The effects of ischemia and cardiac arrest by cardioplegia on the mechanical function and energy metabolism of the ventricular myocardium of the neonatal guinea pig were investigated in the isolated perfused heart preparation and compared with these effects in the adult guinea pig. Whereas reperfusion after ischemia resulted in better recovery of mechanical function and a higher adenosine triphosphase content in the neonatal myocardium than in the adult, recovery from cardiac arrest induced by St. Thomas' Hospital cardioplegic solution was not as good in the neonatal myocardium as in the adult. Contracture developed in the neonatal myocardium on administration of the cardioplegic solution, but did not in the adult. This was considered to be the reason that the protective effect of the cardioplegic solution was inferior in the neonatal myocardium to that in the adult.
We attempted in two types of preparations to delinate the difference in responses to St. Thomas' Hospital cardioplegic solution between the neonatal and the adult guinea pig myocardium. Isolated guinea pig hearts were perfused with Langendorff's method and the tension of the papillary muscle of the right ventricle was recorded. Continuous infusion of St. Thomas' Hospital cardioplegic solution (37 degrees C) for 30 minutes resulted in a significantly higher elevation of the resting tension (development of contracture) in the neonatal myocardium than in the adult. The recovery of normal contractile tension after the resumption of perfusion with normal Krebs-Ringer bicarbonate solution was smaller in the neonate with increases in the myocardial water and calcium contents. The membrane potential of the papillary muscle preparation was recorded by means of conventional glass microelectrodes. There was no significant difference in the control values of the resting membrane potential and in the degree of depolarization during exposure to St. Thomas' Hospital cardioplegic solution between the neonate and the adult. Thus the greater elevation of the resting tension produced in the neonatal myocardium by St. Thomas' Hospital cardioplegic solution was not due to a greater depolarization of the surface membrane.
A patient is reported with IHSS where mitral valve replacement was performed 3 years following myomectomy because of a residual pressure gradient of left ventricular outflow tract. After mitral valve replacement, the left ventricular outflow tract obstruction was almost completely relieved not only at rest but also at provocation by isoproterenol. The patient successfully returned to work with no symptoms.
The protein kinase inhibitors, protein kinase inhibitor isolated from rabbit muscle and isoquinolinesulfonamide, abolished the inward Na current which was elicited by cAMP.
Intracellular injection of the catalytic subunit of cAMP-dependent protein kinase stimulated the outward potassium current associated with endogenous cellular Ca2+ which were abolished by either caffeine or cAMP-dependent protein kinase inhibitor. The principal action of this kinase in current activation may be to release calcium from the intracellular reservoir.
Using BALB/c nu/nu, BALB/c nu/nufC3H (BALB/c nu/nu mice raised by C3H/HeN foster mother), BALB/c thymus-engrafted BALB/c nu/nufC3H, BALB/c nu/+, and BALB/c nu/+fC3H mice, we examined what kinds of cells are carriers of blood-borne mouse mammary tumor virus (B-MMTV). A radioimmunoassay and an immunoperoxidase assay revealed the presence of MMTV-gp52 antigen in the mammary glands of all BALB/c nu/+fC3H and BALB/c thymus-engrafted BALB/c nu/nufC3H mice (more than 60 days old) but only of some BALB/c nu/nufC3H mice (more than 120 days old): those that possessed a significant number of functional T-cells. BALB/c nu/+ mice did not show the antigen expression at any age. Transfer experiments of cells or plasma from young (less than 12 weeks) BALB/c nu/nufC3H to BALB/c +/+ virgins revealed that cells besides T-cells can also become carriers of B-MMTV. This was confirmed by Southern blotting analyses; exogenous provirus DNA sequences were found in B-cells as well as T-cells of BALB/c nu/+fC3H mice. However, when young BALB/c nu/nu mice were inoculated with BALB/c nu/nufC3H blood, they did not show the MMTV-gp52 antigen expression. Transfer experiments of purified T-cells, B-cells, natural killer cells, and macrophages from BALB/c fC3H mice to BALB/c nu/nu mice revealed that only T-cells have the ability to transfer viral activity to the mammary glands. These results suggest that B-MMTV is carried from the gastrointestinal tract to the mammary glands by lymphoid cells such as T-cells and B-cells, then transferred to the mammary gland cells by the T-cells.
