Concentrations of antisecretory factor in eggs and in chicken blood plasma.

1. In the present study, the concentration of antisecretory factor (ASF), a lectin with hormone-like properties, and with the capacity to regulate water and electrolyte transport in the small intestine, was higher in hen's egg yolk than in egg white (1.20 v. 0.46 units/ml). 2. The blood plasma concentrations of ASF activity were higher in d-old chicks (1.03 units/ml) than in 7-d-old (0.42 units/ml) or 21-d-old birds (0.18 units/ml); the contents were found to be high again in 35-d-old chickens, but to have decreased by 60 to 90% after transport to slaughter 1 day later. 3. The amount of ASF activity was lower in two groups of chickens manifesting patent loose droppings at slaughter (0.15 units/ml in one group, and 0.08 units/ml in the other), than in two groups with normal faecal consistency (0.65 units/ml in one group, and 0.72 units/ml in the other). 4. The results demonstrate the presence of ASF in eggs, and a variation of ASF activity in chickens blood plasma in relation to age, stress and faecal consistency. The interpretation of these data suggests a regulatory influence of ASF on chicken intestinal transport of water.

Immunopathological features of rat Staphylococcus aureus arthritis.

Staphylococcus aureus is the most common bacterial species found in nongonococcal bacterial arthritis in humans. We present the first description, to our knowledge, of an outbreak of spontaneous staphylococcal arthritis in a rat colony. In a group of 10 rats, 9 displayed arthritis. Clinically, the most obvious findings were arthritis of one or both hindpaws and malaise. Bacteriophage typing showed the common phage type 85 in isolates recovered from the joints, blood, and bedding of rats and from the nose and cheeks of one person from the staff of the animal facility. The S. aureus strain proved to produce staphylococcal enterotoxin A and exhibited strong binding to collagen types I and II and bone sialoprotein, which are potentially important virulence factors. When the recovered S. aureus strain was injected intravenously into healthy rats, severe septic arthritis was induced in almost all of the animals. The arthritic lesions were characterized by infiltration of phagocytic cells and T lymphocytes into the synovium. Many of the synovial cells strongly expressed major histocompatibility complex class II molecules. Increased levels of interleukin 6 in serum as well as a prominent polyclonal B-cell activation were noted throughout the disease course. Pretreatment of S. aureus-injected rats in vivo with an antibody to the alpha beta T-cell receptor significantly decreased the severity of the arthritis. Our results indicate that alpha beta + T lymphocytes contribute to an erosive and persistent course of S. aureus arthritis.

Evans blue permeation of intestinal mucosa in the rat.

The azo dye Evans blue (EB; molecular weight, 960.83) is widely used as an indicator of increased capillary permeability. In the present study, however, rat gut absorption of EB was investigated after dye instillation in either the small or large intestine. During a brief period of ether anaesthesia, EB was injected either into jejunal loops with a challenge period of 30 or 60 min or into a proximal and a distal colon loop with a challenge period of 30, 60, or 120 min. After the rats had been killed the intestinal specimens were washed with 6 mM acetylcysteine dissolved in phosphate-buffered saline, which efficiently cleared the tissues of mucus, and thus of EB trapped in mucus. Only EB absorbed by the gut wall remained to be estimated, and this absorption was found to be both dose- and time-dependent in the jejunum and the colon. After instillation in the colon, but not in jejunum, EB could be detected in the blood. EB absorption from the jejunum remained unaffected by the addition of either ouabain (1 mM) or lidocaine (0.38 mM). Either of these compounds inhibited EB uptake in the proximal part of the colon, while enhancing it in the distal part. Fluorescence microscopy showed penetration into the intestinal wall to be a prerequisite for EB to become fluorescent, and EB fluorescence increased with time. It is proposed that EB is transported over the mucosa by the paracellular route and that the amount of absorbed EB reflects epithelial permeability differently in different parts of the gastrointestinal tract.(ABSTRACT TRUNCATED AT 250 WORDS)

Antisecretory factor enhances in vivo internalization of cholera toxin and of horseradish peroxidase into rat enterocytes.

