[Effect of nm23H1 on proliferation, tumor formation and metastasis of hepatocarcinoma].

OBJECTIVE: To study the effects of nm23H1 on in vitro proliferation, in vivo tumor formation and metastasis of hepatocarcinoma. METHODS: Vectors containing insert of either sense or antisense nm23H1 cDNA under the albumin enhancer/promoter for liver cell-specific expression were constructed. The expression vectors were transfected into hepatocarcinoma cell line SMMC-7721. Stable cell clones with maximum and minimum expression of nm23 were obtained. Cell growth was measured in vitro and metastasis was observed in vivo by injecting cells into nude mice, subcutaneously or under the spleen capsule. RESULTS: In vitro, cell clone expressing antisense nm23H1 grew faster than that expressing sense nm23H1. In nude mice, the subcutaneously inoculated tumor grew faster and more aggressively in mice bearing antisense nm23H1 transfected hepatoma. So did metastasis of the antisense nm23H1 transfected hepatoma under the spleen capsule. CONCLUSION: Apart from suppressing metastasis, nm23H1 significantly suppresses tumor cell proliferation in vitro and tumor growth in vivo.

[Troponin T during cardiopulmonary bypass].

OBJECTIVE: To evaluate the effect of cardiopulmonary bypass (CPB) on myocardial troponin T and its clinical significance and relationship to myocardial cell injury. METHODS: Thirty-seven patients underwent open heart surgery under CPB. The procedures included correction of congenital heart disease in 15 patients, valve replacement in 15, and coronary artery bypass grafting in 7. Blood samples were taken at four different time intervals for assessing a troponin T. Correlative factors including duration of clamping of aorta, dosage of cardioplegic solution, and metabolic acidosis were studied. RESULTS: When clamping time > 60 minutes, dosage of cardioplegia solution < 10 ml/kg and existing metabolic acidosis, the level of troponin T was significantly increased (P < 0.01). CONCLUSION: The elevation of troponin T is closely related to cardiopulmonary bypass, especially the duration of clamping of aorta, insufficiency of cardioplegia, and metabolic acidosis.

[Cost-effectiveness analysis on two different measles immunization strategies].

OBJECTIVE: To seek the most economic and effective strategy in measles elimination in China, data of cost - effectiveness was analyzed to compare two different strategies of measles immunization (mass campaign and 2 - dose schedule) based on the data of population, economy and epidemiological features of measles in Henan province. METHODS: DALY was used as the measurement of effectiveness. Taking period from 1997 to 2020, the cost - effectiveness ratio (CER) was defined as the cost difference between mass campaign and 2 - dose schedule divided by the decline of DALYs if 2 - dose schedule had been substituted by mass campaign. RESULTS: Along with the increment of time, CER showed a descending trend then the mass campaign will have positive benefits after year 2013. CONCLUSION: Mass campaign is more cost - effective than 2 - dose schedule.

[A study on methods to measure the social burden of disease and their significance].

OBJECTIVE: To explore the effects of social burden of disease (BOD) on the total BOD. METHODS: With stroke as an example, BOD was measured using the index, disability adjusted life years (DALY), and partial social BOD was measured using another index, time lost for nursing care of hospitalized patients. RESULTS: The burden of stroke was 11.70 DALYs per thousand in men and 9.38 in women, respectively, and the burden for caring of patients with stroke during hospitalization accounted for 2.2% and 2.8% of the total BOD for men and women, respectively. CONCLUSION: Effects of social BOD should be taken into consideration in measurement of total BOD.

[The expression of cell adhesion molecules on lymphatic endothelial cells in rectum adenocarcinoma of human].

This study was conducted to explore the mechanism of lymphatic dissemination and metastasis of cancer cells; the specific aim was to investigate the expression of cells adhesion molecules on lymphatic endothelial cells. Immunohistochemical method was used. The rectum tissues and lymphnodes were taken from five normal human subjects. The cancer peripheral tissues and lymphnodes, and the metastasized lymphnodes were taken from eight patients with rectum adenocarcinoma. The result showed that the intercellular adhesion molecule-1 (ICAM-1) and carcinoma embryonic antigen (CEA) were expressed on lymphatic endothelial cells in cancer peripheral tissues and metastasized lymphnode. The expression of ICAM-1 and CEA in cancer peripheral tissues and lymphnodes was much more remarkable than that in metastasized lymphnodes. These data suggest that ICAM-1 and CEA be cell adhesion molecules between cancer cells and lymphatic endothelial cells.

[Transcription regulation of intercellular adhesion molecule (ICAM-1) on lymphatic endothelial cells of rectum adenocarcinoma of human].

