Structure of an amniotic fluid component, 7-(4,5-cis-dihydroxy-1-cyclopenten-3-ylaminomethyl)-7-deazaguanine (queuine), a substrate for tRNA: guanine transglycosylase.

A compound present in bovine amniotic fluid, and which is a substrate for tRNA:guanine transglycosylase, has been structurally characterized as queuine, a hypermodified base which occurs in transfer RNA. The structure assignment is based on mass spectrometry, proton nuclear magnetic resonance, and ultraviolet absorbance data.

Studies of negative ions by collision-induced decomposition and hydrogen-deuterium exchange techniques.

Development of two new techniques for studying the gas phase chemistry of negative ions is reported. Collision induced dissociation (CID) of (M-1)- ions has been accomplished in a newly constructed triple stage quadrupole mass spectrometer. This instrument was assembled by adding two additional Finnigan quadrupole mass filters to a Finnigan Model 3200 CI mass spectrometer. Generation of (M-1)- ions is accomplished by allowing OH- and sample to react under CI conditions in the ion source. The first quadrupole mass filter, Q1, is then employed to selectively pass the (M-1)- ion into a second quadrupole filter containing argon or neon at 10(-3) torr. On collision with the inert gas the (M-1)- ions dissociate into fragments which are then mass analyzed in the third quadrupole filter, CID spectra of (M-1)- ions from twelve carbonyl compounds are presented in this paper. Ion molecule isotope exchange reactions in the CI ion source can be used to count the number of hydrogen atoms in many different chemical environments. Collisions between sample (M-1)- ions and deuterium-labeled reagent gases (ND3, D2O, EtOD) facilitate incorporation of deuterium into the negative ion if the basicities of the sample and reagent anions are similar. Thus it is possible to selectively incorporate deuterium into many organic samples by controlling the exothermicity of the acid base, ion-molecule chemistry.

Reproducible plaquing system for rhinovirus serotypes in HeLa cells--agarose suspension.

HeLa cell--agarose suspension method provides an excellent plaquing system for assay of rhinovirus serotypes which ordinarily produce indistinct plaques on cell monolayers. The variable plaque size of cloned virus progeny does not involve antigenic change in phenotype as shown by plaque neutralization test. The system has proved useful for accurate and reproducible plaque assay of rhinoviruses, assay of serum neutralizing antibodies and cloning of viruses. The plaques produced are large, clear and reproducible. The factors which influence the plaquing of rhinoviruses in this system are the age of the cells, cell density and pH of the medium.

Cross-neutralization reactions of Escherichia coli and Vibrio cholerae enterotoxins as studied by rabbit skin inoculation test.

Neutralization of enterotoxins of Vibrio cholerae 569 B and Escherichia coli 10407 by antitoxins to V. cholerae 569 B, E. coli 334, 408-3 and 10407 was studies by intradermal inoculation test in the rabbit. Neutralization of V. cholerae enterotoxin by homologous as well as heterologous antisera of E. coli was observed, except that there was no neutralization of the enterotoxin by antiserum to E. coli 408-3 enterotoxin. Neutralization of E. coli enterotoxin to a varied extent by homologous as well as all heterologous antisera, including that of V. cholerae 569 B antitoxin, was also observed.

Resistance of Salmonella serotypes to chloramphenicol.

A total of 704 Salmonella strains isolated from various sources were tested for their resistance to chloramphenicol. The results indicate that 36 out of 46 serotypes, comprising 671 strains (95.4%), were sensitive to chloramphenicol up to a level of 10 mug per ml of medium. Most of the strains were sensitive at concentrations between 2 mug and 4 mug of chloramphenicol per ml of medium. None of the strains isolated from animals, including poultry, was resistant. The results indicate that in India resistance of Salmonella strains to chloramphenicol has not increased over the years though this may have occurred in other parts of the world.

Effect of cholera enterotoxin preparations on cutaneous response in rabbit under varied conditions.

Enterotoxic activity of two preparations obtained from Vibrio cholerae, B-53-6 Inaba and B-53-10 Ogawa was tested in ligated ileal loops of rabbit. The biologically active enterotoxic preparations were further used to study the permeability reaction in rabbit skin. Cutaneous response tended to be linear only with higher concentrations of the toxin and showed maximum blueing intensity between 16 and 24 hrs of intracutaneous inoculation. Exposure of enterotoxin preparations to elevated temperatures greatly reduced the cutaneous response and the activity was completely lost at 100 degrees C. Change in pH towards alkaline side lowered the permeability activity to lesser extent as compared to a shift on acidic side. However, a residual activity could still be detected when pH of the enterotoxins was lowered to 3 at 30 degrees C for 4 hrs.
