Isolation and characterization of guamerin, a new human leukocyte elastase inhibitor from Hirudo nipponia.

A new human leukocyte elastase inhibitor was extracted and purified from a Korean native leech Hirudo nipponia. The inhibitor, called guamerin, has a molecular weight of 6,110 and shows inhibition constant (Ki) of 8.1 x 10(-14) M. It is stable at a wide range of pH from 1 to 11 and heat-stable up to 90 degrees C. The complete amino acid sequence of guamerin reveals a cysteine-rich polypeptide of 57 amino acid residues that shows no similarity to any known elastase inhibitors but has 51% sequence homology with hirustasin. Guamerin has identical spacing of 10 cysteine residues as antistasin-type serine proteinase inhibitors, but the P1 reactive site residue is Met36 instead of Arg. The neighboring sequence of the reactive site consists primarily of hydrophobic amino acid residues. Based on examinations of the target proteinases and the reactive site specificity, guamerin is a new low molecular weight protein that inhibits elastases.

In vitro and in vivo antitumoral phenanthrenes from the aerial parts of Dendrobium nobile.

Two phenanthrenes were isolated from the aerial part of Dendrobium nobile Lindl. and their structures were identified to be 4,7-dihydroxy-2-methoxy-9,10-dihydrophenanthrene (1) and denbinobin (2), among which the former has been first isolated from this plant. These two compounds were found to be cytotoxic against A549 (human lung carcinoma), SK-OV-3 (human ovary adenocarcinoma), and HL-60 (human promyelocytic leukemia) cell lines. Compound 1 also showed antitumor activity on the life span of ICR mice intraperitoneally implanted with 1 x 10(6) cells of sarcoma 180.

Discrepancy of recovery times related to potency between atracurium and mivacurium simultaneously administered in isolated forearms.

Recovery from potent non-depolarising muscle relaxants is slower than from the less potent agents. However, recovery from mivacurium, which is more potent than atracurium, is faster than from atracurium following systemic administration. In an attempt to confirm this discrepancy we compared recovery times following simultaneous administration of equipotent doses of atracurium and mivacurium into the isolated forearms of human volunteers (n = 10). This method enabled us to study the interaction of muscle relaxants with receptors at the neuromuscular junction separated from the effects of plasma drug concentration. In these experiments, the recovery times from maximum block to 50% recovery of control twitch height were significantly longer with mivacurium than with atracurium (mean 25.2(SD 4.7) versus 22.6(3.1) min, p < 0.01). We found that the evidence that mivacurium has a slower recovery than the less potent atracurium may be true using the bilateral, isolated forearm technique and that the discrepancy might be due to a difference in the pharmacokinetic variables of the two drugs.

Measuring the success of implementation of information system for health center.

The purpose of this study is to analyze the effects of the Health Management Information System (HMIS) on the productivity and adoption process of health center staff as well as the satisfaction with the services provided by the Kwonsun health center located in Suwon city as a study subject. Three surveys were conducted to measure the changes in productivity and adoption process (knowledge, persuasion, decision, implementation, and confirmation) of health center staffs over time. In addition, the effects of HMIS on the level of satisfaction with the services perceived by the visitors were also measured by comparing the satisfaction level between the study health center and a similar health center as a control group. The results suggest that HMIS increased productivity and satisfaction of staffs but did not increase persuasion and decision level, and that it succeeded in increasing the satisfaction with the services for the visitors.

Characterization and primary specificity of an extracellular metalloproteinase from Serratia marcescens.

An extracellular endopeptidase (proteinase) from Serratia marcescens (Serratia marcescens extracellular proteinase, EC 3.4.24.4), purified to homogeneity, was analyzed for enzyme properties. The enzyme has a polypeptide chain molecular mass of 52 kDa as determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The enzyme has an optimal temperature of 40 degrees C and an optimal pH of 7.0. Enzyme activity was enhanced over two times by the addition of Ca2+ and Mg2+ ions and eliminated almost completely by the presence of 0.2% SDS. The enzyme has broad substrate specificity and contains neither cysteine nor methionine. Low homology was found between the NH2-terminal amino acid sequence of the enzyme of this study and the NH2-terminal sequence of a proteinase from another strain of S. marcescens. Chemical modification with N-bromosuccinimide, 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide, and 8-anilino-1-naphthalene sulfonic acid and by photooxidation with methylene blue reduced enzyme activity considerably. The enzyme was shown to have broad peptide bond specificity judging from the contribution of 11 amino acids to the carboxyl side of the peptide bonds hydrolyzed.

Isolation and characterization of cDNA clones encoding class I chitinase in suspension cultures of rice cell.