This study inquired into the mechanism of cancer lymphatic metastasis. The immunohistochemistry method and DNA hybridization in situ were employed in examining the peritumoral rectum tissues and metastatic lymph nodes from 8 rectum carcinoma patients and the rectum tissues and lymph nodes from 5 normal subjects. The results showed that the proteins of ICAM-1 and nuclear factor kappa B (NF kappa Bp65) were expressed on the lymphatic endothelial cells of the rectum adenocarcinoma patients, and there was a NF kappa B binding consensus sequences on ICAM-1 promoter in lymphatic endothelial cells of rectum adenocarcinoma patients. When DIG-AKP labeled 38 bp oligonucleotide probe was used, there was no the expression of ICAM-1 and NF kappa Bp65 on the lymphatic endothelial cells of the normal human lymph node and rectum tissues. It is suggested that the activation of ICAM-1 promoter may critically depend on NF kappa Bp65 homodimers or heterodimers binding to a variant kappa B site on the lymphatic endothelial cells of the rectum adencarcinoma of human. These observations may indicate the potential roles of NF kappa B in cancer metastasis, thus giving clues to facilitate the development of a novel anti-metastasis strategy, that is obstruction the NF kappa B-activation pathways.

[Expression of simian virus 40 large T-antigen and p53 protein in chinchilla middle ear epithelial cell line and its significance].

OBJECTIVE: To identify the expression of simian virus 40 large T-antigen (SV40TAg) and p53 protein in the cell nuclei of chinchilla middle ear epithelial cell line and explore the significance of the expression. METHODS: To examine the expression of SV40TAg and p53 protein in cell nuclei of chinchilla middle ear epithelial cell which had been infected and had not been infected with Ad 12-SV40 hybrid virus by immunocytochemistry method. RESULTS: All of the cell nuclei in 1 of 8 culturing dishes which infected with the Ad12-SV40 virus expressed the SV40TAg and p53 protein and had cultured over 46 passages. They have become a cell line. All of the cells in rest 7 culturing dishes and the control cells did not express the SV40TAg and p53 protein in their nuclei and died after culturing 6-8 passages. CONCLUSION: The SV40TAg and p53 protein play some important role in process of cell immortalization. The expression of SV40TAg and p53 protein in cell nuclei of CMEE-1 cell line has close interrelations with establishing the CMEE-1 cell line.

[The noise-filtering of chemiluminescence spectra with wavelet multiresolution analysis].

In this paper, the wavelet multiresolution signal decomposition was successfully applied to the filtering of high-frequency noise in chemiluminescence spectra. The spectra signal-to-noise ratio was greatly improved while the noise was suppressed effectively. The effect of wavelet basis and time of decomposition are also discussed. It is shown that the wavelet analyses have many advantages in processing of discrete signals.

A new monoclonal anti-idiotypic catalytic antibody with a CPA-like activity.

IIF9D8, a new monoclonal anti-idiotypic catalytic antibody with a CPA esterase-like activity was elicited by ID11D7, the monoclonal competitive inhibitory antibody to CPA. The hydrolysis of hippuryl-DL-phenyllactic acid by McAb IIF9D8 follows the Michaelis-Menten kinetics. The Km value and kcat are 0.036 M and 0.598 min(-1), respectively, and the rate acceleration (kcat/kuncat) is 30500. Compared with the previous McAb 32C3 induced by polyclonal antibodies to CPA, McAb IIF9D8 shows higher catalytic efficiency The catalytic antibodies with the catalytic properties similar to natural enzymes could be obtained by this approach.

[Healthy life years(HeaLY): a new indicator for measuring the burden of diseases].

Healthy life years(HeaLY) is a new indicator for measuring the burden of diseases. The data of Beijing MONICA were analyzed and reevaluated by using this indicator, and were compared with another indicator, disability adjusted life years (DALY), for the burden of diseases provided by WHO and World Bank several years ago.

[Impact of maternal occupation on the dietary and nutritional status of preschool children. A case study in 8 provinces of China].

This analysis is based on the data from China Health and Nutrition Survey (undertaken in 8 provinces of China from 1989 to 1998) in collaboration with the University of North Carolina at Chapel Hill. Totally, 1461 and 1087 mother-child pairs under age 6 were selected to investigate the impact of maternal occupation on the dietary and nutritional status of children in 1991 and 1993. The results show that the nutrients intake and nutritional status of children whose mothers worked in administrative, scientific, technological and other professional units were better than any other groups. The status of children whose mothers were workers, waiters etc. in factories or service sectors was better than that of children whose mothers were farmers or self-employed laborers. The children of non-working mothers were better nourished than the children whose mothers were farmers, or self-employed laborers, but were more undernourished than the children whose mothers worked in administrative, scientific, technological and other professional units. The associations among maternal occupation, income and education level of mothers were significant. Mothers working in administrative, scientific, technological units etc. had higher yearly income and education level than other groups. But the income and education level of farmers and self-employed laborers were the lowest. The time allocation of mothers was also studied and the results suggested that maternal occupation affected the time spent for child care and food preparing.