The in vivo effect of antisecretory factor (ASF, derived from pig plasma) on the ability of cholera toxin (CT) and of horseradish peroxidase (HRP) to bind to and penetrate into epithelial cells of the rat small intestine was evaluated in the absence of anesthetics. The potency of intravenously administrated ASF was demonstrated by some 70% inhibition of CT-induced secretion in ligated small intestinal loops. Using immunohistochemical methods for visualization, we found ASF to enhance internalization of both CT and HRP after 30 to 60 min of challenge, without interfering with the initial binding to the enterocyte brush border region. The internalization process started in the upper 2/3 of the villus region. After 5 h, no CT or HRP could be seen bound to the enterocytes. The results suggest that ASF might enhance small intestinal absorption.

The effect of taprostene in patients with acute myocardial infarction treated with thrombolytic therapy: results of the START study. Saruplase Taprostene Acute Reocclusion Trial.

Taprostene is a prostacyclin analogue that inhibits platelet aggregation and thus might be a useful adjuvant to thrombolytic agents in acute myocardial infarction. In a placebo-controlled dose rising study, taprostene or placebo was intravenously infused in 80 patients treated with the thrombolytic agent saruplase (rscu-PA) for acute myocardial infarction. Three doses of taprostene were used: 6.25; 12.5; or 25.0 ng.kg-1 x min-1. Taprostene or placebo was infused for 48 h, followed by a 24 h tapering period. All 80 patients had short symptom-to-treatment delay and marked ST segment elevation. Patency at 90 min was documented in 58/78 patients (two patients had no angiography). Success rate varied from 67-82% in the four treatment arms (P = 0.33). Patency after rescue PTCA was seen in 10 out of 13 patients. Of the 58 patients having a patent artery at 90 min, none of the 43 taprostene patients and one of the 15 placebo patients had a re-occluded artery at the second angiography at 32-48 h (5/58 patients had no recatheterization). Conversely, of nine patients who had successful rescue PTCA, three of four placebo patients had a re-occluded artery at the second angiography compared to one of five taprostene patients (one placebo patient had no recatheterization) (P = 0.33). Safety evaluation revealed no major difference between the placebo plus saruplase and the taprostene plus saruplase groups. Taprostene was well tolerated up to 25 ng.kg-1 x min-1. Although taprostene did not affect 90 min patency, there was a trend to better maintenance of patency after rescue PTCA.

Plasma level of antisecretory factor (ASF) and its relation to post-weaning diarrhoea in piglets.

Antisecretory factor (ASF) is a regulatory peptide which counteracts diarrhoea in the pig; ASF is rapidly absorbed from the pig intestine, and significantly reduces the incidence of neonatal diarrhoea in the suckling offspring. ASF is synthesized in the central nervous system, and released to the blood stream via the pituitary gland. In two different experiments (n = 8 and n = 4), the blood concentration of ASF was followed in 5-weeks old piglets from day 7 before weaning up to day 12 days after weaning. In both experiments ASF concentrations were significantly (p < 0.01) lower on day three post-weaning, than either before weaning or on days 7 and 12 post-weaning. In another experiment, where plasma ASF activity was determined in relation to clinical signs of diarrhoea seven days post-weaning, it was found to be 0.87 +/- 0.08 units/ml (mean +/- SEM) in healthy weaners (n = 15), but only 0.22 +/- 0.05 units/ml in piglets suffering from diarrhoea (n = 15), the difference being significant. The faecal flora both of healthy weaners and of their matched controls suffering from diarrhoea were subjected to bacteriological examination before and after weaning, and found to be similar in both groups, namely a mixture of aerobic and anaerobic Gram negative rods, Campylobacter jejuni, Staphylococcus aureus/epidermidis, and Enterococcus faecalis. No particular pathogen was predominant in any of the diseased animals.

Feed-induced lectins in piglets. Feed-induced lectins and their effect on post-weaning diarrhoea, daily weight gain and mortality.

