[Tissue distribution of cefminox in beagle dogs].

A new cephamycin antibiotic, cefminox (MT-141, CMNX), was intravenously infused into Beagle dogs at a dose of 40 mg/kg in order to study it's distribution to various tissues. The following results were obtained. The maximum serum concentration of CMNX observed at the end of the infusion period was 102.3 micrograms/ml, and then the concentration decreased. The biological half-life of CMNX in serum was 37.0 minutes. This half-life was similar to the results of previous studies with Beagle dogs and rabbits. The maximum concentrations in tissues and body fluids were highest in B-bile followed by kidney, urinary bladder, serum, liver, vagina, uterus, pericardiac fluid, trachea, ovary, lung, gallbladder, parotid gland, heart, tonsil, thymus, spleen, pancreas, aqueous humor and cerebrospinal fluid, in that order and not detected in brain. The maximum concentrations in gallbladder, B-bile, pericardiac fluid and cerebrospinal fluid were found at 1-2 hours after administration. In other tissues and body fluids, they were obtained at the end of the infusion period. The area under the tissue concentration curve (AUC) was highest in the urinary bladder followed by the kidney, vagina, liver, uterus, gallbladder, trachea, ovary and lung, in that order. These results suggest that CMNX is useful for various infectious diseases in these tissues. The pharmacokinetic parameter (K1i/K2i) derived from serum and tissue concentrations using the deconvolution method well correlated to maximum tissue concentrations.

A novel method to predict the elimination half-lives and the renal excretion mechanisms of cephalosporins.

A novel method was proposed to predict the elimination half-lives of cephalosporins from plasma protein binding (unbound fraction, f) and fraction of the dose excreted into urine (f*) on the basis of the following four assumptions. 1) The drug is only distributed to the extracellular fluid, 2) the bound fraction of the drug in plasma is independent of the plasma drug concentration, 3) the binding protein of the drug is albumin, 4) the unbound drug in plasma is excreted by the glomerular filtration and the contribution of active secretion and reabsorption is negligible. The VSS's and t1/2 beta's of MT-141, one of cephalosporins, in rabbits, dogs and healthy human subjects were well predicted, whereas in rats, the prediction of the both values was failed. The t1/2 beta's of various cephalosporins in healthy subjects were calculated from f and f*, in reasonably good agreement with the observed ones, except for some cephalosporins which have been reported to be secreted actively in the renal tubules. Thus, the comparison of the calculated t1/2 beta's with the observed ones makes it possible to presume the renal excretion mechanism. Moreover, this method will be applicable to other drugs which satisfy the above four assumptions.

Liposomal membranes. VI. Polysaccharide-induced aggregation of multilamellar liposomes of egg lecithin.

Induced aggregation of multilamellar liposomes of egg lecithin with several polysaccharides, such as pullulan, dextran, hydroxyethylstarch, and amylopectin, has been investigated in conjunction with structural characteristics of polysaccharides, incubation temperature, ionic strength, and surface charge of liposomes. Aggregation of liposomes as a function of time was followed by monitoring turbidity of the liposome suspension at 600 nm. The inverse relationship between the minimal concentration and molecular weight of polysaccharides for the induced aggregation of liposomes was observed for all the polysaccharides employed. Polysaccharides having larger excluded volume more easily bind to liposomes, leading to the acceleration of the aggregation of liposomes. Hence, the order of efficiency of polysaccharides for the promotion of liposome aggregation was pullulan greater than dextran greater than hydroxyethylstarch congruent to amylopectin. Increasing the surface charge of liposomes brought about a decrease in aggregation irrespective of the sign. Even when negatively charged liposomes were incubated with the cationic DEAE-dextran, the liposome aggregation was depressed. These results including salt effect and temperature dependency of the induced aggregation of liposomes supported a plausible mechanism for the reaction, the polysaccharide-bridging mechanism, which has been provided for the polysaccharide-promoted agglutination of human erythrocytes.

Radical mechanisms for 1,5-dihydroflavin reduction of carbonyl compounds.

