In the usual canine heart-lung preparation the coronary blood flow does not remain constant, but increases steadily ; usually, at the moment of the set up of the preparation the coronary blood flow was about 20 ml/min/100 g heart. However, in the course of 2 approximately 3 hours it becomes more than 150 ml/min/100 g heart and the responsiveness of the coronary artery is completely lost under this condition. Concurrently with the increase in the coronary flow, cardiac functions tend to deteriorate. In order to eliminate those experimental disadvantages, we added a donor dog to the heart-lung preparation. It was found that the support by the donor dog resulted in the complete improvement of the above-mentioned deterioration ; the coronary circulation (coronary blood flow and coronary artery and sinus blood oxygen saturation), cardiac function (cardiac output, right atrial pressure, heart rate and others) and cardiac metabolism (myocardial oxygen consumption, mycardial redox potential and others) were kept constant at a favorable state for more than 10 hours.

The prognosis of skin carcinoma and malignant melanoma was studied statistically. One hundred and one cases of basal cell epithelioma (BCE), 93 cases of squamous cell carcinoma (SCC), and 34 cases of malignant melanoma (MM) were studied. No cases of death due to basal cell epithelioma were found. The squamous cell carcinomas and malignant melanomas were classified according to the TNM system into three classes with different prognoses. The first class represents the initial stage, in which cure is expected if treatment is adequate. The second class is the intermediate stage, in which cure is uncertain and the prognosis depends upon the degree of extension. The third class is the late stage, in which distant metastasis is found and in which cure might not be anticipated. Furthermore, the prognosis of previously treated cases was poorer than that of untreated cases. Our findings indicate that early diagnosis and appropriate treatment in the initial stage results in the best prognosis and that the success or failure of the initial treatment has an appreciable influence on the prognosis of skin malignancy.

The question of whether the coronary blood vessels contain an intrinsic adrenergic mechanism for vasodilatation of physiological significance has been examined in the canine heart-lung preparation with a donor by studying the response of the coronary vessels to epinephrine, norepinephrine, isoproterenol and salbutamol in combination with practolol. To differentiate the vasodilatation mediated through adrenoceptors in the coronary vessels from that resulting from an increase in the myocardial O2 consumption, a special method of analysis was developed based on the linear relation between the coronary flow and the myocardial O2 consumption. It was found that all four compounds produced an increase in the coronary flow attributable to an increased myocardial O2 consumption. Epinephrine and norepinephrine produced a decrease in the coronary flow after practolol which completely abolished the increase in the myocardial O2 consumption as well as the positive inotropic and chronotropic effects produced by these compounds, while isoproterenol and salbutamol produced an increase. These results indicate that adrenergic beta-receptor exists in the coronary subserving a vasodilatation. However, the vasodilatation through this mechanism is of minor importance under physiological conditions and becomes completely masked in the presence of an overwhelmingly strong vasodilatation consequent to an increase in the myocardial O2 consumption.

Using the canine heart-lung preparation supported by a donor, the effects of ehtyl adenosine-5'-carboxylate (EAC) on the heart and coronary circulation were studied and compared with those of adenosine. EAC produced qualitatively similar effects to adenosine in this preparation. Aminophylline inhibited the effects of EAC as well as those of adenosine. Dipyridamole did not potentiate the effects of EAC, while it produced a definite potentiation of the effects of adenosine.
