Agrobacterium tumefaciens mediated transformation of Orychophragmus violaceus cotyledon and regeneration of transgenic plants.

Excised cotyledons of Orychophragmus violaceus were used as explants for tissue culture. They were cultured on the MS medium supplemented with BA (3 mg/L) and NAA (0.2 mg/L). When the regenerated buds were 2 cm long, they were excised and transferred onto 1/2 MS medium with IBA (0.03 mg/L), then the whole plants were regenerated. The frequency of plant regeneration was 100%. Subsequently, the genetic transformation of O. violaceus was studied. After 2-3 days of cocultivation with Agrobacterium tumefaciens strain A208se (pTiT37, pROA93), the cotyledons were transferred onto the selection medium containing 25 mg/L Km and 250 mg/L Ap. After shoots emerged, they were excised and transferred onto the rooting medium containing 25 mg/L Km and 100 mg/L Cef. The roots were formed within 4-5 weeks. The whole plants were transplanted into pots and grew well. The frequency of plant regeneration was about 51%. The regenerated plants showed high enzymatic activities of beta-glucuronidase and neomycine phosphotransferase II. Southern blot analysis confirmed that NPTII gene had been stably integrated into the chromosomal genome of O. violaceus. The transformation frequency was 5.6%. The first transgenic plant of O. violaceus is being reported.

[Effect of Chinese herbs on the circadian rhythm of body temperature and heart rate in rabbits with hypothyroidism (yang deficiency)].

The rectal temperature (Tr) and heart rate (HR) of three groups of New Zealand white rabbits, i. e. thyroidectomized (G1), thyroidectomized and treated with Chinese herbs (a decoction composed of tonifying kidney-yang herbs) (G2) and pseudothyroidectomized for control (G3), were observed dynamically for 26 hours. The results indicated that the Tr and HR in G1 were decreased, and the circadian rhythms of the Tr and HR disordered significantly, while these changes in G2 were corrected obviously.

[The changes in firing rates of ventromedial hypothalamic neurons and its modulations by electroacupuncture, arcuate nucleus and locus coeruleus during the immune response].

30 adult SD rats were divided into 3 grous: normal control goup; SRBC immune group (on 5th day after SRBC injection) and MSG(MSG injection at newborn stage) + SRBC group. The changes of the spontaneous firing rates of ventromedial hypothalamus (VMH) neurons and evoked responses by electroacupuncture (EA) and locus coeruleus (LC) stimulation were recorded. The results were as follows: (1) The spontaneous firing rate of VMH neurons was significant increase in the SRBC group. The firing rate in the MSG + SRBC group was higher than that in the control group, and lower than that in the SRBC group. There were statistical differences between them. (2) EA can increase the firing rates in all groups. The effect of EA was the most significant in the SRBC group, and more significant in the MSG + SRBC group. (3) The firing type of most VMH neurons showed first excited then inhibited biphasic response following stimulation LC in the SRBC and the MSG + SRBC groups. EA showed some increase in effect of the excited modulation from LC to VMH in the SRBC group. The results suggest that VMH might be an important neuroendocrinal link in immunoresponse. EA can increase the immune function of SRBC on VMH. LC and beta-endorphine neurons of arcuate nucleus might participate in the EA mediated immunoresponse.

[Extremity gangrene caused by rare co-infection of multiple bacteria: a case report].

UNLABELLED: Because of progressive infectious necrosis on right hand and forearm for 3 months and the necrosis on left mid-finger for 2 months, a girl patient, Yang Xiaoxia was admitted on 15th, Nov. 1994. Systemic intoxicative symptom was slight and the necrotic tissue presented black scar. After 3 months treatment by application of multiple antibiotics, local dressing changes and an amputation of right froearm, the necrosis could not be stopped but keeping deteriorating. Through experts' consultation, systemic surpportive therapy, wide-spectrum antibiotics cilastatin sodium and complete debridement were adopted. One week later, the fresh surface of granulation tissue was seen, and skin grafting was performed. The wound healed 3 months later. After further rehabilitation, the left thumb, index and fifth finger showed normal function. A mechanical motional artificial limb was installed on right forearm and discharged. The deep necrotic tissue of left mid-finger was examined. Results of culture from several hospital presented 2 kinds of aerobic bacteria and 10 kinds of the anaerobic, of which, the names of 3 kinds are still unknown. Pathological examination showed chronic pyogenic necrotic infection, spreading from skin to deep muscles and interosseous membrane. And infiltration of eosinophilic granulocytes was found in the infectious region. CONCLUSION: This patients is progressive gangrene of the extremity caused by rare co-infection of multiple bacteria. The characteristics were slight systemic intoxicative symptom, local progressive necrosis and formation of black scar. By means of systemic surpportive therapy, application of wide-spectrum antibiotics cilastatin sodium, complete debridement and skin grafting, the patient was cured.

