Interaction of porcine von Willebrand factor (platelet aggregating factor) with human platelets.

The binding of 125I-labeled porcine von Willebrand factor to washed human platelets was examined. In the absence of ristocetin, binding was found to be rapid, specific and saturable and was decreased in the presence of urea or at high ionic strength. Platelets were found to contain approx. 4760 binding sites for porcine von Willebrand factor with an average binding constant of 2.92 x 10(-7) M assuming the von Willebrand factor to be a tetramer with a molecular weight of 9 x 10(5). Although ristocetin was not absolutely required for binding, in its presence binding was increased.

The PO glycoprotein of peripheral nerve myelin.

The PO glycoprotein, the major protein of peripheral nerve myelin, is a hydrophobic glycoprotein which can be isolated in soluble and insoluble forms from rabbit sciatic nerve myelin following extensive defatting and mid acidic extraction. The PO glycoprotein was localized exclusively in peripheral nervous system (PNS) myelin of sciatic nerve and rootlets by the immunofluorescent technique using goat anti-PO serum which showed a single precipitin band in double diffusion and did not cross-react with the myelin basic protein or P2 protein. Central nervous system (CNS) myelin from brain and spinal cord was negative by the immunofluorescent procedure. The major glycoprotein bands in PNS myelin, in addition to the PO glycoprotein at 28K, exist at 23K and 19K, as shown by gel electrophoresis in dodecyl sulfate. These glycoproteins, isolated by gel filtration in 2% dodecyl sulfate, show identity to the PO glycoprotein in their monosaccharide profile and overlapping tryptic peptides on peptide mapping. We conclude that both the 23K and 19K glycoproteins are derived from the PO glycoprotein by in situ proteolysis; the 23K glycoprotein has the identical amino terminal sequence. The 19K glycoprotein, beginning with amino-terminal methionine, is identical with the TPO glycoprotein, shown previously to originate from tryptic hydrolysis of the PO glycoprotein in isolated myelin. A tryptic glycopeptide containing 27 amino acids was isolated from the PO glycoprotein and sequenced. It contained a relatively high proportion of aspartic acid (four residues) and glutamic acid (two residues), thus exhibiting a high negative charge. We conclude that the total carbohydrate of the PO, 23K, and 19K glycoproteins does indeed exist as a single nonasaccharide moiety linked through N-acetylglucosamine to Asp-14 of the glycopeptide in a N-glycosidic linkage. These results further support the role of the PO glycoprotein as a typical amphipathic membrane protein.

Synthesis of auxins from tryptophan and tryptophan-precursors by fungi isolated from mycorrhizae of pine (Pinus silvestris L.).

Fungi isolated from mycorrhizae of pine required tryptophan for auxin synthesis. More auxins were found in culture grown with pyrogallol than in those without this compound. The fungi studied produced also auxins from other than tryptophan compounds. Indole employed with serine was more suitable for the production of auxins than indole or anthranilic acid used separately. The active compounds showing auxin activity were located on the chromatograms at Rf 0.2--0.4 and 0.3--0.5 with the solvent system isopropanol, ammonia, water (10:1:1 v/v).

The effect of three rad genes on survival, inter- and intragenic mitotic recombination in Saccharomyces. I. UV irradiation without photoreactivation or liquid-holding post-treatment.

The effect of UV irradiation on the survival, inter- and intragenic mitotic recombination of 3 diploid UV sensitive Saccharomyces mutants was studied and compared with the wild type RAD. These strains, homozygous for either the RAD, r1s rad 9-4, or rad 2-20 gene, have DRF values for survival of 1:1.6:3:20.6 respectively, at LD1. Their recombination behaviour is not correlated to their survival characteristics. The RAD, r1s, and rad 2-20 strains showed UV induced mitotic inter- and intragenic recombinants; the induction in the r1s diploid is ca. 100 times greater for both the inter- and intragenic recombinants than in the RAD strain. The rad 9-4 diploid produced no UV induced mitotic recombinants whatsoever, and is therefore considered to be a rec- mutation.

Production of gibberellin-like substances by fungi isolated from mycorrhizae of pine (Pinus silvestris/L.).

Gibberellin-like substances were found to be produced by fungi isolated from mycorrhizae of pine. Among the isolates studied gibberellin-like substances producers were most numerous among the Basidiomycetes forming ectotrophic mycorrhiza and among non-sporulating fungi forming no mycorrhiza. In most strains the gibberellin-like subtances were elaborated after 10--20 days of incubation. The Rf values of the gibberellin-like compounds were different in various strains, but in most of them substances showing gibberellin activity appeared on the chromatograms run with benzene, acetic acid (10:3 v/v) at Rf 0.2--0.6.

Nucleotide sequence of the "denaturable" leucine transfer RNA from yeast.

The nucleotide sequence of " denaturable"leucine acceptor transfer RNA (tRNA(Leu)(3)) from baker's yeast was determined on (32)P-labeled material. The molecule is 85 nucleotides long and can be folded into the "cloverleaf" model for secondary structure. The basis on which the sequence was deduced from the products of complete enzymatic digestion, prior to its unambiguous determination, is presented.