[Studies on the factors affecting the individual's animal protein intake by a multi-level model].

In this article, the influence of factors related to different levels (seg, provincial, county and household) on the proportion of protein intake from animal food was evaluated by using multi-level modeling. It was found that after being adjusted by individual factors, the factors related to the household, such as household income, dietary habit and knowledge of nutrition were the most important factors affecting the individual's showed animal protein intake. In addition, the results showed that the difference of animal protein intake in various places depended not only on the increase of household income, but also dietary behavior and agriculture crops in different areas. Comparison of different models indicates that in analyzing the data from large-scale health survey with hierchical structure, multi-level modeling is strongly recommended.

Mass spectral fragmentation of cis- and trans-1a,3-disubstituted 1, 1-dichloro-1a,2,3,4-tetrahydro-1H-azirino[1,2-a] [1, 5]benzodiazepines.

The mass spectrometric behaviour of six cis- and trans-1a, 3-disubstituted 1,1-dichloro-1a,2,3,4-tetrahydro-1H-azirino[1,2-a][1, 5]benzodiazepines has been studied with the aid of mass-analysed ion kinetic energy spectrometry and exact mass measurements under electron impact ionization. All compounds show a tendency to eliminate a neutral propene or styrene molecule from the diazepine ring and to simultaneously eliminate a chlorine atom from the aziridine ring to yield azirino[1,2-b][1,3]benzimidazole ions, and to lose hydrogen chloride plus propene or styrene to give quinoxaline ions. All compounds also show a tendency to eliminate N=CCl(2) or HN=CCl(2) to produce dihydro- or tetrahydroquinoline ions, and to eliminate sequentially hydrogen chloride and a chlorine atom to yield pyrrolo[1,2-a][1, 3]benzimidazole ions.

Electron impact mass spectra of 1a,3-disubstituted 1,1-dichloro-1,1a, 2,3-tetrahydroazirino [2,1-d][1,5]benzothiazepines.

The mass spectrometric behaviour of eight 1a,3-disubstituted 1, 1-dichloro-1,1a,2,3-tetrahydroazirino[2,1-d][1,5]benzothiazepines has been studied with the aid of mass-analyzed ion kinetic energy spectrometry and exact mass measurements under electron impact ionization. All compounds show a tendency to eliminate a chlorine radical from the aziridine ring and then eliminate a neutral propene or substituted/unsubstituted styrene from the thiazepine ring to yield 1,4-benzothiazine ions. They can also lose hydrogen chloride and a chlorine radical, or lose two chlorine atoms, to undergo a ring enlargement rearrangement to produce 1,6-benzothiazocine ions, which can further eliminate propyne or phenylacetylene, or small molecular fragments, to yield some abundant fragment ions.

Mass spectral fragmentation patterns of 1a,3-diaryl-1,1,4, 4-tetrachloro-1a,2,3,4-tetrahydro-1H-azirino

The mass spectrometric behaviour of three 1a,3-diaryl-1,1,4, 4-tetrachloro-1a,2,3,4-tetrahydro-1H-azirino-[2,1-e][1, 6]benzothiazocines has been studied with the aid of mass-analyzed ion kinetic energy spectro--metry and exact mass measurements under electron impact ionization. All compounds show a tendency-to eliminate a neutral substituted/unsubstituted 1,1-dichlorostyrene from the eight-membered thiazocine ring to yield azirino[2,1-c][1, 4]benzothiazine ions, which can then lose a chlorine radical and a sulfur-atom to form azirino[2,1-c][1,4]benzothiazine ions and dihydroquinoline ions. All compounds could also eliminate a chlorine radical plus hydrogen chloride, stepwise or consecutively, to undergo a ring enlargement rearrangement to produce nine-membered 1, 7-benzothiazonine ions. The [M(+) - Cl] ions could undergo a four-membered ring rearrangement to yield 1,5-benzothiazepine ions by loss of 1,1-dichlorostyrene or styrene ions and HCl, and undergo alpha, alpha-cleavage to form benzothiazine ions by loss of arylchlorocyclopropane. The [M(+) - Cl - HCl] ions could further eliminate some atoms, small molecules or molecular fragments to form some abundant fragment ions. Copyright 1999 John Wiley & Sons, Ltd.

Increased expression of a plant actin gene during a biotrophic interaction between round-leaved mallow, Malva pusilla, and Colletotrichum gloeosporioides f. sp. malvae.