[Cross-section anatomy of the subphrenic spaces].

The cross-section anatomy of the subphrenic spaces were investigated in 20 Chinese cadavers. On the horizontal sections, the subphrenic spaces consist of the perihepatic and perisplenic spaces. The fissure for ligamentum venosum was separated into two parts by the lesser omentum, the anterior part communicates with gastrohepatic recess, the posterior is the superior recess of the omental bursa into which the caudate lobe of the liver projected. The bare area of the stomach encroaches upon the posterior surface of the gastric fundus between the right and left layers of the gastrophrenic ligament and lies between the superior and splenic recesses of the omental bursa. Left subphrenic retroperitoneal space is between the bare area of the stomach and the diaphragm.

[Expression of retroviral transduced human beta-globin gene in mouse fetal liver cells].

A replication-defective retrovirus vector containing the human beta-globin gene and 36-base-pair HS2 enhancer (N2A beta E36) was constructed and transferred into psi-2 and PA317 packaging cell lines. Titers of amphotropic recombinant retrovirus ranged from 10(3) to 10(4) CFU/ml. Southern blot analysis indicated that intact human beta-globin gene was stably integrated into the genome of the target cells. Northern blot hybridization analysis showed that human beta-globin gene could be expressed in mouse fetal liver cells.

[Evaluation of clinical and pathological factors relating to lymph node metastasis in carcinoma of the tongue].

In this paper we adopt various clinical and pathological parameters to evaluate the factors relative to the lymph node metastasis. We are of the view that the TNM classification can not indicate the presence of the lymph node metastasis clearly. When the size of tumor is less than 2.0 cm in diameter there is almost no metastasis. Using computer, we analyse the factors and found that only the depth, pattern of growth and the degree of lymphoid cell infiltration are relative to metastasis. The coincidence is 78.6%.

[Effects of hypertonic sodium lactate dextran resuscitation in severely burned dogs].

12 dogs with 35% TBSA third degree burns received HLD resuscitation (HLD group, n = 6) or LR resuscitation (LR group, n = 6). Fluid resuscitation started one hour postburn. The amount of fluid infused with HLD resuscitation was calculated by that after giving HLD 19.6 ml/kg in 3 hours and 6 ml/kg/% TBSA lactate Ringer's solution followed. The amount of fluid infused with LR resuscitation was calculated by 8 ml/kg/% TBSA lactate Ringer's solution. Infusion of lactated Ringer's solution in both groups was adjusted by maintaining urinary output 0.5-1 ml/kg/h. The volume of fluid infused in HLD group (5.05 +/- 1.11 ml/kg/% TBSA) was much less than that of LR group (10.03 +/- 1.30 ml/kg/%TBSA) (P < 0.01). There was no significant difference in urinary output, serum Na+ and albumin, and plasmacrystalloid osmolarity between two groups. Plasma level of MDA decreased after resuscitation with HLD, which (0.81 +/- 0.20 mmol/g Hb) was much lower than that (1.39 +/- 0.44 mmol/g Hb) of LR group 4 hours postburn (P < 0.05). Plasma SOD activity (7.22 +/- 0.68 u/g Hb) of HLD group were much higher than that of LR group (4.86 +/- 0.53 u/g Hb) 4 hours postburn (P < 0.05). HLD resuscitation could significant reduce the amount of fluid infused comparing with lactate Ringer's solution. HLD resuscitation could attenuate postburn damage to tissue induced by lipid peroxide by elevating plasma SOD activity.

[Effect of epidermal growth factor and dexamethasone on blast hearing loss].

