Both B151-T cell replacing factor 1 and IL-5 regulate Ig secretion and IL-2 receptor expression on a cloned B lymphoma line.

In the present study, we have demonstrated that both B151-T cell-replacing factor 1 and rIL-5 are responsible for the activity to partially induce CL-3 cells into IgM-synthesizing cells and also to synergize with IL-2 to augment IL-2R expression on and IgM synthesis in CL-3 cells. These actions of rIL-5 on a homogeneous cloned line (BCL1-CL-3 cells) allow us to identify and characterize the two alternated B cell developmental pathways. One is an IL-2-independent, IL-5-driven differentiation pathway without preceding up-regulated IL-2R expression, and the other is an IL-5 plus IL-2-dependent augmented differentiation pathway with preceding up-regulated IL-2R expression. We have also demonstrated the functional difference of two distinct B cell growth-promoting factors, B cell-stimulating factor 1 (rIL-4) and rIL-5. CL-3 cells are equally stimulated to grow by rIL-4 and rIL-5, whereas only rIL-5 can render CL-3 cells responsive to rIL-2, indicating that these two lymphokines affect B cells in a strikingly different manner.

An unusual neurological disorder with abnormal copper metabolism.

Twin males aged 24 years showed dementia, dysarthria, gait disturbances and involuntary movements, with slightly low levels of serum copper and ceruloplasmin, and markedly low excretion of urinary copper. We propose that the unique combination of dementia, dysarthria, gait disturbances, involuntary movements and abnormalities of copper metabolism does not fit any known nosological entity and constitutes a "new" syndrome different from Wilson's and Menkes' diseases.

Gallium-67 scintigraphy in patients with hemochromatosis treated by deferoxamine.

Gallium scintigraphy was performed as an aid for determining the presence or absence of malignant neoplasm in two patients with hemochromatosis treated by deferoxamine. However, gallium scan images could not be obtained. So gallium scintigraphy was performed once more to investigate the cause of low activity. Both patients had heavy urinary excretion of gallium in the first 24 hrs after the injection, and activity was very low on the day of examination. This phenomenon may be attributed to the effect of deferoxamine which is highly bound to the gallium.

Increased dermal collagen density in amyotrophic lateral sclerosis.

We examined specimens of skin overlying the sacral region, among the most common sites of bedsores, from patients with amyotrophic lateral sclerosis (ALS) and controls, and found that in ALS patients, collagen fibrils had a greater density and became more tightly packed with the duration of illness. Our results suggest that the increased density of collagen fibrils may protect the skin of ALS patients from pressure ischemia, a major cause of bedsore formation.

Both amino- and carboxy-terminal portions are required for insertion of yeast porin into the outer mitochondrial membrane.

Yeast porin, the major outer mitochondrial membrane protein, is synthesized without a cleavable extension peptide and post-translationally inserted into the membrane. When inserted into the membrane, it acquires resistance to externally added trypsin. To locate the sequences responsible for membrane insertion and topogenesis in the primary structure of yeast porin, we constructed several deletion and chimeric mutants of the porin cDNA. These cDNAs were expressed in vitro and the products were assayed for capacity to be correctly inserted into isolated mitochondria. It was thus found that deletion of the segment spanning residues 37-98 did not appreciably impair the insertion competence and the inserted protein became resistant to trypsin. On the other hand, the porin mutant lacking the segment consisting of residues 17-98 did not acquire the trypsin resistance, though it could bind to mitochondria specifically. Deletion of the carboxy-terminal 62 amino acid residues also abolished the capacity to be correctly inserted into mitochondria. We conclude that information required for membrane insertion and intramembranous topogenesis of the porin molecule is stored not only in the amino-terminal region but also in the carboxy-terminal portion.

"Delayed return phenomenon" in amyotrophic lateral sclerosis.

When the skin of patients with amyotrophic lateral sclerosis (ALS) is stretched, it returns only sluggishly to its original position. We term this property of skin the "delayed return phenomenon" (DRP). The reticular dermis was examined in patients with ALS and controls with or without DRP, respectively. On light microscopy, ALS patients with DRP showed collagen bundles which were reduced in amount, more loosely woven, and separated by wide space, while controls with DRP revealed decrease of dermal thickness, without abnormalities of collagen bundles. Electron microscopy disclosed a markedly large amount of amorphous material positive for ruthenium red, separating collagen fibrils and bundles in ALS patients with DRP. These observations suggest that DRP in ALS is a specific feature not previously reported in cases of ALS.

Hexacosanoate contents in Japanese common foods.

Recently, a diet enriched in oleate and moderately restricted in hexacosanoate (C26:0) was found effective to reduce the plasma very long chain fatty acid (VLCFA) levels in patients with adrenoleukodystrophy (ALD), an X-linked disorder characterized by demyelination of the adrenal cortex and cerebral white matter, and accumulation of saturated VLCFA, particularly C26:0, in tissues of the demyelination. The information about the C26:0 content in Japanese food was, however, almost nil except for one report about foods in the USA, but this did not include some Japanese common foods. With the hope of treating an ALD patient in our hospital, C26:0 contents in Japanese common foods (42 items) were measured. In our case, a one-hour direct transesterification method was used to obtain methylesters of total fatty acids in foods and they were applied directly to a selected ion monitoring gas chromatography-mass spectrometry for the quantitative C26:0 analysis. The C26:0 content in nuts and seeds as well as in fats and oils was found to be significantly higher than in other foods; the content was highest in peanuts. The content in almost all kinds of examined fishes, the common protein foods in Japan, was relatively low. From these data and that in the national nutrition survey in 1986, the daily intake of C26:0 from the average Japanese diet could be estimated to be 12-36 mg. It can be recommended, therefore, that nuts and seeds as well as fats and oils should be restricted as severely as possible from the diet of ALD patients in Japan in order to keep daily C26:0 intake below 10 mg as recommended in the USA.

[Detection of varicocele by using 99mTc-diethylenetriamine pentaacetic acid human serum albumin (99mTc-DTPA human serum albumin)].

In order to detect the varicocele, scrotal scintigraphies by using 99mTc-HSA-D were performed in 14 patients with male infertile or palpable mass in left scrotum on physical examinations. Abnormal pooling of 99mTc-HSA-D, indicative of varicocele lesion, could be found in left scrotum in 9 cases, confirmed surgically or clinically. Compared with 99mTc-HSA, 99mTc-HSA-D was superior in high uptake ratio of varicocele to soft tissue and in nonvisualization of bladder. Thus, 99mTc-HSA-D scrotal scintigraphy seemed to be of a great use to detect the varicocele.

[Clinical usefulness of a dual photon absorptiometry system using X-ray for peripheral bone--comparison with a single photon absorptiometric system].

A newly developed dual photon absorptiometry system (DCS-600, ALOKA Co., LTD.) using two monochromatic X-rays for the determination of bone mineral in the peripheral bone was fundamentally studied, in comparison to single photon absorptiometry system (SPA). The accuracy and precision in DCS-600 were equal to or better than SPA system. Furthermore, in DCS-600 the forearm could be imaged, and the manipulation was easy. Thus DCS-600 was found to be a good instrument as a quantifying system of bone mass.
