Pathology of atomic bomb casualties.

Thirty seven years ago, 6 August 1945 marks the date of the first atomic bombing never experienced in human history. It was dropped on Hiroshima and this was followed by a second bombing three days later on Nagasaki. The total deaths following exposure to the bomb by the end of 1945 totalled 140,000 (+/- 10,000) in Hiroshima and 70,000 (+/- 10,000) in Nagasaki. The present article described and outline of the physical effects of the atomic bomb and injury to the human body by exposure to the bomb.

Anticomplement immunofluorescence for the titration of antibody to varicella-zoster virus.

Anticomplement immunofluorescence (ACIF) was tested for its use for the titration of antibody against varicella-zoster virus (VZV). ACIF antibody responses of patients with VZV infection were specific for VZV antigen and heterotypic responses to herpes simplex virus type-1 and cytomegalovirus antigens were not observed. Comparative studies of ACIF, membrane immunofluorescence (MIF) and indirect immunofluorescence (IF), using acetone-fixed antigen, were carried out with nonimmune sera and convalescent sera of patients who had recovered from varicella, herpes zoster and Rumsey Hunt disease. Nonspecific staining occurred with some nonimmune sera at a 1:4 dilution in the MIF and IF tests, after freezing and thawing of the serum, but not in the ACIF test. The antibody titers in convalescent sera agreed well in these three methods and the highest titer was obtained by MIF. The titers in ACIF and IF were similar but the ACIF antibody decreased earlier than the IF antibody during convalescence. On the other hand there was a discrepancy between the titers of ACIF and those of MIF and IF antibody in the sera of healthy adults, all sera with titers higher than 10 in the MIF and IF tests had titers below 10 in the ACIF test. The average titer of ACIF antibody declined to less than 10 with increasing age (13 to more than 20 years), whereas the MIF antibody increased during the same period of life.

Physicochemical and catalytic properties of thermostable malate dehydrogenase from an extreme thermophile Thermus flavus AT-62.

Physicochemical and catalytic properties of thermostable malate dehydrogenase (L-malate:NAD+ oxidoreductase, EC 1.1.1.37), isolated from an extreme thermophile, Thermus flavus AT-62, were studied. The enzyme had a molecular weight of 67,000 and consisted of two subunits with an identical molecular weight. The helical content of the enzyme was estimated to be about 25% from the circular dichroism spectrum. The amino acid composition of the thermophilic enzyme was similar to that of its mesophilic counterparts. Titration with 5,5'-dithiobis(2-nitrobenzoic acid) showed that the enzyme contained only one sulfhydryl group per subunit. Substrate inhibition by oxaloacetate was observed. The inhibition decreased with increasing temperature, but was still significant at 60 degrees C. The enzyme was remarkably heat stable, without losing activity after incubation at 90 degrees C for 60 min. The melting temperature of the secondary structure of the enzyme was 96 degrees C.

Adjuvant action of capsular polysaccharide of Klebsiella pneumoniae on antibody response. IX. Its effect on the histology of the regional lymph node and other lymphoid organs.

The sequence of histological changes in the regional lymph node and other lymphoid organs of mice injected with the capsular polysaccharide of Klebsiella pneumoniae (CPS-K) or bacterial lipopolysaccharide (LPS) was followed. Injection of CPS-K, but not LPS, induced the following characteristic histological changes in the regional lymph node. In the early stage there was a marked decrease in the number of small lymphocytes, accompanied by the appearance of scattered fragmented nuclei and infiltration of polymorphonuclear neutrophilic leukocytes, and in the late stage there was marked proliferation of macrophage-like cells and pyroninophilic cells. Histological changes in the thymus and spleen and changes in cell populations in the bone marrow and peripheral blood after CPS-K injection were essentially the same as after LPS injection. Since CPS-K has a much stronger adjuvant action on antibody response than does LPS, it is suggested that the characteristic histological changes in the regional lymph node after injection of CPS-K are closely related to its extraordinarily strong adjuvant action.

Therapeutic effects of tracheostomy in two cases of hypersomnia with respiratory disturbance during sleep.

Recently the association of hypersomnia and respiratory insufficiency without lesion in the respiratory organ has attracted attention of many investigators. Obese patients with such a condition have been called the Pickwickian syndrome. In this report, two non-obese patients with a similar condition were presented, one with micrognathia and frequent apneic episodes during sleep, and the other with laryngeal stenosis due to paralysis of the bilateral laryngeal nerves and chronic laryngitis. Tracheostomy had a prompt and long-lasting therapeutic effect to make their sleep stable and also to relieve their excessive daytime sleepiness. These findings suggest that the obstruction or stenosis of the upper airway during sleep disturbed their nocturnal sleep, and that their excessive daytime sleepiness was a phenomenon compensating for their disturbed nocturnal sleep.

Teratogenic effects of nickel chloride on embryonic mice and its transfer to embryonic mice.

Administration of nickel chloride to the pregnant mice on the seventh to eleventh day of their gestational period, resulted in significant embryotoxic effects in terms of an increased resorption rate, a decreased fetal weight, delay in skeletal ossification and high incidence of malformation. Among the cases of fetal malformation, the following malformations were observed to occur at a higher rate of incidence: acephalia, exencephaly, cerebral hernia, open eyelid, cleft palate, micromelia, ankylosis of the extremity, club foot and skeletal anomalies. Most skeletal anomalies were in the form of vertebral and/or rib fusions and were found mostly at thoracic and lumbar levels. The concentration of nickel retained in embryonic tissues was 800 times higher in the exposed compared to control groups and indicated that increased tissue levels of nickel chloride had a toxic influence on the developing embryo.
