Do brine shrimp diagnose cystic fibrosis?

The nauplii of the brine shrimp Artemia salina are dependent upon the function of their salt gland to maintain osmotic pressure within narrow limits. A number of drugs interfere with this function and are lethal to the nauplii. Saliva and serum from normal persons, patients with cystic fibrosis, and obligate heterozygotes were tested for lethal effect against brine shrimp nauplii. At salt concentrations between 100 mM and 2.5 no difference was found among the phenotypes. At lower concentrations a difference was noted occasionally between some normal subjects and some individuals carrying one or two genes for cystic fibrosis. Data from an independent series of experiments indicate that the naupliar deaths result from distorted ratios of Na+/K+ and not from a specific gene product. No difference was noted in the O2 uptake of nauplii treated with saliva or serum obtained from normal subjects, patients with cystic fibrosis, or obligate heterozygotes.

The polyamine content of the tRNA of E. coli.

A sensitive method of polyamine estimation has been adapted to the study of the organic cations of small amounts of nucleic acid. A procedure utilizing phenol extraction, alcohol precipitation, and separation on Sephadex G100 has been devised for the isolation of tRNA at low ionic strength. The procedure is applicable to the isolation of tRNA from liter batches of bacterial culture. With these methods we have examined the polyamines of tRNA isolated from polyauxotrophic strains of E. coli incubated under various physiological conditions and have found the following: (1) The tRNA from relaxed bacteria (TAU rel) harvested during exponential growth is heterogeneous with respect to polyamine content. Some portions of the population contain about one mole of spermidine per mole of tRNA. Some putrescine and an unknown amine are also present in low concentration. (2) After exponential TAU rel is incubated with thymine and uracil in the absence of arginine, the tRNA population is far more homogeneous with respect to polyamine content. The various fractions contain two moles of spermidine per mole of tRNA and a small amount of putrescine. (3) After exponential TAU rel is incubated in the absence of both arginine and uracil, the polyamine pattern of the tRNA resembles that isolated from exponential cells. (4) The tRNA from stringent bacterias harvested during exponential growth is heterogeneous with respect to polyamine distribution and some fractions contain relatively high concentrations of the unknown amine.

Significance of common acute lymphoblastic leukemia antigen (cALLA) positive cells in bone marrow of children off therapy in complete remission.

Bone marrows from 21 children with acute lymphoblastic leukemia in complete remission (CR) and off therapy for 14 months to 10 years were examined by flow cytometry with a panel of monoclonal antibodies. Significant percentages of cALLA+ cells of low fluorescence intensity were present up to 9 years after CR. These results emphasize the need to interpret flow cytometry results in light of the findings that low intensity cALLA+ cells represent a normal population of immature, non-malignant lymphoid cells.

Surgical treatment of severe postshunt hepatic encephalopathy.

Hepatic encephalopathy is a major complication of portal-systemic shunts with an incidence ranging up to 52%. A small fraction of these patients are refractory to medical therapy. Shunt ligation and colonic procedures are the main surgical approaches. The goal of the latter is to diminish the colonic absorption of nitrogenous substances which are involved in the pathophysiology of hepatic encephalopathy. Six patients, whose average age was 55.7 +/- 2.6 years, were operated for severe postshunt encephalopathy requiring 4.3 +/- 0.9 admissions for a total duration of 76 +/- 26 days over 1-11 years. One patient had undergone a splenoral shunt and 5 had a portacaval shunt. One ligation of the shunt and 5 colon exclusions were performed. The average postoperative hospital stay was 21.5 +/- 3.9 days. The mean follow-up was 47 +/- 20 months. The patient with the shunt ligation remains free of encephalopathy 94 months after the procedure and has not bled from his esophageal varices. Among the 5 colon exclusion patients, there were 1 death and 3 complications. Three patients were completely relieved of their hepatic encephalopathy. One of those 3 died of a subarachnoid hemorrhage 28 months after the surgery. The fourth still needs medication to control a persistent, although improved, encephalopathy that required 2 further hospitalizations. Colon exclusion is a useful intervention in very selected cases. It has a lower operative mortality than total colectomy and the advantage over shunt ligation of not reestablishing hypertension in the portal system.

Self-diffusion and molecular mobility in PVA-based dissolution-controlled systems for drug delivery.

Nuclear magnetic resonance (NMR) microscopy has been used to monitor the hydration of poly(vinyl alcohol) (PVA) samples of varying molecular weight. One-dimensional profiles weighted to predominantly show the variation of water concentration were acquired every 3 min during the first 30 min of hydration and subsequently at 1 and 2 h. Diffusion-weighted profiles obtained after 30 min and 1 and 2 h were used to calculate the spatial variation of the water self-diffusion coefficient. The resulting data provide supporting evidence for the hypothesis that phenomena such as reptation are important near the glassy/rubbery interface of polymers during dissolution, while the diffusion gradually changes to Zimm type near the rubbery/solvent interface.
