Contribution to the amygdaloid projection field in the rat. A quantitative autoradiographic study.

Light microscopic autoradiography and quantitative photometric studies of autoradiographs were performed subsequent to injection of tritiated amino acids into various parts of the amygdala of the rat. The course of amygdalofugal fibers in the stria terminalis (bilaterally), longitudinal association bundle, medial amygdalo-hypothalamic pathway, and medial forebrain bundle is described. Evidence is provided for both additional transcapsular and paracapsular fibers and early descending stria terminalis fibers. Labeling of the ipsilateral main olfactory bulb and the bilateral acessory olfactory bulb was observed following injection into the cortical amygdaloid subnucleus. The termination of amygdalofugal fibers in the preoptic area and especially the subnucleu of the ventromedial and premamillary hypothalamic nuclei bilaterally were analysed by photometric measurement. A caudal amygdaloid projection system arises from the central amygdaloid nucleus. This system extends to the substantia nigra, lateral terminal nucleus of the accessory optic tract, central grey in the midbrain, and to the parabrachial nuclei, nucleus of the mesencephalic trigeminal tract, nucleus of the solitary tract in the hindbrain.

[Photomicrography of autoradiographs (author's transl)].

It is often desirable to delineate the areas of autoradiograms where accumulations of silver grains are observed in order to establish topographic landmarks. This is prevented by either bright field or dark field illuminatiated photographs, where the topography of the silver grains is neglected despite the high resolution obtainable by autoradiography, but may be done by taking advantage of a modification of dark field illumination thus generating an additional bright field effect. This modification also enables to correlate selective histochemical reaction sites with the area of silver grain accumulation.

Evidence for amygdaloid projections to the contralateral hypothalamus and the ipsilateral midbrain in the rat.

Light microscopic autoradiography was performed subsequent to injection of tritiated amino acids into various parts of the amygdaloid body of the rat. Evidence is provided for two hitherto unreported projections of the amygdala: from the medial amygdaloid nucleus to the contralateral premamillary nuclei and from the central amygdaloid nucleus to the mesencephalic central grey. The functional implications of these findings are discussed.

Improvement of hepatic microhemodynamics by N-acetylcysteine after warm ischemia.

In this study we investigated the influence of N-acetylcysteine (NAC) on the hepatic microcirculation after warm ischemia by intravital fluorescence microscopy. Clamping of the left liver lobe was performed in 20 male Wistar rats for 70 min. The treatment group (n = 10) received 400 mg NAC/kg body weight 20 min prior to clamping. After reperfusion, acinar and sinusoidal perfusions were observed as well as the leukocyte-endothelium interaction. Phagocytic activity was assessed after application of latex beads. NAC reduced the number of nonperfused sinusoids in all acinar zones. A reduction in zone 1 (portal) was achieved from 15.5 to 7.1% (p < 0.0001), in zone 2 (midzonal) from 14.6 to 6.1% (p < 0.0001) and in zone 3 (central) from 11.9 to 2.9% (p < 0.0001). There were no significant differences in leukocyte adherence as well as in phagocytic activity detectable. We conclude that NAC improves hepatic microcirculation after warm ischemia by increasing sinusoidal blood flow.
