Eosinophilia in mouse regulated by multiple factors following repeated stimulations with ascaris eggs into the peritoneal cavity.

Repeated injections of viable eggs of Ascaris suum into the peritoneal cavity of BALB/c mice induced marked peritoneal eosinophilia (9.3 X 10(6) cells/mouse, 52.8%) without infection. Extracted antigen (Asc) and eggs that had been killed also were able to induce marked eosinophilia; however, their levels were significantly lower (p less than 0.001). This experimental system was able to demonstrate how various multiple factors cumulatively affected the degree of peritoneal eosinophilia. These factors included: mechanical stimulation, caused by the repeated peritoneal lavages (4.4 X 10(5) cells/mouse), against a background level of peritoneal eosinophils of 4.5 X 10(4) cells/mouse; T-cell-independent stimulation, observed when the viable eggs were injected repeatedly into athymic nude mice (nu/nu; 1.6 X 10(6) cells/mouse), while on the other hand, neither eggs that had been killed nor Asc displayed any T-cell-independent augmentation of eosinophilia; and, T-cell-dependent augmentation of peritoneal eosinophilia, observed in heterozygous athymic mice (nu/+), in thymocyte-reconstituted nu/nu, and in normal BALB/c mice. Egg culture supernatant could not act on bone marrow cells to support eosinophil colony formation. In conclusion, only viable eggs were considered prime inducers, consisting of T-independent and T-dependent parts exerting cumulative effects.

Hormonal changes in major depressive disorder.

The dexamethasone suppression test (DST) was performed in 38 patients satisfying the Research Diagnostic Criteria (RDC) for cortisol, human growth hormone (hGH), prolactin, triiodothyronine (T3), thyroxine (T4) and thyroid-stimulating hormone (TSH) were determined at 4:00 p.m. before the administration of dexamethasone, whereas cortisol, hGH and TSH were determined at 4:00 p.m. the day after. Among hormonal variables, only cortisol index (the ratio of the pre-DST cortisol to the post-DST cortisol) was barely useful for discriminating depression. The cortisol index was also correlated with RDC incapacitating subcategory and with symptoms of affect such as depressed mood and psychic anxiety.

Proliferative responsiveness of B cells from autoimmune NZB mice to anti-immunoglobulin and interleukin-4.

Proliferative responses of B cells to anti-IgM antibody and recombinant interleukin-4 (rIL-4) were studied in autoimmune NZB mice. While anti-IgM antibody and rIL-4 act synergistically on resting B cells to induce proliferation in BALB/c and C57BL/6 mice, they failed to produce synergistic effect on NZB B cells, although these NZB B cells responded better to the individual stimulus with anti-IgM antibody or rIL-4. Cell-fractionation analysis of NZB splenic B cells using Percoll density centrifugation showed marked increase in low-density B cells and decrease in high-density B cells. Proliferative response patterns of each subpopulation of NZB B cells were not different from those of control BALB/c B cells, except for higher response of NZB B cells to anti-IgM antibody. Thus, the accumulation of large low density B cells, probably resulting from in vivo activation, may partly account for the altered responsiveness of NZB B cells as a whole.

Possible catabolism of paraquat in mouse brain microsomes.

Whether formalin would be formed from paraquat (PQ) was studied in vitro using subcellular fractions of the mouse tissues. When Nash's method was used, a detectable increase of the OD at 412 nm was obtained only in the brain microsomal fraction in the presence of NADPH. The OD was increased by the addition of 0.1 mM EDTA. It disappeared when NADPH was omitted or when microsomes were denatured. However, when the brain microsomes were incubated with [3H]PQ in the presence of NADPH and EDTA, no [3H] formalin was detected. These results suggest that PQ may be catabolized in the mouse brain microsomes, though not demethylated, resulting in the production of the Nash reagent-reactive substance.

[Interference in double-determinant monoclonal antibody-based assay for carcinoembryonic antigen (CEA)].

We investigated the false positive phenomena in immunoassays of carcinoembryonic antigen (CEA) using mouse monoclonal antibodies, and studied the properties of interfering substances and the method to eliminate non-specific interference. Serum samples from 2,250 patients, which indicated more than 10 ng/ml of CEA with EIA test kit (Boehringer Mannheim Yamanouchi; BMY) in about 25,000 samples of CEA tests, were subjected to extraction by heating at 70 degrees C followed by CEA measurement of the supernatant. The overall correlation was good between CEA values of the original and extraction methods, but CEA values in 55 cases were found to be remarkably higher in the non-extracted method than the extraction method. CEA titers of 20 samples of the discrepant cases were measured by various commercial kits, and the result indicated that non-specific interference was unavoidable in any kit using a monoclonal antibody-based double-determinant immunoassay, in spite of difference in degree and frequency of interference. The interfering activity of two patients serum with remarkably discrepant values eluted at the void volume of a large molecular size of more than 1,000 kDa in gel chromatography of Sephacryl S-300. The non-specific interference was reduced by addition of mouse gamma-globulin to the buffer of BMY kit and abolished by pretreatment of the serum with heat-killed cells of Staphylococcus aureus, suggesting that interference might be caused by some species of immunoglobulin being able to bind mouse gamma-globulin.(ABSTRACT TRUNCATED AT 250 WORDS)

[Prognosis of chronic respiratory diseases in children].

