Study of estimation of color recognition on the dentist. On the ability of subjects to discriminate color in terms of hue, value and chroma.

The tooth crown color space model was manufactured and the improvement of the color representation and communication on the dental clinic have been trying. The study of color discrimination by dentist and student viewing with the use of tooth crown color chips reported here in basic knowledge on the color sensitivity of human eye. The test card were 55 x 90 mm in size and the color chip, 5 x 10 mm. The paired color chips were placed on the test card to have a 2 mm distance between them. Color difference of two tooth crown color chips on the test cards was 3.5 (delta E*ab), the combination of hue, value and chroma was constant of all cards. The subjects were 22 people, 6 dentists and 16 dental school students with normal color sensibility. In proportion of method of comparison for surface color (JIS Z 8723), the subject observed the test card which were different of hue, value and chroma on the their naked-eye, were investigated to evaluate their judgement of the test cards. The following results were obtained. The standard of judgment for the color discrimination was made firstly in terms of hue, secondly in terms of value, and thirdly in terms of chroma.

[Effects of epidermal growth factor (EGF) on prostaglandin E2 synthesis by cultured human endometrial cells].

Prostaglandins are considered to participate in the nidation of the embryo. However, so far little is known about the regulation of prostaglandin synthesis in endometrial tissue. In this study, we examined the effect of epidermal growth factor (EGF) on the release of PGE2 with primary cell culture of human endometrium. EGF stimulated PGE2 release dose-dependently in the range between 0.1 ng/ml and 100 ng/ml. PGE2 release was augmented significantly as early as 30 minutes after the addition of EGF, reaching a plateau at 3 hours. The stimulatory effect of EGF was eliminated by the concomitant addition of lipocortin, an inhibitor of phospholipase A2. However, the effect of EGF was not influenced by cycloheximide. Estradiol alone had a weak effect on PGE2 release. However, the stimulatory effect of EGF was more pronounced in cells pretreated with estradiol. In conclusion, this report offers evidence that EGF stimulates the release of PGE2 from human endometrial cells, thus implying the involvement of EGF in implantation.

Molecular analysis of T cell receptor and CD3 genes in CD3- large granular lymphocytes (LGLs): evidence for the existence of CD3- LGLs committed to the T cell lineage.

Seven patients with CD3- lymphoproliferative disorder of granular lymphocytes (LDGL) were analyzed for rearrangement of the T cell receptor (TCR) delta and alpha as well as gamma and beta genes. Among those patients six showed the germline configuration of all known rearranging TCR genes. Two analyzed patients of them lacked CD3-gamma transcripts and expressed nonfunctional TCR-beta transcripts. Meanwhile, TCR-delta gene rearrangement accompanied by expression of full-length TCR-delta transcripts was observed in one patient with CD3- LDGL. In addition, the cells from this patient transcribed the CD3-gamma gene which is one of the earliest events restricted to cells committed to the T cell lineage. These findings indicate that CD3- LGLs include not only cells belonging to the NK cell lineage but also precursor cells committed to the T cell lineage.

Metastatic renal tumor from the lung with regional lymph node involvement: a case report.

We report a case of metastatic renal tumor from lung cancer. A 53-year-old man presented with gross hematuria 2 years after treatment of the primary lesion. Investigations suggested a metastatic tumor in the right kidney from the lung which was accompanied with regional lymph node metastasis. Surgical treatment was not performed because of his poor condition and the lymph node involvement. Therefore, the combination chemotherapy of 5-fluorouracil, vincristine and doxorubicin, OK-432 was applied. Although this conservative management was effective, he died of progression 6 months later and an autopsy confirmed the diagnosis of metastatic renal tumor.

[A case of granulomatous orchitis in a hemodialysis patient].