We have cloned and determined the nucleotide sequences of full-length cDNA clones for chitinase, CH6 and CH16, whose sizes were about 1.1 kb with signal peptides, from rice cell suspension cultures. Northern blot analysis with CH16 as a probe showed that the chitinase transcript reached maximum at 8 h after elicitor-treatment and their sizes were about 1.1 kb, demonstrating that these clones were induced by elicitor.

Nucleotide sequence homology of cDNAs encoding soybean Bowman-Birk type proteinase inhibitor and its isoinhibitors.

The sequences of cDNA clones encoding the Bowman-Birk proteinase inhibitor (BBPI) and its isoinhibitors (D-II and C-II) have been identified. Nucleotide sequence homologies among these clones and between the two reactive domains in each clone were very high. These homologies suggest that the BBPI and its isoinhibitor genes may have evolved from a common ancestral inhibitor. Also, clone pB2 (D-II) had two identical reactive sites that inhibit trypsin only, and three repetitive regions that are about 81 bp and initiated with ATG. It was assumed from these results that isoinhibitor D-II was the most primordial type of these inhibitors.

[Immune reactions between excretory-secretory antigens and specific antibodies of Clonorchis sinensis before and after praziquantel treatment in experimentally infected rabbits].

This study was designed to evaluate the humoral immune reactions in clonorchiasis before and after praziquantel treatment. Rabbits were infected with 150 or 450 metacercariae, treated on 4 and 8 1/2 months after infection, and observed for 13 months of posttreatment. Infection controls were maintained for 22 months. Antigen was the metabolic product of worms incubated in physiologic saline. The immune reactions of anti-Clonorchis IgG were observed using SDS-PAGE/immunoblot. During the infection and posttreatment, the antigenic proteins of 66, 63, 54, 52, 50, 47, 42, 40, 38, 34, 33, 30, 27, 25, 23, 20, 19, 18, 17, 16, 15, 14, 13, 12.5 and 11.5 kDa were detected. Of them, 33, 27, 13, and 12.5-kDa antigens were highly antigenic and observed predominantly in infection controls. After the treatment, 13 and 12.5-kDa antigens faded in 6 months after the second treatment, but 33 and 27-kDa antigens were detected until 13 months of posttreatment. The results clearly demonstrate that 13 and 12.5-kDa antigens represent attenuated host immune reactions by praziquantel treatment. As the 12.5-kDa antigen had a large amount of protein in SDS-PAGE, it was designated as 'K2-Ag' of C. sinensis.

Protein composition and antigenicity of the tegument from Paragonimus westermani.

To ascertain that tegument of Paragonimus westermani has specific antigenic proteins, the tegumental fraction was isolated from 10-month-old worms by 0.1% digitonin solution, and subjected to SDS-PAGE and immunoblot. Component proteins of tegumental syncytium comprised of 94, 74 (76-66), 62, 54, 44, 42, 38, 28, 26, 25, 24, 17, 15.5 and 13. 5 kDa proteins. Of them, the 94, 44 and 42 kDa proteins were more specific to tegument, especially the 94 kDa protein was the most prevailing one. In immunoblot, antigens of the 94, 90, 78, 76, 74, 68, 65, 62, 60, 59 and 54 kDa proteins were commonly detected by 7 sera of 10 human paragonimiasis, but none of them reacted with 5 sera of clonorchiasis. In conclusion, the 94 kDa protein was the major tegumental protein, as well as the specific antigen. The 76 and 66 kDa proteins were the minor components of tegument, which were also specific antigens of P. westermani.

Mucosal morphology in isolated bowel segments: importance of exposure to luminal contents.

An isolated bowel segment (IBS) is a loop of intestine that has been freed from its mesenteric attachment after the development of vascular collaterals between the antimesenteric surface of the gut and the host organ. Surgical creation of such artificially vascularized isolated bowel segments is of interest to researchers for a variety of studies, and may be useful in the treatment of short bowel syndrome, allowing longitudinal division of the remaining small bowel to double its length. We created four surgical variants to study the ability of the collateral blood supply to maintain mucosal integrity in the presence or absence of normal luminal contents. In all groups, a collateral blood supply was created in a 5- to 7-cm segment of adult rat jejunum by hepatoenteropexy (Iowa model II). In Thiry-Vella (T-V) and isolated bowel segment (IBS) rats, this segment was exteriorized at both ends to exclude luminal contents. Control and IBS in continuity (IBS-C) loops were left in continuity. The mesentery of IBS and IBS-C rats was divided 5 weeks later, leaving the experimental segment entirely dependent on the collateral circulation. All animals were harvested at 7 weeks after the initial surgery. Tissues were analyzed for mucosal weight, protein content per centimeter of bowel, length of villi, depth of crypts, DNA content, and sucrase activity. We found that segments retaining luminal continuity had significantly higher mucosal weight and DNA content per centimeter of bowel compared with exteriorized loops.