The expression of epidermal growth factor receptor (EGFR) in cochlear hear cells was studied in normal and guinea pigs exposed to blasts. The effects of epidermal growth factor (EGF) and dexamethasone (DXM) treatment for blast hearing loss and the underlying mechanisms were also explored. Scattered expression of EGFR was seen in IHCs and OHCs in normal guinea pigs. Segmentally distributing positive reaction was also located in stereocillia of hair cells. Distribution of EGFR reaction was seen in the cytoplasm of IHC 24 hours after exposure to blasts, and in the stereocillia of IHC and the cuticular plate of OHC 72 hours postexposure. At one week EGFR reaction in hair cells increased obviously and part of OHC stereocillia also showed positive reaction. EGFR reaction reduced at two weeks, though positive reaction could still be found in the stereocillia of hair cells at one month. Combination of EGF and DXM administrations promoted hearing recovery significantly. Our results suggest that healing of injured hair cells may be to EGF.

[Experimental study of streptomycin on 45Ca intake of otolithic membranes].

45CaCO3 was injected into guinea pigs, and the radioactivity in otolithic membranes of utriculi and sacculi, auditory ossicles and femora was determined by liquid scintillation spectrometry, to study the dynamic effect of streptomycin on calcium content of otolithic membranes, as a means to explore the mechanism of streptomycin ototoxicity. The results showed that otoconium was a dynamic structure which took up 45Ca in a time course generally comparable to that of bone. Saccular otolithic membranes showed greater intake than utricular membranes. The calcium ion concentration of saccular endolymph was greater than that of the utriculus and the two maculas were not identical. Streptomycin may influence 45Ca uptake in the two macular otolithic membranes, causes calcium metabolic disorder and a fall of otolithic calcium content. It is possible that the fluid environments of the endolymph have changed, therefore dysfunction of otolithic organ may occur. This hypothesis may be one of the explanation of the mechanism of streptomycin ototoxicity.

Targeting c-erbB-2 expressing tumors using single-chain Fv monomers and dimers.

Single-chain Fv proteins containing a COOH-terminal cysteine (sFv') were constructed by using an antidigoxin 26.10 sFv and an anti-c-erbB-2 741F8 sFv. The fully active sFv' proteins were prepared by expression in Escherichia coli as insoluble inclusion bodies, followed by in vitro refolding using glutathione redox buffers and purification. The COOH-terminal cysteines of the refolded sFv' proteins were protected by a blocking group presumed to be the glutathionyl peptide, which was easily and selectively removed by gentle reduction. Air oxidation of the reduced sFv' monomers resulted in the efficient formation of disulfide-linked sFv' homodimers, designated (sFv')2, which were stable under oxidizing conditions and relatively slow to be disrupted under reducing conditions. The (26-10-1 sFv')-(741F8-1 sFv') heterodimer was prepared and possessed dual-antigen specificity; the active bispecific (sFv')2 dimerized under native conditions, apparently as a manifestation of self-association by the 741F8 sFv' subunit. Biodistribution and imaging studies that were performed on mice bearing human SK-OV-3 tumor xenografts that express the c-erbB-2 as a cell surface antigen were reviewed. Radioiodinated 741F8-2 (sFv')2 homodimer localized to the tumors with high specificity, as evidenced by excellent tumor:normal tissue ratios. Sagittal section autoradiography of whole animals 24 h after administration of antibody species revealed that 741F8 (sFv')2 produced a stronger tumor image than comparable doses of the 741F8 Fab, monomeric sFv', and the 26-10 (sFv')2 control without the high nonspecific background distribution of the 741F8 IgG.

Interaction of substrate and effector binding sites in the ArsA ATPase.

The ars operon of plasmid R773 confers resistance to antimonials and arsenicals in Escherichia coli by encoding an ATP-dependent extrusion system for the oxyanions. The catalytic subunit, the ArsA protein, is an ATPase with two nucleotide binding consensus sequences, one in the N-terminal half and one in the C-terminal half of the protein. The ArsA ATPase is allosterically activated by tricoordinate binding of As(3+) or Sb(3+) to three cysteine thiolates. Previous measurements suggested that the intrinsic fluorescence of tryptophans might be useful for examining binding of Mg2+ ATP and antimonite. In the present study an increase in intrinsic tryptophan fluorescence was observed upon addition of Mg2+ ATP. This enhancement was reversed by addition of antimonite. The ArsA protein contains four tryptophan residues: Trp159, Trp253, Trp522, and Trp524. The first two were altered to tyrosine residues by site-directed mutagenesis. Cells expressing both the arsAW159Y and arsAW253Y mutations retained resistance to arsenite, and the purified W159Y and W253Y proteins retained ATPase activity. While the intrinsic tryptophan fluorescence of the W253Y protein responded to addition of Mg2+ ATP, intrinsic tryptophan fluorescence in the purified W159Y protein was no longer enhanced by substrate. These results suggest that Trp159 is conformationally coupled to one or both of the nucleotide binding sites and provides a useful probe for the interaction of effector and substrate binding sites.

