Emissions of gaseous and particulate pollutants in a port and harbour region in India.

Ports can generate large quantity of pollutants in the atmosphere due to various activities like loading and unloading, transportation, and construction operations. Determination of the character and quantity of emissions from individual sources is an essential step in any project to control and minimize the emissions. In this study a detailed emission inventory of total suspended particulate matter (TSP), particulate matter less than 10 microm (PM10), sulfur dioxide (SO2) and nitrogen oxides (NOx) for a port and harbour project near Mumbai is compiled. Results show that the total annual average contributions of TSP and PM10 from all the port activities were 872 and 221 t yr(-1), respectively. Annual average emissions of gaseous pollutants SO2 and NOx were 56 and 397 t yr(-1), respectively, calculated by using emission factors for different port activities. The maximum TSP emission (419 t yr(-1)) was from paved roads, while the least (0.4 t yr(-1)) was from bulk handling activity. The maximum PM10 emission (123 t yr(-1)) was from unpaved roads and minimum (0.2 t yr(-1)) from bulk handling operations. Similarly the ratio of TSP and PM10 emission was highest (5.18) from paved roads and least (2.17) from bulk handling operations. Regression relation was derived from existing emission data of TSP and PM10 from various port activities. Good correlation was observed between TSP and PM10 having regression coefficient >0.8.

Dark neurons in the ageing cerebellum: their mode of formation and effect of Maharishi Amrit Kalash.

Dark neurons are considered a manifestation of neuronal injury and although they cover various grades of damage their mode of formation is not yet clear. Age-dependent alterations in a dark purkinje neuronal population of guinea pigs (10 months and 32 months old) and rats (3 months, 6 months, 12 months, 15 months and 28 months) were studied. Light microscopical and electron microscopical observations revealed a significant increase (P < 0.05) in the number of dark purkinje neurons with age in both the guinea pigs and rats. Extraction of lipids from the cerebellum sections before processing for histochemical reaction resulted in a reduction of the dark neuronal population. In an other set of experiments, significant age-dependent increase in the cathepsin-D activity and lipid peroxidation was documented in the guinea pig cerebellum. Treatment of guinea pigs with Maharishi Amrit Kalash (MAK) (500 mg/kg body wt/day, for two months) significantly inhibited (P < 0.05) the activity of cathepsin-D and lipid peroxidation, and decreased the number of dark neurons. These findings suggest that the number of dark neurons increases with age and MAK prevents the conversion of light to dark purkinje neurons due to its inhibitory effects on cathepsin-D activity and antioxidant properties. We suggest that the conformational changes in the normal protein structure due to higher proteolytic activity and peroxidation of lipid in the aging cerebellum endangers a redundant capability for various staining agents and the Osimic acid molecules to react with proteins, lipids and other molecules, leading to an intensified cyto- and karyoplasms electron density.

Effects of alternative carbon sources on biological transformation of nitrophenols.

The removal of nitrophenols under denitrifying conditions was studied in bench-scale upflow anaerobic sludge blanket (UASB) reactors (R1, R2, R3 and R4) using three different carbon sources. Initially acetate was used as carbon source (substrate) in all the four reactors followed by glucose and methanol. Reactor R1 was kept as control and R2, R3, R4 were fed with 30 mg/l concentration of 2-nitrophenol (2-NP), 4-nitrophenol (4-NP), and 2,4-dinitrophenol (2,4-DNP), respectively. Throughout the study the hydraulic retention time (HRT) and COD/NO3-N ratio were kept as 24 h and 10, respectively. 2-Aminophenol (2-AP), 4-aminophenol (4-AP) and 2-amino,4-nitrophenol (2-A,4-NP) were found as the major intermediate metabolites of 2-NP, 4-NP and 2,4-DNP degradation, respectively. Methanol was found to be a better carbon source for 4-NP and 2,4-DNP degradation as compared to acetate and glucose, while 2-NP degradation was not influenced much by the change of substrate. Nitrate nitrogen removal was always more than 99%. COD removal efficiency of the nitrophenol fed reactors varied from 85.7% to 97.7%. The oxidation-reduction potential (ORP) inside the reactors dropped, up to -300 mv, with glucose as carbon source. As the reactors were switched over to methanol, ORP increased to -190 mv. The granular sludge developed inside the reactors was light brown in colour when acetate and glucose were used as substrate, which turned dark brown to black at the end of methanol run. Biomass yield in terms of volatile suspended solids was observed as 0.15, 0.089 and 0.14 g per gram of COD removal for acetate, glucose and methanol, respectively.

