Infiltration of Leu-3a+3b-positive lymphocytes and expression of human leukocyte antigen DR on tumor tissue induced by radiation therapy.

Tumor infiltrating lymphocytes induced by radiotherapy were analyzed by the biotin-avidin-horseradish peroxidase method. At the early dose of 4 Gy (400 rad) of irradiation (2 Gy X 2), remarkable infiltration of lymphocytes was observed surrounding cancer cells. This infiltration was mainly composed of Leu-3a+3b-positive lymphocytes, and human leukocyte antigen DR-positive tumor and interstitial cells were remarkably observed.

Autoantibody to human c-myc oncogene product in autoimmune patients' sera.

Sera from autoimmune diseases including systemic lupus erythematosus, dermatomyositis, progressive systemic sclerosis and Sjögren's disease, were examined for reactivity against human c-myc oncogene product. We first found the presence of the autoantibody to human c-myc oncogene product in sera from autoimmune diseases. No significant correlation between the presence of the anti-c-myc product antibody and disease activity, clinical disease types and other autoantibodies such as antinuclear factor, anti-DNA antibody, anti-RNP antibody and anti-Sm antibody were shown in this study.

Expression of novel interleukin 2 binding molecules and their functional roles in human B cell differentiation.

Expressions and functional roles of novel IL-2 binding molecules (p70, 75) in the differentiation of B cells into Ig secreting cells were explored by using human several B cell lines and tonsillar B cells. Affinity-crosslinking studies revealed that five of nine B cell lines expressed p70 and p75 without detectable Tac antigen (p55) expression and the expression was associated with B cell maturation. In tonsillar B cells, small high-density B cells did not express p70 and p75, whereas large low-density B cells, which were thought to be activated in vivo, expressed them. Binding assays of radiolabeled IL-2 showed that the affinity of these molecules was intermediate (kD = 1-3 nM, 700-3,000 sites/cell). Furthermore, high concentrations of IL-2 (greater than 100 U/ml) induced Ig productions in large B cells and two of five cell lines. These results taken together suggest that B cells may express novel IL-2 binding molecules, associated with B cell differentiation and differentiate into Ig secreting cells by IL-2 through novel IL-2 binding molecules.

Autocrine regulation of cell proliferation by estradiol and hydroxytamoxifen of transformed mouse Leydig cells in serum-free culture.

We have previously reported that the cloned cell line (B-1-A-2) derived from an estrogen-responsive mouse Leydig cell tumor shows an estrogen-dependent enhancement of cell proliferation in medium supplemented with charcoal-dextran-stripped fetal bovine serum. To avoid the involvement of unknown factors present in the serum in the pathway for estrogen-dependent cell growth, the present study was designed to establish a serum-free culture system to which growth factors could be added. To this end, we subcloned B-1 cells from the parental tumor cell line. The proliferation of B-1 cells was markedly stimulated by the addition of 10(-11)-10(-8) M estradiol into the serum-free medium [Eagle's Minimum Essential Medium-Ham's F-12 (1:1, vol/vol) containing 0.2% (wt/vol) BSA]. Epidermal growth factor (0.1-50 ng/ml) or insulin (0.1-50 micrograms/ml) alone or in combination with 10(-8) M estradiol did not affect the proliferation rate of B-1 cells. In contrast, a greater than 10-fold molar excess of 4-hydroxytamoxifen blocked estradiol-induced cell proliferation, while 4-hydroxytamoxifen alone failed to show a stimulatory effect on cell multiplication. Additionally, the conditioned medium collected from estradiol-stimulated cells was found to contain a growth-promoting factor(s) whose activity was not antagonized by 4-hydroxytamoxifen. Nonstimulated cells secreted a significant but low level of the growth-promoting factor. Finally, B-1 cells were found to be estrogen dependent for cell proliferation in BALB/c mice. Their growth was markedly inhibited by the administration of tamoxifen to the host mice. These results indicate that the serum-free culture system presented here is suitable for studying the autocrine mechanisms of cell growth regulated by estrogens as well as triphenylethylene compounds.

