Superior cervical ganglionic origin of sympathetic fibers in the facial nerve, and their preganglionic spinal inputs: a WGA-HRP study in Macaca fuscata.

The origin of sympathetic fibers in the facial nerve of Macaca fuscata was demonstrated using the HRP method. WGA-HRP injection into the facial nerve resulted in heavy labeling in the ipsilateral superior cervical ganglion. The majority of labeled neurons in the superior cervical ganglion had sizes ranging from 20 to 35 microns. In the present study it was of particular interest that labeled neurons are also found in the intermediolateral nucleus and lateral funiculus ipsilaterally at the level from the C8 to T4 spinal cord segments. Many labeled neurons in the lateral funiculus were located in close vicinity to the intermediolateral nucleus, and, characteristically, a few of these had a long dendrite extending to the lateral surface of the spinal cord. The labeling in the spinal cord appears to be caused by transneuronal transport of WGA-HRP.

Simple purification method for a Vibrio vulnificus hemolysin by a hydrophobic column chromatography in the presence of a detergent.

A Vibrio vulnificus hemolysin (VVH) was purified by two steps of hydrophobic column chromatography on Phenyl-Sepharose HP. The first chromatography was carried out at pH 6.0. In this pH condition, VVH efficiently bound to the column, but the hemolysin fraction eluted was accompanied with colored substance(s). To eliminate this colored substance, the second chromatography was carried out at pH 9.8 in the presence of 1% 3-[(3-cholamidopropyl) dimethylammonio]-1-propanesulfonate (CHAPS), a zwitterionic detergent. Homogeneity of the hemolysin thus obtained was shown by polyacrylamide gel electrophoresis. The specific activity increased 33,600 times and the yield was 35%. The method is simple and useful to supply enough VVH for study of the role of the hemolysin in the infection by V. vulnificus or on the mechanism of action of the hemolysin.

Studies on hemolytic action of a hemolysin produced by Vibrio mimicus.

Some properties and mechanism of action of a hemolysin (VMH) produced by an enteropathogenic Vibrio mimicus strain was examined. VMH was heat-labile and inhibited by addition of divalent cations, including Ca2+, Mg2+ and Mn2+. The hemolysis by VMH was inhibited by incubating with gangliosides, suggesting that the ganglioside was the binding site on the erythrocyte membrane for VMH. Existence of a galactose moiety on reducing end of the ganglioside molecule and a sialic acid on the galactose moiety was suggested to be important for the binding of VMH molecule. Colloid osmotic manner of the hemolysis by VMH was demonstrated.

Father-son testicular cancer.

Testicular tumors in closely related family members are rare. We report a case of familial testicular cancer occurring in father-son pairs. The father had teratocarcinoma and his son developed pure seminoma. The association of a genetic factor in the etiology of testicular malignancy is discussed.

Postoperative improvement in blood lactate threshold during exercise in a patient with pulmonary arteriovenous fistula.

A 13-year-old female patient with a pulmonary arteriovenous fistula (PAVF) of 24 percent shunt fraction (SF) underwent resection of segments 8 and 9 of the left lower lobe. Three months after the operation, the shunt became insignificant and PaO2 was normalized. In terms of the maximum work rate, maximum VO2 and the VO2-blood lactate relationship, this patient showed a remarkable postoperative improvement in the exercise capacity.

Double left renal veins and multiple right renal veins found in Japanese adults.

A case of circumaortic renal venous collar, the first such collar to be observed in a Japanese female cadaver, is presented, and its anatomical organization is compared with that in another case in which four right renal veins accompanied four right and three left renal arteries in a Japanese male cadaver. Double left renal veins are formed by persistence of a more central retroaortic venous anastomosis, in contrast to multiple right renal veins, which are formed by persistence of some embryonic renal veins arranged in ladder-like patterns. In our case, the dorsal limb of the renal collar communicated with the azygos system veins and lumbar veins. The azygos system veins were considered to have atrophied as a result of some persisting retroaortic venous anastomoses located in the thorax. Therefore, we believe that the renal collar was related to the atrophy of the azygos system veins. To determine whether there are racial differences in incidence of renal collar, further studies in Asians are required.

[Primary mediastinal tumors in children--comparison with mediastinal tumors in adults].