Electron microscopic observation of substance P-like immunoreactive nerve fibres and innervation in the anterior pituitary of macaques.

The anterior pituitary has recently been confirmed to be innervated by substance P-immunoreactive nerve fibres in the monkey and human. The present study investigated the morphology of the nerve fibres and their relationship with anterior pituitary gland cells in the macaque by means of immunoelectron microscopy. Many substance P-immunoreactive and unlabelled nerve fibres were distributed among the gland cells or formed nerve fibre bundles. These nerve fibres were unmyelinated and varicose, and contained a great many clear and/or large dense-cored vesicles. Substance P-immunoreactive cells were seen in the pars distalis of the anterior pituitary. Direct contacts between these nerve fibres and every cell type of the gland could be ascertained, including substance P-immunoreactive cells. Synaptoid contacts were seen among the substance P-immunoreactive varicosities and somatotropes, opiocortico-melanotropes and folliculostellate cells. Typical synapses were identified in somatotropes and opiocortico-melanotropes.

Renal cell carcinoma. Cytogenetic analysis of tumors and cell lines.

Successful cytogenetic analysis was performed on 27 samples from 25 patients with RCC, including 7 of 11 tumors studied and 20 cell lines. Clonal chromosomal abnormalities were detected in all 27 samples. The most frequently involved chromosomes were 7, 1, 3, 9, and the Y (20, 17, 17, 14, and 10 cases, respectively). Polysomy 7 or rearrangement of 7q was seen in 80% (20/25) of the patients, and loss or rearrangement of 3p was seen in 48% (12/25); of the latter, four patients had loss of the whole chromosome and 10 patients had deletions or translocations involving 3p, with breakpoints at either 3p11-14 or 3p21-23 (5/7 translocation breakpoints were at 3p21-23). Loss of the sex chromosomes was seen in 15 patients, including -Y in 10/22 males. Other clonal changes included structural abnormalities of chromosome 1 centromere and the long arm, breakpoints at or near the centromere of chromosome 9 (10 patients), polysomy 16, monosomy 17, polysomy 20, and monosomy 22. With the exception of chromosome 3p loss, which was primarily confined to the nonpapillary cases, no specific clonal abnormality was noted for any particular subtype of RCC. Trisomy or tetrasomy 7 and -Y were seen in all subtypes of renal cell carcinoma.

Effects of ligand homologation and ligand reactivity on the apparent kinetic specificity of papain.

Papain, a prototype cysteine proteinase, shows a pronounced kinetic preference for substrates and inhibitors based on the Ac-L-Phe-Gly-structural motif. Replacing the L-Phe at position P2 with D-Phe, or with a less hydrophobic residue such as Leu or Met, results in decreases of substrate or inhibitory activity of up to 400-fold. In this study we examined the effect of homologating the P1 glycine moiety to beta-alanine in the context of specific ester and amide substrates, peptidyl nitrile and -aldehyde transition state analog inhibitors, and peptidyl Michael acceptors as irreversible affinity labels. Papain discriminates extremely strongly (i.e., from 1000-fold to > or = 29,000-fold) against the 'homologs' based on beta-alanine at P1 compared to 'analogs' based on glycine at P1. However, with highly reactive ligands such as p-nitrophenyl esters, homolog/analog discrimination is greatly reduced (i.e., < or = 10-fold). These observations are interpreted in terms of (1) cooperativity between several non-covalent enzyme-ligand interactions and the covalent interaction of the ligand P1 moiety with Cys-25 of papain, (2) the decreased ability of homologs to utilize these cooperative interactions optimally because of their extended size, and (3) a decrease in the importance of the cooperative interactions as the intrinsic chemical reactivity of the ligand increases. Some implications of this analog vs. homolog discrimination for peptidyl disulfide and peptidyl chloromethane probes of protease specificity and mechanism are discussed.
