Light- and electron-microscopic studies on the secretory cytology of the adenohypophysis of the Japanese quail, Coturnix coturnix japonica.

The effects of thyroidectomy, adrenalectomy, and castration on the pars distalis of male Japanese quail, and of injection of LH-RH on sexually inactive females, were investigated by light and electron microscopy. Correlation between light and electron microscopy was attained by use of alternate thin and thick sections. Six types of secretory cells were identified and the ultrastructural characteristics described. Putative endocrine functions have been designated on the basis of responses to experimental interventions and on other criteria. The putative STH cells are characterized by the presence of large dense secretory granules (250-300 nm) that are stained with orange-G by the trichrome method. They occur only in the caudal lobe and appear to be unchanged by castration, thyroidectomy, adrenalectomy and LH-RH injection. The putative prolactin cells are characterized by large (400-600 nm), spherical or polmorphic, dense secretory granules stainable with acid fuchsin and aniline blue; prominent Golgi apparatus and well developed endoplasmic reticulum with densely packed, regularly parallel lamellae. They are found mainly in the cephalic lobe. The prolactin cells develop some vacuolization after adrenalectomy and undergo some degeneration after castration. The ACTH cells, which are restricted to the cephalic lobe, are identified by the dense, spherical granules (250-300 nm) that are stained with acid fuchsin. After adrenalectomy, they lose their secretory granules and are transformed into large, chromophobic adrenalectomy cells. TSH cells are so designated by their response to thyroidectomy including loss of their fine secretory granules and transformation to large, vacuolated thyroidectomy cells. We have found TSH cells and thyroidectomy cells only in the cephalic lobe. Basophilic cells, considered to be gonadotropes, occur in both the cephalic and caudal lobes. The gonadotropes of the cephalic lobe appear to have slightly larger (120-200 nm) granules than the caudal lobe (120-150 nm). However, after castration, the gonadotropes in both lobes become hypertrophied and vacuolated and are transformed into mutually indistinguishable castration cells. Twenty minutes after injection with LH-RH, the gonadotropes of both lobes increase in size and number, degranulate, develop vacuoles in the cytoplasm, and appear very similar to castration cells.

Adenylate cyclase activation promotes the recruitment of coronary vasodilator reserve and improves subendocardial contractility during coronary hypoperfusion.

This study was designed to examine the effects of an adenylate cyclase activator, NKH477, on epicardial and endocardial contraction and coronary blood flow (CoF) in the presence or absence of ischemia and to compare it to those of adenosine. We measured coronary pressures (CoP), coronary blood flow, epicardial and endocardial wall thickening (i.e., %EPWT and %ENWT, respectively, by sonomicrometry) in 18 anesthetized dogs. The left circumflex coronary artery was perfused with arterial blood using a pressure controlled servo pump. Propranolol (0.5 mg/kg) and atropine (0.25 mg) were used to minimize the neurogenic effects. CoP decreased from 100 mm Hg to 40 mm Hg with and without drugs. At CoP of 100 mm Hg, intracoronary infusion of NKH477 (10(-8) M/kg/min) produced a two-fold increase in CoF, but there were no changes in either the %EPWT or the %ENWT. During coronary hypofusion at coronary pressures equal to 40 mm Hg, NKH477 increased CoF from 16 +/- 2 to 28 +/- 4 mL/min (p < 0.05) and improved %ENWT significantly from 6 +/- 7 to 23 +/- 7% (p < 0.05). However %EPWT was not improved by NKH477. On the other hand, the intracoronary infusion of adenosine (10 micrograms/kg/min) increased CoF from 16 +/- 5 to 21 +/- 6 mL/min (p < 0.05) at CoP of 40 mm Hg. However, this dose of adenosine failed to improve %ENWT (16 +/- 10% vs. 14 +/- 10%, n.s.). Thus, the improvement of subendocardial function by NKH477 might be related to the improvement of subendocardial perfusion which could be induced by the potentiation of endogenously released adenosine as well as the direct vasodilator effect. This contrasts with the effects of exogenously administered adenosine, which failed to improve subendocardial contractility.

An early gallstone clearance following repeat piezoelectric lithotripsy.

Piezoelectric extracorporeal litotripsy was performed in 128 symptomatic patients with radiolucent gall-bladder stones to assess the significance of disintegration in fragment clearance. Up to 10 repeat lithotripsy sessions were scheduled to achieve a fragment target size of < 3 mm. Fragmentation assessed by the size of the largest fragments after the last session was graded into three classes. I: sludge-like disintegration, 18%; II: < 3 mm (mean +/- s.d., 1.7 +/- 0.5 mm), 56%; and III: > or = 3 mm (3.3 +/- 0.6), 26%. All patients were initially subjected to lithotripsy alone. Bile acid dissolution therapy was started only when ultrasonography failed to show the evidence of decrease in the < 3 mm fragments during a 1 month follow up. Finally, 69 patients (54%) were treated by lithotripsy alone, and the remaining 59 received additional dissolution therapy at a mean period of 2.5 months after the initial lithotripsy. The rate of complete clearance in class I, II and III patients was 91, 42 and 10% at 6 months and 100, 68 and 49% at 18 months, respectively. Significant differences were noted between the three fragmentation grades (I vs II, III, P < 0.0001; II vs III, P < 0.02). The patients with complete clearance within 6 months were seen only in those treated by lithotripsy alone, while the majority (87%) of patients with complete clearance during the later period were seen in those treated by additional dissolution therapy. We conclude that a high degree of fragmentation appears to lead stones to an earlier period clearance, and reduce the need for dissolution therapy.