During the past 43 years, 628 patients with a primary mediastinal tumor underwent surgical operation in our institution. Of those patients, 106 patients (16.9%) were children of 15 yr of age or less and 522 patients (83.1%) were adults. 47 (44.3%) of the 106 children had neurogenic tumors, while 21 patients (19.8%) had a teratoma. 189 (36.2%) of the 522 adults had thymoma and 89 patients (17.1%) had teratoma. In the 106 children there were 71 benign tumor patients (66.9%) and 35 malignant tumor patients (33.1%). There were 277 (53.1%) benign and 245 (46.95) malignant tumors in the 522 adults. The adults had significantly more malignant tumors than the children (p < 0.01). In our series of malignant mediastinal tumors in children, 14 patients (13.3%) had a lymphoma and 11 patients (10.5%) had a neuroblastoma. On the other hand, in the adults, 127 patients (24.3%) had a thymoma and 66 patients (12.6%) had a lymphoma. Regarding benign mediastinal tumors, in children, 30 patients (28.5%) had a ganglioneuroma and 19 patients (18.0%) had a teratoma. In the adults, 68 patients (13.0%) had a teratoma and 62 patients (11.9%) had a thymoma. In the clinical manifestation, 51 child patients (48.1%) and 191 adult patients (36.6%) were asymptomatic. As symptoms due to compression or direct invasion to adjacent structures, dyspnea was seen in 16 child patients (15.1%), and chest pain occurred in 59 adult patients (11.3%). Dyspnea was significantly more common in children than in in adults (p < 0.01). Emergency operations were performed in children more than in adults.

[Surgical resection of pulmonary metastases from colorectal cancer].

Fourteen patients with known primary colorectal adenocarcinoma underwent pulmonary resection for its pulmonary metasoasis. The cumurative survival rate at five years was 25.7% in all patients and 37.5% in 9 patients with a solitary metastasis. One of two patients with both pulmonary and hepatic metastases survived 33 months with no evidence of recurrence after pulmonary and hepatic resection. Although none of seven patients with tumor smaller than 3.5 cm in diameter had regional lymph node metastasis, two of three patients with tumors larger than 3.6 cm in diameter had hilar lymph node metastases. Lobectomy and mediastinal lymph node dissection may be recommended for the patient with large tumor.

[Thoracoscopic partial resection of the lung for peripheral lung tumor].

We report a thoracoscopic partial resection of the lung for the peripheral lung tumor in a 66-year-old woman. Her chest roentgenogram and computed tomographic scan showed a single tumor 1.9 cm in diameter within the periphery of the right lower lobe. A solitary metastatic lung tumor of colon cancer, which was resected one year ago, was suspected. Partial resection of the lung involving this tumor was performed using the MULTIFIRE ENDO GIA 30 disposable surgical stapler under thoracoscopy. There was neither air leak nor bleeding at the resection line. Pathological examination of the tumor revealed benign localized pleural mesothelioma. Her postoperative course was uneventful and she was discharged from the hospital on the eighth postoperative day. Thoracoscopic pulmonary resection using EndoGIA is a safe technique for a peripheral lung tumor.

Activation mechanism of human Hageman factor-plasma kallikrein-kinin system by Vibrio vulnificus metalloprotease.

Vivrio vulnificus, an opportunistic human pathogen, secretes a metalloprotease (VVP). The VVP inoculated into a guinea pig is known to generate bradykinin through activation of the Hageman factor-plasma kallikrein-kinin system. VVP was shown to possess the ability to activate the human system through the same mechanism as that clarified in the guinea pig system, namely, VVP converted both human zymogens (Hageman factor and plasma prekallikrein) to active enzymes (activated Hageman factor and plasma kallikrein), and the then generated kallikrein liberated bradykinin from high-molecular-weight kininogen. However, in the presence of plasma alpha 2-macroglobulin (alpha 2M), the VVP action was drastically decreased. This finding suggests that the human system might be activated only at the interstitial-tissue space which contains negligible amounts of alpha 2M or in the bloodstream of the individuals whose plasma alpha 2M level is extremely reduced.

Occurrence of unusual heterogeneous lipid-containing granule-storing cells in the main excretory duct epithelium of the male mouse submandibular gland.

The fine structure of the main excretory duct epithelium of the male mouse submandibular glands was investigated by scanning and transmission electron microscopy. Three principal cell-types were observed: type I and II, and basal cells. This epithelium was characterized by the presence of intercellular canaliculi. Type-I cells were the most numerous. They had an abundance of mitochondria, well-developed Golgi apparatus, a few electron-lucent lipid-containing granules and poorly developed basal infoldings. These cells were also characterized by many glycogen granules throughout the cytoplasm and abundant smooth endoplasmic reticulum in the apical cytoplasm. Type-II cells were the second most numerous. Their most characteristic feature was the presence of abundant heterogeneous lipid-containing granules having acid phosphatase activity at the periphery. They were concentrated in the infra- and supra-nuclear cytoplasm. The granules may be derived from mitochondrial transformation and seem to be a special kind of secondary autolysosome. Type-II cells also contained abundant mitochondria throughout the cytoplasm, much smooth endoplasmic reticulum in the apical cytoplasm, a well developed Golgi apparatus adjacent to the heterogeneous lipid-containing granules and no basal infoldings. Basal cells were situated adjacent to the basal lamina. They had a large nucleus and the cytoplasm was filled with glycogen granules.