Epstein-Barr virus (EBV), a ubiquitous human herpesvirus and an aetiological agent of infectious mononucleosis, has a unique tropism for B lymphocytes. Clinical and laboratory features of chronic active EBV infections are chronic or persistent infectious mononucleosis-like symptoms and high antibody titre against early antigens (EA). Kawasaki disease (KD), aetiology unknown, is thought to be self-limited immunologically mediated vasculitis. Clinical features of KD are fever, rash, mucositis, lymphadenopathy and coronary artery damage. We report here a child with chronic active EBV infection accompanied by dilatation of coronary arteries. All the EBV-determined nuclear antigen (EBNA)-positive lymphocytes had exclusively CD4 antigen, as revealed by dual staining immunofluorescence analysis. Southern blot hybridization showed that the purified CD4+ cells harboured EBV genome.

The spleen cells, thymocytes, and bone marrow cells of autoimmune MRL/MP-lpr/lpr (MRL/lpr) mice do not constitutively produce interleukin 3 (IL-3), but these mice had IL-3-like activity in their sera. MRL/lpr sera supported the growth of the IL-3-dependent cell lines FDC-P2 and DA-1 but not the growth of IL-2-dependent T-572 cells. This IL-3-like activity increased with age. Biochemical analysis of the MRL/lpr sera by anion-exchange chromatography, gel filtration on a Superose 12 column, the binding to protein-A and sodium dodecyl sulfate-polyacrylamide gel electrophoresis showed that the serum factor with the IL-3-like activity was not IL-3 itself but was associated with IgG. Flow cytometric analysis also showed that the serum level of the Ig capable of binding FDC-P2 cells was high in MRL/lpr but not in MRL/+ mice. We suggest that IL-3 is not responsible for lymphoid hyperplasia, contrary to a previous report; rather auto-antibodies directed toward the IL-3 receptor may act pathogenically in MRL/lpr mice.

In experimental studies, 0.6 N HCl-induced gastric mucosal injury was significantly severe in submandibularectomized rats (SMR rats) than that in either SMR rats receiving exogenous mouse EGF (SMR + EGF rats) or controls. This was also true in gastric injury induced by 0.4 N HCl under pretreatment with indomethacin to reduce gastric mucosal prostaglandins (PGs). Somatostatin (SLI), PGE2, and PAS-stained mucus in the corpus were significantly reduced in SMR rats in comparison to SMR + EGF and control rats. In clinical studies, salivary EGF secretion was much higher in peptic ulcer patients than healthy controls. beta-Urogastrone was effective in the treatment of gastric ulcers. On the basis of experimental studies, we conclude that the protective effect of EGF on the gastric mucosa is, in part, mediated indirectly by increases in SLI, PGE2, and mucus production. However, endogenous, as well as exogenous, EGF has an important direct, cytoprotective effect on the gastric mucosa. From the clinical studies, we also conclude that salivary EGF secretion in ulcer patients increases in a homeostatic response to the presence of an ulcer, facilitating ulcer healing. Furthermore, we believe that beta-urogastrone, human EGF, might prove to be an effective drug in the clinical treatment of gastric ulcers.
To elucidate the role played by catecholamines in the early phase of ischemic derangement of the myocardial energy metabolism, effects of beta-blockers (propranolol, pindolol, and celiprolol) and isoproterenol, and of pretreatment with reserpine and 6-hydroxydopamine (6-OHDA), were studied using the isolated perfused rat heart. beta-Blockers and isoproterenol were infused for 12 min prior to induction of global ischemia. The myocardial energy consumption rate was assessed by calculating the product of left ventricular pressure and heart rate (LVP x HR). The myocardial cell creatine phosphate (CP), ATP and pH were determined with 31P nuclear magnetic resonance (31P-NMR). Global ischemia induced by cross-clamping of the aortic inflow line for 15 min resulted in falls in CP, ATP, and pH. Propranolol (6 x 10(-8) and 1.8 x 10(-7) mol/min) and pindolol (6 x 10(-7) mol/min) produced a decrease in LVP x HR and suppressed the pH fall during ischemia. Celiprolol was without significant effects on these two parameters. Isoproterenol (6 x 10(-12) mol/min) produced an increase in LVP x HR and tended to accelerate the pH fall. Catecholamine depletion with reserpine or 6-OHDA produced no beneficial effects on the pH fall. It was concluded that the beneficial effects of beta-blockers on the pH fall during early ischemia were not due to the specific beta-blocking action but to the nonspecific cardiodepressant effects of these compounds.
