Association of HLA-B40 and DRW9 with Japanese alcoholic liver cirrhosis.

Seventy-seven chronic alcoholics with liver disease were studied to evaluate the HLA antigen association. There were no significant differences of HLA antigen phenotype frequencies (PF) between the patients and controls regarding A and C loci, (62 healthy Japanese). Prevalences of HLA-B40 complex (B40 . 48 . 13) and DRW9 tended to increase among chronic alcoholics. When chronic alcoholics were divided according to whether they had liver cirrhosis or not, the cirrhosis group (42 cases) revealed a significantly higher frequency of HLA-DRW9 (chi 2 = 10.88, p less than 0.001, corrected p less than 0.05, relative risk (R.R.) = 4.17) as compared to controls. There was also a tendency of B40 complex to increase in frequency (chi 2 = 5.51, p less than 0.05, R.R. = 2.65) in the cirrhosis group. Haplotype frequency and linkage disequilibrium parameters of HLA-B40 . 48-DRW9 were significantly higher than those of controls. Moreover, the increased frequency of DRW9 in the cirrhosis group was similar to that in autoimmune disease like ulcerative colitis or SLE. These data suggest that HLA-DRW9 and/or HLA-B40-DRW9 might be closely associated with susceptibility to developing alcoholic cirrhosis and that autoimmune mechanisms might be involved partly in its etiology.

Purification and some properties of nitrite reductase from Clostridium perfringens.

Nitrite reductase from Clostridium perfringens was purified by chromatographies on DEAE-cellulose, DEAE-Sephadex, Sephadex G-150, and hydroxylapatite and by isoelectric focussing to a homogeneous state, showing essentially a single protein band in disc gel electrophoresis and a single immuno-precipitation line in double diffusion against antiserum obtained from immunized rabbits. The reductase was induced in the presence of nitrate. It had a molecular weight of 54,000 and showed no absorption peak in the visible region. The pH optimum was 6.2 and Km for nitrite was 5 mM. Ferredoxin, as well as viologen dyes, was found to be an electron donor. The product of nitrite reduction was hydroxylamine. This reductase was inhibited by o-phenanthroline and azide but not by cyanide or diethyldithiocarbamate.

[Influence of the donor's smoking on P50 of the banked blood].

Influence of the donor's smoking on the banked blood P50 was investigated. Twenty four male and 19 female donor's CPD blood were measured P50 immediately after donation with new P50 determing system. This system ( [P50-H] ) consists of a fully automatic blood gas analyser and Co-oxymeter and the oxygen equilibrium curve and P50 are mathematically determined with microcomputer. P50 of eleven male non-smokers was 24.8 +/- 1.9 mmHg at pH 6.98 +/- 0.06 and P50 of 16 male smokers was 22.9 +/- 1.2 mmHg at pH 6.97 +/- 0.08. On the otherhand, P50 of 13 female nonsmokers was 24.3 +/- 1.3 mmHg at pH 6.91 +/- 0.05 and P50 of 3 female smokers was 23.4 +/- 1.7 at pH 6.93 +/- 0.03. These data showed that P50 of smoker's blood was significantly low compared with non smoker's blood.

Association of the human lymphocyte-DR2 antigen with Japanese ulcerative colitis.

The frequency deviation of human lymphocyte-DR antigens as well as human lymphocyte A, B, and C antigens was assessed in 40 Japanese patients with ulcerative colitis and compared with 51 ethnically matched healthy Japanese. The antigen that was of significantly higher frequency in patients with ulcerative colitis was HLA-DR2. This antigen was found in 70% of the patients vs. 31% of controls [chi 2 = 13.4, p less than 0.001, corrected p less than 0.01, relative risk = 5.1). In addition, an increased frequency of HLA-B5-DR2, which is the most common haplotype in Japanese populations, was found in 55% of the patients with ulcerative colitis compared with 22% of the controls (chi 2 = 10.84, p less than 0.001).

[A case of hereditary spherocytosis--role of complement system in hemolysis (author's transl)].

A female patient, 27 years old, with the chief complaint of anemia and jaundice, was diagnosed as hereditary spherocytosis with a complication of cholelithiasis and was examined before and after the operation of cholecystectomy and splenectomy. The surgery was followed by an increase in red cells, decrease in reticulocytes and normalization of serum bilirubin level. Both prothrombin time and partial thromboplastin time were normal before and after the surgery. The serum level of complement 4 was 11.5 mg/dl before and 20 days after the surgery it rose to 37 mg/dl. Similarly the serum level of complement 3c was 30 mg/dl before and 72 mg/dl 4 days after the surgery. From these results it was inferred that red cells of the patient were liable to hemolysis by the action of the complement system which was favored by an intrasplenic hemostasis without incurring intravascular coagulation.

[Simulation of K-contracture curve in smooth muscle (author's transl)].

40 mM K-induced isometric contracture of guinea pig taenia coli in the presence of 30 microM dantrolene showed a rapidly rising peak followed by a plateau and then a low sustained tonic contraction. The plateau was delayed by low Ca so that two phasic contractions, fast and slow, were separated from each other. In K-contracture after 15 sec contact with normal Ca following low Ca condition, the fast phasic contraction but not the slow one regained its tension depressed by preceding low Ca, while the slow phasic contraction recovered from its delay. In the presence of 0.2 microM verapamil, K-contracture consisted of the fast phasic contraction without plateau and of the low tonic contraction. The results suggest that K-contracture in the normal state consists of three components, the fast and slow phasic contractions and the tonic contraction, and that dantrolene inhibits a tonic contraction, whereas verapamil inhibits the slow phasic and the tonic contraction. Thus, the contraction curve of each component was tentatively expressed by exponential function, and for simulation of the observed curves a computer was utilized to synthesize the curves using the components of varying parameters. The simulation was successful when based on the above suggestion.

HLA and mycobacterial infection: increased frequency of B8 in Japanese leprosy.

A total of 60 leprosy patients, 28 of lepromatous and 32 of tuberculoid form, and 70 active tuberculosis patients was compared with a control of 184 for 34 HLA specificities. The most interesting finding was an increased frequency (10.0%) for HLA-B8 (corrected P = 0.062, relative risk = 20.3) in the leprosy patients as compared with the control group, despite the fact that the frequency of HLA-B8 was extremely low in Japanese. Furthermore, all leprosy patients with B8 had leprous member(s) in their family.

Potassium contracture of smooth muscle in the presence of dextran.

The apparent diffusion coefficient of Ca ions in the extracellular space of guinea pig taenia coli was estimated from model experiments to be 3.2 x 10(-6) cm2 sec -1, while the relationship between Ca concentration in the medium and tonic tension in 40 mM K-contracture was measured in the same muscle both in the presence and absence of dextran 10. On the basis of these experiments and certain assumptions, the time course of tension decline by Ca withdrawal during K-contracture was calculated. Under all dextran concentrations tested (0--15%) the calculated time course of tension decline was in good agreement with one actually observed except in a short period of time immediately after Ca withdrawal. The results suggested that Ca distribution in the extracellular space during the loss of Ca is in good agreement with the diffusion theory in cylinder, and that each muscle fiber shows its tension without delay in response to the change in Ca concentration in the vicinity of the fiber according to the tonic tension-Ca relationship mentioned above. The discrepancy between calculation and observation was partially explained on an experimental basis.