A case of granulomatous orchitis in a 47-year-old patient is described. He had a history of recurrent pyelonephritis and subsequent chronic renal failure, and was under treatment with hemodialysis therapy. He presented with painful swelling of the right scrotal contents accompanied by fever. After the unsuccessful treatment with antibiotics, an exploration of the right scrotal contents revealed the testis replaced by a hard yellow mass. An orchiectomy was performed and granulomatous orchitis was diagnosed pathologically. The clinical and etiological aspects of this rare inflammatory disease of the testis are reviewed. It is important to recognize this disease, because this is often initially thought to be neoplastic. Although the cause of this disease is still controversial, it is supposed to be associated with sperm extravasation caused by urinary tract infection or trauma.

Analysis of molecular events in leukemic cells arrested at an early stage of T-cell differentiation.

We analyzed the rearrangement and expression of T-cell receptor (TCR) genes, including the recently identified TCR delta gene, in 21 patients with T-lineage leukemia/lymphoma. Among 8 patients with CD3-, CD4-, and CD8- (group I), 2 patients showed germline configuration of the TCR delta, gamma, beta, and alpha genes and 1 patient demonstrated only TCR delta gene rearrangement. All nine patients with CD3-, CD4+, and/or CD8+ (group II) showed concomitant rearrangements of the TCR delta, gamma, and beta genes. TCR alpha gene rearrangement was also observed in two patients. Three of four patients with CD3+ (group III) showed rearrangement of the TCR alpha gene with deletion of both alleles or of a single allele of the TCR delta gene. With Northern blot analysis, full-length transcripts of the TCR delta gene were detected in 3 of 15 examined patients. All were restricted to group I or group II. In contrast, full-length transcripts of TCR beta and alpha were observed mainly in samples from groups II and III. Based on these findings, rearrangement of the TCR delta gene may be the earliest event in T-cell differentiation, preceding rearrangements of the other TCR genes.

Demonstration of clonal selection in a child with non-Hodgkin's lymphoma.

A 2-year-old boy with B-lineage non-Hodgkin's lymphoma is described. He presented with growing skin tumors on his head, and biopsy specimens showed a malignant lymphoma of diffuse lymphoblastic type. Sixty-four percent of bone marrow cells were replaced with lymphoblasts, and they expressed B-lineage markers (CD19 and HLA/DR). Southern blot analysis demonstrated immunoglobulin heavy chain gene rearrangements (two rearranged and one germline) with the germline configuration of the T-cell receptor beta chain gene. Ten months later he relapsed with blasts of M5 morphologic type and a myeloid phenotype with the germline configuration of the immunoglobulin genes. During the next 2 months, myeloid blasts with immunoglobulin gene rearrangement which was identically rearranged with one of the two rearranged bands detected at diagnosis appeared. The most likely explanation for these findings is that initially the patient seemed to have at least two different clones of blasts, and clonal selections occurred during the treatments.

Low testosterone levels in diabetic men and animals: a possible role in testicular impotence.

Poorly controlled NOD spontaneously diabetic mice were proven to have significantly less plasma and testicular testosterone than well-controlled diabetic mice (489 +/- 15 ng/dl and 3.89 +/- 0.79 micrograms/100 g tissue, vs. 176 +/- 24 and 9.00 +/- 1.24, respectively), and these in turn had significantly less than NOD non-diabetic control mice. These data were consistent with our previous observation of a decrease in total plasma and testicular testosterone levels in streptozotocin diabetic rats. A greater difference between total plasma testosterone levels and free testosterone levels was found in streptozotocin diabetic rats (17 +/- 4 ng/dl vs. 91 +/- 7 ng/dl) than in control rats (660 +/- 141 vs. 352 +/- 77). Fat droplets, depicting blocked testosterone synthesis, were found in the testicular Leydig cells of streptozotocin diabetic rats and disappeared with insulin treatment. No difference was found among plasma total testosterone concentrations in people in different stages of diabetes, as had been previously reported. However, human diabetic males, free of complications but poorly controlled, had less free testosterone than those without complications but well controlled (18.0 +/- 2.0 pg/ml vs. 22.8 +/- 1.3), who in turn had significantly less than age-matched controls (25.3 +/- 1.1 pg/ml). These data suggest gonadal dysfunction in diabetes mellitus.