The effect on various biological variables of a special weaner feed (FD) with the capacity to induce endogenous synthesis of feed induced lectins (FIL), and a commonly used commercial diet (CD) was evaluated in 891 piglets in four different experiments. The FD diet was found to induce a high activity of FIL (FIL = 0.94 +/- 0.03 units/ml. blood), as compared to CD (FIL < 0.20 units/ml. blood). The FIL values did not decrease after weaning. The incidence of diarrhoea in piglets fed FD 0-14 days post-weaning was 12 +/- 5 per cent, as compared to 41 +/- 9 per cent in the CD group, the difference being significant. Daily weight gain 0-35 days after weaning was 67 +/- 15 grams greater in the FD group than among those fed CD. The consumption of FD was double that of CD in the period before weaning (at 5 weeks), and some 40 per cent more in the postweaning period up to 9 weeks of age.

Antisecretory activity in a lectin fraction of plasma from patients with acute diarrhea.

The present study describes the first attempt to detect antisecretory activity in a lectin fraction of plasma from patients with acute diarrhea. The plasma antisecretory protein (ASP) was purified by affinity chromatography in agarose, and its antisecretory activity tested in rats subjected to intestinal challenge with cholera toxin. During the first 24 h of the diarrheal episode, antisecretory activity in patients (median 0, range 0-25%) was lower than that seen in the asymptomatic group (median 10, range 0-30%); 3 days later, when diarrhea ceased in most of the patients, the ASP activity increased significantly (median 30, range 0-75%). However, 5 days later the activity decreased again (median 0, range 0-55%). No differences in ASP levels were found between cases associated with an enteropathogen and those where no pathogen was identified. These findings reveal an inverse relationship between the increase in ASP and the patient's intestinal secretion; suggesting that ASP plays a role in the compensatory mechanisms that occur in diarrhea in humans.

[Seroepidemiological studies of the detection of leptospires of the sejroe group in cattle in middle Thuringia].

From 1983 to 1989 14,361 head of cattle from the middle Thuringia region were investigated by means of microagglutination test for the presence of Leptospira serovars hardjo, saxkoebing and sejroe. 4,484 samples from cattle with abortion (1983-1985) and a random sample of 5,284 cattle sera (1985) were investigated giving priority to L. hardjo. Furthermore 3,293 samples from cattle with abortion (1986/87) and a random samples test of 1,300 cattle sera (1989) were tested giving priority to L. saxkoebing. 2.5 percent of the cows having aborted and 10.3 percent of cattle tested out of the whole livestock showed antibodies against L. hardjo. The test for presence of L. saxkoebing demonstrated in cattle with abortion a seroprevalence of 14 percent and in the random samples tests a portion of reagents of 11.3 percent. The degree of infection differed regionally, the highest degree was found in livestocks of the northern part of the Thuringia forest. The prevalence of L. saxkoebing in cattle with abortion from large dairy farms was 18.6 percent, which is higher than the average in the region examined. Antibodies against hardjo and sejroe were interpreted as caused by cross reactivity.

[Seroepidemiological studies on the detection of Q fever in sheep in middle Thuringia].

In a random samples test altogether 4337 sheep of varying ages from several herds of different sizes in the middle region of Thuringia were investigated with complement fixing test for the existence of Q fever. In 47.1 per cent of the tested herds and in 5 from 8 of the included districts Q fever reagents were provable. The serological detection quota of all tested sheep amounts to 1.11 per cent, but the percentage of serological reagents in mother-sheep was 1.36 per cent, in the female young sheep 1.04 per cent and in the lambs 0.74 per cent. The investigations of rams and sheep for slaughter were negative. Serological testing in 7 sheep showed a Q fever antibody persistence about 6 until 10 months. Increased animal concentration and adverse conditions led to an increase of the Q fever seroprevalence in the herds.

[Q fever antibody titer--follow-up study in cattle with special reference to pregnancy].

