Structural and biosynthetic characterization of the fibroblast growth factor receptor 3 (FGFR-3) protein.

Recently an additional member of the fibroblast growth factor receptor family, FGFR-3, was isolated. In this report, the structure and biosynthesis of the FGFR-3 protein product are investigated. In vitro transcription and translation of the three immunoglobulin-like domain form of FGFR-3 demonstrated the primary translation product to be approximately 97 kDa. However when analysed in COS-1 cells, this form of the receptor directed the expression of three polypeptides with apparent molecular weights of 97 kDa, 125 kDa and 135 kDa. Pulse-chase analysis, treatment of the cells with N-linked glycosylation inhibitors and digestion of these proteins with endoglycosidases demonstrated that the difference in molecular weights was the result of varying degrees of glycosylation. The 97 kDa protein was determined to be a non-glycosylated, cytoplasmic protein, whereas the 125 kDa protein was found to be a membrane-associated glycoprotein that is the biosynthetic precursor of the mature 135 kDa glycoprotein.

Parasites of South African wildlife. VIII. Helminth and arthropod parasites of warthogs, Phacochoerus aethiopicus, in the eastern Transvaal.

Helminth and arthropod parasites were collected from 41 warthogs, Phacochoerus aethiopicus, in the Hoedspruit Nature Reserve, eastern Transvaal. This reserve consists of a military base, which is a restricted area and is surrounded by a reserve, which is open to the public. Eleven nematode species, 1 or 2 cestode species and the larvae of 2 cestode species were recovered from the animals in the reserve, and 8 nematode species and 1 or 2 cestode species were recovered from those in the military base. Oesophagostomum spp. were generally most abundant in warthogs in the reserve during the cooler months of the year, while Probstmayria vivipara also occurred in peak numbers during the cooler months, with an additional peak in October and November 1988 in warthogs in the reserve and the base, respectively. No pattern of seasonal abundance could be determined for the other helminth species. The warthogs also harboured 8 ixodid and 1 argasid tick species, 3 flea species and 1 louse species. Adult and immature Haematopinus phacochoeri were most numerous during August and September, and the largest numbers of adult Rhipicephalus simus were present from December to April.

Derangement of adipose tissue: a case report of multicentric retroperitoneal liposarcomas, retroperitoneal lipomatosis and multiple subcutaneous lipomas.

No aetiological factors are at present known in the pathogenesis of liposarcomas. Benign lipomatous disorders, such as lipomas and lipomatosis, generally do not predispose to the development of liposarcomas. A case is presented of a patient with multicentric retroperitoneal liposarcomas, associated with retroperitoneal lipomatosis and preceded by a long history of multiple subcutaneous lipomas. In the literature this combination has not been described. This rare combination of lipomatous disorders seems to indicate a common derangement of adipose tissue.

Magnetic resonance imaging of the wrist.

Magnetic resonance imaging is one of the newest diagnostic techniques for evaluating the complex anatomy of the wrist. It is the only diagnostic modality that can directly depict abnormalities within the ligaments of the wrist and the TFC complex. Pathologic processes involving bone and soft tissue can also be demonstrated.

Crystals of complexes mimicking protein biosynthesis are suitable for crystallographic studies.

A complex of 70S ribosomes from Thermus thermophilus together with an average of 1.5-1.8 equivalents of PhetRNA(Phe) and a short mRNA chain, composed of 35 +/- 5 uridines, was crystallized under the conditions used for the growth of crystals of isolated ribosomes from the same source. Considering the reproducibility of their growth, their internal order and their shape, the crystals of the complex are superior to those of isolated ribosomes. In accord with previous three-dimensional reconstruction and modeling experiments, we conclude that the complex is less flexible and that an average population of complexes is more homogeneous than that of isolated 70S ribosomes. The crystals of the complex diffract to higher than 15 A resolution and can be irradiated with synchrotron X-ray beam at cryo-temperatures for days without noticeable decay. Since the crystals of the complex are apparently isomorphous with these of the isolated 70S ribosomes (P4(1)2(1)2; a = b = 526; c = 315 A), they should provide tool for phasing as well as for locating the mRNA and tRNA binding sites.

Localization of nerve growth factor receptor mRNA in the rat basal forebrain with in situ hybridization histochemistry.

1. In situ hybridization histochemistry was used to localize nerve growth factor receptor (NGFR) mRNA in the adult rat basal forebrain. 2. In emulsion-dipped sections 35S-labeled RNA antisense probes produced a high density of silver grains over cells located in the medial septum, vertical and horizontal limbs of the diagonal band of Broca, and nucleus basalis. 3. This distribution of NGFR mRNA overlaps with the distribution of NGFR protein localized using immunocytochemical techniques. 4. No hybridization signal was detected when sections were hybridized with a 35S-labeled RNA sense (control) probe. 5. We suggest that NGFRs are synthesized in these basal forebrain nuclei and transported to terminal areas where NGF is thought to be bound and internalized, an initial step in the many actions of this neurotrophic factor.

The effects of ethanol and diazepam on reactions to predatory odors.

In a straight alley containing a cat odor stimulus rats show high rates of risk assessment, including flat back approach and stretch attend behaviors oriented toward the threat stimulus and contact with the stimulus. In this situation, diazepam (2.0 and 4.0 mg/kg) significantly reduced risk assessment measures (flat back approach + stretch attend), while not reliably altering control locomotion (curved back). In combination with earlier findings that the same doses of diazepam reliably increased risk assessment from a movement arrest baseline, these results strongly support a behavioral model of anxiety involving predictable nonmonotonic changes in risk assessment as a function of anxiety reduction. In comparison to diazepam, ethanol had less pronounced effects on the Cat Odor Test, as in earlier tasks of reactivity to potential threat. However, reliable dose x time interactions for risk assessment measures suggest ethanol effects similar to those of diazepam but most pronounced in initial stages in the test session.

