Effect of verapamil on the lower urinary tract.

The effect of verapamil on the in-vivo bladder and urethra was investigated in female dogs. The bladder and urethral pressure responses to electrical stimulation of the pelvic and hypogastric nerves were decreased by verapamil.

Application of ultrasonic M-mode scanning and computer evaluation on urodynamics.

The M-mode scanning images of urethra, detrusor pressure and urine flow rate during voiding were recorded together on the line scan recorder, and allowed for plotting and calculation of some valuable urodynamic parameters by computer evaluation. This procedure will permit first application of M-mode scanning and new direction on urodynamics.

Role of renal prostaglandin in renin release during renal nerve stimulation in anesthetized dogs.

The relationship of prostaglandin (PG) release to changes in renin release and renal blood flow (RBF) to renal nerve stimulation (RNS) was investigated in pentobarbital anesthetized dogs. Experimental values were obtained before and during RNS (3 or 5 Hz, 10 to 20 V, 1 msec, for 10 min) in the absence and/or presence of indomethacin (5 mg/kg, i.v.), respectively. Plasma renin activity and plasma PGE2 concentration were measured by radioimmunoassay. RNS increased PGE2 secretion rate and renin secretion rate. RBF was initially reduced by RNS, then partial recovery of RBF occurred despite the continuous stimulation. In the presence of indomethacin, the recovery response of RBF and the increase in PGE2 secretion rate during RNS were not observed. Indomethacin also suppressed the increase in renin secretion rate. These results suggest that the renin and RBF responses to RNS are modulated by concomitantly released PG.

Stimulation of prostaglandin E2 biosynthesis in rat dental pulp explants in vitro by 5-hydroxytryptamine.

When excised rat dental pulp tissue was incubated in Krebs solution, prostaglandin E2 (PGE2) was released into the medium. The release was completely blocked by the PG synthetase inhibitor, indomethacin (0.1 mM). Exogenous arachidonic acid (0.01 mM), a substrate of PG synthetase, increased the amount of PGE2 production and the endogenous PGE2 biosynthesis was strongly stimulated by 5-hydroxytryptamine (5-HT) at concentrations higher than 0.01 mM. Bradykinin (0.1 mM) and histamine (0.1 mM) had no effect.

Effect of acute and chronic treatment with tricyclic antidepressants on 3-methoxy-4-hydroxyphenylethyleneglycol sulfate (MHPG-SO4) contents in various regions of rat brain.

1. Effects of acute and chronic treatment with desipramine (DMI) and amitriptyline (AMI) on 3-methoxy-4-hydroxyphenylethyleneglycol sulfate (MHPG-SO4) levels in eight brain regions were examined in rats. 2. A single injection of DMI decreased MHPG-SO4 levels in all brain regions except the midbrain, whereas the effect disappeared after the chronic treatment except in the septal area. MHPG-SO4 levels in the hippocampus and thalamus were increased after the chronic treatment with the drug. 3. A single and chronic administration of AMI decreased MHPG-SO4 levels in the thalamus and hippocampus, respectively. 4. These results indicate that the effects of acute and chronic treatment with the tricyclic antidepressants on the central noradrenergic systems differ in different regions of rat brain.

Anti-warfarin antibody preparation and its characterization for radioimmunoassay.

Anti-warfarin antiserum was prepared in rabbits by immunization with a synthesized warfarin antigen, 4'-azo-warfarin human serum albumin, which possesses two enantiomorphic haptenic sites of warfarin on the molecule. The antiserum recognized both R- and S-warfarin to the same degree, 50% cross reactivities of racemic warfarin, respectively. One of the warfarin metabolites, racemic warfarin alcohol, showed 1% cross reactivity, and R- or S-warfarin alcohol have half the reactivity of the racemic alcohol. Rabbit plasma warfarin levels were determined by radioimmunoassay using this antiserum and racemic [14C]warfarin and by fluorometric assay after isolation by thin layer chromatography. After a single administration of warfarin (2 mg kg-1 orally or 500 microgram kg-1 i.v.), the plasma levels determined by both assay methods showed a good correlation (r = 0.97, P less than 0.001, Y = 1.04-0.09). The results show that the radioimmunoassay can determine total plasma warfarin without interference of plasma metabolite. The applicability and limitation of the radioimmunoassay for pharmacokinetic study are discussed.

The acoustic characteristics of the left ventricle.

A new method was developed to describe the instantaneous resonance curve (IRC) of the left ventricle (LV), which represents the acoustic characteristics and instantaneous viscoelastic properties of LV. This method was evaluated using coronary-perfused isolated canine LV preparations.
