OBJECTIVE: To evaluate the relationship of p53 alteration and myc family gene overexpression with the clinical characteristics of lung cancer. METHODS: A series of 59 resected primary lung cancer specimens was analyzed for p53 gene by DNA/PCR sequencing and immunohistochemistry technique, and for myc family genes by RT-PCR methods. RESULTS: Thirty-seven of 57 tumors were found to have p53 mutations or/and p53 protein accumulation. The presence of p53 alteration was not related to tumor size, lymph node metastasis, stage and relapse. Forty-seven cases were analyzed for myc family genes. The results showed that there was a positive correlation between unregulative expression of myc genes and the above mentioned clinical parameters. Our finding also showed that 19 of 30 cases (63%) with p53 alteration had myc gene overexpression which occurred in 63% and 76% cases with stage III and relapse, respectively, which was higher than 27% and 22% with p53 alteration but no myc gene overexpression and 50% and 71% with p53 negative but myc gene overexpression. CONCLUSIONS: p53 alteration is a vital genetic event in the earlier stage of lung carcinogenesis, but not a prognostic marker. myc family genes overexpression may be regarded as one of the independent prognostic determinants in lung cancer. The cooperation between p53 alteration and myc gene overexpression may occur during progression of lung cancer, but prognostic determinant is myc gene overexpression.
OBJECTIVE: To compare the efficacy of transcranial Doppler ultrasound (TCD) and digital subtraction angiography (DSA) in identifying the clinical and hemodynamic relationship between intra- and extra-cranial circulation and subclavian steal syndrome. METHODS: Sixteen patients were collected. The intra- and extra-cranial arteries were insonated with Doppler ultrasound. DSA and brain CT were performed in all patients. The relationship between the grade of arterial stenosis showing in DSA and the flow direction showing in TCD was studied. RESULTS: TCD showed reversal flow of the vertebrobasilar artery in 13 patients, normal flow in 3, which was influenced obviously by fist-clenching. In the hyperemia test, the flow velocity and the direction changed significantly in all patients. The steal phenomenon of the middle cerebral artery (MCA) was coincident with that of the arterial cerebral artery (ACA) in 2 cases. Stenosis of MCA or ACA was found in 5 cases (31%). Nine patients were treated with arterial bypass or angioplasty. TCD was performed during the follow-up. CONCLUSIONS: Reversal flow of the vertebral artery may occur in rest or only in exercise, depending on the grade of subclavian arterial stenosis. Hyperemia test is the most sensitive index. It is necessary to monitor blood flow in the MCA and basilar artery in hyperemia test. The coexistence of intra- and extra-cranial arterial stenosis is common.
OBJECTIVE: To measure serum 7S collagen (7S-C) and type VI collagen (VI-C) levels by radioimmunoassay (RIA) in Chinese patients with various liver disorders and in CCl4-treated SD rats, and to investigate the significance of the elevated levels of serum 7S-C and VI-C. METHODS: Serum 7S-C and VI-C levels were measured in 40 healthy control subjects, 168 patients with various liver disorders and non-hepatic diseases, and 52 CCl4-treated SD rats by using RIA which was developed in our hospital. RESULTS: Serum 7S-C and VI-C were significantly elevated in patients with chronic active hepatitis (CAH), liver cirrhosis (LC), hepatic cellular carcinoma (HCC) (P < 0.01 respectively), chronic persistent hepatitis (CPH), and some with non-hepatic diseases (P < 0.05). Serum 7S-C, serum laminin and hyaluronic acid were well correlated. Serum 7S-C and VI-C were not closely correlated. Both collagens were correlated with serum albumin/globulin ratio, aminotransferase and total bilirubin, not with alkaline phosphatase. In CCl4-treated SD rats, serum 7S collagen and type VI collagen levels were correlated with the degree of hepatic fibrosis. CONCLUSIONS: Serum 7S collagen and type VI collagen are useful markers for diagnosing liver fibrosis. And the combined measurement of IV-C, VI-C and other markers of connective tissue metabolism or biochemical data seems to provide additional information to predict progressive hepatic fibrosis.
OBJECTIVE: To assess the value of endometrial transvaginal sonography in diagnosis of patients with postmenopausal bleeding. METHODS: High resolution transvaginal ultrasonographic examination was performed in 192 patients with postmenopausal bleeding before fractional uterine curettage. The results of endometrial sonogram were compared with endometrial histological findings. RESULTS: Seventy-seven cases had endometrial thickness < or = 4mm. 74 of them (96.1%) showed physiological changes by pathological examination. Only 3 were endometrial cancer. In contrast, in 115 patients who had endometrial thickness > or = 5mm, 59 (51.3%) had endometrial cancer. CONCLUSION: Transvaginal ultrasonography may be used as a screening procedure in patients with postmenopausal bleeding. Diagnostic uterine curettage may be avoided in patients who had endometrial thickness < or = 4mm.