A discussion of the expected mechanism of radical reduction of carbonyl compounds by dihydroflavin as it relates to the structure of the carbonyl compound is provided. Factors which must be taken into account are the free energies of formation of the radical anions (CR2O), THE ACIDITY OF THE CONJUGATE ACID OF THE CARBONYL FUNCTION (R2C negative charge OH), and the stability of the carbanion species of the product [(-)CR2(OH)]. It is proposed that in those instances where the product alcohol can flavin radical to CR2O or CR2OH occurs, otherwise H transfer is the terminal step for dihydroflavin reductions. The free energy of formation of the radical species CH2OH obtained by acid-catalyzed electron transfer from dihydroflavin (Fred) to formaldehyde is shown to be less than the experimentally determined free energy of activation (increment F is not equal to exp) for the reduction of formaldehyde by dihydroflavin (i.e., Fred plus CH2O leads to Fox plus CH2OH). Therefore, the radical pair composed of dihydroflavin radical species serve as an intermediate in the reduction. Our proposed mechanism is: Fred plus CH2O ka[H3O] forms k-a[H2O] Frad CH2OH leads to kc Fox plus CH3OH. The value of kaah/k-a has been obtained from the calculated standard potential Eo' for Fred plus CH2O plus H forms Frad plus CH2OH. Assuming ka to represent a two-step process (i.e., Fred plus CH2O plus H k'a forms k'-a Fred plus CH2OH kb forms k-b Frad CH2OH, where ka equals (k'akb)/(k'-ak-b), the value of k'-a will equal 10-10 M-minus 1 sec-minus 1 and kb 10-9 M-minus 1 sec-minus 1. From k'a/k'-a and kb there can be computed the expected value of increment F is not equal to calc as a function of pH. Comparison of increment F is not equal to calc to increment F is not equal to exp reveals that increment F is not equal to calc varies from increment F is not equal to exp by only about 2kcal mol-minus 1 (8.4 kJ mol-minus 1) between pH 5 and 9. Similar considerations establish that radical intermediates should serve eminently well in dihydroflavin reduction of ethyl pyruvate, pyruvic acid, etc. In these cases, 1 e transfer should compete with H transfer to yield the carbanions as the immediate products. Similar comparisons suggest that dihydronicotinamide reduction proceeds via RPyH plus C negative charge O forms RPyH plus C-0minus +H forms -H RPyH plus C-OH forms RPy plus HC-OH.

Incorporation of an artificial receptor into a native protein: new strategy for the design of semisynthetic enzymes with allosteric properties.

The sugar-facilitated structure and enzymatic activity change of engineered myoglobins bearing a phenylboronic acid moiety, which were semisynthesized by a cofactor reconstitution method, were studied by the denaturation experiment, spectrophotometric titration of the pKa shift of the axial H2O, circular dichloism (CD), and the kinetics of the myoglobin-catalyzed-aniline hydroxylation reaction. Both boronophenylalanine-appended myoglobin [Mb(m-Bphe)2] and phenylboronic acid-appended myoglobin [Mb(PhBOH)2] were stabilized by approximately 2 kcal/mol upon complexation with D-fructose. CD spectral changes and the sugar-induced pKa shift suggested that the microenvironment of the active site of these myoglobins was re-formed from a partially disturbed state to that comparable to the native state upon D-fructose binding. The correlation of pKa with kcat (for the aniline hydroxylase activity) and the delta GDH2O-kcat profile showed that these structural changes of Mb-(m-Bphe)2 and Mb(PhBOH)2 were closely related to their sugar-enhanced aniline hydroxylase activity. Thus, the results established that an incorporation of the artificial receptor molecule can be a valid methodology for the design of stimuli-responsive semiartificial enzymes.

Coenzyme models. 10. Rapid oxidation of NADH by a flavin immobilized in cationic polyelectrolytes.

The title flavin rapidly oxidizes NADH, the observed enhancement being of 460--4650-fold compared with 3-methyltetra-O-acetylriboflavin. Analysis of the kinetic data at 30 degrees C established that the reaction proceeds according to the Michaelis-Menten kinetics at low ionic strength (mu = 0.02: Km = ca. 10(-5) M, kact. = 0.06-0.37 s-1) and according to the simple second-order kinetics at high ionic strength (mu = 0.3), the largest rate difference being 342-fold. On the other hand, the oxidation of 1-benzyl-1,4-dihydronicotinamide, a NADH model compound, was not accelerated by the use of the title flavin. It is concluded, therefore, that the formation of the polymeric flavin-NADH complexes due to the electrostatic interaction is responsible for the marked rate enhancement. This is the first example that the oxidation of NADH is facilitated in the nonenzymatic system.
