Human calcitonin has the same inhibitory effect on osteoclastic bone resorption by human giant cell tumor cells as salmon calcitonin.

Human calcitonin (hCT) has been reported to have a less hypocalcemizing effect on rats and to have a lower binding affinity for the receptor of mouse osteoclasts than salmon CT(sCT). In this study we comparatively examined the effect of hCT and sCT on osteoclastic bone-resorbing activity of unfractionated cells obtained from human giant cell tumor of bone and from rabbit and mouse long bones. We found that hCT had the same inhibitory effect as sCT on the bone-resorbing activity of human and rabbit osteoclastic cells, but a different one on that of mouse cells. These results indicate that the activity of drugs should be assayed using human cells if possible.

Interaction of calcium ions and polyelectrolytes.

Interaction of Ca(2+) ions with poly(acrylic acid) (PAA) or poly(methacrylic acid) (PMA) was studied using a Ca(2+) ion sensitive electrode. When the PAA solution was neutralized with Ca(OH)(2), the Ca(2+) activity had a maximum around the degree of neutralization 0.5 and decreased with further increase of Ca(OH)(2), being similar to the behavior of the Ca(2+) activity in a maleic acid copolymer solution as reported previously. When the polymer concentration was as low as 0.1 mM, this peak in the Ca(2+) activity was not observed and the counterion condensation theory held. The decrease of the Ca(2+) activity in PAA solution at the degree of neutralization unity was depresseed by the presence of several millimolar KCl. The Ca(2+) activity in the PAA solution at the low degree of neutralization was increased by the presence of dilute KCl and decreased by the presence of concentrated KCl. The decrease of the Ca(2+) activity in PMA solution was observed also at the degree of neutralization unity, but its extent was small compared with that of the PAA solution and the maximum of the Ca(2+) activity was shifted to the degree of neutralization 0.75. The effects of KCl on the Ca(2+) activity in the PMA solution were almost the same as those in the PAA solution. Interpretations of the behavior of the Ca(2+) activity were discussed.

Acylated pelargonidin glycosides in red-purple flowers of Ipomoea purpurea.

Four acylated pelargonidin glycosides were isolated from the red-purple flowers of Ipomoea purpurea. The acylated anthocyanins were all based on pelargonidin 3-sophoroside-5-glucoside, acylated with caffeic acid and/or glucosylcaffeic acids. Three novel anthocyanins were elucidated to be pelargonidin 3-O-[2-O-(6-O-(trans-3-O-(beta-D-glucopyranosyl)-caffeyl)- beta-D-glucopyranosyl)-6-O-(trans-4-O-(6-O-(trans-caffeyl)-beta-D- glucopyranosyl)-caffeyl)-beta-D-glucopyranoside]-5-O- [beta-D-glucopyranoside], pelargonidin 3-O-[2-O-(6-O-(trans-caffeyl)-beta-D-glucopyranosyl)-6-O- (trans-4-O-(6-O-(trans-caffeyl)-beta-D-glucospyranosyl)-ceffeyl )-beta-D- glucopyranoside]-5-O-[beta-D-glucopyranoside] and pelargonidin 3-O[2-O-(2-O-(6-O-(trans-caffeyl)-beta-D-glucopyranosyl)-6-O- (trans-caffeyl)-beta-D-glucopyranoside]-5-O-[beta-D-glucopyoside]. Another anthocyanin was pelargonidin 3-O-[2-O-(glucosylcaffeylglucosyl)-6-O-(caffeyl)-glucoside]-5-gluc oside, which was reported to be present in the red-purple flowers of Pharbitis nil. Apart from the second pigment cited, these are analogous pigments of the violet-blue Ipomoea anthocyanins which are composed of acylated cyanidin glycosides, instead of pelargonidin.

Age-related changes in Meissner corpuscles in the mouse palate: a histochemical and ultrastructural study.

Meissner corpuscles in the palatine mucosae of ddY-mice of various ages were studied histochemically and electron microscopically. As the age of the animals advanced, regressive Meissner corpuscles with lessened or depleted activity of non-specific cholinesterase (ChE) increased in number. Immunohistochemistry for Na+/K(+)-ATPase showed that the complex ramification of axon terminals in Meissner corpuscles attained at maturation gradually changed to a sparse distribution as the animals aged. Only a few putative rudiments of Meissner corpuscles, which were completely devoid of nerve terminals, were encountered. Electron microscopy showed that the regressive Meissner corpuscles contained atrophic axons and lamellae in a disordered arrangement, although the severity of the atrophy differed depending on the degree of the regression. Furthermore, the Meissner corpuscles were found to progressively accumulate fibrous long-spacing fibers having a periodicity of 160 nm and unique spherical corpuscles 150-300 nm in diameter in their intercellular spaces. From these results we concluded that: 1) the atrophy of the axon and lamellae proceeds almost simultaneously during the age-related regression of Meissner corpuscles; and 2) not only the atrophy of the axon and the lamellar cell but microenvironmental changes such as the accumulation of intercellular substances is also involved in the regression of these corpuscles.

