Fatty acyl-CoA synthetase purified from rat liver microsomes was immobilized on either CNBr-activated Sepharose 4B or activated CH-Sepharose 4B, and the enzymatic activities of the syntheses of CoA esters from lignoceric acid (C24:0) and palmitic acid (C16:0) were studied and compared. The ratio of activities of the synthesis of lignoceroyl-CoA to palmitoyl-CoA increased 4.5 fold with CH-Sepharose, but only slightly with CNBr-Sepharose. The effects of a detergent and chaotropic agent on both substrates were significantly altered by the immobilization. The results of this study thus indicate that the stability and fatty acid specificity of fatty acyl-CoA synthetase are significantly affected by the physical state of the enzyme.
As part of a long-term study of sphingolipid metabolism in brain, we have purified and partially characterized a long-chain acyl-CoA synthetase from microsomes of developing rat brain and compared it with the hepatic microsomal enzyme from the same animals. Both enzymes were solubilized from microsomes by treatment with Triton X-100 and then chromatographed successively on Blue-Sepharose and DEAE-Sepharose. Blue-Sepharose chromatography yielded a single peak with acyl-CoA synthetase activity, whereas DEAE-Sepharose chromatography of both brain and liver preparations yielded two peaks. Elution patterns of lignoceroyl-CoA synthetase and palmitoyl-CoA synthetase activities were identical throughout these steps and were similar in brain and liver. Gel filtration of each DEAE-Sepharose fraction on Sephadex G-200 also yielded two peaks of activity. The more rapidly eluted material contained much more lignoceroyl-CoA synthetase activity, while the activity for palmitoyl-CoA synthetase was higher in slower eluting peaks. In all preparations the ratio of lignoceroyl-CoA synthetase activity to palmitoyl-CoA synthetase activity was much higher in brain than in liver. These results suggest that although the brain acyl-CoA synthetase is chromatographically similar to the liver enzyme, there are differences in substrate specificity.
Two cases are presented in whom full-thickness defects of the abdominal wall were repaired by combined groin-tensor fasciae latae flaps nourished only by the superficial circumflex iliac artery. The dissection of the flap is simple and it will probably cover even the upper parts of the flank.
This case report illustrates the use of a combined scapular and parascapular flap to cover a large defect of the lower leg. This flap has great coverage potential and a constant vascular anatomy.

The authors report a case of solitary metastasis to the pancreas from an intracranial angioblastic meningioma (hemangiopericytoma) and discuss secondary (metastatic) tumors of the pancreas. A 65-year-old man developed a pancreatic tumor 15 years after the craniotomy was performed for a parasagittal meningioma. Histological investigation of the pancreatic tumor excised as a biopsy on exploratory laparotomy revealed a tumor identical to the intracranial meningioma. It was concluded that the intracranial angioblastic meningioma had recurred and metastasized to the pancreas.

Of 16 human malignant tumor cell lines established in our laboratory, seven lines, including three gastric cancer cell lines derived from patients with cancer, were found to carry carcinoembryonic antigen on their cell surface, as determined by radioimmunoassay and indirect immunofluorescence. The existence of antibodies (IgG class) against the gastric cancer cell lines (HPE-GAC-T, -2, -3) and lung cancer cell line (HPL-Ad-K) in the sera of patients with gastrointestinal cancer (incidence 70.8%) was demonstrated by indirect immunofluorescence. In nonmalignant cases and healthy controls, the incidence was 7.7 and 3.2%, respectively. Specificity of the antibodies detected in the sera of patients with gastric cancer was examined by absorption and blocking test methods of immunofluorescence. Even though there was an apparent heterogeneity of the specificity among the antibodies, the detection of such antibodies may be a feasible and practical approach to a clinical diagnosis of malignancy.
Using abdominal skin flaps in experimental rats, we created flaps that exhibited relatively constant necrosis in the distal side. The existence of arterial inflow had a marked effect on the survival of the distal part of these flaps. We postulate that the so-called delay mechanism produces the arterial inflow as a consequence, although how this occurs is still unsolved. We suggest the possibility that a large flap may be transposed in one stage with vascular anastomosis in the distal part--preferably artery and vein, or artery only.
Using the abdominal skin flap of rats, we showed that the arterial inflow through the venous system nourished the distal part of a flap and also worked as a pedicle of an island flap and a free flap. The position of the arteriovenous anastomosis and the draining vein seems important. This study implies that delay produces arterial inflow as a consequence.