Four monoclonal antibodies against factor VIII, NMC-VIII/1, NMC-VIII/2, NMC-VIII/3 and NMC-VIII/4, were produced. The first three antibodies were of the IgG1 immunoglobulin subclass, while the fourth was IgM. The affinity of each antibody for factor VIII was high and anti-factor VIII clotting activity was detected in NMC-VIII/2, NMC-VIII/3 and NMC-VIII/4. NMC-VIII/1 had no inhibitory effect on factor VIII clotting activity. Immunoblotting using purified intact and thrombin-treated factor VIII identified the antibodies' factor VIII polypeptide specificities. With thrombin-treated factor VIII the factor VIII fragments for NMC-VIII/1 and NMC-VIII/2 were 80 kDa and 54 kDa, respectively, while both NMC-VIII/3 and NMC-VIII/4 recognised a 44 kDa fragment. With intact factor VIII, antibodies NMC-VIII/2-4 bound to polypeptides larger than 90 kDa, especially NMC-VIII/2, which reacted with a chain of 330 kDa thought to be a mature form of factor VIII protein. We also detected a mature form of factor VIII in hepatic sinusoidal endothelial cells by immunoperoxidase staining. Identification of the factor VIII polypeptides bearing these fragments enabled us to clarify the localization of the factor VIII thrombin cleavage site, and suggested the existence of factor VIII in hepatic sinusoidal endothelial cells.
1. To elucidate the functional consequences of phosphatidyl inositol (PI) breakdown produced by activation of the muscarinic receptor of the atrial muscle, and to clarify the subtypes of the muscarinic receptor involved, the effects of muscarinic agonists and antagonists on mechanical function were studied in atria isolated from rats given intravenous islet-activating protein (IAP; 50 micrograms kg-1 body weight) 48-72 h before the experiments. 2. The negative chronotropic and inotropic actions of carbachol (CCh) were attenuated and positive inotropic effects (62.5 +/- 5.8% above basal level) were observed with 10(-5) -10(-3) M CCh. Oxotremorine did not produce positive inotropic effects even in doses as high as 3 x 10(-4) M High doses of carbachol produced positive chronotropic effects, although the effects were weak. 3. Propranolol (10(-7) M) did not modify the positive inotropic effect of carbachol observed in IAP-treated atria, nor was there any change in the tissue cyclic AMP levels after carbachol. 4. High doses (10(-5)-10(-3) M) of carbachol produced PI breakdown in the absence and presence of IAP. Oxotremorine (3 x 10(-4) M) did not produce PI breakdown. In the presence of oxotremorine, the positive inotropic effects and PI breakdown by carbachol were abolished. 5. The positive inotropic effect of carbachol was readily antagonized by atropine but pirenzepine and gallamine exhibited only weak antagonist effects. 6. These results suggest that a muscarinic agonist such as carbachol can produce a positive inotropic effect in IAP-treated atria, in association with PI breakdown, through activation of a muscarinic receptor which shows some similarity to that previously identified in smooth muscles.
The composition of the fauna of rumen ciliates in zebu cattle in Kenya was surveyed and 13 genera containing 51 species with 19 formae were identified. Four new species were recognized, then described as Diplodinium africanum n. sp., Diplodinium namum n. sp., Eudiplodinium kenyensis n. sp., and Ostracodinium iwawoi n. sp. In addition, Buetschlia triciliata and Ostracodinium stokyi were found for the second and third times and described as Hsiungia triciliata n. comb. and Enoploplaston stokyi n. stat., with two formae, respectively. The species composition was similar to that of Indian zebu but several species were considered to originate from African native ruminants. In the percentage composition of genera, the numbers of Entodinium, which normally predominate in rumens in other areas, were very low.
The adoral ciliary zone of Cycloposthium spp., inhabiting the large intestine of the horse, was studied by scanning electron microscopy. It could be divided into four parts: outer, inner, left, and right zones. The outer zone, extending on the anterior periphery of the apical cone of the body, had 20 tuft-like syncilia arranged radially around the longitudinal axis. Each ciliary tuft consisted of about 170 cilia, and in cross section it had a rectangular shape. The cilia of the inner zone, situated at the top of the apical cone, were aggregated irregularly to form shorter bundles than the tufts of the outer zone. The innermost ciliar of this zone were shorter than the outermost. There was a distinct non-ciliated border between the outer and inner zones. A horseshoe-like operculum having no cilia was present at the center of the adoral ciliary zone, and the opening of the vestibulum was situated as a cleft crossing from the center to the right periphery of this zone. No cilia extended onto the vestibular wall. The left ciliary zone was situated beneath the outer zone and consisted of five short rows of barren kinetosomes of which only the central row possessed very short cilia. The right ciliary zone, consisting of a few rows of cilia situated at the bottom of the inner adoral lip, was also easily distinguished from the other ciliary zones. This zone was interpreted as an extension of the outer adoral zone passing along the right side of the apical cone.(ABSTRACT TRUNCATED AT 250 WORDS)
