Functional association of idiotypic and I-J determinants on the antigen receptor of suppressor T cells.

The serological characteristics of the antigen receptor on 4-hydroxy-3-nitrophenylacetyl (NP) specific suppressor T cell hybridomas were analyzed. Three T-cell hybrids could be lysed with anti-idiotype and complement. The reactivity pattern observed from a panel of anti-idiotypic reagents indicated that NPb determinants were detected on all three hybrid lines. NP conjugates with bovine serum albumin or caproic acid specifically inhibited the complement-mediated lysis of these cells by both anti-NPb idiotype and anti-I-J antisera. These hapten conjugates failed to block lysis by anti-Thy 1 or anti-H-2K antisera on the same target cell populations. The data indicate that both I-J and Igh variable region gene products are intimately involved in the recognition of antigen by suppressor T cells. Finally, the suppressor cell hybrids produce soluble factors that mediate antigen-specific suppression. The characteristics of the cells and their factors indicate that the hybrids correspond to the Tsi or first-order suppressor cells in the suppressor cell pathway.

Idiotypic analysis of anti-GAT antibodies. VIII. Comparison of interstrain and allotype-associated idiotypic specificities.

A guinea pig anti-idiotypic antiserum made against pooled specifically purified A/J anti-GAT antibodies was characterized. This antiserum contains anti-idiotypic antibodies specific to interspecies, interstrain, and allotype-linked idiotypic determinants. These idiotypic determinants are associated with the combining sites of idiotypic antibodies that are induced by GT-related but not GA-related antigenic moieties. Genetic and strain distribution studies indicated that the shared allotype-linked idiotypic determinants are controlled by Igh-1e- or Igh-1b-linked genes. The interrelationships of the allotype-linked and interstrain CGAT idiotypic specificities are described using monoclonal anti-GAT hybridoma antibodies. Four of 7 hybridoma anti-GAT antibodies of C57BL/6 origin expressed a major fraction of the idiotypic specificities of A/J anti-GAT antibodies. These 4 hybridoma antibodies also carried the common interstrain idiotype, termed CGAT, but not all CGAT-bearing anti-GAT hybridoma antibodies expressed the allotype-linked idiotypic specificities. The Ig-1b-positive, F17-167.1 hybridoma anti-GAT antibody was used as a ligand to selectively identify the major allotype-linked idiotypic specificities, which were designated Gte idiotype.

Cross-induction of predominant NPb idiotypic antibodies with derivatives of (4-hydroxy-3-nitrophenyl) acetyl.

Immunization of C57BL/6 mice with bovine gamma-globulin (BGG) conjugated with (4-hydroxy-3-nitrophenyl) acetyl (NP) induced a population of anti-NP antibodies that bear predominantly lambda light chain, exhibit heteroclitic affinity for heterologous NP derivatives, and share NPb idiotype. The present study analyzes the idiotypes of antibodies induced with BGG conjugated with the iodo-, bromo-, or nitro-NP derivatives (NIP, NBrP, and NNP). NIP-BGG, NBrP-BGG, and NNP-BGG, induce specific antibodies bearing NPb idiotype in C57BL/6 or C57BL/10 congenic mice, but not in many other inbred strains. Furthermore, the quantity of NPb idiotype in immune sera from various mouse strains immunized with NIP-BGG, NBrP-BGG, and NNP-BGG was similar to that in sera from mice immunized with NP-BGG. Anti-idiotypic antisera against C57BL anti-NP, anti-NIP, or anti-NBrP antibodies exhibit extensive idiotype binding of specifically purified B6 anti-NP, -NIP, -NBrP, and -NNP antibodies. These purified antibodies contain a high percentage (greater than 70%) of lambda-chain-bearing molecules. The data indicate that an extensively shared repertoire composed of predominantly lambda-bearing NPb-positive idiotypic antibodies is used in response to NP and its derivatives in C57BL mice.

Idiotypic analysis of anti-GAT antibodies. IX. Genetic mapping of the Gte idiotypic marker within the Igh-V locus.

The gene controlling the expression of the murine Gte idiotype marker was mapped within the Igh-V gene complex. This idiotypic marker was identified in B6, C.B-20 and A/J and C.AL-20 anti-GAT antisera, but either was not detectable or was present in very low quantities in the anti-GAT antisera obtained from BALB/c, BAB-14, SM/J, NZW, and NZB mice. The data are consistent with the interpretation that the latter 5 strains do not process Igh-Gte genes and the Igh haplotypes of these strains represent examples of crossing over within the Igh complex. Furthermore, the expression of the Gte idiotype(s) is not influenced by the Lyt-3 linked genes that control the kappa light chain polymorphism. Segregation analysis with 52 recombinant inbred strains confirmed the close linkage of Igh-Gte idiotype with Igh-1b locus. The recombination frequency between these loci was calculated to be 1 +/- 0.7 percent. Furthermore, by using B X D recombinant inbred strains, the Igh-Gte gene was mapped within the Igh-V region gene closely linked to the Igh-Np, Igh-Nbp, and Igh-BgI genes.