[CD antigens and the differentiation of human lymphocytes].

Many monoclonal antibodies directed against differentiation antigens expressed on human lymphocytes have been produced to study the development and functional difference of human lymphocytes. These monoclonal antibodies were classified to the group of "cluster of differentiation (CD)". We reviewed the functional roles of CD antigens on the activation, proliferation and differentiation of human lymphocytes.

Proto-oncogene expression and nuclear factor specifically bound to c-myc gene in peripheral blood mononuclear cells from progressive systemic sclerosis patients as the indicator of clinical activity.

In the present study, we examined the expression of various protooncogenes and the specific binding of nuclear factor to c-myc gene in peripheral blood mononuclear cells (PBMC) from progressive systemic sclerosis (PSS) patients. We demonstrated first amplified expression of c-myc and c-myb gene and the existence of a nuclear protein specifically bound to 5'-non-coding fragment of c-myc gene. We then found a positive correlation between the degree of expression and/or affinity for the c-myc gene fragment of the nuclear protein and clinical disease activity. The amount of the factor was significantly reduced along with or prior to the amelioration of clinical symptoms and laboratory abnormalities with treatment. The significance of c-myc and c-myb proto-oncogene expression and nuclear factor specifically bound to c-myc gene is discussed.

Effect of sofalcone on gastric mucous glycoprotein in experimental gastritis induced by sodium taurocholate.

Changes in gastric mucous glycoprotein (GP) and effects of the anti-ulcer agent, sofalcone, on experimental gastritis induced by 3 or 6 months administration of sodium taurocholate (TCA) were investigated. Macromolecular mucous GP determined as the hexose content, which was fractionated by gel filtration, was decreased with the development of gastritis. This was also demonstrated by the histochemical observations. Soluble mucus, which is easily released into the incubation medium, was significantly decreased, and the insoluble mucus remaining in the gastric mucosa was not changed in 3-month-old gastritis. On the other hand, insoluble mucus was significantly decreased in 6-month-old gastritis. Synthetic activity of mucous GP, which was determined by incorporation of [3H]-glucosamine and [35S]-sulfate, was not changed in 3-month-old gastritis, but, it was markedly decreased in 6-month-old gastritis. The decrease in the content and the synthetic activity of macromolecular mucous GP in 6-month-old gastritis were significantly normalized by 3 weeks administration of sofalcone. The histochemical study also supported the therapeutic effect of sofalcone. These findings suggest that the therapeutic effect of sofalcone on experimental gastritis is closely associated with increased mucus synthesis.

[Application of the immunohistochemical method as predictive assay in radiotherapy of squamous cell carcinoma].

We analysed various subsets of lymphocytes infiltrated into cancer tissues from 15 patients (10 cases of oropharyngeal cancer and 5 of hypopharyngeal cancer) at pretreatment and at about 10 Gy and 30 Gy of irradiation by the method of biotin-avidin-horseradish peroxidase using mouse monoclonal antibodies. In 3 cases, tumor infiltrating lymphocytes were remarkably increased at the delivery of small dose of irradiation: Two of them showed remarkable radiosensitivity, and the lymphocyte subpopulation of the cancer tissue was mainly composed of Leu3a + 3b positive, Leu-8 negative and Leu-HLA-DR positive lymphocytes (activated helper inducer T lymphocytes). On the other hand, the third one proved to be rather radioresistant and the lymphocyte subpopulation was mainly composed of Leu-14 positive lymphocytes (B lymphocytes). These findings indicate that the analysis of the lymphocyte subpopulation infiltrating into cancer tissues at the delivery of small dose of irradiation is applicable as a predictive assay in radiotherapy of squamous cell carcinoma of oropharynx and hypopharynx.

Reduction of doxorubicin-induced cardiotoxicity in mice by taurine.

