Diazepines. 5. Synthesis and biological action of 6-phenyl-4H-pyrrolo[1,2-a][1,4]benzodiazepines.

A series of 6-phenyl-4H-pyrrolo[1,2-a][1,4]benzodiazepines (2) has been prepared with 2-phthalimidomethylfurans (12) and 1-phthalimidoalkane-2,5-diones (15) or 2,5-dimethoxy-2-phthalimidomethyltetrahydrofurans (16) as the key intermediates and subsequently evaluated for CNS activity. The structure-activity data generated indicate that, in general, introduction of the methyl and/or ethyl group(s) in the pyrrole ring and a chlorine atom at the ortho position of the 6-phenyl group increases the activity and that substitution of the above chlorine atom for a fluorine atom decreases the activity. 8-Chloro-6-(2-chlorophenyl)-1,3-dimethyl-4H-pyrrolo[,2-a][1,4]benzodiazepine (2p), the most potent among the compounds synthesized, was equipotent in taming and sedative activities to diazepam. The acute LD50 of 2p in mice was larger than 3000 mg/kg po.

[Biological activity of 1alpha-hydroxycholecalciferol (1). Effect on intestine and bone in rats (author's transl)].

Biological activity of 1alpha-hydroxycholecalciferol was studied in rats. 1alpha-Hydroxycholecalciferol was found to be more potent and rapidly active than vitamin D in stimulating intestinal calcium transport and calcium mobilization from bone both in normal and vitamin D deficient rats. 1alpha-Hydroxycholecalciferol was also active in nephrectomized and/or thyroparathyroidectomized rats both in intestine and bone. Although it is well known that 1alpha-hydroxycholecalciferol is metabolized to 1alpha, 25-dihydroxycholecalciferol in the liver, there is the possibility that the former is active without further metabolism. In rats in which hepatitis was induced by CCl4, 1alpha-hydroxycholecalciferol was active both in the intestine and in the bone, while it was inactive in hepatectomized rats. These data clearly demonstrate that 1alpha-hydroxycholecalciferol is not active by itself and must be metabolized in the liver. This idea also shows the lag time in response of rats to 1alpha-hydroxycholecalciferol has more potent antirachitic activity than vitamin D and does not lose its activity with chronic oral administration. In view of these findings, 1alpha-hydroxycholecalciferol appears to have a good potential for clinical application in cases of renal failure and metabolic bone diseases.

Reconstruction of intact gamma-globulin from S-sulfonated gamma-globulin in vivo.

The conversion of the S-surfonate group in a peptide to a disulfide bond has been observed in vitro. This paper reports that the conversion of this group in human S-sulfonated gamma-globulin (S-GG) appeared to occur in vivo judging from the change in molecular weight of S-GG observed in the presence of sodium dodecyl sulfate (SDS) and the restoration of hemolytic activity.

Therapeutic plasmapheresis for liver failure after hepatectomy.

Therapeutic plasmapheresis (PP) for liver failure following the resection of primary liver cancer was evaluated retrospectively. PP was repeated two to three times per week using plasma exchange (PE) with fresh frozen plasma as a substitution fluid in all eight cases and direct hemoperfusion (DHP) in two cases. PE effectively decreased serum bilirubin and/or improved hepatic encephalopathy in six of eight patients and prolonged survival time (p less than 0.05, chi-square test). DHP was not effective. Liver failure following the resection of damaged liver differs from acute liver failure in both the condition itself and the reaction to PP. Fractionation of serum bilirubin using high performance liquid chromatography showed differences in the ratio of delta bilirubin in one survivor versus two who died and was effective in selecting a patient who would respond to PE. In conclusion, PE is effective in prolonging survival time after liver failure in hepatectomized patients compared to conventional therapy.

Innominate artery erosion complicating use of tracheal tube with adjustable flange.

A tracheal tube having an adjustable flange (AF tube) can be adjusted in cases of abnormal depth from skin to trachea. In a 43-year-old man with squamous cell carcinoma of the floor of the mouth, an AF tube was inserted because of severe edema of pretracheal tissues. He developed innominate artery erosion on the 6th postoperative day caused by the malfitted AF tube and died of recurrent hemorrhage 5 days after a surgical repair of the wound. The AF tube should be utilized not routinely but only in individuals with abnormal anatomy of the neck or trachea. Use of the AF tube requires extra care in addition to routine care required in the case of a conventional tracheal tube. Its correct fit should be verified initially by radiography. Fiberoptic examination should be employed within days after tracheostomy for patients with an AF tube to determine whether the tracheal mucosa is intact.

Effect of methionine-deprived nutrition on cell growth and cell kinetics in cell cultures and experimental tumors.

The effect of methionine-deprived nutrition on cell growth and cell kinetics was investigated in cell cultures and in tumor-bearing rats using the total parenteral nutrition (TPN) technique. A simultaneous flow cytometric measurement of the cellular DNA content and the amount of 5-bromodeoxyuridine incorporated into cellular DNA was performed for analysis of cell kinetics. The methionine-free medium demonstrated a cytocidal effect on the growth of SLC cells after 6 hours of culturing. It decreased viability from 80% in the control medium to 23%, and it decreased the S phase and increased the G0/G1 phase of the cell cycles. The methionine-deprived medium showed a concentration-dependent inhibition in cellular growth. Methionine-deprived TPN was seen to inhibit AH109A and SLC tumor growth compared with conventional TPN and decreased the S phase and increased the G0/G1 phase of cell cycles. These results confirm that methionine deprivation blocks cells from processing into the G1 phase and recycling, and that it is effective in inhibiting tumor growth in cultures and in vivo.

Some physiological characteristics of (SLN X C3H/He)F1 obese mice.

(SLN x C3H/He)F1 mice of both sexes showed significantly heavier body weights than the parental strains associated with the higher food intake and an accumulation of adipose tissue in the subcutaneous and the abdominal regions. Furthermore, serum levels of free fatty acid and growth hormone of F1 mice were higher and lower, respectively, than those of the parental strains. The results indicate that the obesity of these F1 mice is ascribed to excess food intake and metabolic disorders and they are promising as an animal model for the study of several diseases related to obesity.
