[Quantitative determination of dracorhodin in Daemonorops draco B1. and traditional Chinese medicines containing Daemonorops draco B1. by HPLC].

A reversed phase high performance liquid chromatographic mertod for the determination of Dracorhodin in Daemonorops draco B1. and Traditional Chinese Medicines containing Daemonorops draco B1. has been developed so as to set the quality standard for Sichuan Jinyao. The result showed that the components of Dracorhodin were separated by ODS column, with MeOH-0.05 mol/L NaH2PO4 (51:50) (pH = 3.10-3.15, adjusted with H2PO4) acting as the mobile phase. The other high retentive components were eluted by methanol; thus the analytical time was reduced. The method is simple and accurate.

[Perinatal monitoring in intrahepatic cholestasis of pregnancy].

The sensitivity of meconium stain in amniotic fluid for prediction of fetal well-being in intrahepatic cholestasis of pregnancy(ICP) was evaluated. The study consisted of an ICP group(n = 30), and a control group (n = 30) and the umbilical arterial pH value(< 7.2) was used as a standard. The positive and negative predicttive valus of meconium-stained amniotic fluid in ICP group were 80.0%; the positive and negative predictive values in control group were 60.0% and 92.0% respectively. There was no significant difference (P > 0.05) between the two groups in positive and negative predictive values. However, the positive and negative predictive values of the two groups were high, which indicated that meconium-stained amniotic fluid related to fetal hypoxia. Moreover, the incidence of meconium-stained amniotic fluid in ICP was higher than that in control (40.0%: 16.70%, P < 0.05). Therefore, we suggest that the amniotic fluid of patients with ICP should be observed very closely. When meconium-stained amniotic fluid is discovered, delivery by cesarean section is imminent.

[Teniposide, cyclophosphamide and dexamethasone combination treatment for stage III multiple myeloma: a report of 21 cases].

OBJECTIVE: To evaluate the efficacy and toxicites of combination chemotherapy of teniposide, cyclophosphamide, and dexamethasone (VCD) for stage III multiple myeloma (MM). METHODS: Twenty-one patients with stage III MM were treated with VCD regimen. Among 21 evaluable patients, 16 were in stage IIIA; 5 in stage IIIB. Teniposide was given 30 mg/m2 by intravenous(i.v.) infusion on days 1-3; cyclophosphamide 200 mg/m2 i.v. on day 1, 3, 5; dexamethasone 10-20 mg daily p.o. on days 1-7. The treatment was recycled every 28 days. RESULTS: The overall response rate was 76%(16/21), including partial response 38%(8/21); improvement 38%(8/21) and failure 24%(5/21). The survival time was 3-31 months; medium survival time (MST) 13 months; MST of MM patients in stage IIIA, IIIB were 11, 12 months, respectively. The medium duration of response was 5 months; the medium cycle for initial response was 2. Among 21 cases, leukopenia and thrombocytopenia were detected respectively in 10%, 54%, 31%, 5% and 67%, 17%, 13%, 3% according to WHO grade 0, I, II, III criteria. The gastrointestinal reaction was mild. No cardiovascular, renal and hepatic toxicites were observed. CONCLUSION: VCD regimen is an effective treatment for stage III multiple myeloma.

[Management and prognosis of patients with locally recurrent soft tissue sarcomas].

OBJECTIVE: The results and prognositic factors in treating locally recurrent soft tissue sarcoma (STS) of the extremities and trunk were identified. METHODS: The clinical, pathologic, and therapeutic characteristics of 96 STS patients were investigated using univariate and multivariate analyses. Seventy-six patients (79.2%) had high grade sarcoma and 20 patients (20.8%) had low grade sarcoma. Operative procedures were wide excision (surgical margin > or = 2 cm) in 66, local excision (surgical margin < 2 cm) in 6, and amputation in 24 patients. Adjuvant radiotherapy and chemotherapy was administered in 24 and 36 patients respectively. RESULTS: After reoperation of the recurrent tumor, the 5-year local control and overall survival rates were 70.1% and 59.1%, respectively. High grade, large size, and tumor in the upper extremities were associated with decreased overall survival. CONCLUSION: The authors emphasize that the initial management of primary STS should be adequate and suggest that safty margin of > or = 2 cm should be adhered to. In patients with local recurrence only, the overall survival may not be adversely affected if the recurrent lesion is properly treated. Investigations of more effective multimodality therapy needs to be done.

[Immunohistochemical study on the expression of c-erbB2 oncoprotein in breast tumors].