The effect of taurine on doxorubicin-induced cardiotoxicity was examined in mice. A single intraperitoneal injection of doxorubicin (15 mg/kg) produced a significant elevation of calcium and lipoperoxide content at 72 hr, as well as a significant depletion of creatine phosphokinase, glutamic oxaloacetic transaminase and lactate dehydrogenase activities at 48 hr and glutathione peroxidase activity at 24 hr in the myocardium. These results suggest that a substantial myocardial damage had occurred. All biochemical alterations except depletion of glutathione peroxidase, were markedly attenuated by the combined oral and intraperitoneal administration of taurine. Taurine significantly improved the survival rate of the mice treated with doxorubicin. These results demonstrate that taurine antagonized doxorubicin-induced cardiotoxicity.

c-fos expression in human skin fibroblasts by reperfusion after oxygen deficiency: a recovery change of human skin fibroblasts after oxygen deficiency stress.

A dramatic and specific induction of c-fos mRNA was observed in human skin fibroblasts in vitro culture by oxygen reperfusion after oxygen deficiency treatment. C-fos mRNA reached a maximum about 30-60 min after oxygen reperfusion and declined to basal level after 120 min. And this phenomenon was duration of oxygen deficiency-dependent, and remarkably observed for 0.5-2 hr of oxygen deficiency. More long duration of oxygen deficiency induced a decreasing tendency of c-fos mRNA overexpression due to essential and irreversible cellular damage. Thus increased c-fos gene expression might be an early event in cellular recovery process in particularly human skin fibroblasts with oxygen deficiency.

Heat-shock protein synthesis by human peripheral mononuclear cells from SLE patients.

Peripheral blood mononuclear cells (PBMC) from systemic lupus erythematosus (SLE) patients have heat shock protein (hsp) 70 and hsp 85 as well as the response to various temperatures of normal PBMC and are inducible for these proteins over about 5-8 levels with heat exposure in short-term culture. Synthesis of hsp 70 and hsp 85 as well as the response to various temperatures and the time course of induction were typical for mammalian cell systems. This enhancement with heat-shock treatment was blocked by actinomycin D added before heat exposure. This demonstration that hsp genes are activated in PBMC from active SLE patients without heat exposure supports the hypothesis that gene activation can serve some roles in these cells and be related to the PBMC function in SLE patients. These observations are at least correlative and consistent with a possible homeostatic function for hsps in this system.

Vitamin D-binding protein synthesized by a human hepatocellular carcinoma cell line.

The binding protein for 25-hydroxycholecalciferol was studied in the medium spent for the culture of HuH-7 cells, which were originally derived from a human hepatocellular carcinoma tissue. The binding protein for 25-hydroxycholecalciferol synthesized by HuH-7 cells was immunologically similar to vitamin D-binding protein in human serum and had an inter-alpha mobility. A sedimentation coefficient of 4.1 S was found on sucrose density gradient analyses. The molecular weight was estimated to be approximately 58,000 by gel filtration on a standardized column of Sephadex G-150. When mixed with filamentous actin purified from rabbit skeletal muscle, it depolymerized filamentous actin and bound to monomeric, globular actin to make a 5.5 S 1:1 molar complex with a molecular weight of approximately 100,000. These results support the conclusion that HuH-7 cells produce a functional vitamin D-binding protein.

Start of mu-chain production by the further two-step rearrangements of immunoglobulin heavy chain genes on one chromosome from a DJH/DJH configuration in an Abelson virus-transformed cell line: evidence of secondary DJH complex formation.

A variant was selected from an Abelson virus-transformed cell line with identifiable and distinguishable DJH rearrangements on both chromosomes. Southern blotting experiments suggested that this variant resulted from a secondary DJH complex formation accompanied with the deletion of one of the DJH rearrangements on the same chromosome. Furthermore, this variant started making mu-chains by the further functional join of a VH gene to the secondarily formed DJH complex. Thus, we describe here the first evidence of a secondary DJH complex formation.
