Type VIII collagen is upregulated after vessel injury, and this collagen has been implicated in both smooth muscle cell migration and angiogenesis. This study examines the temporal and spatial pattern of expression of type VIII collagen in porcine coronary vessels at specific time points after balloon angioplasty. In situ hybridization studies demonstrated that collagen VIII messenger ribonucleic acid (mRNA) was markedly elevated in the neoadventitia at 3 days post-angioplasty. By 14 days, elevated collagen VIII message was seen mainly in the neointima and this expression decreased to background levels by 90 days. The distribution of collagen VIII protein, detected using immunohistochemistry, was similar but the up-regulation lagged behind the mRNA increase by a few days. Pre-treatment of sections with pepsin highlighted variations in the organization and appearance of extracellular collagen VIII containing structures in both injured and normal vessels. New vessel formation was evident in the neoadventitia after 3 days, but there was no colocalization of type VIII collagen immunostaining with that of von Willebrand factor (a marker of endothelial cells) in the neoadventitia. These data show that up-regulation of collagen VIII in the neoadventitia is an important early marker of the coronary arterial response to injury, and is not associated with new vessel formation.
Fifty patients underwent a variety of hand operations and were randomized for wound closure either with tissue adhesive (Indermil) or sutures. The two treatment groups had similar demographic characteristics and similar outcomes at the 2 and 6 week postoperative assessments which were performed by a designated tissue viability nurse blinded to the method of closure. Five minor wound dehiscences occurred: three in the adhesive group and two in the suture group. No infection occurred in either group. In conclusion, the study demonstrates tissue adhesive is as effective as suture in this type of hand surgery.
Dysembryoplastic neuroepithelial tumor (DNT) is a recently recognized tumor entity with distinctive clinicopathological features and an excellent long-term prognosis. We report 10 cases of DNT out of neurosurgical specimens sent for histopathological examination since 1994. Epilepsy surgery is not done at our center, and all the cases were sent with a clinical diagnosis of glioma. On histopathological examination, all the 10 cases contained the specific glioneuronal element. In 6 cases, glial nodules were noted in addition. This study therefore reports 4 cases of simple DNT and 6 cases of complex DNT. There was no nuclear atypia or mitosis in any of the cases. Proliferative index by PCNA varied from negative to <1%. It is very important to recognize this entity, as surgery cures the patient, and radiotherapy or chemotherapy is not required.
The methanol extract of the rhizome of Bergenia ciliata Sternb. (Saxifragaceae) has been evaluated for its potential in a cough model induced by sulphur dioxide gas in mice. The extract exhibited significant anti-tussive activity in a dose-dependent manner, as compared with control. The antitussive activity of the extract was comparable to that of codeine phosphate (10 mg/kg body wt.), a standard anti-tussive agent. The extract at doses of 100, 200 and 300 mg/kg body wt. (p.o.) showed significant inhibition of cough reflex by 28.7, 33.9 and 44.2%, respectively, within 90 min of the experiment.
The typical output of many computational methods to identify binding sites is a long list of motifs containing some real motifs (those most likely to correspond to the actual binding sites) along with a large number of random variations of these. We present a statistical method to separate real motifs from their artifacts. This produces a short list of high quality motifs that is sufficient to explain the over-representation of all motifs in the given sequences. Using synthetic data sets, we show that the output of our method is very accurate. On various sets of upstream sequences in S. cerevisiae, our program identifies several known binding sites, as well as a number of significant novel motifs.
HYPOTHESIS: High-resolution magnetic resonance imaging (MRI) at 1.5 T preferentially enhances the perilymph over endolymph after administration of contrast with gadodiamide, which allows for differentiation of the membranous labyrinth. Furthermore, this imaging allows for the detection of endolymphatic hydrops. BACKGROUND: Endolymphatic hydrops is believed to be associated with a number of ear diseases, including Ménière's disease. Although the pathologic changes of overaccumulation of endolymph in the inner ear are obvious on postmortem histologic examination of the temporal bone, they have yet to be observed in a living organism. Previous attempts to visualize this condition with high-resolution contrast-enhanced computed tomographic imaging and MRI have been unsuccessful. METHODS: Healthy pigmented guinea pigs underwent a unilateral surgical ablation of the endolymphatic sac to create endolymphatic hydrops in the ear. High-resolution temporal bone imaging was performed by use of a 1.5-T MRI system. Two-dimensional images were acquired by a spin-echo technique with and without contrast enhancement by gadodiamide. RESULTS: T1-weighted gadodiamide contrast-enhanced MRI of the midmodiolar level of the cochlea demonstrated that the perilymph appeared to be preferentially enhanced relative to the endolymph, resulting in a clear distinction between the scalae of the inner ear. The contrast-enhanced T1-weighted MRI of the midmodiolar level of the hydropic cochlea demonstrated a significantly enlarged scala media in comparison with the normal cochlea in the same animal. CONCLUSION: The demonstration of endolymphatic hydrops has been possible for the first time in vivo by the use of a standard 1.5-T MRI system. This research has important clinical implications: Specifically, this technique may allow for the noninvasive diagnosis of Ménière's disease.

