The behavioral effects of ethyl loflazepate and its metabolites were investigated in mice and rats, and they were compared with those of diazepam, nitrazepam and lorazepam. Locomotor activity of rats in open-field situation was increased with a wide range of doses of ethyl loflazepate and with large doses of nitrazepam. The anticonflict effect of ethyl loflazepate was slightly more potent than that of diazepam and much more potent than that of lorazepam. In suppressing muricide of both olfactory bulbectomized and raphe lesioned rats, ethyl loflazepate was approximately as potent as diazepam and much less potent than nitrazepam and lorazepam. Ethyl loflazepate was more potent than diazepam, nitrazepam and lorazepam in preventing pentetrazol-induced convulsion. In potentiating thiopental anesthesia in mice and impairing rotarod performance in mice and rats, ethyl loflazepate was less potent than diazepam, nitrazepam and lorazepam. The pharmacological activities of CM6913 and CM7116, metabolites of ethyl loflazepate, were approximately as potent as that of ethyl loflazepate. The duration of action of ethyl loflazepate was longer than those of the metabolites. These results indicate that ethyl loflazepate possesses pharmacological properties characteristic to benzodiazepines, and it is approximately equal to diazepam in potency and has a longer duration of action than diazepam, nitrazepam and lorazepam.
A combination immunochemotherapy regimen containing cisplatin (20 mg, days 1-5), peplomycin (20 mg, days 2, 9, 16), +/- vinblastine (5 mg, days 1, 2), and picibanil (3-5 KE/week) was performed in twelve patients with advanced or recurrent uterine and ovarian cancers under intravenous hyperalimentation (IVH), except one patient receiving peplomycin by a continuous infusion method using IVH bag (10 mg/day for 5 days). This regimen was repeated every three weeks. Five (71.4%) of seven evaluable patients showed partial response. No patients yielded the complete disappearance of disease. No severe and lethal pulmonary or renal dysfunction occurred and all was well tolerated. In a regimen without vinblastine, myelosuppression, especially thrombocytopenia, occurred later compared to the regimen including vinblastine.
High-metastatic murine fibrosarcoma cells readily attached to Type IV (basement membrane) collagen, whereas low-metastatic cells isolated from the same tumor did not. The addition of laminin--a glycoprotein that facilitates the adherence of epithelial cells to their basement membranes--enhanced the attachment of the low-metastatic cells, but not the high-metastatic cells. Using anti-laminin antibodies and a laminin-binding lectin as probes, the authors were able to identify by immunofluorescence a moiety associated with the high-metastatic cells, but not the low-metastatic cells, which cross-reacted with murine laminin purified from the EHS sarcoma. When extracts from the high-metastatic cells were separated by affinity chromatography, with the laminin-binding lectin as the affinity substrate, a substance was isolated that had an apparent molecular weight of 56,000 daltons. The affinity-purified material reacted strongly with anti-laminin antibodies by enzyme-linked immunosorbent assay.

Local cerebral glucose utilization (LCGU) was studied using the carbon 14-deoxyglucose method in rats with local freezing lesion in the left parietal cortex. A depression of LCGU developed with time after the lesion, being most prominent throughout the cortical areas of the lesioned hemisphere 3 days after lesion. Corresponding results in other regions were contralateral cortical areas, ipsilateral and contralateral subcortical structures. Brain stem structures were not affected. In white matter bilateral depression of LCGU reached its peak 24 hours after the lesion. LCGU returned to normal within 5 days in all affected areas. The areas affected and the time-course of the observed changes did not correlate with the location and known time-course of development of cerebral edema. Local cerebral blood flow (LCBF) was measured 1 and 3 days after a freezing lesion using the carbon 14-iodoantipyrine method. Twenty-four hours after a freezing lesion normal LCBF were seen throughout the traumatized brain. Three days after the lesion an increased LCBF involved all cortical areas, with the hyperemia being more pronounced in the hemisphere contralateral to the lesion. No corresponding changes in LCBF were observed. Evans blue dye was injected intravenously before the start of the barrier (BBB) at each time period following the lesion could be determined. Blue staining was seen in the area of the lesion in all animals killed 4 or 24 hrs after the lesion was made, indicating a BBB permeable to the Evans blue-serum protein complex.(ABSTRACT TRUNCATED AT 250 WORDS)

An enzyme linked immunosorbent assay (ELISA) was introduced for the detection of nephritogenic glycopeptide, nephritogenoside and Heymann's antigen. In the fraction of crude nephritogenoside which induced membranous glomerulonephritis (besides proliferative glomerulonephritis) in rats, both nephritogenoside and Heymann's antigen were detected by ELISA. On the other hand, in the sample of pure nephritogenoside which induced only proliferative glomerulonephritis, Heymann's antigen was not detected at all. These results indicate that crude nephritogenoside preparation contains Heymann's antigen as an inducing factor of membranous glomerulonephritis in homologous animals. In addition to TCA treatment or DEAE-cellulose column chromatography, gel filtration on Bio-Gel P200 was a good tool for the removal of Heymann's antigen from nephritogenoside.
