Detailed data are presented concerning the relative amounts of Hb A and two alpha chain variants (Hb Duan with alpha 75 Asp----Ala, and Hb Westmead with alpha 122 His----Gln), and the occurrence of an alpha-thalassemia-2 heterozygosity in five members of a small Chinese family. The three children who have the three abnormalities inherited the alpha-Duan and alpha-thalassemia-2 heterozygosities from their father, and the alpha-Westmead heterozygosity from their mother. The base substitution which leads to the synthesis of the alpha-Duan chain occurred at codon 75 of the alpha 1 globin gene of the chromosome which also carried the alpha-thalassemia-2 deletion; the concentration of alpha-Duan (37% of total alpha) is similar to that observed for other alpha chain variants, linked to an alpha-thalassemia-2 condition.
DNA amplification combined with hybridization with 32P-labeled synthetic oligonucleotide probes has been used to identify base substitutions in the 5' promoter region of the A gamma globin gene in members of eleven families from China, Sardinia, Canada, and the United States who had a heterozygosity for the A gamma-beta+-hereditary persistence of fetal hemoglobin (HPFH), and in members of six black families with a possible G gamma-beta+-HPFH heterozygosity. All three known A gamma types were observed, ie, the British type (-198, T----C), the Chinese type (-196, C----T), and the Green type (-117, G----A); the latter has been found in a black family. Of the six families with G gamma-beta+-HPFH, three had C----G at -202 and none T----C -175. Conditions for hybridization of amplified DNA with the specific probes are provided and the usefulness of the technique is discussed. The increase in numbers of A gamma(G gamma)-beta+-HPFH heterozygotes with specific base substitutions greatly enhances the probability of a direct correlation between these substitutions and the increase in the production of a specific gamma chain.

Sympathetic sprouting in the hippocampus following septal denervation is thought to involve nerve growth factor (NGF). This sprouting response is dramatically reduced in aged rats, but immunological assays reveal no age-related decline in hippocampal NGF levels. In the present study, both a bioassay and an immunoassay were used to examine the effect of a medial septal lesion on hippocampal NGF levels in young adult (2-5 months) and aged (24 months) Fischer 344 rats. No significant differences were detected between normal young and aged rats, in agreement with earlier results. Following medial septal lesions, however, only young rats demonstrated significant increases in hippocampal NGF-like activity. These results support the hypothesis that the age-related deficit in sympathetic sprouting results from an attenuated neurotrophic response to hippocampal denervation.
A protocol was created for performing geographically randomized snail surveys for schistosomiasis research using the global positioning system (GPS). This protocol differs from traditional surveys in its ability to accurately map and measure the spatial distribution of snail habitat. The protocol was used to map irrigation ditches, the primary habitat for Oncomelania hupensis, in two residence areas in Sichuan Province, China. From the 7,450 meters of mapped ditches, snail surveys were performed at 203 random sites along the ditch network. Of these, 116 (57.1%) sites had snails. The total number of living snails captured was 2,014, resulting in an average snail density of 0.27 snails per linear meter of potential habitat.
The present study is part of a program to optimize the cryopreservation ligament and tendon allografts for the biological remodeling that occurs following transplantation. The osmotic behavior of fibroblasts from the medial collateral ligament (MCL) and the patellar tendon (PT) of new zealand white rabbits was measured to obtain the hydraulic conductivity (and its temperature dependence) as well as the osmotically inactive volume. MCL fibroblasts were found to have an isotonic cell volume of 3800 (m(3), an osmotically inactive volume of 1300 (microm(3), and a hydraulic conductivity of 0.56 (microm/min/atm at 20 degrees C with an activation energy of 10.9 kcal/mol. PT fibroblasts were found to have an isotonic cell volume of 6300 (microm(3), an osmotically inactive volume of 2000 (microm(3), and a hydraulic conductivity of 0.71 (m/min/atm at 20 degrees C with an activation energy of 10.1 kcal/mol.