OBJECTIVE: To examine the relationship between oncoprotein c-erbB2 over-expression and prognosis of primary breast tumor patients, and find if it can be used as a prognostic indicator. METHODS: Eighty-five cases of paraffin-embedded specimens were used for immunohistochemical study. RESULTS: Over-expression of c-erbB2 was observed in none of the 7 benign breast tumors. Twenty-three out of 78 cases of breast cancer showed over-expression with a positive rate of 29.4%. Over-expressed c-erbB2 oncoprotein was not detected in 80% of patients with negative lymph node. In clinical stage III patients, 71% of them over-expressed c-erbB2. The expression of c-erbB2 and estrogen receptor(ER) was inversely correlated in 67% of breast cancer patients. Oncoprotein c-erbB2 over-expression had no correlation with lymph node status(P > 0.05), but was closely correlated to clinical staging(P < 0.05). CONCLUSION: Patients with over-expression of c-erbB2 oncoprotein showed poor prognosis. The c-erbB2 may be used for predicting the prognosis of breast cancer patients.

[Color television microscope with an automatic stage].

To mount a group of stepper motors on a television microscope automates the instrument in coarse focussing and equi-spaced moving a specimen. A microprocessor 8031 application system is used to control the stepper motors. For enhancing the properties and reliability of the system, we select RIF 530, a field effect transistor for the power amplifier of the motor. Combination PC with the instrument becomes basic hardware of a universal microscopic image processing system.

[Mutiple factors analysis of the effects of excimer laser photorefractive keratectomy].

PURPOSE: To observe the effect of excimer laser photorefractive keratectomy (PRK) correcting myopia and astigmatism, and to study the related factors. METHOD: PRK was performed with the aim to correct myopia errors from 1.00 to 16.00 diopters on 529 myopia eyes (306 patients). All eyes were divided into two groups: Group I: -1.00-6.00 D(294 eyes); group II: -6.25-16.00 D(235 eyes). All the patients were followed up for 3 to 12 months. RESULTS: At postoperative 12 months the rates of uncorrected visual acuity more than or equal to 10/20 or 20/20 were 97.5%, 77.5% in group I, and 80.3%, 32.8% in group II respectively; 92.5% of eyes were within +/- 1.00 diopter of the desired emmetropia in group I, but 42.6% in group II; The influential factors of changes of postoperative diopters was the severity of corneal haze, methods of removing corneal epithelium, intraocular pressure, the central corneal thickness. CONCLUSION: The excimer laser PRK is regarded to be a very predictable, safe, stable, and effective method to correct myopia up to -6.00 D. Prior to operation, the plan would be disigned according to diopters, ages, corneal thickness intraocular pressure. During operation, the corneal epithelium bad better to be removed by laser.

[Experimental study of artificial bone composite with high biological induction in repairing bone defect].

On the basis of the animal model of segmental ultra defects in rabbits, two different composite artificial bones (HA-bBMP-Co and HA-Co) were first used to repair the segmental bone defect respectively in 12 rabbits. The results of X-ray, gross, stereomicroscopic and histological examination were found that HA-bBMP-Co was much faster and much better than HA-Co, involving induction of new bone formation in quality and quantity. It is suggested that there is a widespread prospect in use of HA-bBMP-Co as a bone substitute material.

[Ultramicrostructure study of artificial bone composite with high biological induction in repairing bone defect].

The scanning electric microscopy (SEM). X-ray energy graph and Ca, P contents analysis were adopted in studying the mechanism and superiority of two different composite artificial bones (HA-bBMP-Co and HA-Co) to repair segmental bone defect in rabbits. The results indicated that HA-bBMP-Co had a double effects of bone induction and bone conduction. No difference of P content in two weeks after reconstructive surgery was found, but in other periods, there was an obvious difference in Ca and P content between HA-bBMP-Co group and HA-Co group (P < 0.05).

[Changes in morphology and calcium content of otoconia in rats after 120 d tail-suspension].

The morphology and calcium content of otoconia in rats after long term (120 d) tail-suspension were studied using scanning electron microscopy (SEM) and X-ray microanalysis, respectively. The results showed that after 120 d simulated weightlessness otoconia were round, irregularly shaped, or with fissures, and there were rough and fine granular or small globular substances on the surface. X-ray microanalysis showed that the calcium content in the otoconia of both utricule and saccule was significantly decreased in the 120 d tail-suspended rats than that in control (P < 0.01). These results suggest that a long term(120 d) simulation of the headward distribution of blood volume and hindlimb underloading effect induced by weightlessness may cause the morphological changes and lower calcium content of the otoconia. Finally, the possible mechanism and the physiologic meaning of these findings are discussed.

[Character of emergence for the seeds of Ephedra sinica].

The experimental results on the germinating of Ephedra sinica seeds and sowing show: the seeds of E. sinica are not sensitive to illumination, but very sensitive to humidity; the effects of soil, the depth of sowing and irrigation on emergence are very notable, but not remarkable to covering which can be used to prevent measure.

[Study on T lymphocyte recognition of tumor antigens on autologous CML cells].

