[Cerebral blood flow and oxygen metabolism in patients with Parkinson's disease].

The distribution of cerebral blood flow and metabolism is related to neuronal activity. Cerebral blood flow (CBF), cerebral metabolic rate of oxygen (CMRO2), and oxygen extraction fraction (OEF) in ten patients with Parkinson's disease and five age-matched normal control subjects were measured with positron emission tomography (PET) using 15O2, C15O2 steady state inhalational technique to investigate functional changes of the cortex and the basal ganglia in Parkinson's disease. All patients had no history of cerebrovascular disease and CT scan showed no abnormal findings except for moderate cerebral atrophy in only one patient. When the level of clinical disability was related on the scale of Hoehn and Yahr, one patient was stage I, four were stage II, four were stage III, and one was stage IV. Psychic symptoms which include hallucination, depression, and dementia were recognized in four patients. One of these four patients was mildly demented. Four patients were newly diagnosed and had never been treated with antiparkinsonian medication before. Before the patients had their PET study their antiparkinsonian medication was discontinued for more than three days. But in two patients PET study was performed without discontinuity of antiparkinsonian medication. The values for regional CBF and regional CMRO2 were lower in the patients than in the normal control subjects, especially in the frontal cortex there was a significant decrease of CBF and CMRO2. There was no discrepancy between CBF and CMRO2 both in the patients and the normal control. CBF and CMRO2 in the cortex and the basal ganglia were not correlated with the severity of tremor, bradykinesia, and rigidity.(ABSTRACT TRUNCATED AT 250 WORDS)

Three-dimensional ultrastructure of the Golgi region of the formative chondrocyte in newborn rat mandibular condyle.

The three dimensional ultrastructure of Golgi region of formative chondrocytes from newborn rat mandibular condyle was examined by means of the method based on dilute osmium maceration with a high-resolution scanning electron microscope. The large area of formative chondrocytes was occupied by the cytoplasmic organelles related to matrix-synthesis such as Golgi apparatus, rough endoplasmic reticulum (RER) and mitochondria. The Golgi region of formative chondrocytes was composed of the limiting membranous structures such as some groups of the stacks of Golgi cisterns, vesicles, vacuoles, secretory granules, reticular constituents and small tabular or tubular RER. The Golgi cisterns were composed of both small tabular parts and rod-like parts arising from tabular parts. Small tabular parts were furthermore subdivided into three types of many, few and no penetrated ones. Reticular constituents were made up of limiting membrane visible as smooth image by scanning electron microscopy, therefore, are assumed to be one part of the GERL (Golgi apparatus-endoplasmic reticulum-lysosome).

[Concentrations of steroid hormones in maternal vein, amniotic fluid and chorionic tissue during early stages of pregnancy].

To study the steroid concentration in human amniotic fluid in early stages of pregnancy between 6-10 weeks, progesterone (P4), 17 alpha-OH-progesterone (17P4), estrone (E1), estradiol (E2) and estriol (E3) levels in amniotic fluid (AF), maternal vein (MV) and chorionic tissue (CH) were measured by RIA. The levels of P4, 17P4, E1, E2 and E3 in CH were significantly higher than in AF and MV. The levels of P4 and E1 in AF were significantly higher than in MV, but the levels of 17P4 in MV were higher than in AF between 6-10 weeks of gestation. Although a peak at 7th week of gestation in AF was noticed, the levels of P4 increased gradually between 6-10 weeks of gestation in AF,MV and CH. There was no significant change in 17P4 levels in AF and CH, except for a slight decrease between 6-8 weeks of gestation in MV. The levels of E2 in MV increased after the 9th week of gestation and the levels were significantly higher than in AF. All estrogens (Es) in CH significantly increased at 10th week of gestation. The levels of P4 in AF and that in CH correlated well. A significant correlation was observed between the levels of E1 and E3 in MV and in CH. These results indicate the possible transference of P4 from CH to AF rather than that to MV. The transference of Es from CH to AF seems to be less than that of P4.(ABSTRACT TRUNCATED AT 250 WORDS)

Leukotriene A4 hydrolase in the human lung. Inactivation of the enzyme with leukotriene A4 isomers.

Leukotriene A4 hydrolase from the human lung was purified to apparent homogeneity. The molecular weight (68,000-71,000), the amino acid composition, and the N-terminal amino acid sequence were similar to those of the human neutrophil enzyme but different from those of human erythrocyte enzyme. The lung enzyme was inactivated by its substrate, leukotriene A4. To elucidate the substrate and the inactivator specificity of this enzyme, we synthesized various geometric and positional isomers of leukotriene A4. 14,15-Leukotriene A4, leukotriene A4 methyl ester, and geometric isomers of leukotriene A4 could not serve as substrates, but they inactivated the enzyme. On the other hand, styrene oxide and (5S)-trans-5,6-oxide-8,10,14-cis-12-trans-eicosatetraenoic acid neither served as substrates nor inactivated the enzyme. These results indicate that whereas allylic epoxide structures of arachidonic acids are responsible for inactivation of the enzyme, the free carboxylic acid, 5,6-oxide, and the tetraene structure with the 7,9-trans-11,14-cis configuration are required as a substrate for leukotriene A4 hydrolase.
