Infection status of dragonflies with Plagiorchis muris metacercariae in Korea.

Plagiorchis muris has been found in both house and field rats as well as in humans. The infection status of the second intermediate hosts of P. muris is prerequisite in understanding their biological features in an ecosystem. Six species of dragonflies were caught in a wide range of areas in Korea; and they were Sympetrum darwinianum, S. eroticum, S. pedomontanum, S. infuscatum, Pantala flavoscens, Calopteryx atrata, and Orthetrum albistylum speciosum. The occurrence of P. muris metacercariae in dragonflies was nationwide with various infection rates. The metacercarial burden of P. muris in the surveyed areas was the highest in S. eroticum followed by S. darwinianum, S. pedomontanum, and C. atrata. The highest infection rate by P. muris metacercariae was found in S. darwinianum followed by S. eroticum. The metacercarial burden was particularly heavy in the dragonflies found in Hamyang-gun and Kosong-gun, Kyongsangnam-do. It is, therefore, likely that dragonflies play a significant role as the second intermediate host in the life cycle of P. muris in Korea.

A cytogenetic study on human intestinal trematodes of the genus Metagonimus (Digenea: Heterophyidae) in Korea.

In order to analyze chromosome numbers and karyotypes of intestinal trematodes belonging to the genus, Metagonimus, the gonad tissues of M. takahashii, M. miyatai, and M. yokogawai were prepared and examined. The number of bivalents in the first meiotic division of M. takahashii was nine (n = 9). The diploid number of M. miyatai was observed to be eighteen (2n = 18) and their chromosomes consisted of one pair of metacentric, 7 pairs of submetacentric, and one pair of telocentric chromosomes. The diploid number of M. yokogawai was thirty-two (2n = 32) and the chromosome complements were composed of two pairs of metacentric, 11 pairs of submetacentric, and three pairs of subtelocentric chromosomes. These results could be a supporting evidence for the validity of the new species, M. miyatai, distinct from M. yokogawai.

Venous dynamics in leg lymphedema.

To determine whether there is anatomical and/or functional impairment to venous return in patients with lymphedema, we examined venous dynamics in 41 patients with unilateral leg lymphedema. A Volometer was used for computer analysis of leg volume, a color Duplex Doppler scanner was used to determine deep vein patency and skin thickness, and Air-plethysmography was used to assess ambulatory venous pressure, venous volume, venous filling index and the ejection fraction. In the lymphedematous leg, volume and skin thickness were uniformly increased (126.4 +/- 21.3% and 156.9 +/- 44.5%) (mean +/- S.D.), respectively. The ambulatory venous pressure was also increased (134 +/- 60.7%) as was the venous volume (124.5 +/- 37.5%), and the venous filling index (134.5 +/- 50.5%). The ejection fraction was decreased (94.9 +/- 26.1%). Greater leg volume correlated with increased venous volume and venous filling index (values = 0.327, 0.241, respectively) and decreased ejection fraction (r = -0.133). Increased subcutaneous thickness correlated with increased venous filling index and venous volume (r = 0.307, 0.126, respectively) and decreased ejection fraction (r = -0.202). These findings suggest that soft tissue edema from lymphatic stasis gradually impedes venous return which in turn aggravates the underlying lymphedema.

An infestation of the mite Sancassania berlesei (Acari: Acaridae) in the external auditory canal of a Korean man.

We here report the case of a storage mite, Sancassania berlesei, infestation in the external auditory canal of a 46-yr-old male. He complained of feeling a foreign body and itching in the left external auditory canal for 1 mo, with accompanying otalgia for 3 days. Considering the duration of the patient's complaint and the 8-9-day life cycle of the mite, the mites are believed to have lived in the patient's ear for more than 3 generations.

Unsupervised connectivity-based thresholding segmentation of midsagittal brain MR images.

In this paper, we propose an algorithm for automated segmentation of midsagittal brain MR images. First, we apply thresholding to obtain binary images. From the binary images, we locate some landmarks. Based on the landmarks and anatomical information, we preprocess the binary images, which substantially simplifies the subsequent operations. To separate regions what are incorrectly merged after this initial segmentation, a new connectivity-based threshold algorithm is proposed. Assuming that some prior information about the general shape and location of objects is available, the algorithm finds a boundary between two regions using the path connection algorithm and changing the threshold adaptively. In order to test the robustness of the proposed algorithm we applied the algorithm to 120 midsagittal brain images and obtained satisfactory results.

Karyotype analysis of Neodiplostomum seoulense.