Poor prognosis of mediastinal non-Hodgkin's lymphoma with an immature phenotype of CD2+, CD7 (or CD5)+, CD3-, CD4-, and CD8-.

Nine children with mediastinal non-Hodgkin's lymphoma (NHL) were treated according to our new regimen which is characterized by intensified therapy with high-dose cytosine arabinoside (HDCA). After induction therapy with a combination of five drugs, such as vincristine, doxorubicin, cyclophosphamide, 1-asparaginase, and prednisolone, intermediate dosages of methotrexate (MTX) (1 g/m2) and HDCA (1.5 g/m2 x 12 doses) were administered. All but one patient (88.9%) achieved complete remission and then received this intensified therapy. With a median follow-up period of 25.5 months, five patients are still in complete remission, but three patients have relapsed. From the phenotypic point of view, these relapsed patients showed only very immature T-cell differentiation antigens such as CD2 and CD7 (or CD5). These results suggest that HDCA as intensified therapy for children with mediastinal NHL seems to be effective. However, for patients with an immature phenotype of T-lineage cells, more sophisticated regimens should be prepared.

In vivo effects by estrone sulfate on the central nervous system-senile dementia (Alzheimer's type).

Seven women with senile dementia-Alzheimer's type (SDAT) were treated with conjugated estrogen [main content: estrone sulfate (E1-S)], at a dose of 1.25 mg/day over a 6-week period. A New Screening Test for Dementia developed by Japanese National Institute of Mental Health (NS) and the scores of Hasegawa Scale for dementia (HS) were performed every 3 weeks. Six women showed improvements in NS (P less than 0.05) and 5 women showed improvements in HS. Untreated women with SDAT did not show any improvement. Serum E1-S was measured by a direct radioimmunoassay. Serum E1-S was 911 +/- 156 pg/ml in 7 women with SDAT and lower than that of 7 normal women (1020 +/- 216 pg/ml). Following the treatment, serum E1-S increased to a level of 21.1 +/- 8.1 ng/ml. Estrone and estradiol-17 beta also increased. The results suggest a possibility for the future clinical use of estrogen for senile dementia, after careful clinical research trials including the side effects.

Magnetocardiographic studies of ventricular repolarization in old inferior myocardial infarction.

Isomagnetic maps of 50 normal subjects (control group) and 23 patients with old inferior myocardial infarction (IMI group) were recorded in order to analyse T wave abnormalities in inferior myocardial infarction. The T wave of the magnetocardiogram (MCG) in the control group showed negative deflections in the left upper portion and positive deflections in the right lower portion, thus resulting in a T vector directed leftward and inferiorly. The T wave of the IMI group was flat or positive in the left upper portion and flat or negative in the right lower portion, suggesting a T vector directed superiorly. In addition, opposing dipoles were observed in 36.4% of the IMI group; i.e. one directed superiorly, presumably due to inferior myocardial ischaemia, and the other directed inferiorly due to normal repolarization. Localized T vector abnormalities could be detected by the MCG in some cases, in which coronary T waves of the standard electrocardiogram had returned to normal. Furthermore, multiple dipoles were more frequently observed in the isomagnetic map than in the isopotential map (5 vs. 15; P less than 0.01). These results suggest that the MCG is helpful in diagnosing myocardial ischaemia when this is not detectable on the electrocardiogram.

Clonal T-cell lymphoproliferation containing Epstein-Barr (EB) virus DNA in a patient with chronic active EB virus infection.

A 10-year-old boy with chronic active Epstein-Barr virus (EBV) infection developed T-cell lymphoproliferation in the terminal stage of hepatic failure. The phenotypes of the proliferating lymphocytes were CD3+, CD4+ and HLA-DR+. The genomic DNAs from these cells demonstrated two rearranged T-cell receptor beta-chain genes and contained the EBV genome. These findings indicate that EBV can infect T lymphocytes and might cause clonal T-cell lymphoproliferation.

CD3-negative lymphoproliferative disease of granular lymphocytes containing Epstein-Barr viral DNA.

