The effect of thrombin activated factor XIII, thrombin and plasmin on the chemiluminescence produced by human neutrophils stimulated by opsonized zymosan (Mannozym).

Coagulation factor XIII formed by thrombin activation from zymogen factor XIII decreases the chemiluminescence (CL) of human neutrophils stimulated by opsonized zymosan (Mannozym). At high concentrations, thrombin and plasmin also decreased the CL induced by opsonized zymosan. The inhibitory effect of all the three enzymes was due to their influence on the cell membrane receptors (C3b and Fe) and not to their direct effect on opsonized Mannozym. The potential clinical role of factor XIII, thrombin and plasma in the regulation of neutrophil functions is assumed.

Stimulation of phagocytosis by human lymphokines.

Supernatants of concanavalin A stimulated human lymphocytes increased the phagocyting and intracellular killing activities of human macrophages and neutrophils tested with particles of Saccharomyces cerevisiae.

Effects of lymphokines on fibrinolysis and kinin generation in human plasma.

Supernatants from Con-A-stimulated human lymphocytes containing leukocyte migration inhibitory factor, skin reactive factor and procoagulant activities were tested for their effect on fibrinolysis and kinin generation. The supernatants of stimulated lymphocytes activated the fibrinolysis and generated kinin in human plasma. These results are discussed from the aspect of cell-mediated (type IV) immune inflammation.

Macrophage disappearance reaction induced by lymphokines.

Macrophage disappearance reaction was induced in normal (non-sensitized) guinea pigs by human lymphokines deriving from Con A-stimulated lymphocytes, by ellagic acid (an activator of intrinsic coagulation) and by thromboplastin (an activator of extrinisic coagulation). The results are discussed from the aspect of the pathomechanism of macrophage disappearance reaction.

Macrophage disappearance reaction in Rana esculenta induced by specific antigen and rabbit lymphokines.

Macrophage disappearance reaction was induced by intraperitoneal injections of specific antigens in Rana esculenta sensitized by bovine gamma globulin and Mycobacterium bovis BCG. Rabbit lymphokines derived from concanavalin A stimulated blood lymphocytes injected intraperitoneally were able to induce macrophage disappearance reaction in normal Rana esculenta. This suggests that mammalian lymphokines are capable of acting in amphibia. Mammalian lymphokines have not an exclusive class-specificity in vertebrates.

The effect of colloidal gold on blood coagulation.

Coagulation tests were carried out in rabbits after the intravenous administration of 2 mg/kg colloidal gold. Among the clotting factors, platelet Factor 3 and Factor XIII were found to be inhibited by the gold preparation. Gold at 2 mg/ml concentration induced an overall antithrombotic state in spite of an increased tendency to platelet aggregation. These experiments suggest a new possibility for the inhibition of Factor XIII and platelet Factor 3.

Local effect of single stranded polyadenylic acid on passive cutaneous anaphylaxis in mice.

Single stranded polyadenylic acid (Poly A) administered locally inhibited passive cutaneous anaphylaxis in mice. In experiments performed by equilibrium dialysis Poly A was able to bind histamine. The association constant of the reaction was determined, Ka = 1.3 +/- 0.2 x 10(5) I/M. One Poly A molecule can bind maximally two molecules of histamine dichloride. Poly A inhibited the antigen-induced release of histamine from the peritoneal rat mast cells when it was given together with sensitizing antibodies.

Effect of hypothermia and endotoxin on phagocytosis.

Experiments with colloidal 198Au indicated that gold clearance highly decreased in rabbits cooled to 29-30 degrees C. The capacity of storing colloidal gold decreased especially in the spleen and liver. Endotoxin reduced the gold clearance in normothermic rabbits; the decrease was more marked in hypothermic animals.

Effect of aurothioglucose on some allergic and non-allergic skin reactions in animals.

The antiphlogistic effect of aurothioglucose was examined in reversed passive Arthus reaction, delayed-type hypersensitivity and carrageenan oedema. A close association existed between the antiphlogistic effect of aurothioglucose and the pathogenetic role of polymorphonuclear leukocytes. In reversed passive Arthus reaction and in carrageenan oedema the drug decreased the inflammation considerably, whereas in delayed hypersensitivity, where the leukocytes play a less important part, it exerted only a mitigating effect.