Wrist injuries: correlation of clinical and arthroscopic findings.

Wrist arthroscopy was done for one hundred nine patients with chronic wrist pain, averaging 22.8 months in duration. History regarding mechanism of injury, painful crepitus, pain with activity or at rest, localization of tenderness, visible swelling, and instability with examination was compared with arthroscopic findings of ligament damage, articular cartilage damage, and synovitis. Both pain at rest and swelling correlated significantly with synovitis. Tenderness did not correlate with specific ligament injury. Wrist ligament injuries and/or cartilage damage were noted in 96.3% of these wrists. Ligament injuries were frequently multiple, averaging 2.6 ligament injuries per wrist. No significant association was found between the presence of chondromalacia, synovitis, or specific ligament tears and the mechanism of injury, duration of symptoms, presence of clicking, or pain with activity.

Increased genetic risk of hypertension in glomerulonephritis?

The present study was carried out to test the hypothesis that the familial risk of hypertension is elevated in individuals with glomerulonephritis. Sixty-three parents of 39 consecutive patients below the age of 50 years who were admitted with biopsy-confirmed primary glomerulonephritis (excluding familial glomerulonephritis) were examined. Of these parents, 57% had blood pressure greater than or equal to 160/95 mmHg and/or were taking antihypertensive medication (81% of hypertensives). In 138 parents of 87 controls (age- and sex-matched patients admitted to a surgical ward for trauma), hypertension was found in 32.6% (of whom 73% were on antihypertensive medication). Age, sex ratio and body mass index were comparable in both groups of parents. A similar proportion of parents of patients with glomerulonephritis (27%) and parents of controls (25%) had already died. There was no evidence of excess cardiovascular or renal mortality in either group of parents. Urinary abnormalities were found in a similar proportion of the parents of patients with glomerulonephritis (5%) and controls (8%). Although the effects of a shared environment are not formally excluded, the data are consistent with a possible increased genetic risk of hypertension in patients with primary chronic glomerulonephritis.

Identification of fox rabies by a monoclonal antibody directed against nucleocapsid of a street rabies virus.

Hybridomas were prepared by fusion of spleen cells from BALB/c mice immunized with dog rabies isolates from Nigeria with P3x63Ag8 myeloma cells. More than 69 hybridomas secreted antinucleocapsid (antiNC) antibodies when tested with homologous viruses by indirect immunofluorescence. One hybridoma (Z144-88) was found which secreted antiNC antibody that reacted negatively with fox rabies isolates from the Federal Republic of Germany, Switzerland and France and with rabies-related viruses and European bat isolates. It reacted positively with other strains/isolates of rabies virus. It is possible to use this antiNC monoclonal antibody (mab) for the investigation of fox rabies outbreaks in Europe.

[Anamnestic, clinical and urodynamic comparative data from a group of patients with urinary stress incontinence].

Two hundred and seventy seven patients presenting genuine USI were divided into 3 groups based on a positive history for USI (group I), positive history and clinic for USI (group II), and positive history, clinic and urethral profile of USI (group III). When compared to a group of women without USI, the values for the FL and MUCP diminish progressively from group I to group III, giving evidence of increasingly severe impairment of the muscular-elastic environment in the urethra. Only in group III is the transmission factor (F) reduced, attesting a loosening/distension of the posterior pubo-urethral ligaments. The tonomeric values calculated for the patients with recurring USI following previous surgical correction (group IV) are comparable to values for group III. A scoring of severity of USI is proposed serving as a basis for establishing the criteria determining the indication for surgery and the post-operative prognosis before an intervention to correct USI is undertaken.

Histochemical and biochemical studies on the effect of the prostacyclin derivative iloprost on CCl4-induced lipid peroxidation in rat liver and its significance for hepatoprotection.

In the present study, it was investigated whether the prostacyclin derivative Iloprost would protect hepatocytes against CCl4-induced liver injury and which mechanism(s) of hepatocellular pathogenesis might be affected by it. Rats were treated with a single oral dose of CCl4 (2 ml per kg); Iloprost was infused continuously from 2 to 4 hr before intoxication until killing. The following results were obtained. The CCl4-induced release of AST, lactate dehydrogenase and alkaline phosphatase into the serum was reduced by 50 to 70% in rats treated with doses of 0.1 and 0.5 micrograms Iloprost per kg per min. Infusion of 0.02 and 0.004 micrograms Iloprost per kg per min did not affect the CCl4-induced enzyme release into the blood. CCl4 induced the occurrence of aldehydes (products of lipid peroxidation), which were detected by histochemical and biochemical means. At 12, 48 and 72 hr after CCl4, the aldehyde-positive liver section area was about 58, 69 and 16% in rats treated with CCl4 alone, but only about 18, 13 and less than 1% in rats treated additionally with Iloprost. The aldehyde-positive hepatocytes were located predominantly in the centrilobular zone of the liver. At 24 hr the extent of the aldehyde-positive section area was the same in rats with or without Iloprost treatment (about 90%). Biochemical determination, however, revealed that at this time point the malondialdehyde content after Iloprost in rats was about 70% lower than without Iloprost treatment.(ABSTRACT TRUNCATED AT 250 WORDS)