Genetic control of a shared idiotype among antibodies directed to distinct specificities.

We developed an idiotypic radioimmunoassay system that detects shared idiotypic determinants, termed GTGL idiotype, on antibodies bearing distinct antigen-binding specificities in various mouse strains. Either poly-(Glu, Tyr) (GT)- or poly-(Glu, Lys) (GL)-related determinants are able to induce anti-GT and anti-GL (GTGL)-idiotypic antibodies. Strain distribution studies indicate that GTGL-idiotypic antibodies are readily induced and frequently expressed in antisera obtained from 25 different mouse strains immunized either with GT-related or GL-related polymers. The ability to express GTGL-idiotypic antibodies is a dominant trait and is controlled by Igh-linked gene(s). In addition, we demonstrated that in anticopolymer of L-glutamine acid60- L-alanine30-L-tyrosine (GAT) and anticopolymer of L-glutamic acid54-L-lysine35-L-phenylalanine11(GLphi) antisera, both antibodies uniquely specific to GAT or GLphi, respectively, and antibodies bearing dual specificities for GAT and GLphi, expressed GTGL idiotype. The genetic implications of these findings are discussed. X

Idiotypic analysis of anti-GL phi antibodies. I. Identification and strain distribution of GL-1 idiotypes.

We utilized both the inhibition of antigen binding and direct idiotype binding methods to identify a new set of common idiotype determinants on anti-GL antibodies of various mouse strains. Three anti-idiotypic antisera, each prepared against individually purified B10.WB anti-GL phi antibodies, were able to detect antibody-combining site-associated common idiotypic determinants, designated GL-1 idiotype(s), in antisera with GLT-binding activity obtained from all mouse strains except strains bearing Igh-1e allotype. Anti-GL phi antisera obtained from rabbits, guinea pigs, and rats did not express detectable levels of GL-1 idiotypes. Nonresponder mice to GL phi, upon immunization with GL-F gamma G or GL phi-F gamma G produced anti-GL antibodies expressing GL-1 idiotypes. Although the magnitude of the immune response to various GL-containing polymers is controlled by distinct Ir genes, the common GL-related antigenic determinants on these polymers are able to induce anti-GL antibodies with GL-1 idiotypic specificities.

Idiotypic analysis of anti-GAT antibodies. VII. Common idiotype on hybridoma antibodies to poly(Glu60 Ala40)

A single DBA/2 mouse, immunized with L-glutamic acid60-L-alanine40 (GA), was used to produce hybridoma cell lines. Seven hybridoma anti-GA antibodies were obtained for idiotypic analyses. Two hybridoma anti-L-glutamic acid60-L-alanine30-L-tyrosine10 (GAT) antibodies, preferentially reactive to GA, were studied in parallel. Anti-idiotypic antisera to purified anti-GAT and anti-GA serum antibodies and to hybridoma anti-GA antibodies were analyzed by idiotype binding and inhibition of idiotype binding assays. Five of the nine hybridoma antibodies exhibited common GA-1 idiotypic specificities previously demonstrated on the majority of anti-GA antibodies of inbred mouse strains of differing immunoglobulin heavy chain linkage groups; these hybridoma antibodies also possessed private idiotypic determinants. Two GA-1 negative hybridoma anti-GA antibodies appeared identical by immunochemical criteria, arguing that somatic hybridization does not artifactually generate private idiotypic determinants. The results demonstrate that the common GA-1 idiotype system is associated with a family of nonidentical but idiotypically related antibody molecules present in a single DBA/2 mouse, and these antibodies are part of the "GA-1 idiotypic family".

Hapten-specific T-cell responses to 4-hydroxy-3-nitrophenyl acetyl. II. Demonstration of idiotypic determinants on suppressor T cells.

The ability of NP-coupled syngeneic spleen cells to induce antigen-specific T-suppressor cells capable of binding to NP-BSA-coated Petri dishes and mediating transfer of specific suppressive activity to NP was demonstrated. Furthermore, in strains of mice bearing the Ig-1b allotype, including SJL, and in (non-Ig-1b x Ig-1b)F1 hybrids, the NP-specific suppressor cells also interferes with expression of immunity after priming with NIP-BGG. Anti-NPb anti-idiotype antiserum plus complement treatment effectively abrogated the ability to transfer suppression. Formal genetic mapping of the fine specificity of cross-reactivity with Ig-1 allotypic congenic mice implies that expression of this trait is linked to the Ig-1b heavy chain linkage group. The sensitivity of NP-suppressor cells of appropriate strains to anti-idiotype treatment was also consistent with the formal mapping data. These experiments suggest that there are shared V-region structures on antibody and T cells that are crucial in the suppression pathway for the same antigen.

