[Electron microscopic studies of cell cultures of various origins].

Thirty-four cell cultures of various origins were examined in the electron microscope. Mycoplasma were found in 31 cultures, A, C and D particles of oncornaviruses in 27 cultures, adenovirus in one culture and paramyxovirus in 8 cultures. Two cultures were found to be virus- and mycoplasma-free. A stringent control of cell cultures for the presence of viruses, mycoplasma or other microbiological agents is recommended.

[The vaccinal prophylaxis of hepatitis B in Russia--the achievements, problems and outlook].

Certain specific features of the present epidemic situation with hepatitis B (HB) in Russia were established: significant growth of HB morbidity, starting from 1995; the prevalence of persons aged 15-29 years among HB patients, which was linked with the sharp activation of the sexual route of the transmission of HB virus in recent years; an essential increase in the number of patients having contacted this virus in the process of the intravenous use of drugs. The results of the use of vaccine "Engerix B" among persons belonging to different risk groups were considered (a decrease in HB morbidity among them by 8-19 times was noted), the study demonstrated high immunogenicity anti-HBs antibodies on protective titers were determined in 92.3-95.7% of the vaccinees) and low reactogenicity of the vaccine, as well as stable postvaccinal immunity (5 years after the course of vaccination was completed anti-HBs antibodies were retained in 70.6-74% of the vaccinees). The study showed that only the vaccination of adolescents in combination, in the presence of opportunity, with the immunization of newborn infants and young children in the first year of their life made it possible to produce an essential effect on the activity of the epidemic process. Already in 2 years such organization of work on the prophylaxis of HB in one of the cities of the Sverdlovsk region led to a decrease in HB morbidity by 2.9 times, and among adolescents 9 times.

[Morphogenesis of Okhotsky virus from the genus orbisvirus].

Morphogenesis of Okhotsky virus isolated in the USSR was studied. Virions of 50-65 nm in diameter were found in the peripheral zone of cytoplasmic matrices. Crystalline-like arrays of virions were observed not reported by other investigators studying morphogenesis of orbisviruses. Besides, not directly associated with matrices, crystalline arrays of some proteins were found in infected cells for the first time with orbisviruses. Nevertheless the main stages of morphogenesis of Okhotsky virus were similar to those of other members of the orbisvirus group.

[The characteristics of human immunodeficiency virus strains isolated from HIV-infected persons on the territory of the USSR].

Fifty-seven HIV-1 strains were isolated from peripheral blood lymphocytes of 102 HIV-infected persons involved in epidemic outbreaks in different cities of the USSR. The effectiveness of isolation was 29.1% in asymptomatic infection, 51.7% in cases with generalized lymphadenopathy, and 82.6% in persons with severe clinical manifestations. Identification of the isolates by indirect immunofluorescence, ELISA, reverse transcriptase activity, Western blot, electron microscopy, and polymerase chain reaction showed them to belong to HIV-1 type. Reproduction of the isolates in cell cultures was accompanied by cytopathic effect and syncytium formation. The isolated strains can be divided into two groups: (1) the poorly growing isolates with low or negative RT and ELISA results and (2) the isolates with high infectivity, broad spectrum of cell tropism, and high levels of RT and ELISA. These data show the correlation of biological properties of HIV-1 strains isolated in the USSR with those of HIV strains previously isolated in Europe, USA, and Africa.

[Detection of hepatitis C virus RNA in the liver biopsy specimens from patients with chronic hepatitis C and hepatocellular carcinoma by means of in situ RT-PCR].

The in situ RT-CPR technique has been first adapted in Russia to detect hepatitis C virus (HCV) RNA in the samples from patients with chronic hepatitis C (CHC) and hepatocellular carcinoma (HC). A total of 18 patients with CHC and HC were examined. Fourteen (78%) samples were ascertained to be positive. A positive reaction was found in 2 of 3 patients with HC. A label was revealed in individual hepatocytes without any regularity in the distribution along the tissue section. A reaction was negative in all samples from control groups. The label was observed in both the cytoplasm and the hepatocytic nuclei. There was no correlation between the degree of hepatic lesion, estimated after Knodelle, the results of the classical RT-CPR and the in situ RT-CPR techniques.

[Identification of a hemadsorbing agent discovered in uninfected mouse L cell cultures and also the same cultures chronically infected with Sindbis virus].

