The effect of permethrin impregnated sisal curtains on vector density and malaria incidence: a pilot study.

Impregnation of bednets and curtains with suitable pyrethroids may reduce entomological inoculation rates (EIR) and malaria incidence. We conducted a quasi-experimental pilot study over 3 months in Western Kenya on 20 houses with 54 children. Ten houses in the experimental site received sisal curtains treated with permethrin at either 0.5g/m2 or 0.1g/m2. Control houses had untreated curtains or none. Mosquito vector density (MVD), man biting rates (MBR), and residual insecticidal effects (RIE) of permethrin were determined every two weeks. MVD was reduced by 97.7% and 98.7% in houses from the 2 experimental groups with a 60% reduction with unimpregnated curtains. MBR varied from 6.4 (no curtains), 1.7 (unimpregnated), 0.7 (0.5g/m2 curtains) to 0.4 (1.0g/m2 curtains). RIE begun to decline after the fourth month. Malaria incidence remained similar at the two sites. We conclude that covering of eaves and windows with permethrin impregnated sisal curtains can reduce MVD and the number of mosquito bites to individuals sleeping in protected houses.

Studies on Japanese encephalitis vector mosquitoes in Selangor, Malaysia.

Mosquito collections were carried out from May to June 1992 and from September to December 1992 in an area where a case of Japanese encephalitis was confirmed. A total of 40,072 mosquitoes belonging to 35 species and 8 genera were collected. The dominant species in that locality were Culex vishnui, Culex tritaeniorhynchus, Culex pseudovishnui, Culex gelidus, Aedes butleri, and Mansonia uniformis.

Studies on the effect of lambdacyhalothrin on Anopheles maculatus Theobald and its response to residual spraying at Jeram Kedah, Negeri Sembilan, Malaysia.

A field trial was carried out to study the effect of lambdacyhalothrin on Anopheles maculatus in trap huts in Jeram Kedah, Negeri Sembilan, Malaysia. Two trap huts were built, of which one was sprayed with lambdacyhalothrin at a dosage of 25 mg ai/m2 and the other served as control. Eight collectors commenced collecting mosquitos from 1900 to 2400 hours, two each indoors and outdoors. Bioassay was also carried out in the treated and control huts to determine susceptibility of adult mosquitos to lambdacyhalothrin. In the treated hut more mosquitos were present during the pre- spraying period. Lambdacyhalothrin gave a mortality of 100% against An. maculatus for 8 months.

Glutathione conjugation and pharmacokinetics of 2-bromo-3-phenylpropionic acid in vitro and in the rat in vivo.

Glutathione (GSH) conjugation of the chiral compound 2-bromo-3-phenylpropionic acid (BPP) was studied in vitro and in the rat in vivo. GSH conjugation of BPP, catalyzed by a mixture of glutathione-S-transferases (GST's) from rat liver cytosol in vitro, was stereoselective: at a substrate concentration of 250 microM, (R)-BPP was more rapidly conjugated than (S)-BPP (R/S-ratio = 2.6). The blood elimination kinetics of the separate BPP enantiomers and the biliary excretion kinetics of the corresponding GSH conjugates were studied in the rat in vivo after administration of (R)- or (S)-BPP at a dose level of 50 mumol/kg. Elimination of (R)-BPP from blood was faster than that of (S)-BPP: half lives were 9 +/- 2 min for (R)-BPP and 13 +/- 1 min for (S)-BPP. The biliary excretion rate of the GSH conjugate of (R)-BPP declined monoexponentially, while that of the GSH conjugate of (S)-BPP displayed a biphasic profile. Half lives of excretion were 13 +/- 1 for the GSH conjugate of (R)-BPP, and 11 +/- 2 for the GSH conjugate of (S)-BPP (second phase). The first phase in the biliary excretion of the GSH conjugate of (S)-BPP could not be attributed to capacity limitation of biliary transport carriers as higher excretion rates were attained upon administration of higher doses (100 and 200 mumol/kg) of (S)-BPP). The blood elimination profiles of (R)- and (S)-BPP differed greatly from the biliary excretion profiles of the corresponding GSH conjugates. This suggests that the kinetics of BPP conjugate excretion are determined by other processes than hepatic GSH conjugation.

Effects of bentonite on wool growth and nitrogen metabolism in fauna-free and faunated sheep.

Two experiments were carried out with sheep that originated from a fauna-free flock and were fed a soybean meal-corn silage diet with or without a bentonite supplement. One-half of the sheep fed each diet in each experiment were faunated with a mixed population of ruminal protozoa, whereas the other half of the sheep remained fauna-free until the end of both experiments. Wool growth and daily gain were measured in Exp. 1. (eight rams per treatment), which lasted 110 d, and the metabolic effects in the rumen and intestinal tract of protozoa and dietary bentonite supplement were tested with cannulated wethers (four wethers per treatment) in Exp. 2. The results of Exp. 1 showed decreased wool growth (P less than .05) due to the presence of protozoa in the rumen. Dietary supplementation with bentonite partly offset the decreased wool growth in sheep with protozoa, but there were no effects of dietary bentonite and no protozoa x bentonite interaction (P greater than .05). Daily gain was decreased by the dietary bentonite (P less than .05) supplement but was not affected (P greater than .05) by the ruminal presence of protozoa. In Exp. 2, protozoa increased (P less than .01) the ruminal concentrations of ammonia and decreased (P less than .05) the acetic:propionic acid molar ratio. Fractionation of N in the duodenal digesta flowing from the stomach to the small intestine showed that protozoa decreased (P less than .05) the flow of nonammonia N and bacterial N, and there was a protozoa x bentonite interaction for these effects (P less than .05).(ABSTRACT TRUNCATED AT 250 WORDS)

Comparative determination of Plasmodium falciparum sporozoite rates in Afrotropical Anopheles from Kenya by dissection and ELISA.