Shared idiotypic determinants on antibodies and T-cell-derived suppressor factor specific for the random terpolymer L-glutamic acid60-L-alanine30-L-tyrosine10.

T-cell derived suppressor factors (TsF) specific for the random copolymers L-glutamic acid60-L-alanine30-Ltyrosine10 and L-glutamic acid60-L-alanine40, referred to as GAT and GA, respectively, were prepared and partially purified on the approprate antigen immunoadsorbents. GAT-TsF obtained from nonresponder DBA/1 (H-2q) and SJL (H-2s) mice were passed over immunoadsorbents prepared from normal guinea pig serum (NGPS) or guinea pig anti-idiotype antiserum (anti-CGAT) specific for a common cross-reactive idiotype found on most anti-GAT antibodies in all mouse strains tested. Both the directly suppressive activity of the GAT-TsF and the ability of GAT-TsF to induce new suppressor T cells (Ts2) in vitro were adsorbed to and fully recoverable from the guinea pig anti-CGAT-Sepharose immunoadsorbent, while the TsF passed through the control NGPS-Sepharose without appreciable binding. The SJL GAT-TsF specifically eluted from anti-CGAT-immunoadsrobents was shown to still posses I-J determinants. These data provide evidence suggesting a sharing of V region structures between B-cell antibody and T-cell suppressor factor specific for an antigen (GAT) under Ir gene control, in agreement with earlier studies on T and B-cell alloreceptors, T-cell helper factors, and T and B-cell receptors for conventional antigens.

Idiotypic analysis of antibodies against the terpolymer L-glutamic acid60-L-alanine30-L-tyrosine10 (GAT). IV. Induction of CGAT idiotype following immunization with various synthetic polymers containing glutamic acid and tyrosine.

The immune responses of all inbred strains of mice specific to the synthetic terpolymer poly(LGlu60LAla30LTyr10), referred to as GAT10, are characterized by the presence of anti-GAT antibodies which share a common (CGAT) idiotype. In this report, we describe the ability of the synthetic polymers, LGlu33LAla33LTyr33, LGlu51-LAla34LTyr15 and poly-L(Tyr, Glu)-DLAla--LLys [(T,G)-A--L] to induce antibodies with CGAT idiotypic specificities. All of these polymers contain "GT"-related determinants. Following immunization with these polymers, antisera from responder mice bind the corresponding 125I-labeled antigen and 125I-labeled poly(LGlu50LTyr50) or GAT10. These antisera shared the CGAT idiotype which is associated with the antibody fraction with binding specificity for GAT10. Collectively, the present results indicate that GT-related determinants are required for the induction of the CGAT idiotype. Moreover, since the immune responses to these synthetic polymers are under distinct H-2-linked immune response (Ir) gene control, a mouse strain can be nonresponder to one polymer and responder to another; in this case, only the latter polymer induces CGAT idiotype. Thus, although the immune responses of inbred strains of mice to different polymers are under distinct Ir gene control, the antibody responses can be idiotypically related.

Fine specificity of antibodies to poly(Glu60Ala30Tyr10) produced by hybrid cell lines.

The polyethylene glycol-mediated cell fusion technique has been used to analyze the diversity of the antibody response to the terpolymer poly(Glu60Ala30Tyr10)(GAT). Nine stable clones (all producing IgM K anti-GAT antibodies) were isolated from a fusion between P3-X63-Ag8 myeloma cells and spleen cells from a DBA/2 mouse sensitized to GAT 5 days earlier. Seven other clones (producing IgG K anti-GAT antibodies) were derived from another fusion between NSI myeloma cells and spleen cells of (C57BL/6 X DBA/2)F1 hybrid mice hyperimmunized with GAT. These 16 anti-GAT antibodies were grouped according to their pattern of reactivity with GAT and the two related polymers of poly(Glu60Ala40) (GA) and poly(GLU50Tyr50) (GT). Two monoclonal anti-GAT antibodies (IgM F9-102.2 and IgG F17-148.3) demonstrated crossreactivity with GA but failed to crossreact with GT determinants. In contrast, the remaining 14 hybridoma antibodies demonstrated preferential reactivity with GAT but also exhibited crossreactive binding to GT and in some cases GA. There was a correlation between the fine specificity pattern and the presence of a common anti-GAT idiotype on these antibodies. Thus, the hybridoma anti-GAT antibodies which reacted with GT shared crossreactive idiotypic determinants (CGAT) present in mouse anti-GAT immune sera. In contrast, the monoclonal F9-102.2 and F17-148.3 antibodies that failed to bind to GT lacked the major CGAT idiotypic determinants.

