Renin activity determination using human plasma as a substrate.

A method for human renin activity determination using human plasma as the substrate is described. Angiotensin I is generated by incubating renin with human plasma, which is available along with the commercial radioimmunoassay kit for angiotensin I. The method obviates the need to isolate and purify the substrate, human angiotensinogen, from human plasma. In addition, the assay is highly renin specific, sensitive, and convenient to use for the routine determination of active human renin during its isolation and purification from tissue extracts or from genetically engineered bacterial and nonbacterial expression systems.

Transient increase in intracellular concentration of adenosine 3':5'-cyclic monophosphate results in morphological and biochemical differentiation of C6 glioma cells in culture.

Incubation of C6-BU1 glioma cells in the presence of isoproterenol and Ro20-1724--a potent cAMP phosphodiesterase inhibitor--results in a transient increase in intracellular cAMP levels, followed by a rapid efflux of cyclic AMP from the cells into the media. Two distinct types of morphological changes could be seen: rounded cell bodies with multipolar processes and beadings after 30 minutes of incubation--this period coincides with a 70-80-fold increase in intracellular cAMP levels, and elongated cell bodies with extended bipolar processes after 24-48 hours. By this time the intracellular cAMP concentration dropped to a low level, which was only three- to four fold higher than that in control. The transient increase in intracellular cAMP concentration results in retardation of cell growth, diminished uptake of 3H-2-deoxyglucose, and abolition of enhanced synthesis of cyclic AMP by concanavalin A.

Effect of undernutrition on developing rat cerebellum. Some electrophysiological and neuromorphological correlates.

Electrophysiological studies of Purkinje cells from the developing normal and undernourished rat cerebellar cortex were correlated with the neuromorphological studies. In undernourished animals the firing pattern of Purkinje cells was simpler as compared to complex electrical activity in normal animals. The firing frequency of Purkinje cells remained reduced in all the age-matched undernourished groups. The duration of a spike was also prolonged as compared to their normal age mates. Similarly the mean amplitude was also relatively smaller in the undernourished animals when compared with the normal litter mates. Undernourished animals exhibited typical increased stimulus thresholds, prolonged N-2 peak wave latencies, reduced number of functional components, amplitude and increased duration of mossy fiber response (MFR). The mossy fiber (MF) activity exhibited a sluggish rise in its amplitude, when stimulus intensity was increased in undernourished animals. A phenomenon of fatigue was observed with a significant reduction in the amplitude and frequency of Purkinje cell unit discharge on repeated MF stimulation in the undernourished animals. However, repeated MF stimulation provided a high frequency discharge in the normal as well as undernourished adult rats. Neuromorphological studies at light-microscopic level exhibited delayed cell migration from the external granular layer in the undernourished rats. At electron-microscopic level, intracellular morphology exhibited almost similar ultrastructural details except for a few minor alterations. A free ribosomal pool, immature development of E.R., increased incidence of lysosomes and electron-dense membrane stacks were observed in the Purkinje cells at 30 days in the undernourished animals. The results of the present study indicated that undernutrition affects the maturation of Purkinje cell regional neurocircuitry.

Haemodynamic dose-response actions of cicloprolol in left ventricular dysfunction due to ischaemic heart disease.

Cicloprolol is a cardioselective beta-1 partial agonist; its haemodynamic and radionuclide (nuclear stethoscope) effects were determined in 22 patients with impaired left ventricular function due to coronary artery disease. Following a 20 min stable control period, the effects of four doses of cicloprolol (0.025, 0.025, 0.05 and 0.1 mg/kg at 10 min intervals) were measured at rest 5-10 min after each intravenous injection. The effects of the cumulative 0.2 mg/kg dosage were assessed during supine bicycle exercise and compared with a control exercise period. At rest there were significant increases in systolic arterial without change in mean blood pressure. The heart rate and cardiac index were unchanged. There was a significant increase in left ventricular ejection fraction with a reduction in filling pressure and volume. Patients with resting heart rate below 75 beats/min and with ejection fraction greater than 35% showed the greatest improvement. During supine bicycle exercise, ejection fraction was increased compared to control (31 +/- 2 to 36 +/- 2; P less than 0.01), cardiac volume reduced and exercise tachycardia attenuated. These data suggest that cicloprolol may be of value where beta-blockade is considered in the presence of underlying left ventricular dysfunction due to ischaemic heart disease.

Incomplete epispadiac urethral duplication with dorsal penile curvature.

Incomplete urethral duplication, although not uncommon, usually is associated with insignificant chordee of the penis and it is clinically asymptomatic. We report a case of incomplete urethral duplication with marked dorsal curvature of the penis and other congenital anomalies. Excision of wedge-shaped ellipses of the tunica albuginea from the ventral aspect of the corpus provided an excellent result.