Sonographic patterns of radiolucent gall-bladder stones for predicting successful shock-wave lithotripsy.

The aim of this study was to examine whether the sonographic patterns of gallstones are useful for predicting the outcome of piezoelectric shock-wave lithotripsy. Pretreatment analysis of gallstones based on our sonographic classification was conducted on 115 patients with radiolucent solitary stones of 10-30 mm in diameter, monitored for at least a year after the first lithotripsy. All stones were categorized as type I with gradual attenuation of echoes: type Ia, the stone echo appears as a full moon, usually accompanied by comet-tail artifacts beyond the stone itself (n = 55); type Ib, the stone echo showing the anterior half of the stone, seen as a half moon (n = 29); and type Ic, the stone echo seen as a crescent (n = 31). The most complete fragmentation, 'pulverization', was achieved at a significantly higher rate for type Ia (51%) than for type Ib (14%, P < 0.005) and type Ic (7%, P < 0.0001) after significantly fewer shock-waves (vs type Ib, P < 0.01; vs type Ic, P < 0.0001). The rate of complete clearance at 12 months after lithotripsy was significantly greater for type Ia (91%) than for type Ib (62%, P < 0.01) and type Ic (45%, P < 0.0001). Comparison of the sonographic and computed tomography (CT) patterns of stones revealed a close relationship between the two: the vast majority (98%) of type Ia showed the iso- or hypo-dense, and the majority (90%) of type Ic the rimmed.(ABSTRACT TRUNCATED AT 250 WORDS)

Morphogenesis of craniopharyngeal derivatives in the neurohypophysis of Fisher 344 rats: abnormally developed epithelial tissues including parotid glands derived from the stomatodeum.

Morphogenesis of craniopharyngeal derivatives of the neurohypophysis found in 14 Fischer 344 (F344) rats was studied. The incidence of the craniopharyngeal derivatives was 0.17% in male (7 out of 4,200) and 0.16% in female (7 out of 4,450) F344 rats. Neither a sex-related difference in their incidence nor a strain-related difference in their morphological features was observed. Craniopharyngeal derivatives were composed of aberrant epithelial structures consisting of serous acinar and tubular and fusiform cell structures, and most of these derivatives were associated with Rathke's cleft cysts, which are suggestive of a congenital background. The acinar structures were positive for periodic acid-Schiff reaction and negative for Alcian blue stain. Immunohistochemically, cells forming these structures were positive for cytokeratin, and basal cells of the acinar or tubular structures and some of the fusiform cells showed positive staining for alpha-smooth muscle actin. Electron microscopically, these spindle-shaped basal cells had intracytoplasmic myofilaments with focal density in their cytoplasm, and they were regarded to be myoepithelial cells. These findings strongly indicate that the craniopharyngeal derivatives are not a neoplastic lesion but rather are a developmental aberration derived from the stomatodeum, which is known to be the origin of both nasal and oral epithelial tissues, including the parotid glands, other than Rathke's pouch.

Eosinophilic granulated cells comprising a tumor in a Fischer rat.

A systemic tumor developed in multiple organs, including spleen, bone marrow, lymph nodes, liver, ovaries, and thoracic and lumbar vertebrae, of a female F344Du/Crj rat. The tumor was composed of round to indented mononuclear cells containing abundant large eosinophilic granules in the cytoplasm. The peripheral blood smear revealed that the large granules in the cytoplasm of the tumor cells were stained basophilic with Giemsa, suggesting a basophil or mast cell origin. However, these granules did not show metachromasia with toluidine blue and were stained blue to dark blue with phosphotungstic acid hematoxylin. Cellular morphology and characteristics in the specific stains of tumor cells suggested the development of a tumor of globule leukocytes in a F344Du/Crj rat.

A congenital ciliated epithelial cyst on the stomach of a B6C3F1 mouse.

Congenital anomalies of the alimentary tract are rare lesions in laboratory animals. We describe a congenital cyst attached to the greater curvature of the forestomach in a B6C3F1 mouse. The inner surface of the cyst was mostly covered with cuboidal or pseudostratified ciliated epithelium and was focally covered with the flat cuboidal epithelium. The base of the cyst appeared to be inserted between the layers of the outer longitudinal muscle layer of the forestomach, although no smooth muscle layer was evident in the free surface of the cyst wall. The cyst resembled duplication of the alimentary tract, as it was lined with ciliated epithelium and had developed at the greater curvature of the forestomach. Since the smooth muscle layer did not completely cover the whole wall and the cyst did not communicate with the gastric lumen, the cyst was not thought to be a standard duplication but rather a simple congenital cyst.