Genetic differences in steroid-induced protein synthesis in vivo of the liver and magnum in immature chicks (Gallus domesticus).

1. The present study was conducted to investigate whether or not the rate of steroid-induced protein synthesis measured in vivo in the liver and magnum was different between chicks from different genetic backgrounds. 2. Both protein deposition and synthesis in the liver were enhanced after the administration of beta-estradiol with or without testosterone. In the magnum, the combined administration of beta-estradiol and testosterone resulted in significantly higher protein deposition and synthesis than did beta-estradiol alone. 3. After the administration of beta-estradiol and testosterone, protein deposition in the liver and magnum was lower in broiler than in layer and dual-purpose chicks. Protein synthesis in the magnum tended to be highest, though not significantly, in layer, followed by dual-purpose and broiler chicks in decreasing order. 4. Steroid-induced protein deposition in the liver was not changed between chicks genetically selected for low and high albumen contents. In contrast, steroid-induced protein synthesis in the magnum was significantly higher in high-albumen birds than low-albumen chicks. 5. It was concluded that genetic traits for high egg production were well reflected in increased protein deposition and synthesis in the liver and especially in the magnum of immature female chicks stimulated by steroid hormone treatment.

Purification and characterization of a novel dipeptidyl carboxypeptidase from a Streptomyces species.

An extracellular protease derived from the culture broth of a microorganism, a Streptomyces species, produced Boc-Pro-Pro and diproline from Boc-Pro-Pro-Pro-Pro. The enzyme was purified 726-fold, with a yield of 2.6%, by ammonium sulfate fractionation, ion-exchange chromatography, and gel filtration chromatography. The molecular weight of the enzyme was determined to be 65,000 by gel filtration and 70,000 by SDS-PAGE. The enzyme released a C-terminal dipeptide from peptide substrates having a C-terminal proline and a penultimate proline or alanine residue, but did not hydrolyze angiotensin I or bradykinin. When the enzyme hydrolyzed Leu-Pro-Pro-Pro-Pro-Pro, it produced Leu-Pro-Pro-Pro and Pro-Pro before producing Leu-Pro. The enzyme thus seems to be a kind of dipeptidyl carboxypeptidase, its substrate specificity being very different from that of the well known dipeptidyl carboxypeptidases [EC 3.4.15.1] such as the angiotensin-converting enzyme.

Involvement of calcium in germination of coat-modified spores of Bacillus cereus T.

The effect of calcium on germination of coat-modified Bacillus cereus T spores was investigated. Coat-modified spores produced either by chemical extraction (SDS-DTT-treated spores) or by mutagenesis (10LD mutant spores) were unable to germinate in response to inosine. While SDS-DTT-treated spores could germinate slowly in the presence of L-alanine, 10LD mutant spores could not germinate at all. The lost or reduced germinability of coat-modified spores was restored when exogenous Ca2+ was supplemented to the germination media. The calcium requirement of coat-modified spores for germination was fairly specific. The simultaneous presence of germinant with Ca2+ was also required for germination of coat-modified spores. The optimal recovery of germinability was observed in the presence of 1.0 mM of calcium acetate. The calcium requirement itself was remarkably diminished under the condition in which L-alanine and a certain purine nucleoside analog, adenosine or inosine, coexisted. The lost or diminished germinability observed in SDS-DTT-treated spores or 10LD mutant spores may be attributed to the loss of calcium associated with the spore integuments.

Significant role of an exocellular protease in utilization of heme by Vibrio vulnificus.

Clinical and environmental isolates of Vibrio vulnificus could grow in a synthetic medium supplemented with heme protein as the iron source. Protease-deficient mutants of the bacterium could not utilize any of the heme proteins in the synthetic medium, but the addition of purified V. vulnificus protease restored their growth. The protease digested all heme proteins tested and elicited heme liberation from the proteins. Furthermore, the induction of protease production by the heme proteins was demonstrated. These observations suggest that protease contributes to the efficient utilization of heme by V. vulnificus.
