Site-specific mutagenesis of Rhodobacter capsulatus ferredoxin I, FdxN, that functions in nitrogen fixation. Role of extra residues.

One of the two [4Fe-4S]-type clusters of the Rhodobacter capsulatus ferredoxin I, FdxN, was modified through site-specific mutagenesis of the distinctive features of the second cluster-binding motif, Cys38-X2-Cys41-X8-Cys50-X3-Cys54-X4-Cys59. First, various mutagenized products were tested to learn whether they could rescue the decreased capacity of an fdxN-null strain MSA1 to fix nitrogen: the phenotype of MSA1 was reassessed to Nifs (slow growth by nitrogen fixation) from our previous description of Nif- (Saeki, K., Suetsugu, Y., Tokuda, K., Miyatake, Y., Young, D. A., Marrs, B. L. and Matsubara, H. (1991) J. Biol. Chem. 266, 12889-12895). Substitution of Cys59 to Ser yielded an almost fully active product, while that of Cys54 did not. Gradual deletions and deletion-substitution of the 8 residues between Cys41 and Cys50 also yielded active products. Second, three of the modified FdxN proteins were subjected to purification. Only the GA protein, whose 8 residues between positions 42 and 49 were replaced by the Gly-Ala sequence, was purified. The GA protein and the authentic FdxN showed similar optical properties. The two clusters in the former had Em values of -490 and -430 mV, while those in the latter had an identical value of -490 mV, when determined by EPR analysis. It was concluded that: 1) Cys59 is not a ligand to [4Fe-4S] clusters but is important for structural integrity, 2) the residues between positions 42 and 49 may form a "loop-out" from a structure analogous to the Peptococcus aerogenes ferredoxin, and 3) the loop-out region does not have functional significance in nitrogen fixation but may be responsible for maintaining the highly negative redox potential of one of the two clusters.

Ductal Carcinoma In Situ of the Breast: Is Breast Conserving Treatment Feasible?

Between 1968 and 1993, 43 cases of ductal carcinoma in situ of the breast (DCIS) were treated. We examined the extent of the cancer using multiple sections of surgical specimens and considered whether or not breast conserving treatment (BCT) is feasible for treatment of DCIS. In nine out of 40 patients (23%), extent of the cancer was classified as grade III, defined as extension over a 2.5-cm margin around the tumor. Among those nine cases, 67% (6/9) had extensive microcalcification on mammography. With respect to the correlation between the tumor size and the extent of the cancer, 29%(5/17) of the cases with a small size tumor, ie, 1.0 cm or less, showed grade III or greater. All tumors with a size of 1.1-2.0 cm showed grade II or lower, and 50%(4/8) of cases with a tumor size of 2.1 cm or more showed grade III or greater. Concerning the relationship between the histological subtype and the extent of the cancer, 22%(2/9) of comedo carcinomas and 23%(7/30) of noncomedo carcinomas showed grade III or greater. The histological extent thus showed no difference between comedo carcinoma and noncomedo carcinoma. It follows that, when BCT performed on DCIS consists of lumpectomy alone, BCT is feasible when the tumor size is 1.1-2.0 cm. without extensive microcalcification on mammography. However, BCT for comedo carcinoma should be approached with caution because of its malignant behavior, although there was no difference in histological extent between comedo and noncomedo carcinoma.

Electrostatic environment of the tryptophylquinone cofactor in methylamine dehydrogenase: evidence from resonance Raman spectroscopy of model compounds.

Methylamine dehydrogenase (MADH) utilizes its endogenous tryptophan tryptophylquinone (TTQ) as a cofactor in enzymatic catalysis, with the C6 carbonyl of the quinone implicated as the site of attack by substrates and other nucleophiles. Resonance Raman (RR) spectroscopy provides an ideal method for investigating the state of this carbonyl group whose C==O stretch is distinct from other vibrational modes of the cofactor and is readily identified by its shift to lower energy in H218O. In a series of indole 6,7-quinone models for TTQ, the in-phase stretching vibration of the two C==O groups occurs at 1650 cm-1 in nonpolar solvents and shifts to 1638 cm-1 in H2O. The absorption maximum undergoes an analogous shift from 400 to 425 mm. The spectral properties of the indole quinones in H2O approach the corresponding values in Thiobacillus versutus MADH (C==O stretch at 1612 cm-1, lamdamax at 440mm) and are indicative of strongly hydrogen bonding of the C==O and NH groups of the cofactor in the native enzyme. Addition of monovalent cations [NH4+,Cs+, and (CH3)3NH+] to MADH causes further increases in the lamdamax and decreases in the frequency of the C==O stretch[1590 cm-1 with (CH3)3NH+]. This implies a strong electrostatic interaction between monovalent cations and a carbonyl oxygen (most likely at C6) in TTQ. The fact that these cations behave as competitive inhibitors of the methylamine substrate suggests that methylamine binds to the same location in the enzyme prior to its covalent reaction with the cofactor. Addition of monovalent cations to the one-electron-reduced semiquinone form MADH results in RR spectral shifts for a number of vibrational modes of the cofactor. Thus, the ability of monovalent cations to promote and stabilize the formation of the semiquinone intermediate is also due to their direct electrostatic interaction with the TTQ cofactor.