Idiotypic analysis of monoclonal antibodies to poly(Glu60Ala30Tyr10).

Fifteen hybridoma anti-poly(Glu60Ala30Tyr10) (anti-GAT) antibodies were analyzed for the presence of a common set of idiotypic specificities associated with murine anti-GAT antibodies, termed CGAT idiotype, which are present on the anti-GAT antibodies of all mouse strains. Thirteen of these monoclonal anti-GAT antibodies expressed a major fraction of CGAT idiotypic specificities. However, the remaining fraction of CGAT idiotypic specificities were not detected in individual or pooled hybridoma anti-GAT antibodies. Anti-idiotypic antisera made against each of the 15 hybridoma anti-GAT antibodies preferentially bound homologous ligand and showed minimal binding activity to specifically purified serum anti-GAT antibodies. Furthermore, the diversity of the hybridoma anti-GAT antibodies was demonstrated by the presence of individual idiotypic specificities on each of the hybridoma anti-GAT antibodies. However, relatedness among some of the hybridoma antibodies was also apparent since idiotypic analysis revealed that some hybridoma anti-GAT antibodies shared cross-reactive idiotypic specificities not associated with CGAT idiotype. The genetic mechanisms which could account for the generation of such antibody diversity are discussed.

Idiotypic analysis of anti-GAT antibodies. III. Determinant specificity and immunoglobulin class distribution of CGAT idiotype.

The humoral response to poly-(L-glutamic acid60, L-alanine 30, L-tyrosine10), GAT, in mice is further characterized by both idiotype and fine specificity analyses. The common idiotype on murine anti-GAT antibodies (CGAT) was identified in anti-GAT antisera from seven additional strains of mice. These data confirm that the CGAT idiotype can be induced in all inbred strains of mice. Using a partially inbred strain of mice selected for their ability to respond to poly-(L-glutamic acid50, L-tyrosine50), GT, we demonstrated that the GT copolymer is capable of inducing antibodies that express the CGAT idiotype. In contrast, antisera directed against poly-(L-glutamic acid60, L-alanine40), GA, bind GAT but lack CGAT idiotype. These results indicate that GAT molecules contain determinants either similar or identical to those on GT molecules, which are responsible for the induction of CGAT idiotypic antibodies. We also demonstrated that both GAT responder and nonresponder strains of mice can produce anti-GAT antibodies with similar fine specificity patterns and CGAT idiotype. In addition, we demonstrate that antibodies of the IgM, IgG1, and IgG2 immunoglobulin classes express the CGAT idiotype.

Idiotypic analysis of antibodies to poly(Glu60Ala30Tyr10): interstrain and interspecies idiotypic crossreactions.

A common idiotype was defined by a guinea pig anti-idiotypic antiserum made against D1.LP antibodies specific to the synthetic terpolymer poly(Glu60Ala30Tyr10), referred to as GAT. This idiotype was found in the anti-GAT antibodies of all individuals of all inbred strains of mice tested. Due to its common (C) occurrence in the anti-GAT antibodies of all mouse strains, it was termed the CGAT idiotype. The presence of CGAT idiotype in mice is independent of the expression of various Ig-1, H-2, or Vg genetic markers. In contrast, antibodies produced in response to the crossreactive poly(Glu60-Ala40), termed GA, bound to the GAT ligand but did not bear the CGAT idiotype. This suggests that GAT, although bearing GA determinants, preferentially stimulates clones of CGAT idiotype. Some of the CGAT idiotypic specificities were also identified in sera of WF inbred rats after immunization with GAT. However, no significant levels of CGAT idiotype were detected in LEW rats, F344 rats, guinea pigs, or rabbits, although measurable quantities of anti-GAT antibody were present.

Idiotypic analysis of anti-GAT antibodies. I. Presence of common idiotypic specificities in both responder and nonresponder mice.

The synthetic terpolymer of L-glutamic acid60-L-alanine30-L-tyrosine10 (GAT) induced anti-GAT antibodies in D1.LP mice. Some of these antibodies shared common idiotypic specificities. The interaction between idiotypic antibody and anti-idiotypic antiserum was specifically inhibited by GAT and the closely related polymer of L-glutamic acid and L-tyrosine (GT) but not by the copolymer of L-glutamic acid and L-alanine (GA). The binding of labeled GAT by the anti-GAT antibodies was also specifically inhibited by the anti-idiotypic antiserum. These results indicated a close association of idiotypic determinants with the antibody-combining sites. Furthermore, such active site-related common idiotypic specificities were found in individual sera of both responder mice (D1.LP) and nonresponder mice (DBA/1) immunized with GAT and GAT-MBSA, respectively, indicating that both responder and nonresponder mice carry B cells with common clonotypes.
