Transplantation of cryopreserved pancreatic islets into the portal vein.

Pancreatic islet cells of rats obtained by the collagenase digestion method were preserved from 7 to 10 days at -80 degrees C in a deep freezer or at -196 degrees C in liquid nitrogen with Dulbecco modified Eagle's medium, supplemented with 10% or 20% DMSO (dimethyl sulfoxide); thawing was performed in a 37 degree C water bath. Cryopreserved islets were morphologically almost intact, and possessed approximately 50% of the insulin secretion activity of the control groups. About 300 pancreatic islets preserved at -196 degrees C with 20% DMSO were transplanted into the portal veins of streptozotocin-induced diabetic rats. Rats recovered from the diabetic state, and the normalized condition was maintained up to 20 weeks, although 4 weeks were needed before blood and urine glucose reached normal levels after transplantation. Intact B cells were found in the transplanted islet cell masses in the liver of the recipients, but B cells of the recipient's pancreases (streptozotocin-treated rats) showed a marked decrease, as well as degenerative changes.

[Studies of interventricular septal motion after heart valve replacement by two-dimensional echocardiography and thallium 201 scintigraphy (author's transl)].

Etiology of abnormal interventricular septal motion detected by M-mode echocardiography after heart valve replacement was studied by two-dimensional echocardiography and thallium 201 scintigraphy in three patients with valvular heart disease (two with mitral regurgitation, and one with aortic regurgitation). M-mode and two-dimensional echocardiograms were recorded before and after heart valve replacement, using Aloka SSD-800. After intravenous injection of 2.0 mCi of ionic thallium 201, myocardial images were recorded with a scintillation camera (Searle PHO Gamma V) with converging collimator. Abnormal interventricular septal motion (systolic anterior motion) was observed echocardiographically in all of three patients after mitral or aortic valve replacement. Two-dimensional echocardiogram showed systolic anterior motion of the whole heart towards the anterior chest wall, though the left ventricle itself contracted normally. Thallium 201 scintigram showed no significant perfusion defects of the left ventricular wall including ventricular septum. No findings showing conduction disturbance, right ventricular volume overload or pericardial effusion were observed after heart valve replacement.

Characteristics of the binding component for testosterone and androstenedione in rat liver cytosol.

A study was made of the nature of a macromolecular component in the hepatic cytosol of male rats which binds with testosterone and androstenedione. The saturation analysis revealed that this component has binding affinity with these androgenic steroids and a limited number of binding sites. This component was inactivated by incubation with proteolytic enzymes and by heating, but was unaffected following incubation with RNase. Analysis by sucrose density gradient centrifugation revealed that this component had a sedimentation coefficient of approximately 10 S. On the basis of the elution profile of the component on gel chromatography, the molecular weight and the similarity of the structure of these androgens, it seems most likely that one binding component is responsible for both testosterone and androstenedione. It was postulated that the only difference in binding for these androgens is that testosterone, showed a more rapid association rate with this component, than androstenedione