Micronucleus induction by chromium and selenium, and suppression by metallothionein inducer.

The clastogenic effects of chromium compounds (CrCl3 and K2CrO4) and selenium compounds (H2SeO3 and Na2SeO4) in mouse bone marrow cells have been investigated. K2CrO4 induced significant, dose-related increases in micronuclei. H2SeO3 also showed a significant micronucleus induction at the highest dose. CrCl3 and Na2SeO4 were negative for micronuclei induction in mice. The suppressive effect of Bi(NO3)3, a metallothionein inducer, on the micronucleus induction by K2CrO4 and H2SeO3 has been also investigated. Pretreatment with Bi(NO3)3 suppressed the micronucleus induction by K2CrO4 and H2SeO3. In addition, the incidence of micronucleus induction seemed to be reduced by double dosing with K2CrO4 and H2SeO3, compared to single dosing. It is generally recognized that a number of metal compounds induce metallothionein synthesis. These results suggest that the mechanism of suppression of micronucleus induction by K2CrO4 and H2SeO3 involve possible participation of metallothionein in mouse bone marrow.

Interleukin 1 (IL-1) production is not essential for acquired resistance of human A375 melanoma cells to anti-proliferative effect of IL-1.

The proliferation of human melanoma cell line A375-6 is inhibited by interleukin l (IL-l). However, the cells acquired resistance to IL-l after a long period of culture. We have reported that 2 resistant subclones, A375-R8 and -R19, produced IL-l alpha constitutively and that IL-l induced IL-6 production in an autocrine manner. Therefore, we supposed that IL-l alpha production renders the cells resistant to IL-l. To investigate the relationship between IL-l alpha production and IL-l resistance, we transfected the IL-l alpha expression plasmid to the IL-l-sensitive clone, A375-6, and the anti-sense mRNA expression plasmid to IL-l-resistant cells, A375-R8 and -R19. A375-6MS, a transfectant of mature IL-l alpha expression plasmid, expressed IL-l alpha mRNA and produced IL-l activity at a level comparable to the resistant cells. The transfectant also produced IL-6 and exhibited augmented expression of Mn-SOD mRNA. However, IL-l sensitivity of this transfectant was not affected. With respect to sensitivity to anti-proliferative effects of other cytokines, such as IL-6 and TNF alpha, there was no difference between the transfectant and parent cells. Although A375-R8PH10 and -R19PH10, transfectants of IL-l alpha anti-sense mRNA expression plasmid, exhibited a decrease in the level of IL-l production, their IL-l sensitivity did not differ from parent cells. These results, therefore, suggest that IL-l alpha production is not essential or sufficient for the acquisition of resistance to the anti-proliferative effect of IL-l.

Effects of new quinolone antibacterial agents on mammalian chromosomes.

The clastogenic effects of several new quinolones (ciprofloxacin, enoxacin, levofloxacin, nalidixic acid, ofloxacin, pipemidic acid, and N1-cyclopropyl quinolones for drug candidate) were studied cytogenetically using Chinese hamster lung cells (CHL) and the mouse micronucleus test. Some N1-cyclopropyl quinolones strongly induced chromosomal aberration on CHL cells, and some, but not all, were also capable of inducing micronuclei in mouse bone marrow cells. Levofloxacin showed weak clastogenicity in CHL cells but did not induce either micronuclei in mouse bone marrow or unscheduled DNA synthesis (UDS) in rat hepatocytes when administered to intact live animals. The lack of concordance between in vitro and in vivo assays could reflect the differences in the tissue levels of the drugs and the in vitro conditions.

Experimental study on nerve regeneration through the basement membrane tubes of the nerve, muscle, and artery.

We evaluated neurotization after transplantation with lyophilized nerves, muscles, and arteries, and examined the possibility of practical application of long bridging grafts. Grafts of 10 mm and 25 mm of lyophilized nerves, muscles, and arteries harvested from Fisher rats were transplanted to the sciatic nerves of recipient Lewis rats. The histological changes undergone by short grafts were observed at weekly intervals. The sham-operated and isograft groups were used to compare the results of long grafts. In both the nerve and muscle-graft group, regenerated axons grew out through the residual basement membrane tube. But in the muscle graft group, phagocytosis of myofibril debris took longer than that of degenerated axons. No statistical differences were found between results of TSI, induced EMG, and quantitative analysis of myelinated axons in the nerve and muscle graft groups. No neurotization was noted in the long artery graft. In long grafts, laminin found on the basement membrane may not be sufficient to accelerate neurotization, and arteries should not be used for tubulization.