Electron microscope examinations of continuous lines of mouse L cells, both uninfected (L-init) and chronically infected with Sindbis virus (L-SV) revealed accumulations of ribonucleoprotein strands and virions corresponding by their parameters to paramyxoviruses in the cytoplasms of the cells. Further studies showed L-init and L-SV cell lines to have a manifest hemadsorption effect which could be completely inhibited by antiserum to parainfluenza SV5 virus. Immunofluorescence procedures detected intensive fluorescence in the cytoplasm of these cells which was observed only after treatment of the cells with antiserum to SV5 virus. In response to inoculation of cell homogenates of continuous L-init and L-SV cultures guinea pigs developed antihemagglutinating antibody to simian parainfluenza SV5 virus. On the basis of these results it may be assumed that virus-specific structures and viruses identical by their parameters to paramyxoviruses observed in electron microscope examinations of continuous mouse L-init and L-SV cells are simian parainfluenza SV5 virus.

[Interaction of Sindbis virus with a culture of cells producing oncornavirus].

Electron microscopy and biophysical methods were used for examinations of primarily trypsinized chick fibroblast cell culture spontaneously producing C-type oncornavirus at 1, 6, and 24 hours after inoculation with Sindbis virus. During the first 6 hours rapid maturation and release of oncornavirus from cells were observed. At later intervals oncornavirus production was inhibited. It is assumed that in the system under study, biosynthesis of oncornavirus and Sindbis virus occur separately.

[The localization of delta antigen in the hepatocytes in chronic delta hepatitis].

The results of immunohistochemical and electron microscopic examinations of liver biopsies from 47 patients with chronic delta hepatitis are presented. Delta antigen (HDAg) was detected by immunoperoxidase method in the liver of 35 (74.4%) patients. HDAg was demonstrated in hepatocyte nucleus alone in 22 (62.9%) patients, simultaneously in the nucleus and the cytoplasm in 9 (25.7%), in the cytoplasm alone in 4 (11.4%). In nuclear localization, delta antigen may fill the entire nucleoplasm or only the peripheral zone of the nucleus. When delta antigen fills the entire nucleus, hepatocytes undergo more marked pathological changes than those in which HDAg is localized in the nuclear periphery or those where no delta antigen was found.

[The phenomenon of transition of filamentous virions of influenza viruses into spherical virions].

A type of transition of filamentous influenza A virus virions into spherical ones differing from fragmentation is described. The transition occurs through the formation of racket-like particles. Such transitions may be initiated by deformations occurring in filamentous virions upon storage. The stability of filamentous virions depends greatly upon the ionic strength of the solution. The main regularities of this process are described. A possible mechanism of formation of filamentous virions and the factors determining their stability are discussed.

[Isolation of polymeric glycoprotein complexes of enveloped viruses using the Quill A glycoside and a study of their immunogenic activity].

Specific multimer complexes of glycoproteins of enveloped viruses were prepared by treatment of suspensions of purified concentrated virus with a non-ionic detergent MECK mixed with 0.2% glycoside Quil A. Mixed complexes of glycoproteins with glycoside Quil A were formed upon removal of the detergent from the mixture of solubilized glycoproteins and glycoside Quil A. According to the results of electron microscopy, the formed complexes differed morphologically from the conventional micelles of glycoproteins and presented spherical reticular structures 20-30 nm in diameter. The immunogenic potency of the complexes was much higher than that of virus particles and micelles of purified glycoproteins and was comparable to the immunogenic potency of glycoproteins mixed with complete Freund adjuvant. The protective activity of the complex of protein G of rabies virus with glycoside Quil A was higher than that of the subunit rabies vaccine adsorbed on aluminium hydroxide.

[Isolation and characteristics of endocytic vacuoles containing the influenza virus].

Endocytic vacuoles (receptosomes) containing influenza virus were isolated from the cytoplasm of Ehrlich ascitic carcinoma cells and characterized. In the sucrose density gradient, the virus-containing material was detected in two peaks with a buoyant density of 1.175-1.16 and 1.155-1.135 g/cm3 with which the activity of marker enzymes of cell plasma membranes was associated. The virus was present in receptosomes in morphologically and electrophoretically intact condition. Examinations for the lipid composition of endocytic vacuoles showed the presence in their membranes of large amounts of cholesterol and glycolipids, particularly asialo-GM1 which, according to some authors may enhance the fusion of viral and cell membranes.
