The relationship between biochemical changes in bronchoalveolar lavage fluid (BALF), serum and the lung of different dustexposed rats was studied. Wistar rats were divided into 5 groups: 1. Xingkong chrysotile asbestos (CH-As); 2. Dust in a sieve selection workshop of Xingkong asbestos mine (Dust-Wo); 3. Silica group (SiO2); 4. Titanium dioxide (TiO2) and 5. Normal control group (Control). All the rats were killed in three months after experiment. The results showed that the level of alveolar macrophages (AM), lactic dehydrogenase (LDH) and acid phosphatase (AcP) in each group was marked by related to collagen, lung fat, ceruloplasmin (Cp) and hydroxyproline (HoP) by r and t-test. Among the LDH from BALF, culture fluid and serum, there was also a marked relationship. So the authors pointed out that the BALF especially AM and LDH test could serve as a good and valuable index for detection the condition of pneumoconiosis.
A mass survey was conducted to investigate the association between cervical carcinoma and human papilloma virus (HPV) infection in a high-risk area, Xiangyuan County, Shanxi Province. Fifty-four cases of cervical cancer in situ (CIS), including severe dysplasia (CIN III), 14 cases of invasive cervical carcinoma, 28 cases of mild cervical dysplasia (CIN I), and 13 cases of moderate cervical dysplasia (CIN II) were identified among 6710 women examined. One hundred and sixty-nine punch biopsies from abnormal cervix and genital tract were examined for the presence of HPV 6B/11, 16 and 18 DNA sequences by dot blot hybridization: The positive rates of HPV infection in cervicitis, CIN I, II, III, CIS and invasive cervical carcinoma specimens were 35.06% (27/77), 25% (2/8), 33.33% (2/6), 40% (2/5), 70.58% (24/34) and 40% (4/10), respectively. Among these groups, there was no regular distribution of HPV types except for HPV 16, the positive rate of which was increased from 25% in CIN I to 46.15% in CIS: HPV 16 was also the only type seen in the invasive cervical cancer group. Distribution analysis of HPV types in HPV-positive cervical cancer tissues uncovered an HPV 16 positive rate of 59.4%; 3.7 and 2.4 times higher than those of HPV 6B/11 and HPV 18, respectively. These data confirm the close association between cervical cancer and HPV 16 infection.
The lymph drainage of lower lip mucosa was studied in 30 fresh cadavers of fetus and neonatus by submucosa injection of 30% Prussian blue of chloroform solution. The collecting lymphatics in lateral third of both the right and left sides of the lower lip mucosa ran laterally to the angles of the mouth, and then back- and downward between mandible and depressor muscle of angle of mouth, and finally, accompanying the facial vessels, emptied into submandibular lymph nodes. The efferent lymphatics of the latter emptied into angular lymph nodes and jugulo-omohyoid lymph nodes. The collecting lymphatics in the middle third of the lower lip mucosa went superficially to the mental muscle and downward into submental lymph nodes. The efferent lymphatic of the latter emptied, anteriorly to the body of the hyoid bone, latter- and downward into right and left jugulo-omohyoid lymph nodes. All collecting lymphatics lie in the loose connective tissue between the muscles.

The virA and virG gene products are required for the regulation of the vir regulon on the tumor-inducing (Ti) plasmid of Agrobacterium tumefaciens. VirA is a membrane-associated protein which is homologous to the sensor molecules of other two-component regulatory systems. We overproduced truncated VirA proteins in Escherichia coli by deleting different lengths of the 5'-coding region of the virA gene and placing these genes under lacZ control. These proteins were purified from polyacrylamide gels and renatured. The renatured proteins became radiolabeled when they were incubated with [gamma-32P]ATP but not with [gamma-32P]GTP or [alpha-32P]ATP, which suggests an ATP gamma-phosphate-specific autophosphorylation. The smallest VirA protein, which retained only the C-terminal half of the protein, gave the strongest autophosphorylation signal, which demonstrates that the C-terminal domain has the autophosphorylation site. The phosphorylated amino acid was identified as phosphohistidine, and a highly conserved histidine was found in all of the VirA homologs. When this histidine was changed to glutamine, which cannot be phosphorylated, the resulting VirA protein lost both its ability to autophosphorylate and its biological function as a vir gene regulator. Results of this study indicate that VirA autophosphorylation is required for the induction of the vir regulon and subsequent tumor induction on plants by A. tumefaciens.
Thirty-eight cervical carcinomas and 1 cervical condyloma were examined for the presence of human papillomavirus (HPV) type 16 DNA using 3H-dCTP-labelled HPV-16 DNA probe in paraffin section in situ hybridization and 32P-dCTP-labelled probe in dot blot hybridization (Tm-17). The results showed an HPV-16 DNA positive rate in our series of about 74.4% (29/39) as determined by paraffin section in situ hybridization and about 71.8% (28/29) by dot blot hybridization. There was no significant difference between the two methods. We thus confirmed that paraffin section in situ hybridization is an informative, reliable, and sensitive method for the diagnosis of cervical HPV infection.
In this paper, a track self-tuning control system for fed-batch penicillin fermentation was constructed by using the adaptive control theory. This system, unlike normal control methods can adapt itself to the random change of controller parameters during penicillin fermentation. The order of the system control SISO model is two fold and it is a non-minimum phase system. Therefore, the extended least square self-tuning control strategy was employed. The imitation showed that the biomass growth was controlled by a reference trajectory using this system.