The Plasmodium falciparum rate was determined by microscopical examination of one salivary gland (three lobes) and by enzyme-linked immunosorbent assay (ELISA) of the other salivary gland in each of 1580 Anopheles mosquitoes collected from western Kenya during both the wet and dry seasons. The sporozoite rate in the wet season was much higher than that in the dry season, and the sporozoite rate determined by ELISA was generally lower than that determined by microscopy. The ELISA gave a positive reaction to circumsporozoite protein in some glands whose counterparts did not show the presence of sporozoites by microscopy, thus giving an 'overestimation' of the sporozoite rate. This overestimation was greater in the dry season than in the wet season, and greater in Anopheles gambiae than in An. funestus, but overall it was only 1.2% (19/1580). These results are at variance with reports of other workers, who have shown ELISA overestimation of the sporozoite rate as high as 30%. Our tests indicated that the ELISA sensitivity was 80.6%, its specificity was 98.7%, and its accuracy was 97.5%.

Tc, an unusual promoter element required for constitutive transcription of the yeast HIS3 gene.

Tc is the proximal promoter element required for constitutive his3 transcription that occurs in the absence of the canonical TATA element (TR) and is initiated from the +1 site. The TC element, unlike TR, does not respond to transcriptional stimulation by the GCN4 or GAL4 activator protein. Analysis of deletion, substitution, and point mutations indicates that Tc mapped between nucleotides -54 and -83 and is a sequence-dependent element because it could not be functionally replaced by other DNA sequences. However, in contrast to the behavior of typical promoter elements, it was surprisingly difficult to eliminate Tc function by base pair substitutions. Of 15 derivatives averaging four substitutions in the Tc region and representing 40% of all possible single changes, only 1 inactivated the Tc element. Moreover, the phenotypes of mutant and hybrid elements indicated that inactivation of Tc required multiple changes. The spacing between Tc and the initiation region could be varied over a 30-base-pair range without significantly affecting the level of transcription from the +1 site. From these results, we consider it possible that Tc may not interact with TFIID or some other typical sequence-specific transcription factor, but instead might influence transcription, either directly or indirectly, by its DNA structure.

A mark-release-recapture experiment with Mansonia mosquitos in Malaysia.

An experiment was carried out with Mansonia mosquitos in an area endemic for subperiodic Brugia malayi to assess the applicability of the mark-release-recapture method to these mosquitos. An estimated 17,880 individuals of six species of Mansonia were marked with fluorescent dust and released: 453 Ma. annulata, 305 Ma. annulifera, 6,200 Ma. bonneae, 516 Ma. dives, 3,998 Ma. indiana and 6,408 Ma. uniformis. Twenty-three marked individuals were recaptured. Most recaptures were made one or two nights after their release, but one Ma. annulifera was recaptured five nights later and one Ma. bonneae had been marked and released 6-11 nights previously. The recaptured mosquitos were collected between 0.5 and 2.4 km from their release points.

Typhoid fever below five years.

During a seven year period (1981-87), 53 cases of typhoid fever in children below five years were seen. This accounted for 13.5% of all typhoid admission in pediatrics during the above period. Predominant symptoms were fever (100%), vomiting (52.8%), diarrhea (30.2%) and anorexia (24.5%). Chills and rigor associated with fever was noticed in 38% of the children. Salmonella typhi was isolated from blood in 22 of 53 (41.7%) cases. A significant finding on peripheral smear was eosinopenia (86.8%). Complications like endotoxic shock, enteric encephalopathy and gastrointestinal hemorrhage were noticed in only few cases (7.6%). There were no deaths.

Poliomyelitis: 20 years--the Pondicherry experience.

During a 20-year period, 12,830 cases of acute poliomyelitis in children (aged 1 month-12 years) were seen in a South Indian teaching hospital (Pondicherry). Almost three-quarters (72.4%) of children were below the age of 2 years. The median age at onset of poliomyelitis shifted from 19.5 months (in the period 1968-1977) to 11 months (in the years 1978-1987). After 1981, epidemic peaks were noted every third year. 66.3% of cases had not received even a single dose of oral polio vaccine and 14.7% cases were vaccine failures (in the years 1983-1987). Provocation of poliomyelitis occurred in 66% of children and usually followed intragluteal injections associated with treatment of non-specific fevers. The overall mortality was 2.13%, the major cause of death being respiratory failure (60%). Polio virus type 1 was the major isolate among selected cases studied during 1983-1987. The risk factors identified in our study are lack of immunization and administration of intramuscular injections during the pre-paralytic phase. Ensuring early administration of multiple doses of oral polio vaccine to infants and avoiding intramuscular injections in young febrile children are important measures for the control of poliomyelitis--a continuing problem in Southern India.