Lycopene prevents sugar-induced morphological changes and modulates antioxidant status of human lens epithelial cells.

Cataract is a multifactorial disease. Osmotic stress, together with weakened antioxidant defence mechanisms, is attributed to the changes observed in human diabetic cataract. Epidemiological studies provide evidence that nutritional antioxidants slow down the progression of cataract. The usefulness of lycopene, a dietary carotenoid, in the pathogenesis of human cataracts has not been studied so far. Since the epithelium is the metabolic unit of the lens, the effect of lycopene on galactose-induced morphological changes and antioxidant status of human lens epithelial cells (HLEC) in culture was evaluated in the present study. HLEC of fresh cadaver eyes obtained from an eye bank were cultured in medium supplemented with fetal calf serum (200 ml/l). On confluency, the cells were subcultured in medium containing either 30 mm-d-galactose or 30 mm-d-galactose+lycopene (5, 10 or 20 microm) for 72 h. The cells were observed under the phase-contrast microscope and transmssion electron microscope for any morphological changes and then harvested for the estimation of various biochemical variables. Malondialdeyde, glutathione and antioxidant enzymes were significantly altered in the control as compared with the normal cultures. Vacuolization was also observed in the presence of galactose. Addition of lycopene confers significant protection against these changes in HLEC.

Image-guided fine-needle aspiration cytology of deep-seated enlarged lymph nodes.

PURPOSE: To evaluate the role of image-guided fine-needle aspiration cytology (FNAC) of deep-seated lymph nodes. MATERIAL AND METHODS: Image-guided FNACs were performed on 242 patients of deep-seated lymph nodes which included thoracic, retroperitoneal and abdominal nodes. A sterile 3.5/5-MHz micro convex sector probe was used for localisation of the node. The FNAC was performed using a 0.7- to 0.9-mm needle with the stylet removed and attached to a 20-ml syringe and FNAC handle after the needle was visualised in the lesion. For each case a minimum of 4-5 smears were made, and two observers without bias interpreted the smears. RESULTS: A total of 242 patients were aspirated, of which 216 (90%) aspirations were US-guided and the remaining 26 (10%) were CT-guided. Adequate material for cytologic diagnosis was obtained in 208 (86%) patients with a similar diagnostic accuracy. The aspirate material was non-representative or scanty in 34 (14%) patients. The most common cytological diagnosis was tuberculosis/consistent with tuberculosis seen in 108 (45%) patients followed by metastasis (17%) and reactive hyperplasia (10%). Non-Hodgkin lymphoma was diagnosed in 22 (9%) patients. All patients were briefly followed for a period of 1 1/2 to 2 years (mean 1 year). CONCLUSION: Image-guided FNAC has a pivotal role, and is a cost-effective tool for establishing tissue diagnosis as a primary investigative modality. It is also helpful and accurate in follow-up of patients with a known malignant disease, thereby avoiding surgical intervention.

Biosorption of nitrophenols on anaerobic granular sludge.

The sorption and desorption of 2-nitrophenol (2-NP), 4-nitrophenol (4-NP) and 2,4-dinitrophenol (2,4-DNP) on live anaerobic granular sludge was studied at five different sorbate (nitrophenols) concentrations between 10 x 10(3) - 90 x 10(3) microg l(-1). Experiments were conducted at room temperature (29 +/- 2 degrees C) and at an initial pH of 7.5. The maximum uptake capacity of live anaerobic granular sludge was found to b e 1,427, 1,511 and 1,865 microg g(-1) VSS for 2-NP, 4-NP and 2,4-DNP, respectively. The equilibrium data of three nitrophenols obeyed both Freundlich and linear isotherm models. Sorption was observed to be partially reversible. About 20-89%, 36-90% and 29-80% desorption was observed for 10 x 10(3) - 90 x 10(3) microg l(-1) sorbate concentrations of 2-NP, 4-NP and 2,4-DNP, respectively.