Opioid actions are initiated at membrane receptors which couple to cellular effectors through G protein-mediated pathways. In the central nervous system opioids reduce neuronal activity through the inhibition of voltage-dependent Ca2+ channels, the activation of K+ channels and the inhibition of adenylyl cyclase. A significant clinical limitation to opioid therapy is the development of tolerance, a biological event that has been linked to agonist effects at the receptor level. Molecular studies on the consequences of opioid receptor regulation will provide a better understanding of the cellular mechanisms involved in the agonist-mediated events in tolerance development.
OBJECTIVE: To verify from the aspects of immune senescence and free radical metabolism that "removing blood stasis as well as tonifying kidney is an ideal way to delay aging" and further investigate the anti-aging mechanism of this therapy. METHODS: In the first period, the old mice were separately fed with the herbal decoction of the recipe of tonifying kidney, or of removing blood stasis, or of tonifying kidney and removing blood stasis for four weeks, and then IL-2 production of splenic lymphocytes, IL-1 production of peritoneal macrophages, SOD, CAT and GSH-Px activity in erythrocytes, and LPO contents in liver and brain were compared between the young control group and the old control group, or the old control group and each of the old medication groups. In the second period, the adult mice inhaled ozone or the splenic lymphocytes from adult mice were cultured in the medium containing peroxide hydrogen. The effects of free radicals on immune system and the reversion of the herbal decoction of the recipe of removing blood stasis as well as tonifying kidney were observed. RESULTS: The activities of IL-1, IL-2, SOD, CAT and GSH-PX in old mice were significantly lower than those in young mice, and LPO contents were higher. The three herbal decoctions all have reversing effects on each of these parameters except that the decoction of removing blood stasis has inhibitory effects on IL-1 and IL-2 production. The effect of the decoction of removing blood stasis as well as tonifying kidney was the best among the three decoctions, and it can also reverse the impaired production of IL-2 by ozone in vivo and peroxide hydrogen in vitro. CONCLUSIONS: The therapeutic method of removing blood stasis as well as tonifying kidney is an ideal way to delay aging, and its effects to improve immune senescence is correlated with the decrease of free radical damages.
OBJECTIVE: To investigate the expression of transforming growth factor (TGF)-alpha and myc oncoprotein on human follicular development and atresia in vitro. METHODS: Ovarian tissues were obtained from 36 women with regular menstrual cycles who underwent abdominal hysterectomy for a variety of gynecological conditions. Expression of TGF-alpha and myc protein in the ovarian tissues was examined by immunohistochemical technique. RESULTS: In primordial follicale only the oocyte showed intense immunostaining for both TGF-alpha and myc oncoprotein. Immunostaining for TGF-alpha in granulosa cells and theca interna cells became apparent in the follicles of preantral stage. With the increase in the size of follicles the intensity of immunostaining in the oocyte decreased, whereas the staining intensity of the granulosa and theca cells increased, and persisted in the corpus luteum and further intensifying during the midluteal phase. myc protein expression in granulosa cells was apparent only in the preantral follicle stage. In the regressing corpus luteum, expression of both TGF-alpha and myc oncoprotein was restricted only in the peripheral theca lutein cells adjacent to the central core of scar issue. In atretic follicles theca interna cells exhibited prominent immunostaining for TGF-alpha and myc protein. CONCLUSION: TGF-alpha and myc oncoprotein combination play a role as intraovarian regulators through autocrine and paracrine mechanism and may participate in remodelling the initial growth of the oocyte, follicular growth, differentiate and apoptosis.
OBJECTIVE: To study the effects of oral low-dose magnesium gluconate in prevention of pregnancy induced hypertension (PIH) and its mechanism. METHODS: A prospective randomized double-blind study was carried out in 51 pregnant women as treatment group (including 22 cases as treatment group 1 and 29 cases as treatment group 2) and 51 pregnant women as controls (including 28 cases as controls group 1 and 23 cases as control group 2). Low-dose magnesium gluconate (3 g/day) or placebo was given from the 28th week of gestation to delivery consecutively. RESULTS: 4% of the pregnant women developed PIH after magnesium gluconate treatment, which was substantially lower than that in the control group (16%) (P < 0.05). In the treatment group 2, women showed higher concentration of 6-keto-prostaglandin F1 alpha (PGF1a) and 6-keto-/thromboxane B2(TXB2) (P/T) ratio than that of the control group 2. Moreover, TXB2 level was lower than that in the control group 2. In the treatment group 1 women showed higher ratio of P/T than that of the control group 1. There were no significant differences of serum magnesium concentration among all groups. CONCLUSION: Low-dose magnesium gluconate may efficiently prevent PIH in high risk women. The mechanism of action of magnesium gluconate probably involves to keep the balance of PGI2 and TXA2, but not associates with serum magnesium level.

