Test of leukocyte migration inhibition. 1. Value of the test in the diagnosis of certain immune diseases.

The test of leukocyte migration inhibition (LMI) being a specific and sensitive method for the detection of cell-mediated immunity, its ever increasing use in the clinic necessitates a more accurate estimation of its actual value. The technique used in this study is that of leukocyte migration in capillary tubes according to Soborg and Bendixen. The test was performed in three groups of patients: A--with drug intoxication; B--with chronic post viral hepatitis and C--with alcoholic liver disease. The results obtained showed that in the patients with drug allergy the LMI test using the incriminated drugs as antigens was positive in 83% of the cases, in the patients with chronic post viral hepatitis the cellular migration inhibition to the HBsAg was observed in 35% of the cases and in alcoholic liver disease lymphocyte sensitization to ethanol was present in 28.7% of the cases. It is concluded that in drug allergy the LMI test is useful and has diagnostic value being preferable to other in vitro determinations and even, in certain cases, to the skin tests. In chronic liver diseases the value of the test is limited to investigation purposes as it can reveal the degree of involvement of cell-mediated immunity in the pathogenesis of disease.

Test of leukocyte migration inhibition. II. Comparative study of the techniques in capillary tubes and in agarose.

The object of the paper was to estimate the advantages and disadvantages of two techniques of the leukocyte migration inhibition (LMI) test (in capillary tubes and in agarose) for the in vitro testing of delayed type hypersensitization. The capillary technique was carried out according to Soborg and Bendixen and the agarose technique according to Clausen. The antigen used in both techniques was PPD in a concentration of 10 micrograms/ml. The techniques were performed in parallel in two groups of individuals: 15 healthy controls and 20 patients with chronic hepatitis. It was observed that in the control group sensitization to tuberculin in vitro was detected by the capillary technique in 7 of the 15 subjects (47%) and by the agarose technique in 4 of the 15 subjects (27%). In chronic hepatitis sensitization to PPD was detected, by the capillary method, in 11 of the 20 patients (55%) and by the agarose method, in 9 of the 20 patients (45%). The results were consistent by the two methods in 18 of the 20 patients. In conclusion the LMI test in capillary tubes is preferable to that in agarose being simpler, more rapid, more sensitive and less expensive.

Active rosette-forming lymphocytes in chronic hepatitis.

Lymphocyte subpopulations (total T lymphocytes, active T lymphocytes, late T lymphocytes, B lymphocytes), leukocyte migration inhibition test, serum HBsAg, serum immunoglobulins (IgG, IgA, IgM) were determined in 15 chronic active hepatitis patients and 35 chronic persistent hepatitis patients (diagnosed clinically, biochemically and histologically) and 10 healthy subjects who had acute B viral hepatitis three years ago. A decreased number of total T lymphocytes has been found in HBsAg-seropositive chronic hepatitis. An increase of relative and absolute number of active T lymphocytes was noticed in HBsAg-seronegative healthy persons with a past history of acute viral hepatitis B.

Coronary heart disease and its risk factors in a group of 5,000 middle aged men in urban environment.

The results obtained at entry in the subjects included in a pilot study (Institute of Internal Medicine, Bucharest) for the detection and prevention of coronary heart disease and hypertension, are presented. These data are the prevalences of the risk factors of coronary heart disease (high serum cholesterol, hypertension, smoking, overweight, diabetes, nonspecific minor ECG signs, family history), as well as the prevalences of the various forms of coronary heart disease. The study of the frequency distribution of biologic parameters likely to become risk factors showed that in middle aged subjects the upper limit of the normal should be lowered from the 95th percentile to the 76th one.
