurally with similar granular materials in other liver diseases. OPHM was not found in 15 control livers, 45 metastatic liver cancers or ten cholangiocellular carcinomas. OPHM in HCC was stained with orcein when the tissue sections were preoxidized. In addition, it was positively stained with rhodanine and rubeanic acid for copper. These findings were identical to the OPHM in Wilson's disease, in primary biliary cirrhosis and in neonatal livers. Seven of 12 resected HCC revealed cytoplasmic dense bodies ultrastructurally in which an x-ray microanalyzer demonstrated copper. OPHM in HCC, a granular accumulation, was concluded to be rich in copper-protein complex, and might correspond to dense bodies derived from lysosome ultrastructurally. The pathogenesis and biologic significance of this copper overload in a number of HCC are also discussed.
We investigated the biological activity of IL-4 to murine connective tissue-type mast cells (CTMC). When purified peritoneal mast cells, typical CTMC, were incubated with pokeweed mitogen-stimulated spleen cell-conditioned medium (PWM-SCM) in methylcellulose, about one-fifth of mast cells showed clonal growth. Recombinant IL-4 alone did not stimulate the clonal growth, and purified IL-3 alone induced development of a small number of tiny clusters. In contrast, addition of IL-4 to IL-3 increased the number of clusters by a factor of 10. The number and size of clusters induced by the combination of IL-3 and IL-4 were comparable to those of mast cell clusters induced by PWM-SCM. The present results indicate that IL-4 is an essential factor for in vitro clonal growth of CTMC.
Oxytocin was measured in maternal and fetal plasma, amniotic fluid and neonatal plasma and urine using a specific radioimmunoassay, following extraction procedures with Florisil. Maternal oxytocin levels rose progressively with advancing gestation, but there were no significant differences between oxytocin levels around the onset of labor. No diurnal rhythm of oxytocin was evident in maternal plasma during the third trimester. Maternal and umbilical plasma oxytocin levels at spontaneous delivery were significantly higher than those at elective cesarean section. Maternal oxytocin levels in four cases of post-term delivery were lower than those during normal late pregnancy; all four cases experienced uterine inertia. All amniotic fluid samples had detectable oxytocin levels and there were no significant differences between oxytocin levels in the second trimester and those in the third trimester. Oxytocin levels in neonatal urine were higher than levels in amniotic fluid and lower than in the umbilical artery. Neonatal plasma oxytocin levels gradually decreased and oxytocin levels of 7-day-old infants were significantly lower than those in the umbilical artery, but higher than those in adults. In conclusion, it appears that maternal oxytocin levels may not be involved in triggering the onset of labor but may play a role in the maintenance and reinforcement of labor.
