Clinical studies were conducted on BRL 28500 (a formulation containing 15 parts ticarcillin plus 1 part clavulanic acid). BRL 28500 was administered at doses of 1.6 g or 3.2 g b.i.d., generally for 10 days by drip infusion to patients with pelvioperitonitis or Douglas' abscess. The results obtained were summarized as follows. 1. Clinical efficacy was evaluated in 18 patients (pelvioperitonitis 14, Douglas' abscess 4), but 8 patients out of a total of 26 patients were excluded. 2. In the evaluation of clinical improvement by doctors in charge, clinical improvement rates were 44.4% on day 3, 88.2% on day 5. 3. On the basis of committee judgement, the clinical efficacy rate was 100%. 4. The bacteriological eradication rate of causative organisms was 100% in 11 patients (15 strains). Five strains out of a total of 15 strains produced beta-lactamase. 5. As a side effect, nausea was observed in 1 case. In laboratory examination, liver function abnormalities were observed in 1 case. 6. Regarding usefulness as judged by doctors in charge, the satisfactory rate was 83.3%. From the above results, it has been concluded that BRL 28500 is very useful in the treatment of pelvioperitonitis and Douglas' abscess.

A case of successive metachronous colorectal cancers repeatedly resected 5 times during an 8-year period is reported. The 1st cancer was found in the sigmoid colon when the patient (male) was 32 years old. The 2nd was a mucosal carcinoma of the rectum which was locally excised. The 3rd was found in the ascending colon, and there was detected yet another small cancer in the adenoma in the resected specimen. The 4th and 5th cancers were found in the transverse colon and rectum, respectively. This case is considered to be highly associated with genetic factors. Long-term and careful surveillance is mandatory for early detection of recurrent or additional tumors.

To evaluate the usefulness and the safety of cefuzonam (CZON, L-105) against obstetric and gynecological infections, we conducted clinical trials and obtained the results summarized as follows. 1. Thirteen patients with obstetric and gynecological infections (intrauterine infection 2 cases, adnexitis 7, 1 case each of abscess of adnexa uteri, abscess of the vaginal wall, pyelonephritis, and mammitis) were treated with CZON. The CZON was administered by intravenous injection or intravenous drip infusion, 1 g twice daily for 4 to 7 days (8 g to 14 g in total). The clinical effect was good in 12 and poor in 1. 2. Four cases, on which other antimicrobial agents were ineffective, responded well to CZON. 3. The CZON displayed excellent effects on anaerobic bacteria (Peptostreptococcus anaerobius, Bacteroides fragilis) and Gram negative rods (Escherichia coli, Klebsiella pneumoniae). 4. No side effects or laboratory abnormalities were observed. 5. From the above results, CZON appeared to be an effective and useful drug for obstetric and gynecological infections.
Our previous study with 3H-thymidine autoradiography showed that neurons of the zona spongiosa, the nucleus proprius of the dorsal horn, the zona intermedia and the ventralhorn differentiated earlier than those of the substantia gelatinosa and the neck and the base of the dorsal horn, and that neurons of the substantia gelatinosa which were the last to differentiate reached their final position at stage 36 (Fig. 1). In the upper cervical cord of chick embryos at stage 36 when all spinal neurons finished cell migration and the cytoarchitecture similar to that of the cat spinal cord (Rexed, 1952) could be recognized (cf. Figs. 1, 3B), we studied the distribution of synapses by the electron microphotomontage (Fig. 3 A) and the morphology of axon collaterals coming from the white matter by the Golgi method (Fig. 4), in order to examine i) which spinal neurons have synaptic contacts at this stage and ii) what part of the axon collateral makes synaptic contacts. In the white matter, synapses were numerous around the gray matter and they were few in the peripheral part along the external surface of the cord. The paucity of synapses in the peripheral part was explained by a finding that dendrites reaching the external surface of the cord were few in number at this stage (cf. Fig. 3 C). In the gray matter, synapses were more numerous and denser in the zona intermedia and the ventral horn than in the dorsal horn.(ABSTRACT TRUNCATED AT 250 WORDS)
The addition to culture dishes of 10-50 ng/ml of the essential diterpene ester of Sapium sebiferum, 12-O-hexadecanoylphorbol-13-acetate (HPA), increased colony formation of normal peripheral blood lymphocytes co-cultured with gamma-irradiated HTLV-I-producing HUT 102 cells. The cells in the stimulated colonies showed an approximately 3-fold increase in the expression of interleukin-2 (IL-2) receptors and a 1.5- to 2.0-fold increase in human T-lymphotropic virus type- I (HTLV-I) p19-positive cells. This biological potency was analogous to that induced by the most potent tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA) and stronger than that of 12-O-hexadecanoyl-16-hydroxyphorbol-13-acetate (HHPA) isolated from Aleurites fordii.
Recombinant murine interferon-beta produced in Escherichia coli was purified and crystallized in an orthorhombic space group C222(1) with a = 61.67 A, b = 55.62 A, and c = 92.16 A. The crystals with a slight tendency for orientational disorder around the c axis diffract at least up to 3.3-A resolution. The crystallizability and the fact that the crystallographic asymmetric unit contains only one molecule of murine interferon-beta strongly indicate that the present preparation (Matsuda, S., Utsumi, J., and Kawano, G. (1986) J. Interferon Res., in press) of recombinant murine interferon-beta is predominantly homogeneous with respect to chemical, tertiary, and quaternary structures.
To elucidate the biological mechanisms of 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced phenotypic changes in HTLV-I virus-infected human T-cell line KH-2Lo cells, inhibitors of TPA-induced ornithine decarboxylase (ODC), protein kinases and calmodulin were examined for their effects on TPA-induced multinucleated cell formation and HTLV-I p19 antigen expression. Among the inhibitors of TPA-induced ODC activity, alpha-difluoromethyl ornithine (DFMO), 1,25(OH)2D3 and its analogues, and retinoic acid were tested. As inhibitors of protein kinases, 1-(5-isoquinolinyl-sulfonyl)-2-methylpiperazine (H-7), N-[2-(methylamino)ethyl]-5-isoquinoline-sulfonamide (H-8) and 1,(5-isoquinolynylsulfonyl)-2,3-dimethylpiperazine (H-5) were used. In addition, a calmodulin inhibitor, N-(4-aminobutyl)-5-chlor-2-naphthalenesulfonamide (W13) and its inactive analogue, N-(4-aminobutyl)-2-naphthalenesulfonamide (W12) were also tested. 1,25(OH)2D3 and its active analogues inhibited both TPA-induced HTLV-I p19 antigen expression and multinucleated cell formation after 4 days of culture with TPA. On the other hand, an inhibitor of ODC, DFMO, the protein kinase inhibitors, the calmodulin inhibitor and retinoic acid suppressed TPA-induced HTLV-I p19 expression but did not suppress multinucleated cell formation.