The skull defects with neogenetic bone were repaired with a method in which the neogenetic bone, the cranial periosteum were sutured and fixed to a Ti-net. Totally 60 children were treated. This method has following advantages: 1. Averting trephining of skull to reduce blood loss and the possibility of brain injury: 2. The neogenetic bone is well protected so that it would grow more normally; 3. The method is easy and the operation time is shortened; 4. There is little reaction and postoperative results are satisfactory. The paper describes the principle and technic of the method.
Capture of rats by digging their cavities, identification of rat strains, sex and age, examination of their skin cut or damage, and detection of antigen and antibody of epidemic hemorrhagic fever virus (EHFV) were conducted in epidemic areas of epidemic hemorrhagic fever (EHF) in Jinaghan Plain, Hubei Province, to study prevalence of EHFV infection in rats. Results showed that Apodemus agarius and Rattus norvegius accounted for 69.7% and 23.6% of the total rats captured, respectively, their infection rates of EHFV were 21.8% and 30.9%, respectively. Infection rate of EHFV was significantly higher in adult rats than in sub-adult and young ones, with prevalence of 40.8%, 17.0% and 12.4%, respectively. Infection rate of EHFV was 45.9% in rats with skin cut, significantly higher than in those without it (13.9%). There was an obvious litter-clustering phenomenon in infected rats, but their litters distributed in space randomly. Serotyping of infected rats showed that Apodemus agarius mainly was in a wild rat pattern, Rattus norvegius in a domestic rat pattern, and a small number of rats were in undefined patterns. It indicated that Jinaghan Plain was a mixed epidemic area of EHF with domestic and wild rat patterns and with Apodemus agarius and Rattus norvegius as main reservoir host, and age, skin cut, and close contact with them were important factors contributing to transmit it.
In order to investigate the mechanism of effects of lead on learning and memory, an experiment of glutamate uptake in the hippocampal synaptosome of rats was carried out. Results showed that (1) uptake of 3H-DL-glutamate increased by 70.38% to 193.24% in the synaptosome by exposure to 3.1 to 50.0 mumol/L of lead for 15 minutes in vitro, with a dose-dependent relationship; and (2) uptake of 3H-DL-glutamate increased by 65.73% to 124.93% after exposuring to 10 to 1,600 micrograms/ml of lead in drinking water for 30 days, also in a dose-dependent manner with lead content in the hippocampus. It suggests that lead ion can accelerate the clearance of glutamate transmitter and cause inhibitive effects.
In this paper, the tri-dimensional computer reconstruction and animated display from serial sections of the genuine crude drug Radix Aconitii have been addressed. Accordingly, some tridimensional image technique and information for the teaching and identification of pharmacognosy have also been offered.
The classification of pathogenic yeast, which is one of the most common reasons of opportunistic infection in human body, will be useful for the epidemiological survey and clinical works. The genotypic polymorphism of intergenus, interspecies and intraspecies in 48 isolates, including Candida, Cryptococcus, Torulopsis, Trichosporon, Saccaromyces had been administered carefully with patterns amplified by RAPD (Randomly amplified polymorphic DNA) in 53 primers. The pologenetic affinities had been evaluated by similarity coefficients obtained from these profiles. The results indicated that there are significant difference among intergenus, interspecies and intraspecies in Candida and related yeast. The similarity coefficients among Candida and Crytococcus, Trichosporon were maintained about 80%. The similarity coefficients among interspecies of Candida ranged from 82%-87%, and ones of intraspecies of different Candida species were more than 90%. The genotypic typing of species except C. guilliermondii seems to be related to morphological classification.
Color Doppler flow imaging (CDFI) was performed in 35 patients with portal hypertension and esophageal varix. Thirty subjects were served as control. After esophageal variceal ligation, CDFI observed that the portal-superior vena cava collateral veins were partialy blocked. The esophageal varices disappeared. The esophageal wall thinned. Diameter of left gastric vein enlarged and blood flow velocity decreased. Azygos diameter was reduced and blood flow velocity was decreased. Although diameter of portal vein was not changed, the blood flow velocity slightly increased. The results suggest that ligation treatment can have the tendency to increase blood flow of liver and stomach, which might aggravate gastric mucosal lesion.
The symptoms of prostatic and periprostatic cysts are nonspecific and digital rectal examination has no definite findings detected. The conventional radiological plain film cannot define the relationship of a cyst to the surrounding structures. The use of MRI as a means in detection is too expensive. To solve these problems, we detected 118 cysts in 73 patients by transrectal ultrasound (TRUS) and discussed the TRUS imaging findings of various prostate and periprostate cysts with illustrations from our clinical practice. We suggest that TRUS can be used routinely, because its distinct advantage of high resolution for minute nodular or cystic lesion detection with high quality image demonstrates the relationship of surrounding structures, and provides an outstanding novel diagnostic modality.