We investigated the prognosis of chronic respiratory diseases in children by questionnaires to the pediatric departments of 4 universities and 14 national or public hospitals in Fukuoka Prefecture. The total number of patients was 155, consisting of 77 males and 78 females. These diseases included bronchiectasis, which had the highest incidence (37, 23.9%), prolonged respiratory disorders in the newborn, pulmonary hypoplasia, pulmonary cysts and pulmonary fibrosis, etc. These 5 diseases comprised 65.8% of all cases. The number of cases on oxygen therapy was 9 (5.8%) and 5 of these 9 cases were on home oxygen therapy. A total of 18 cases (11.6%) were surgically treated. Of 155 cases, 91 cases were cured or improved, while 31 cases were unchanged, 6 cases deteriorated and 6 cases died. The prognosis of 21 cases was unknown. The number of cases whose daily activities were limited was 25 (16.1%).

Pulmonary and pleural involvements at initial diagnosis in children with malignant neoplasm.

Seventeen cases of pediatric malignant neoplasm with pulmonary and/or pleural lesions shown by chest radiography at initial diagnosis were reviewed and analyzed. Respiratory symptoms such as dyspnea, tachypnea, and chest pain were observed on admission in approximately one-half of them. The initial chest radiography showed pleural lesions in 7 of the 17, pulmonary lesions in 8, and both pulmonary and pleural lesions in 2. Unilateral or bilateral pleural fluids were observed in all of the seven patients with pleural lesions, and malignant cells were confirmed in the pleural fluid of all patients. The radiographic patterns of the eight patients with pulmonary lesions were solitary nodule in two, multiple nodules in two, diffuse miliary nodules in three, and diffuse honeycomb in one. Histological examination of the pulmonary lesion was performed in six patients at the initial diagnosis or after death, while in the remaining two primary or other metastatic sites were examined. The two patients with both pulmonary and pleural lesions showed multiple nodules or infiltrates with pleural fluid on chest radiography. Increased malignant cells were detected in the pleural fluid of both patients. In all cases, the lesions gradually or rapidly disappeared with multidrug chemotherapy. The review confirms the need for a combination of complete radiographic and pathological analysis at the time of initial diagnosis of malignant neoplasm in children to distinguish other causes such as infectious complications.

Differentiation in vitro of T3+ large granular lymphocytes with characteristic cytotoxic activity from an isolated hematopoietic progenitor colony.

Blast colonies were developed by rIL-3 from the spleen cells of mice pretreated with 5-fluorouracil (5-FU) in the methylcellulose cultures. When such IL-3-induced blast colonies were individually lifted up and recultured in the presence of rIL-3 and recombinant erythropoietin (rEpo), a variety of hematopoietic colonies developed from every single colony, including neutrophils, macrophages, eosinophils, megakaryocytes, mast cells, and erythroblasts. The results indicated that IL-3-induced blast colonies consisted of multipotential hematopoietic progenitor cells. By culturing individual IL-3-induced blast colonies in the presence of rIL-2 and irradiated peritoneal macrophages, on the other hand, the proliferation of homogeneous lymphoid cells was observed in 5 of 24 wells, each of which received a single blast colony. Morphologically, they were typical large granular lymphocytes (LGL), and thus it was indicated that LGL could be differentiated directly from the hematopoietic progenitor cells utterly in vitro by rIL-2 and accessory macrophages. From one of these culture wells, a continuous LGL line, IL3B1, was successfully obtained. The proliferation of IL3B1 was dependent on IL-2 in the presence of accessory macrophages, but they no longer responded to IL-3, nor to another T cell growth factor, IL-4. Flow cytometric analysis indicated that the phenotype of IL3B1 was Thy-1+,T3+,L3T4-,Lyt-2-,T200+ Asialo GM1+, whereas that of original IL-3-induced blast cells was Thy-1+,T3-,L3T4-,Lyt-2-,B220-. The results suggested that the expression of T3 molecules was induced in the process of LGL differentiation from the hematopoietic progenitor cells in vitro. Conforming to this, it was revealed that both gamma and beta chain genes of the TCR were rearranged in IL3B1. Northern blot analysis indicated that IL3B1 had abundant mRNA for gamma chain, while mRNA for beta chain was rather faint. Functionally, IL3B1 exhibited typical NK-patterned cytotoxic activity against a panel of tumor cell targets. In addition, they showed significant cytotoxic activity against normal bone marrow cells, as well as various factor-dependent myelogenous progenitor cell lines, all of which were resistant to endogenous NK activity of the fresh spleen cells. These results indicated that at least a set of T3+ LGL with rearranged TCR genes could be directly differentiated from isolated hematopoietic progenitor cells in vitro. Results also suggested that such a prethymically differentiated subset of T-lineage lymphocytes, namely T3+ double-negative LGL, had particular cytotoxic activity in addition to conventional NK activity, which might well contribute to feedback regulation of hematopoiesis.

Automated determination of drugs in serum by liquid chromatography with column-switching. I. Separation of antiepileptic drugs and metabolites.

This is a fully automated system for determining six common antiepileptic drugs and two principal metabolites of carbamazepine in serum. It is based on "high-performance" liquid chromatography (HPLC), with column switching. TSKprecolumn BSA-ODS and TSKgel ODS-120A (both from Toyo Soda) were used as the precolumn and analytical column, respectively. The former contains octadecylsilyl resins treated with bovine serum albumin (BSA), and does not adsorb macromolecules such as serum proteins but retains small lipophilic molecules such as antiepileptic drugs. Serum samples are directly injected onto the precolumn. After washing out the serum proteins from the precolumn with sodium phosphate buffer, we switch the column connections to introduce the retained substances onto the analytical column and elute with a step-gradient of acetonitrile/sodium phosphate buffer. The high analytical recovery (95-102%) and the reproducibilities (CV less than 5% within-run) indicate that this system is suitable for use in theraputic drug monitoring in clinical laboratories.