Lymphoproliferative disease of granular lymphocytes (LDGL) is a heterogeneous disorder and the pathogenesis is likely to be complex. Some patients with chronic active EBV (CAEBV) infection also have LDGL. To investigate the relationship between EBV infection and the pathogenesis of LDGL, we conducted a survey for EBV DNA sequences by Southern blot analysis of DNA obtained from the peripheral blood of seven patients with LDGL, including one with CAEBV infection. Interestingly, EBV DNA was detected in the sample from the patient with CAEBV infection, and in the samples from four other patients with CD3-LDGL. Moreover, a single band for the joined termini of the EBV genome was demonstrated in two samples, suggesting a clonal disorder of those LDGL. These findings strongly suggest that EBV may play a pathogenic role in some cases of LDGL.

A chemical model of catechol-O-methyltransferase. Methylation of 3,4-dihydroxybenzaldehyde in aqueous solution.

The reaction of 3,4-dihydroxybenzaldehyde (LH2) and dimethylsulfate (DMS) to form the m- and p-O-methylated products (vanillin and isovanillin, respectively) in aqueous 2-(N-morpholino)ethane sulfonate buffer was studied kinetically. The products were determined by means of high-performance liquid chromatography. The O-methylation occurred principally at the p-hydroxyl group in the absence of divalent metal ions. In the presence of Cu(II), the m-methylation was promoted and became predominant. Zn(II) showed a similar but less pronounced effect. The effects were explained in terms of the complex formation of LH2. The second order rate constants for the m- and p-methylation of the species, LH2, CuL and CuL2-2 by DMS were calculated. The values and their ratio for the m-/p-reactions increased in the order of LH2 less than CuL less than CuL2-2. The reaction may serve as a chemical model for catechol-O-methyltransferase, which requires divalent metals and catalyzes the m-methylation.

The T wave abnormality in the magnetocardiogram.

The T wave changes in left ventricular hypertrophy and old inferior wall myocardial infarction were studied by the isomagnetic map to compare with those in the isopotential map. The isomagnetic map could detect the instantaneous current source, and the abnormal repolarization vector was more frequently observed in the isomagnetic map than in the isopotential map. The multiple dipoles were more frequently observed in the isomagnetic map, which has been difficult to detect by the isopotential map. The present study showed that repolarization in ventricular hypertrophy and myocardial ischemia is very complex and both normal and abnormal repolarization vectors are present, which might be opposite in direction. In some cases, these vectors were summated and only one dipole could be shown in the electrocardiogram or isopotential map. These results suggest that the magnetocardiogram is useful in detecting opposing current dipoles tangential to the heart.

HPLC quantitative analysis of plantaginin in Shazenso (Plantago asiatica L.) extracts and isolation of plantamajoside.

Quantitative analytical method by means of high-performance liquid chromatography (HPLC) has been developed for the quality control of Shazenso (Plantago asiatica L.) extracts. Plantaginin (I) which is one of the known constituents of Shazenso, was selected as a standard compound of the analysis. A chemically labile component has been isolated from the methanol extract and identified to be plantamajoside (II). It was found that II was converted into III even in hot water.

[Roles of epidermal growth factor (EGF) in the growth and differentiation of human endometrium].

Endometrial tissues undergo drastic changes during menstrual cycle. After menstruation, they proliferate and differentiate into cells with secretory activity in the preparation for egg implantation. Although sex steroids play an important role in the development of endometrial tissues, sequential events occurring in the endometrium can not be fully explained by the direct actions of sex steroids. In this study, we offer evidences that EGF is released from endometrial cells and they possess the receptor for EGF. These findings prompted us to explore the biological roles of EGF in endometrial tissues. Here we clearly demonstrate that EGF is involved in the proliferation of endometrial cells. Moreover, EGF is found to enhance both glycogenesis and glycogenolysis, thus increasing the supply of glucose for blastocysts. We further set forth that EGF augments the capacity of progestin receptor and release of prostaglandins in endometrial cells. In summary, this study emphasizes that EGF may participate in the development of human endometrial tissues in concert with sex steroids, thus contributing to the acquisition of receptivity of eggs in the endometrium.