Effect of melatonin and phenytoin on an intracortical ferric chloride model of posttraumatic seizures in rats.

The present study was carried out to investigate the effect of melatonin, a potent antioxidant, and phenytoin, a conventional antiepileptic, against FeCl3-induced posttraumatic seizures. Male Wistar rats weighing 200-250 g were implanted with epidural electrodes and allowed to recover. After the recording of baseline EEG, FeCl3 (5 ul, 100 mM) was administered intracortically over a period of 5 min and EEG was monitored for 2 h. Subsequently, rats were sacrificed to estimate oxidative stress, i.e., the amount of malondialdehyde (MDA) in whole brain tissue. A sham group was run parallel with saline (pH adjusted), and a similar protocol for EEG recording and estimation of oxidative stress was followed. FeCl3-treated animals exhibited epileptiform EEG changes (high amplitude sharp waves of increased frequency and polyspikes) within 15 min, which continued throughout the period of observation. MDA levels were found to be significantly elevated as compared to the sham group. Melatonin (50 mg/kg i.p.) administered 30 min before FeCl3 injection delayed the onset of appearance of epileptiform EEG changes, while at a 100 mg/kg dose of there was complete protection, as none of the animal exhibited epileptiform EEG discharge. Brain MDA levels were also significantly reduced in melatonin (50 and 100 mg/kg dose)-treated animals as compared to the vehicle-treated FeCl3-injected rats. In the phenytoin group, all animals showed epileptiform EEG discharge. However, phenytoin at both 50 and 100 mg/kg dose delayed the onset of epileptiform EEG discharge. There were no significant changes in brain MDA levels in the phenytoin-treated group as compared to controls. Melatonin and phenytoin at doses of 100 mg/kg did not show any sign of motor impairment as observed during the rota-rod test. These findings showed a superior protective effect of melatonin over phenytoin in an intracortical FeCl3 model of posttraumatic epilepsy.

Evaluation of the immunocontraceptive potential of Escherichia coli-expressed recombinant dog ZP2 and ZP3 in a homologous animal model.

Dog zona pellucida glycoprotein 2 (dZP2), excluding the N-terminal signal sequence and the C-terminal transmembrane-like domain, was cloned and expressed as a polyhistidine fusion protein in Escherichia coli to evaluate the immunocontraceptive efficacy of ZP glycoproteins. The recombinant dZP2 (rec-dZP2) revealed a 70 kDa band corresponding to the full length transcript, as well as several low molecular mass fragments in western blot analysis. In addition to rec-dZP2, E. coli expressed recombinant dog ZP glycoprotein 3 (rec-dZP3), which has also been evaluated for its efficacy to block fertility in a homologous system. Three groups of female dogs (n = 4 per group) were immunized with rec-dZP2 conjugated to diphtheria toxoid (rec-dZP2-DT), rec-dZP3 conjugated to DT (rec-dZP3-DT) and DT alone. Immunization of female dogs with rec-dZP2-DT and rec-dZP3-DT led to generation of antibodies against the respective ZP proteins as well as to DT. Subsequent to mating, the four female dogs immunized with rec-dZP2-DT all conceived, which is indicative of failure of the anti-rec-dZP2 antibodies to block fertility. In the group of dogs immunized with rec-dZP3-DT, three of four animals did not conceive when mated with males of proven fertility. The block in fertility was associated with anti-dZP3 antibody titres. Ovarian histopathology revealed that the block in fertility in the group immunized with rec-dZP3-DT is probably manifested by inhibition in the development of follicles and is due to atretic changes in the zona pellucida. These results, although preliminary, indicate that immunization with dZP3 may be a feasible proposition to control dog populations provided that adequate antibody titres are achieved.

Expression of Bcl2 proto-oncogene in primary tumors of the central nervous system.