The methanol extract of the rhizome of Bergenia ciliata Sternb. (Saxifragaceae) has been evaluated for anti-inflammatory potential using two acute rat models (carrageenan- and serotonin (5-HT)-induced rat paw oedema) and a chronic rat model (cotton pouch-induced granuloma). Phenylbutazone (100 mg kg(-1)), a non-steroidal anti-inflammatory agent, was used as a standard. The methanol extract (100, 200 or 300 mg kg(-1)) exhibited significant (P < 0.05) anti-inflammatory activity in all the animal models. At 300 mg kg(-1) the methanol extract exhibited maximum inhibition of 32.4+/-2.89% in carrageenan-induced rat paw oedema while the standard showed an inhibition of 44.1+/-2.7% after 3 h of drug treatment. In the serotonin-induced rat paw oedema model, 300 mg kg(-1) methanol extract suppressed oedema by 45.33+/-2.09%, whereas the standard produced an inhibition of 53.5+/-4.3%. In the cotton pouch granuloma model the methanol extract inhibited significantly (P < 0.001) the granuloma weight in a dose-dependent manner. In this model, 300 mg kg(-1) extract produced a maximum inhibition of 31.4+/-1.09% in granuloma weight compared with 41.1+/-1.32% reduction in granuloma weight for the standard. The methanol extract of B. ciliata exhibited significant anti-inflammatory potential at the dose levels examined.
Clostridium septicum gas gangrene (myonecrosis) is an acutely painful and rapidly fatal infection occurring in the absence of trauma. Urgent surgery is essential both to control pain and to ensure survival. Most patients who develop this infection have an underlying malignancy and clinicians should be aware of this association. We present a case of bifocal myonecrosis which to our knowledge has not been reported previously.
For ease of detection, soluble forms of phage-displayed scFv antibodies are usually expressed with a tag, e.g., c-myc or His (Histidine). The binding is then assayed by a monoclonal antibody to the tag. In this article, we describe the use of biotinylated antigen for detecting soluble scFv antibodies without utilizing the peptide tag detection system. The scFv antibodies were against the oncoplacental antigen heat-stable alkaline phosphatase (HSAP). The method essentially consisted of either reverse Western or antigen capture enzyme-linked immunosorbent assay (ELISA). In the reverse Western, periplasmic extract was electrophoresed, and binding to biotinylated antigen was detected by the detection system based on streptavidin-horseradish peroxidase. The antigen capture ELISA utilized the binding of periplasmic extract to a polystyrene plate. We have also demonstrated the use of antigen capture ELISA for studying specificity and affinity of the selected clones. Although these techniques have been developed for antibodies to HSAP, they have general utility for phage expression systems without a peptide tag.
The present paper discusses QSAR studies on antimalarial 2,4-diamino-6-quinazoline sulfonamide derivatives using electronic parameters, namely energy of highest occupied molecular orbitals (EH), energy of lowest unoccupied molecular orbitals (EL) and charge density (CD). The results have shown that better results are obtained by introducing dummy parameters (indicator parameter), Ip. Excellent results are obtained when all the four parameters (EH, EL, CD and Ip) are used in correlation analysis.

A rare case of a young man with a diffusely calcified oligo astrocytoma in right parieto occipital region, which spread along the axonal fibres into right temporal lobe and to the left parietal lobe is presented. Computed tomography (CT) and magnetic resonance imaging (MRI) demonstrated the extent of the lesion. The mass was partially resected.
A case of glioblastoma multiformis (GBM) where lesion appeared within a month of normal CT scan is reported. It emphasises the importance of early CT scan after neurological deterioration particularly in older patients because of rapid growth of glioblastoma.
A case of medulloblastoma, which disappeared following steroid administration and a single sitting of radiotherapy (8Gy) is presented. At surgical exploration tumour was not found, multiple biopsies were negative. Post operative CT scan too did not show the tumour. Five months later, tumour reappeared in left cerebellar lobe in cystic and solid form. Patient was reoperated and tumour was decompressed. The biopsy showed medulloblastoma. To our knowledge, medulloblastomas are not known to be completely responsive to either of these modalities.