Laparoscopic minilaparotomy pancreaticoduodenectomy with lymphadenectomy using an abdominal wall-lift method.

Because of technical difficulties attributable to the lack of appropriate techniques, the high cost of laparoscopic instruments, and the need for numerous disposable stapling devices, laparoscopic pancreaticoduodenectomy has been performed in only two patients. To solve the above problems, we devised a method of laparoscopic minilaparotomy using an abdominal wall-lift method and have successfully used it to perform pancreaticoduodenectomy with lymphadenectomy in a patient with distal choledochal cancer.

[Ipsilateral oculomotor nerve palsy and contralateral downbeat nystagmus due to unilateral paramedian thalamopenduncular infarction--a case report].

A case of midbrain and thalamic infarction which showed complete oculomotor nerve palsy of the ipsilateral eye and monocular downbeat nystagmus, ptosis, upward movement disturbance and adduction disturbance of the contralateral eye was reported. The patient is a 53-year-old woman who was admitted to our hospital because of sudden onset of double vision. Head magnetic resonance imaging showed unilateral midbrain and thalamic infarction. The midbrain lesion was located in the paramedian area and the bilateral ptosis, bilateral upward gaze palsy and adduction disturbance of the contralateral eye were considered to be caused by the lesion involving the unilateral oculomotor nucleus. This case is considered to be important because the association of contralateral monocular downbeat nystagmus is very rare.

[A study on reduction in care burden of the family caregiver of the bedridden and senile elderly--examination of subjective factors about construction in care burden].

The purpose of this study was to develop a supporting system in the community which reduces care burden of the family caregiver members of bedridden elderly age 75 and over. Preliminary study was carried out in order to find out fundamental elements of care burden. Five families were picked up among the families living in Chuo ward in Tokyo who were provided professional nursing care services at home. A nursing intervention was done through home visits and interviews with caregivers and their subjects who need proper care services. 1. fundamental elements of a caregiver's care burden: a) main elements are health and mental condition of elders, b) influential elements are the function of caregivers, the function of assistant caregivers, the function of professionals and the function of care systems. 2. The line of stress defence: All the functions except that of caregivers were positioned at the line of stress defense, and in our view the caregiver's coping capability would be increased as the line of stress defense functions and, hence, the care burden might be reduced. 3. Assessment of fundamental elements shall be determined from 3 aspects a) health and mental condition of elders, b) accessibility to buffers and their quality and c) capacity and skill of a care giver. In order to reduce the care burden of the family caregiver, it is necessary to assess multilaterally the constituents and/or structures of care in the future, and at the same time the additional functions will be required to furnish information and educate the caregivers on how to utilize various social care services, there by enabling them to cope with psychological stress effectively. These findings suggest that it is also necessary to complete the training of professional concerning feasible means that could promote the caregivers' abilities to cope with the bedridden and senile elderly.

Structural analysis of Tpn1, a transposable element isolated from Japanese morning glory bearing variegated flowers.

The 6.4 kb transposable element Tpn1 belonging to the En/Spm family was found within one of the DFR (dihydroflavonol-4-reductase) genes for anthocyanin biosynthesis in a line of Japanese morning glory (Pharbitis nil) bearing variegated flowers. Sequencing of the Tpn1 element revealed that it is 6412 bp long and carries 28-bp perfect terminal inverted repeats. Its subterminal repetitive regions, believed to be the cis-acting sequences for transposition, show striking structural features. Twenty-two copies of the 10-bp sequence motif GACAACGGTT can be found as direct or inverted repeats within 650 bp of the 5' end of the element, and 33 copies of the sequence motif lie within 800 bp of the 3' terminus. All these 22 copies of the sequence motif near the 5' terminus and 30 copies in the 3' terminal region are arranged as inverted repeats and 3-8 bp AT-rich sequences are detected between these inverted repeats. In addition, four copies of 122-bp tandem repeats and six copies of 104-bp tandem repeats are present in the 5' and 3' subterminal repetitive regions, respectively. No large open reading frame characteristic of autonomous elements of the En/Spm family can be detected within the element. The results are discussed with respect to heritable changes in flower variegation in this line of Japanese morning glory.