Vesicovaginal fistulas of uncommon etiology.

We report 2 cases of vesicovaginal fistulas sustained during sexual intercourse. In our first case fistula formation was probably the result of physical disparity of the partners and a frightened, unprepared woman. In the second case increased friability caused by hormonal influence during menstruation and a urinary infection probably contributed to fistula formation. Both patients underwent a successful, transabdominal repair with omental interposition.

A haemodynamic and radionuclide assessment of diltiazem in coronary heart disease.

To obtain multiple dose-response haemodynamic and radionuclide data on i.v. diltiazem, 12 ischaemic patients were studied during routine catheterization. At rest, following a 20 min stable control period, the effects of four doses (0.0625, 0.0625, 0.125 and 0.25 mg kg-1 diltiazem at 5 min intervals) were measured in the 3-5 min following i.v. injection. The exercise effects of the cumulative 0.5 mg kg-1 dosage were assessed by comparing a control and post drug period of supine bicycle exercise. The increase in plasma diltiazem levels correlated linearly with the administered dose and achieved therapeutic levels. There were significant dose-related reductions in systemic arterial blood pressure and vascular resistance index; the heart rate fell and cardiac stroke index increased. The calculated double-product (heart rate X systolic blood pressure) was significantly reduced. The left ventricular filling pressures, ejection fraction and cardiac volumes were unaltered. During supine bicycle exercise, the systemic diastolic blood pressure, heart rate and calculated double-product were reduced without change in other parameters. Over the dose range 0.0625-0.5 mg kg-1, diltiazem acutely increased cardiac stroke index and reduced resting heart rate. These haemodynamic data, taken together with its described coronary vasodilator activity suggest that its role in acute vasospastic angina and during angiographic procedures ought to be explored further.

Lymphoid tissue responses to a novel perfluorochemical emulsion in rats.

The effects of a novel perfluorochemical emulsion on rat lymphoid tissues and antibody production against sheep red blood cells (SRBC) have been studied. The responses were compared with those following injection of identical doses of the proprietary emulsion, Fluosol-DA 20% (F-DA). Liver weight was increased up to 15% at 8 days following intravenous (i.v.) or intraperitoneal (i.p.) injection of the novel emulsion but was unaffected by F-DA injection. Spleen weight also increased by a maximum of 20% in response to i.p. injection of the novel emulsion but this was less than increases of up to 44% which occurred in F-DA-injected rats. Thymus weight decreased (P less than 0.05) following i.p. injection of the novel emulsion whereas mesenteric lymph node (MLN) weight remained unchanged. However, MLN weight was increased in response to i.v. injected F-DA, while thymus weight showed a small increase following i.p. F-DA injection. Mean plasma antibody titres to SRBC were significantly (P less than 0.01) increased at 7 days after immunization in rats pretreated with i.p. injections of either the novel emulsion or F-DA; titres in animals pretreated with i.v. injections of either emulsion were similar to control.

Effect of whole body gamma-radiation on glutathione reductase of rat tissues.

Adult female Sprague Dawley rats approximate ten months old were exposed to whole body gamma radiation of sublethal (4 Gy) and lethal (10 Gy) doses of radiation. Serum glutathione reductase enzyme values of 4 Gy exposed rats did not show any change on first and fourth day but significantly decreased on eighth day, while enzyme values in 10 Gy exposed group remained unaltered throughout. Heart glutathione reductase activities of sublethal group did not show any change on first and eighth day but significantly decreased on fourth day. Rats exposed to lethal doses of radiation, enzyme activities remained unchanged throughout. Liver glutathione reductase enzyme values of 4 Gy exposed group showed significant decrease on first and fourth day but arrived at control values on eighth day while the same in 10 Gy exposed group showed significant decrease from first day till the eighth day. An appreciable diminution was observed in spleen glutathione reductase enzyme activities of sublethal and lethal exposed rats from first day till eighth day.

Folding and activation of recombinant human prorenin.

In vitro folding of mature renin, prorenin, and fused prorenin, all produced in denatured form in inclusion bodies in recombinant Escherichia coli, has been studied in order to evaluate the importance of prosequence in the folding of human renin. These studies have been compared with the in vivo folding and subsequent in vitro activation of recombinant human prorenin secreted by a nonbacterial expression system, namely Chinese hamster ovary (CHO) cells grown in serum-free medium. It is concluded that prosequence is essential in the folding of human renin and, therefore, the DNA coding for this sequence cannot be removed without affecting the recovery of active human renin from recombinant bacterial and nonbacterial systems.