Dipfluzine (Dip) is a new derivative of cinnarizine (Cin) first developed by Department of Chemistry, Beijing University. Dip showed a dose-dependently inhibitory effect on both KCl- and NE-induced contraction in the rabbit aortic rings. It was more effective in suppressing the contractile response evoked by KCl than that by NE. Dip also inhibited the KCl-induced contraction in porcine basilar, coronary and radial arteries. Their pD2' values were 5.7 +/- 0.6, 5.4 +/- 0.4 and 4.6 +/- 0.5 respectively. The selectivity of Dip for vasodilation was proved by higher pD2' value of the basilar artery than that of the coronary and radial arteries, and this selectivity of Dip was more significant than that of Cin. The acute iv LD50 of Dip and Cin in mice were 37 and 36 mg/kg, respectively.
A mathematical model with noises which is suitable for penicillin fed-batch fermentation in factory has been presented. By taking the carbon dioxide production rate as an online measured variable, an extended Kalman filter (EKF) was used for the real-time identification of the state and parameters. The study offers a basis for the adaptive optimal control of fermentation processes. It is shown that the estimated values of state by EKF coincide with the experimental data. The results prove that the filter is a better online observer for the state of fermentation process and dynamical parameters.

This paper addresses the question: in Graves' disease is there a thyroid-growth stimulating IgG (TGI) separate from thyroid-stimulating antibody (TSAb)? Using the functioning rat thyroid line (FRTL5) cells for TGI (incorporation of [3H]-thymidine into DNA) and TSAb (increase in cAMP concentration) assays, we tested IgG from 30 Graves' patients. Positive TGI assay occurred only if cAMP increased in the cells and responses correlated, i.e., r = 0.95, P less than 0.001. With one very potent TSAb-IgG we showed that Fab was active as TGI and TSAb, IgG with pI of 8.5-9.0 was the most potent fraction in both systems and an inhibitory IgG prevented the action of both TSAb-IgG and TSH in both the TSAb and TGI assays. In the last example, the action was on the cell membrane and not on the TSH or IgG. These data are entirely compatible with the view that in Graves' disease, at least as tested in FRTL5 cells, the same IgG is active in stimulating both growth and adenylate cyclase.
Thyroid function, including growth, is TSH dependent, and most metabolic functions of TSH are thought to be mediated by cAMP. Recently, it has been suggested by several groups that growth may be an exception and that it may not be related to cAMP action. In addition, evidence has accrued indicating that the thyroid-stimulating antibody (TSAb) of Graves' disease, the metabolic actions of which are also cAMP mediated, may not be the goitrogenic agent in that syndrome. To evaluate these concepts, we used functioning rat thyroid cells (FRTL5) in monolayer culture and, as indices of growth, the incorporation of [3H]thymidine ([3H]Tdr) into DNA, the concentration of DNA measured directly, and the percentage of cells in S phase, as assessed by flow cytometry, all studied over 72 h of incubation. TSH, forskolin, and cholera toxin enhanced growth by each criterion and increased the concentration of cAMP in parallel; the effect on cAMP occurred rapidly and was maximal well in advance of influences on growth. In all instances, measures of growth promotion were minimal at 24 h and maximal at 48 h, except for [3H]Tdr incorporation, which was greater at 72 h than at 48 h. 3-Isobutyl-1-methylxanthine (IBMX) and (Bu)2 cAMP were also tested. Both enhanced all indices of growth and were as effective as TSH. Maximal responses to TSH were obtained at 100-200 microU/ml, maximal responses to both IBMX and (Bu)2cAMP occurred at 5 X 10(-4) M, and all three stimulators increased the DNA concentration and [3H]Tdr uptake and induced S phase in at least 20% of all cells in culture. The peak effect on DNA and S phase was consistently at 48 h. Epidermal growth factor (EGF) was shown to increase [3H]Tdr incorporation in a nondose-dependent fashion (10(-10) to 5 X 10(-9) M gave approximately 250% of control) over 1, 2, 3, 5, and 7 days, with no increase in DNA and a slight decrement in the concentration of cAMP. A laboratory standard TSAb-immunoglobulin G was shown to parallel TSH in both increasing cAMP (over 2 h of incubation) and growth stimulation (over 72 h). The data are entirely consistent with the view that TSH-stimulated thyroid growth is mediated by cAMP and that the established action of TSAb on adenylate cyclase is sufficient to explain goiter as well as hyperthyroidism in Graves' disease.
Mutagenicity of fasting gastric juice from 108 chronic gastritis patients in high- and low-risk areas of stomach cancer was assayed using in vitro SCE test in cultured human lymphocytes and micronucleus test in cultured Chinese hamster V79 cell. The results showed that mutagenicity could be found in this gastric juice. The positive rate of these two tests, SCE/cell, micronucleated cell rate and micronucleus rate were significantly higher in high-risk area than in low-risk area (P less than 0.01). There was a tendency that the mutagenic activity was higher in high pH than in low pH gastric juice and in males than in females. In high-risk area, the mutagenicity of gastric juice was obviously higher in younger than in elder aged group. Because no S9 was added in this test system, the results seem to suggest that the mutagenicity found in the gastric juice of patients with chronic gastritis might be, at least in part, related to direct-acting N-nitroso-compounds synthesized in the stomach.