Sensitization interval and administration method alter the effect of 15-deoxyspergualin on heart transplantation in sensitized recipient rats.

We evaluated the effect of 15-deoxyspergualin (DSG) on accelerated rejection. Brown Norway rats (BN) served as organ donors and Lewis rats (LEW) as recipients. In an accelerated rejection model, after a LEW rat was sensitized with BN skin, a BN heart was transplanted. Various intervals between sensitization and heart transplantation were examined. The heart allografts in sensitized recipients were rejected earlier than those in unmodified recipients regardless of the sensitization interval. DSG (2.5 mg/kg per day), given to the recipients during the sensitization phase, significantly prolonged graft survival compared with the untreated hosts when the sensitization interval was short. When the recipients were treated with DSG after heart transplantation, heart graft survival was significantly prolonged regardless of the sensitization interval. Flow cytometric analysis and complement-dependent cytotoxicity tests revealed that DSG suppressed antidonor antibody formation and the postoperative administration of DSG significantly decreased the proliferation of B cells when the sensitization interval was short and the proliferation of class II antigen-positive cells when the sensitization interval was long.

Intracaval endovascular ultrasonography for preoperative assessment of retrohepatic inferior vena cava infiltration by malignant hepatic tumors.

The accurate diagnosis of inferior vena cava infiltration is important not only for preoperative staging but also for performing surgery for hepatic tumor involving the inferior vena cava (IVC). We investigated the diagnostic value of intracaval endovascular ultrasonography (ICEUS) in the diagnosis of invasion of the IVC by a prospective study of 30 consecutive patients with hepatic tumors with possible invasion of the IVC. The procedure was performed using an 8F, 20-MHz intravascular ultrasonography (US) from the right femoral vein. Twenty-nine of the 30 patients underwent surgery, and the tumor was resected in 25 cases, including 3 cases of combined resection of the IVC. The diagnostic criterion for IVC infiltration with ICEUS was obliteration of a single echogenic layer of the IVC wall or intracaval tumor mass. The results of ICEUS were compared with those of computed tomography (CT), cavography, conventional US, and intraoperative US. Vascular invasion was identified in 7 of 30 cases and was subsequently confirmed by pathological examination of four resected specimens, including one autopsy and two operative findings. The sensitivity, specificity, and overall accuracy of ICEUS for the diagnosis of the IVC invasion was 100%, 95.8%, and 96.7%, respectively, against 85.7%, 60.8%, and 66.7% for CT, and 71.4%, 73.9%, and 73.3%, respectively, for cavography. The sensitivity, specificity, and overall accuracy of conventional US was 87.5%, 59%, and 66.7%, respectively; those of intraoperative US were 100%, 81.8%, and 86.2%, respectively. ICEUS is a useful technique that can precisely evaluate the IVC for possible hepatic tumor invasion.

Application of primary hepatocytes from p53-knockout mice for studies of expression of Cyp3a.

CYP3A rapidly disappears in primary hepatocytes, although the primary cells are suitable for studies of the regulation of CYP3A genes. In the present study, we found that Cyp3a mRNA could be expressed in the primary hepatocytes from p53-knockout mice for at least 2 weeks when the cells were cultured in the presence of dexamethasone. Propoxycoumarin O-depropylase activity, which is known to be mainly catalyzed by CYP3A, was maintained at a level of 50% of the initial activity even after 5 days of culture, and the activity correlated with the expression level of Cyp3a mRNA in the primary hepatocytes from p53-knockout mice. The cells remained morphologically intact during 4 weeks. These results suggest that hepatocytes from p53-knockout mice are a useful tool for studies of the expression of Cyp3a.

Cerebral energy metabolism in insulin induced hypoglycemia in newborn piglets: in vivo 31P-nuclear magnetic resonance spectroscopy.

The effect of insulin induced hypoglycemia on cerebral energy metabolism was examined in four newborn piglets. Cerebral energy metabolism was assessed using in vivo 31P-nuclear magnetic resonance spectroscopy. It was demonstrated that the normal level of phosphocreatine/inorganic phosphate (PCr/Pi), an indicator of phosphorylation potential, was maintained at a blood glucose level of 40 mg/dL or above, whereas when blood glucose was reduced to less than 40 mg/dL, PCr/Pi rapidly decreased in parallel with this. Below the critical blood glucose level of 40 mg/dL, a positive correlation (y = 0.02x + 0.632; r = 0.668; P < 0.001) existed between blood glucose and PCr/Pi. In the present investigation, a reduction of blood glucose level to 20 mg/dL or lower resulted in a PCr/Pi of less than 1, indicating a state of cerebral energy failure. The intracellular pH (pHi) was 7.08 +/- 0.05 at the onset and 7.15 +/- 0.07 in the hypoglycemic state, indicating no significant difference between the two groups. The present study has clarified that cerebral energy failure occurs when the blood glucose level is about 20 mg/dL or lower. The critical point of blood glucose exists to maintain brain energy metabolism.