The present study was addressed to find out the expression of Bcl2 proto-oncogene in tumor tissues derived from 25 patients with primary central nervous system tumors. Brain parenchyma in 8 cases, with deeply located tumor, was also examined for Bcl2 expression which served as control. Both benign and malignant tumors (confirmed by histopathological examination) expressed Bcl2 gene product. Tumors exhibited 2-6 fold increase in Bcl2 expression as compared to the normal parenchyma adjacent to some of these tumors studied. However, no correlation was found between the histopathological types of tumor, glial fibrillary acidic protein positivity and degree of Bcl2 expression. Based on this study, we propose that the overexpression of Bcl2 gene product found in primary CNS tumors may be an important molecular event which is known to make the various types of tumor resistant to chemotherapy or radiotherapy.

Primary central nervous system lymphoma: experience of 46 cases with review of literature.

Primary central nervous system lymphomas (PCNSL) constitutes only 1.0 to 1.5% of all brain tumors. Their incidence has gone up over tenfold in the last 25 years. Though, there has been an association of PCNSL with acquired immune deficiency syndrome (AIDS), yet the increased incidence of PCNSL appears to be real and unrelated to AIDS and organ transplantation. This increased incidence could be because of improvement in diagnostic technology and practice. The outcome remains gloomy despite surgical resection, radiotherapy and intensive adjuvant chemotherapy regimens, as majority of the patients succumb to the disease, with only 30-40% survival in patients under 70 years of age.

Short-term androgen deprivation does not alter CaR and VDR mRNA expression in duodenal mucosa in male rats.

Androgen deprivation is associated with decline in intestinal calcium absorption. The effect of androgen on CaR and VDR intestinal mucosa has not yet been studied. Calcium homeostasis, a real bone mineral density (aBMD, dual energy X-ray absorptiometry) and expression of CaR and VDR mRNA in duodenal mucosa of orchidectomized (ORX) and sham operated (Sham) adult Sprague Dawley rats at 4 week have been studied. There was no significant difference in serum calcium, alkaline phosphatase, calcidiol and calcitriol levels between both the groups. Serum testosterone (T) (ng/dl) and inorganic phosphorous (iP) (mg/dl) levels were significantly lower in ORX rats. As compared to sham rats, ORX rats had significant decline in in-vitro aBMD at proximal, middle and distal tibia, proximal, mid and distal femur and femoral neck (P < 0.05). Northern blot analysis revealed no significant alteration in the CaR and VDR mRNA expression in duodenal mucosa in ORX rats. CaR and VDR mRNA expression in duodenal mucosa is therefore, not affected by physiological concentrations of testosterone in rats.

Assessment of the quality of sewage effluents from dry weather flow channel, Calcutta.

The quality of sewage effluent of Dry weather Flow channel, Calcutta in respect of salinity, sodicity, chloride, sulphate and bicarbonate toxicity and heavy metal hazards was assessed in order to utilize it for irrigation. Although raw sewage in the winter season was toxic in respect of chlorides, sulphates, bicarbonate, BOD, COD; its dilution in the monsoon decreased the toxicity hazards considerably, making it worth using for irrigation. The sewage effluents were rich in N and K, but poor in P status with marginal concentrations of micronutrients (Fe, Cu, Zn & Mn). Heavy metal contents of the soil treated with effluents were also studied.

Suppression of hadrons with large transverse momentum in central Au+Au collisions at root square[s(NN)] = 130 GeV.

Transverse momentum spectra for charged hadrons and for neutral pions in the range 1 GeV/c<p(T)<5 GeV/c have been measured by the PHENIX experiment at RHIC in Au+Au collisions at root square[s(NN)] = 130 GeV. At high p(T) the spectra from peripheral nuclear collisions are consistent with scaling the spectra from p+p collisions by the average number of binary nucleon-nucleon collisions. The spectra from central collisions are significantly suppressed when compared to the binary-scaled p+p expectation, and also when compared to similarly binary-scaled peripheral collisions, indicating a novel nuclear-medium effect in central nuclear collisions at RHIC energies.
