5-Hydroxy-6-methoxy-2-indolylcarboxylic and 6-hydroxy-5-methoxy-2-indolylcarboxylic acids in the urine of hamsters with melanotic melanoma.

The isomeric metabolites, 5-hydroxy-6-methoxy-2-indolylcarboxylic and 6-hydroxy-5-methoxy-2-indolylcarboxylic acids have been identified in the urine of hamsters bearing transplantable melanotic melanoma. Their identification was carried out by spectral and chromatographic comparison of the two isomers isolated from urine with the same compounds achieved synthetically.

Pineal vasotocin: release into cat cerebrospinal fluid by melanocyte-stimulating hormone release-inhibiting factor.

The intracarotid injection of both synthetic melanocyte-stimulating hormone release-inhibiting factor (MIF) and purified MIF prepared from bovine hypothalami induces arginine vasotocin release into cerebrospinal fluid of cats and significantly decreases the pineal arginine vasotocin content at 5 minutes after the injection. The present results demonstrate an extrapituitary endocrine effect of synthetic and purified bovine MIF.

Chromatographic evidence for vasotocin biosynthesis by cultured pineal ependymal cells from rat fetuses.

Cultured pineal ependymal cells from rat fetuses aged 17 to 19 days post-coitum release into their media a substance that has hydroosmotic, antidiuretic and rat uterine activities. Paper chromatography of concentrates from culture media demonstrates that the substance possessing all the above activities was eluted from a slow moving region (Rf 0.15-0.35) with a peak at 0.20-0.30. This region is not significantly different from the Rf of synthetic arginine vasotocin (AVT) used as standard. Neither hydroosmotic nor rat uterine activities could be detected in the chromatographic eluates from the fast moving region corresponding to the Rf of synthetic oxytocin (0.55-0.65) used as standard. The chromatographic mobility of biological activities from culture media, the ratio of the activities as well as their susceptibility to tryptic digestion, demonstrates the presence of a basic peptide indistinguishable from synthetic AVT. The total amount of AVT released into the medium during 43 days of incubation is about 40 times greater than the amount contained in non-incubated pineal glands of the same age, strongly suggesting de novo synthesis of AVT.

Diurnal rhythm of vasotocin in the pineal of the male rat.

The pineal content of arginine vasotocin in the male rat is subject to diurnal changes, having a high value around noon and a low value around midnight. Since in rats exposed to 24 h constant light the pineal content in arginine vasotocin decreases, and in the rats exposed to 24 h constant darkness the pineal content in arginine vasotocin increases, it appears that during the night both the synthesis and release of arginine vasotocin is enhanced.

Melanogenuria as consequence of elevated tyrosinase activity in melanoma.

The authors point out that melanogenuria, i.e. the excretion of specific phenolic and, particulary, indole metabolites in melanoma, is undoubtedly due to elevated tyrosinase activity in this disease. Other enzymes, in particular glucuronyltransferase, catechol-O-methyltransferase and possibly hydroxyindole-O-methyltransferase, also evidently participate in the further biochemical conversion of the resultant metabolites, however.

Vasotocin biosynthesis by neurohypophysial cells from human fetuses. Evidence for its ependymal origin.

Cultured neurohypophysial cells from human fetuses aged 130-155 days release into the culture medium an active principle which has antidiuretic, hydroosmotic and rat uterine activities. The chromatographic mobility of the active principle, as well as the susceptibility of all of the above activities to tryptic digestion, indicates the presence of a basic peptide identical to arginine vasotocin (AVT). The ability of these cultured cells, which are probably ependymal cells, to release the above activities during 43 days of incubation, suggests that AVT is synthesized and secreted by specialized ependymal cells in the developing human neurohypophysis. Neither in the culture media from anterior pituitaries of the same fetuses nor in control culture media could antidiuretic, rat uterine or hydroosmotic activities be detected. Although the nonincubated neurohypophyses of the same age contain pharmacological activities suggesting the presence of vasopressin, oxytocin and AVT, the neurohypophysial cells cultured in vitro apparently release only AVT into the medium. This supports the suggestion that whereas vasopressin and oxytocin are synthesized in the hypothalamic neurosecretory cells, AVT, on the contrary, appears to be synthesized by ependymal neurohypophysial cells in the developing human neurohypophysis. Consequently, the fetal human neurohypophysis, and presumably the fetal mammalian neurohypophysis, appears not only as a storage site for neurohypophysial hormones but also as an endocrine structure which synthesizes and secretes AVT by ependymosecretion.

Reversal by vasotocin of pinealectomy and constant light effects on the pituitary melanocyte-stimulating hormone (MSH) content in the mouse.

A single injection of 1 pg synthetic arginine vasotocin (AVT) into the third ventricle of the mouse completely prevented the pituitary MSH increase which occurs seven days after pinealectomy or 24 h after exposure to constant light. The same concentration of AVT injected into the third ventricle of normal mice markedly decreased the pituitary MSH content at 20 min following the injection. The present results suggest that AVT injected into the third ventricle of the mouse inhibits synthesis and/or release of MSH-release inhibiting factor (MIF).