In 290 Q fever positive cattle from three 2000 head dairy farms in the former district of Erfurt (Thüringen) the course of titers was examined serologically over several months by means of the complement fixation test (CFT). In 47.2% of the cows serologically observed for 2 up to 28 months complement fixing antibodies against Coxiella burnetii could be demonstrated til the end of the investigation period. Repeated tests during pregnancy showed increase of antibody titers in the first 4 months and after a short decrease again from the 5. til the 7. month. By observing the antibody titers during several pregnancies each time a new increase comparable to a booster immunisation could be found. This may explain the persistence of Q fever antibodies in cows for several years. The results of this investigation suggest that from a high antibody titer it can not be concluded an abortion in a positive cow being caused by a Coxiella burnetii infection.

Induction of anti-secretory factor in mice by the nonintestinal parasite Schistosoma mansoni.

Anti-secretory factor (ASF) was determined by the rat intestinal loop model in the pituitary gland and in the blood at various intervals after experimental infection of mice with Schistosoma mansoni. ASF could be detected in the pituitary gland by 6 hr after percutaneous infection with cercariae and in the blood at 24 hr after infection. A peak level was reached 3 days after infection and, after a decline, 2 more peaks were noted between 4 and 8 wk after infection. Characterization of ASF induced by S. mansoni by means of isoelectric focusing revealed 2 distinct peaks at approximately pH 4 and pH 5, respectively. Upon affinity chromatography on agarose, the activity dissociated in a gradient of methyl-alpha-D-glucoside at 1.0 M but not at 0.1 or 0.3 M concentrations.

[Listeria abortions in cattle--bacteriology, serology and epizootiology].

Listeria abortions were established in foetuses of cattle in 1.2% of the cases investigated in the Erfurt region and 1.8% in the Cottbus region from 1984 to 1989. The isolated strains have been found biochemically to be L. monocytogenes. The serological discrimination revealed a majority of serovar O I, II. Typing by phages of a part of isolated strains showed a clear predominance of code number 000 124 by the 1/2 a isolate (corresponds to O I, II). This type of phages seems to have a epizootiological importance. Listeria abortions were found in (a lot of 63) flocks in the Erfurt region. Nevertheless there were almost no accumulations of listeria abortions and listeria encephalitis at the same time in the analysed flocks.

[Chlamydia as the cause of abortions in horses].

Between 1982 and 1989 59 equine fetuses were investigated for chlamydiae using animal experiments and embryonated eggs. Chlamydiae were isolated from 16 fetuses (27.1%) originating from 8 studs. The macroscopical lesions of the fetal organs were characterized by extensive haemorrhages. The histological picture shows severe lesions of the blood vessels of different organs. In 6 studs in which chlamydiae had been isolated, blood sera of clinically healthy and pregnant mares were investigated for antibodies during 1989 and 1990. Antibody titres between 1:10++ and 1:40 were detected by using complement fixation test in sera of mares of 4 studs.

[Intolerance reactions to ionic and nonionic contrast media in cardiac diagnosis. A randomized double-blind study].

Reactions to an ionic and a nonionic contrast medium were compared in a randomized double-blind trial involving 1153 patients (897 men, 256 women; mean age 56 [18-84] years) who underwent left-heart catheterization with coronary angiography. 584 patients (group 1) received the ionic contrast medium meglumine diatrizoate (Urografin 76%), while 569 (group 2) received the nonionic iopromide (Ultravist -370). Reactions of skin and mucosae, heart and circulation, lungs and respiratory tract, as well as of the central nervous system were noted. There was no significant difference between the two groups with respect to age, weight, height, allergic predisposition, left-ventricular ejection fraction, or serum electrolyte concentrations. Mild reactions, not requiring any treatment, occurred in 108 of the 1153 patients (9.4%), 85 (14.6%) in group 1 and 23 (4%) in group 2 (P less than 0.001). Severe reaction requiring treatment occurred in eight patients of group 1 (1.4%) and 14 of group 2 (2.5%)--difference not significant. Ventricular fibrillation was more frequent among group 2 patients (four compared with one in group 1), while two of three asystoles requiring treatment occurred in group 1 patients. The results show that, while mild reactions not requiring treatment were clearly less frequent after nonionic contrast medium injection, the risk of severe reactions was not reduced by their use.