We performed a cytogenetic analysis of eosinophilic colonies grown from bone marrow culture of a patient with acute myelogenous leukemia (FAB-M2) associated with t(8;21). The bone marrow, in which 50.2% myeloblasts and 5.6% eosinophils were observed, gave rise to eosinophilic colonies, as well as colonies consisting of immature myeloid cells. The cytogenetic analyses of the colonies were performed on two occasions. The first study revealed t(8;21) in 15 of 16 metaphases in randomly plucked colonies from a dish, in which 65% of the total colonies were those consisting of immature myeloid cells and 35% were eosinophilic colonies identified by positive staining with Luxol-Fast-Blue. The second study, in which eosinophilic colonies were selectively plucked for cytogenetic analysis, showed Cq- and Gq+ markers, which were considered to be t(8;21), in all metaphases (6/6). Our present study strongly suggests that an involvement of the eosinophilic series in the leukemic process of this patient.
Recombinant mouse interferon-beta (rMuIFN-beta) produced in Escherichia coli was purified to homogeneity and characterized. The purified protein exhibited a single band of Mr 19,900 on SDS-polyacrylamide gel electrophoresis under both reducing and nonreducing conditions, and also exhibited a single band on native polyacrylamide gel electrophoresis at pH 4.3. The observed molecular weight corresponded to that of the polypeptide moiety of natural MuIFN-beta of Mr 19,700. The amino acid composition and the amino-terminal sequence of the purified rMuIFN-beta were identical to those predicted from cDNA sequence. These results indicate that the purified protein is a nonglycosylated MuIFN-beta, which forms no disulfide-linked dimer and probably exists as a monomeric form.
An outbreak of echovirus type 30 infection associated with aseptic meningitis occurred among newborn babies in a hospital neonatal room at Fukui City in 1983. The isolated virus was identified as an antigenic variant of echovirus type 30 by cross-neutralization tests with antisera against the prototype Bastianni strain and the present isolate. Western blot analysis demonstrated that the antigenic determinants responsible for virus neutralization, some of which were type specific and others strain specific, were present on the capsid protein VP1. The current strain was more thermoresistant than the prototype, suggesting some alterations in virus structural proteins.
