Murine heavy chain disease.

Using cultured mouse myeloma cells, it has been possible to derive cells which are now synthesizing products similar to human heavy chain disease proteins. An initial mutant was isolated which snythesized a heavy chain with an internal deletion and a normal light chain. In a subsequent step, a variant was identified in which the synthesis of the heavy chain with the deletion persisted in the absence of light chain synthesis. These experiments also demonstrate that in the MPC-11-derived cultured cell line, the continued synthesis of heavy chain does not require the synthesis of light chain.

Estimation of antibodies specific for dextran.

Methods are described for the isolation and characterization of picogram quantities of anti-dextran antibodies. 14C-dextrans produced by using the dextransucrases of Leuconostoc mesenteroides strains B1355 and B512 were used in a radioimmunoassay. The specificity of this assay was verified by using cell cytoplasmic lysates from mouse plasmacytomas, J558 (anti-alpha 1 leads to 3 dextran) and W3129 (anti-alpha 1 leads to 6 dextran). Dextran produced by strain B1355 and insolubilized with epichlorohydrin was used as an immunoabsorbent.

Prognosis of new and worsening angina pectoris.

The natural history of new and worsening angina pectoris was studied in 251 men aged under 70 years. Most were ambulant and all were referred by selected general practitioners to a special hospital clinic over two and a half years. Heart attacks developed in 39 patients, nine of whom died. Seventy-two per cent of the attacks occurred within six weeks of the onset or worsening of angina. Of the 212 patients who did not suffer myocardial infarction and who were clinically reviewed six months after their first attendance 66 had been pain free for the previous three months and 14 had experienced only infrequent attacks of angina. Of the 128 men aged under 65 years who were previously in employment 81% had returned to full-time work six months after their first attendance. A discriminant function analysis using many variables was made to develop a predictive index that would allow patients with new or worsening angina who were likely to develop serious cardiac complications to be identified. This did not prove possible, and the only predictive factor of significance was an increased cardiothoracic ratio. The syndrome of new and worsening angina has a low risk of early death, and many patients are symptom free six months later. In general, emergency coronary arteriography and surgery is not indicated.

Heavy chain-producing variants of a mouse myeloma cell line.

Eleven variants producing only heavy (H) chains have been isolated from the P3 cell line after mutagenesis with either ICR-191 or nitrosoguanidine. The H chains produced by these variants are similar both in molecular weight and by peptide maps with the parental H chain. H chains dimerize intreacellularly, are not secreted but persist as stable cytoplasmic proteins. No evidence for the continued synthesis of light (L) chains of L chain fragments has been found in these variants. These studies show that, in the P3 cell line, the synthesis of H chains can continue in the absence of detectable L chain synthesis.

Characterization of the J chain from polymeric immunoglobulins (IgA-IgM-immunological specificity-primary structure).

The J chains isolated from polymeric IgA and IgM were shown to be distinct from the other immunoglobulin subunits, including the secretory component, both on the basis of amino-acid content and antigenic determinants. By the same criteria, the J chains were found to be indistinguishable from each other whether they were derived from pentamer IgM, polydisperse myeloma IgA, or dimer colostral IgA. The only differences were observed in (a) the extent of disulfide bond cleavage required for J chain release from the different polymers, and (b) the three-dimensional arrangement of J chains in the different polymers. These data support the hypothesis that the same J chain joins the monomeric units of IgA and IgM to form their respective polymeric molecules.