OBJECTIVE: To investigate the existence of cytotoxic precursor cells in previously untreated or remissive chronic myelogenous leukemia(CML) patients,and identify their phenotypical and functional characteristics. METHODS: Bone marrow or peripheral blood mononuclear cells from CML patients were stimulated with autologous CML cells by using mixed lymphocyte tumor cell coculture. RESULTS: A kind of cytotoxic T lymphocytes could be generated from bone marrow or peripheral blood of CML patients. These T cells showed differential cytotoxicities against autologous and allogeneic CML cells and no activity to autologous and allogeneic normal bone marrow cells. They also exhibited no inhibitive effect on CFU-GM yields. LAK cells had no effect on autologous CML cells, but showed intensive cytotoxic activity to allogeneic CML cells. The T lymphocytes obtained were CD3+ CD56+ non-MHC restricted or CD3+ CD56- MHC restricted. HLA-DR and CD25 were expressed in a significantly larger proportion of T lymphocytes stimulated with autologous CML cells than those not stimulated. The T lymphocytes showed low proliferative response to autologous CML cell stimulation and no or least response to allogeneic CML cells, they showed also no response to EB virus-transformed autologous B cells(obtained in remission) pulsed with peptides corresponding to the BCR-ABL joining region. CONCLUSION: There is probably a common tumor antigen among CMLs, this leukemia-specific antigen can be recognized by T cells,and it shows no indication to be a p210 fusion sequence.

[Deviation of size-exclusion chromatographic behavior of tryptophan on TOYOPEARL HW-40S column].

A deviation of size-exclusion chromatographic behavior of amino acids on TOYOPEARL HW-40S column is reported. On the column 18 amino acids were not eluted in the same fraction in the total volume, but separated into 5 fractions with tryptophan as the last one far behind the total volume. Interaction between tryptophan and the matrices was investigated under different conditions in order to elucidate the chemical nature of the deviation. It was found that addition of trifluoroacetic acid and sodium chloride of high concentration in mobile phase, dramatically increased the retention time of trypophan, while addition of acetonitrile, sodium chloride of lower concentration and the hydrogen bond breaker urea reduced the interaction. It is concluded that adsorption of tryptophan on TOYOPEARL HW-40S is basically due to the hydrophobic nature with limited influence of hydrogen bond and ionic interaction. The former factor can be eliminated by keeping acentonitrile concentration over 50% (V/V), while the latter two can be reduced to the minimum by adding 0.17mol/L sodium chloride and 3.51mol/L urea. The results not only lead to better understanding of size-exclusion chromatography of this matrices, but also help to apply hydrophobic chromatography of this matrices to the isolation of amino acids and peptides.

[Study on chemical constituents of essential oil of Vitexd rotundifolia L. by gas chromatography/mass spectrometry].

The essential oil of the Vitexd rotundifolia L. was abstracted by steam distillation. The oil obtained was dried with anhydrous sodium sulfate. The chemical constituents were analyzed by GC/MS. More than 30 peaks were separated by GC, and 28 compounds were identified by using NBS mass spectral data, and the Wiley/NBS registry of mass spectral data. The structrure of delta3-carene was further verified by GC/FTIR. The identified constituents represent over 95% of the peak area on TIC of the essential oil. The major chemical constituents of them are delta3-carene, 1,8-cineole, sabinene, beta-phellandrene and isoterpinolene.

[Studies on determinaion of selenium in drink by GFAAS].

Pd(NO3)2-Triton X-100 was used as matrix modifier for determination of selenium in drink. In this way ashing temperature can be increased up to 1200 degrees C and absorbance can increase by 2.92 times. Interferences of Fe, P and other elements were reduced effectively by means of Zeemen effect background correction. Detection limit is 8.0ng/mL and linear range is from 10 to 120ng/ml, and r = 0.9931.

[The influence of KCl on SERS of fuchsin basic in silver colloids].

Surface enhanced Raman scattering (SERS) spectra of fuchsin basic (FB) in silver colloids were measured. The influence of KCl on the SERS of FB was studied systematically with different excitation wavelengths (476.5, 488.0, 514.5, 632.8nm). The dependence of the relative SERS intensity of the 1366cm(-1) and 242cm(-1) bands of FB on KCl mole value at different excitation wavelengths were given. The SERS spectra of FB at 514.5nm and 632.8nm wavelength excitations were very different, and the relative intensity of 242cm(-1) to 1366cm(-1) SERS spectra was changed.

[Study on determination of composition of complex by multiwavelength R factor method].

The pure spectrum of the complex is obtained by R factor method to eliminate the absorbance of equilibrium concentration of residual reagent. The reagent concentration is determined by multiple linear regression method. As initial concentrations of metal and reagent are changed, and the stable constant is not changed, complex composition and its constant are acquired by means of trial method, in which the input parameters, m and n, are selected. Then the K value is calculated and the minimum value of the total derivation in K is found. By this method, the composition of complex in systems such as Pb-XO, Al-CAS and Zn-PAR are all to be 1:2.