Two actin genes, actA from the hemibiotrophic anthracnose fungus, Colletotrichum gloeosporioides (Penz.) Penz. & Sacc. f. sp. malvae, and act1 from its host, Malva pusilla (Sm.) were cloned from a cDNA library developed from infected host tissue. The actin gene, actA, of C. gloeosporioides f. sp. malvae, which is similar to that of other euascomycetes, appears to be expressed constitutively. The actin gene of M. pusilla is most similar to one of the actin genes of Arabidopsis thaliana that is unique in being responsive to environmental stimuli such as wounding. Expression of actA was used to follow the growth of the fungus in the plant tissue. Low actA expression occurred until 72-96 h after inoculation and then increased rapidly, corresponding with the timing of the shift from slower biotrophic fungal growth to much more rapid necrotrophic growth. In contrast, expression of act1 approximately doubled during the biotrophic phase and then rapidly declined during the necrotrophic phase. Increased host actin expression could be due to host cytoskeleton rearrangement in response to biotrophic infection, and the subsequent decrease in host actin expression could be due to host cell disruption resulting from tissue maceration during necrosis. This is the first report of a host actin gene that can increase in expression during a compatible plant-pathogen interaction.

Induction of cytokine production in naive CD4(+) T cells by antigen-presenting murine liver sinusoidal endothelial cells but failure to induce differentiation toward Th1 cells.

BACKGROUND & AIMS: Murine liver sinusoidal endothelial cells (LSECs) constitutively express accessory molecules and can present antigen to memory Th1 CD4(+) T cells. Using a T-cell receptor transgenic mouse line, we addressed the question whether LSECs can prime naive CD4(+) T cells. METHODS: Purified LSECs were investigated for their ability to induce activation and differentiation of naive CD4(+) T cells in comparison with bone marrow-derived antigen-presenting cells and macrovascular endothelial cells. Activation of T cells was determined by cytokine production. LSECs were further studied for expression of interleukin (IL)-12 by reverse-transcription polymerase chain reaction, and the unique phenotype of LSECs was determined by flow cytometry. RESULTS: We provide evidence that antigen-presenting LSECs can activate naive CD62Lhigh CD4(+) T cells. Activation of naive CD4(+) T cells by LSECs occurred in the absence of IL-12. In contrast, macrovascular endothelial cells from aorta could not activate naive CD4(+) T cells. The unique functional characteristics of microvascular LSECs together with a unique phenotype (CD4(+), CD11b+, CD11c+, CD80(+), CD86(+)) make these cells different from macrovascular endothelial cells. Furthermore, LSECs did not require in vitro maturation to activate naive CD4(+) T cells. Most importantly, LSECs failed to induce differentiation toward Th1 cells, whereas conventional antigen-presenting cell populations induced a Th1 phenotype in activated CD4(+) T cells. Upon restimulation, CD4(+) T cells, which were primed by antigen-presenting LSECs, expressed interferon gamma, IL-4, and IL-10, which is consistent with a Th0 phenotype. Exogenous cytokines (IL-1beta, IL-12, or IL-18) present during T-cell priming by antigen-presenting LSECs could not induce a Th1 phenotype, but neutralization of endogenously produced IL-4 during T-cell priming led to a reduced expression of IL-4 and IL-10 by CD4(+) T cells upon restimulation. The addition of spleen cells to cocultures of LSECs and naive CD4(+) T cells during T-cell priming led to differentiation of T cells toward a Th1 phenotype. CONCLUSIONS: The ability of antigen-presenting LSECs to induce cytokine expression in naive CD4(+) T cells and their failure to induce differentiation toward a Th1 phenotype may contribute to the unique hepatic microenvironment that is known to promote tolerance.

Characterization of petroleum-degrading bacteria from oil-contaminated sites in Vietnam.

Four petroleum-degrading bacterial strains, 2TN-NB, 6TBX-CL, MVK2-5, and XCK, were isolated from various oil-contaminated sites in Vietnam. Determination of the nucleotide sequence of the gene encoding 16S rRNA allowed 2TN-NB to be identified as Acinetobacter sp. and the other three stains as Pseudomonas sp. Among the four isolates, 2TN-NB was most effective in degrading crude oil: in 1 d, it degraded 95% of the crude oil in the culture medium (5%, v/v). The isolated strains could also degrade a sulfur-containing aromatic hydrocarbon, dibenzothiophene (DBT), with low efficiency. Except for MVK2-5, which degraded crude oil least efficiently, the isolates produced biosurfactants in amounts sufficient for structural analysis. FT-IR measurement suggested that strains 6TBX-CL and XCK produced glycolipid-type biosurfactants while that produced by 2TN-NB was of the polysaccharide type.
