Venous plasma cyclic AMP in acute cerebrovascular disease.

Antecubital venous blood was sampled from stroke patients in the presence of disodium ethylenediamine tetraacetate. Plasma was analyzed for cyclic AMP applying a competitive protein binding method without any special pretreatment. In mild hemispheric infarction as manifested by moderate hemiparesis and/or dysarthria, plasma cyclic AMP remained in the normal range (8-18 picomoles/ml). In most of the cases with moderate infarction, the cyclic AMP level was distinctly below the normal range several days after the onset of symptoms. However, cyclic AMP remained in the normal range in severe infarction with signs of brain edema, and in two cases with moderately severe symptoms. One of the two cases suffered from later development of brain edema, and the other revealed a large lesion in brain scintigrams. The sizes of the lesion revealed in brain scintigrams were smaller in the moderate cases and larger in the severe cases, except in one of the cases mentioned above. It appeared that with plasma cyclic AMP levels we could predict the extent of the lesion, and perhaps the subsequent development of impending brain edema in a few days after the onset of cerebral infarction. In moderate cases of cerebral hemorrhage, judged from the consciousness, cyclic AMP decreased to a subnormal level 2-4 days after the onset. In severe cases it remained in the normal range. Subarachnoid hemorrhage showed significantly elevated cyclic AMP levels in the early stage.

Immunoglobulin E: raised levels in sera and pleural exudates of patients with paragonimiasis.

Serum IgE concentrations of patients with paragonimiasis were determined by a radioimmunosorbent test. The mean concentration was 3,462.3 IU/ml in a group of 13 cases of paragonimiasis miyazakii in which patients showed clinical symptoms and/or positive immunological diagnostic tests, and 1,026.6 IU/ml in a control group of 13 individuals who had eaten uncooked freshwater crabs, Potamon dehaani, but had been found to be free from the infection. Moreover, the IgE level of the pleural exudates obtained from four patients with paragonimiasis miyazakii on the day of bleeding or within several days after was significantly higher than that of their sera, ranging between 4,200 IU/ml and 10,000 IU/ml. This was true also in a case of paragonimiasis westermani. Sera and pleural exudates of patients with both forms of paragonimiasis were applied to immunosorbent columns of Sepharose 4B beads coupled with saline extracts of Paragonimus miyazakii, P. weetermani, or P. ohirai. IgE eluted from the corresponding column was considered to be specific, being around 5% to 10% of the total IgE.

Mechanism of suppressive effect of basic cupric acetate on rat liver carcinogenesis by ethionine.

The rate of ethylation of t-RNA in vivo by L-ethionine[ethyl-1-14C] markedly lowered in the liver of rats fed a diet containing copper than that of control. In corporation of the labeled compound into the t-RNA of liver by a single injection. of L-ethionine[ethyl-1-14C] was inhibited as much as 50% by the preceding concurrent administration of copper and ethionine added in the diet to rats. Direct interaction of copper ion with ethylation by ethione in the specific t-RNA of the rat liver was examined as an important biochemical explanation in molecular level for the inhibitory mechanism of copper on rat liver carcinogenesis by ethionine. Profiles of normal t-RNA on methylated albumin-kieselguhr column chromatography revealed three components for leucine, two of which were found at the tube number 40 to 50 and disappeared in the leucyl t-RNA treated with ethionine. The components for leucine that disappeared were normalized in the liver of rats by the concurrent administration of copper.

[Aneurysm of the great vein of Galen--report of a case (author's transl)].

A case of aneurysm of the great vein of Galen was reported in which craniotomy and clipping of a feeding artery arising from the left posterior cerebral artery was successfully performed. This 5-month-old girl developed progressive hydrocephalus from 2 months after birth. At 5 months the head circumference was 50 cm with tense enlarged fontanelle. Both eye balls were deviated downward. The deep tendon reflexes were hyperactive with bilateral ankle clonus and positive Babinski's sign. Mild cardiomegaly and hepatomegaly were also noted. Cerebral angiograms showed a large aneurysm of the great vein of Galen fed by a single enlarged arterial branch from the left posterior cerebral artery. After the evaluation of systemic circulatory status and under strict control of fluid transfusion craniotomy was performed. The feeding artery was clipped at its entrance to the aneurysm via left parieto-occipital interhemispheric approach. A rise in the mean arterial blood pressure of 20 mmHg was observed immediately following clipping. The postoperative course was satisfactory except for a left subdural fluid collection which required subduro-peritoneal shunt. The aneurysm was completely disappeared on the postoperative angiograms and the child is regaining the normal development. This is the first case of aneurysm of the great vein of Galen successfully operated in Japan. Clinical symptoms and signs, radiological features and operative treatment of the disease were reviewed from a total of 46 cases reported in the world literature.

The distribution of serotonin-containing cells in the human alimentary tract.

The distribution of serotonin-containing cells (SC cells) in the human alimentary tract was studied by means of the fluorescent histochemical method of Falck et al. on surgically resected specimens. No SC cells could be observed in the nonmucosal parts of the alimentary tract and in the squamous cell epithelium of the esophagus. The richest SC cells were observed in the upper portion of the small intestine (namely the duodenum and the jejunum). The rectum was relatively rich in the distribution of SC cells. They moderately distributed in the terminal ileum and the pyloric and cardiac gland region of the stomach. The ascending, transverse, descending and sigmoid colon and the fundic gland region of the stomach showed poor distribution of SC cells. The basal-layer of the mucosa contained generally richer SC cells than the superficial-layer of the mucosa throughout the alimentary tract except for the esophagus. In the pyloric gland region of the stomach, the rectum and the upper portion of the small intestine, often a few SC cells were observed in the basal part of several glands in the transverse section. In contrast, SC cells could be rarely identified in other parts of the alimentary tract even in the basal part of the mucosa. And it was characteristic that SC cells distributed fairly even in the mucosa of the sigmoid colon.

PCA reactions with mouse antibodies in mice and rats.

Mouse IgE was titrated in rats. The sensitization period was 2 h. The results were consistent and corresponded to the titers obtained in young adults SJL mice using a sensitization period of 48 h. With longer sensitization periods in rats, higher antibody titers were obtained. The optimum sensitization period in rats was found to be 48 h. Old CFW mice are inadequate for mouse IgE titration. IgG1 will not give a PCA reaction in rats. IgG1 titers are higher in SJL or A/J mice than in young CFW mice, and markedly higher than in old CFW mice.

Induction of esophageal cancer associated with gastric cancer in a dog by N-ethyl-N'-nitro-N-nitrosoguandine.

The experimental induction of gastric cancer was studied in four dogs given oral administration of N-ethyl-N'-nitro-N-nitrosoguanidine (ENNG) in solution. Esophageal cancer with the regional lymph node metastasis was found in one dog at autopsy, with concomitantly existing gastric cancer. This dog, which ingested a total amount of about 38 g of ENNG, died of weakness on the 513th experimental day. Three remaining dogs are still living and under observation.