A karyotype analysis of the chromosome of Neodiplostomum seoulense, one of causative agents of human intestinal trematodiasis, was done from the gonad tissue by the squashing method. The chromosome number was n = 10 and 2n = 20. Chromosome length was 1.30-4.0 microns. Chromosome pairs in the complement consisted of two pairs of metacentric, five pairs of submetacentrics/subtelocentric and three pairs of telocentric chromosomes. These data were comparable with those of other intestinal trematodes.

Excision of subcutaneous tissue and deep muscle fascia for advanced lymphedema.

In advanced peripheral lymphedema particularly involving the legs, complex physical therapy (massage/bandage-wrapping compression, remedial exercises, and maintenance use of a low stretch elastic compression garment), is not uniformly successful. In six patients with morbid lymphedema of the legs, we described our positive experience using a modified Auchincloss/Homans excisional operation.

Identification of YH439 and its metabolites in rat urine by gas chromatography-mass spectrometry and liquid chromatography-mass spectrometry.

YH439 is a potential drug candidate for the treatment of various hepatic disorders. YH439 and its three metabolites have been identified in rat urine by liquid chromatography-mass spectrometry (LC-MS) and by gas chromatography (GC)-MS. Identification of YH439 and its metabolites was established by comparing their GC retention times and mass spectra with those of the synthesized authentic standards. Both electron impact- and positive chemical ionization MS have been evaluated. The metabolism study was performed in the rat using oral administration of the drug. A major metabolite (YH438) was identified as the N-dealkylation product of YH439. Other identified metabolites were caused by the loss of the methyl thiazolyl amine group (metabolite II) from YH439, the isopropyl hydrogen malonate group (metabolite IV) and the decarboxylated product (metabolite III) of metabolite II.

Localization of worm antigen in Neodiplostomum seoulense by immuno-electronmicroscopy.

The localization of worm antigen of Neodiplostomum seoulense was examined by immuno-electronmicroscopic observation. Not only the immunized serum of mice with crude worm extract of N. seoulense but also serum of infected mouse were reacted to the worm section. Using immunized serum as primary antibody, the gold particles were deposited on the rough endoplasmic reticulum of the cell of tribocytic organ, spermatozoa in the seminal vesicle, microvilli of the caecum and vitelline follicle. Using infected serum, gold particles were deposited only on the vitelline follicle prominently. This finding suggested that the tribocytic organ, seminal vesicle, caeca and vitelline follicles may play a role of antigen to immunized serum with crude worm extract of N. seoulense, whereas the vitelline follicle, to the infected serum.

Chemotherapeutic trial to control enterobiasis in schoolchildren.

To assess several chemotherapeutic schemes for control of enterobiasis, 738 children in five primary schools in Chunchon, Korea, were studied from May 1994 to June 1995. They were divided into 6 groups by the schemes: treatment of once or twice a year; treatment of positive cases or of whole class students; treatment with or without family members. The overall egg positive rate before intervention was 17.5% out of 789 children. Treating all individuals in a class together with family members of positive cases brought better control efficacy than other schemes (p = 0.000). However, when egg positive rate is less than 30%, treating only egg positive cases also can reduce egg positive rate. The confounding factors for the enterobiasis control in primary schoolchildren were new-comer to a class and familial infection.

PCR-RFLP pattern of three kinds of Metagonimus in Korea.

We tried to compare the three kinds of Metagonimus species, M. yokogawai, Metagonimus Miyata type, and M. takahashii, which were known to be distributed in Korea with polymerase chain reaction based-restriction fragment length polymorphism (PCR-RFLP) patterns. We amplified the internal transcribed spacer 1 (ITS1) site of ribosomal RNA and mitochondrial cytochrome c oxidase I (mCOI) gene. The restriction patterns of ITS1 gene with Rsa I, Alu I and Msp I showed multiple fragmented bands of different sizes between three kinds of Metagonimus. In case of mCOI gene, Rsa I and Alu I enzymes produced differentially fragmented band patterns. According to the parsimony analysis of PCR-RFLP patterns, the estimated genetic divergence between M. yokogawai and Metagonimus Miyata type was 0.034880, between Metagonimus Miyata type and M. takahashii was 0.028098, between M. yokogawai and M. takahashii was 0.018179. It is suggested that Metagonimus Miyata type may be separate species and evolutionize at the older time than the other two species.

Genetic comparison between Spirometra erinacei and S. mansonoides using PCR-RFLP analysis.

The only observed morphological difference between Spirometra erinacei and S. mansonoides is the uterine shape of the mature proglottid. Two species of worms are thought to be evolutionarily closely related. Biomolecular comparison of the two worms by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis was conducted to observe the genetic distance. The 28s rDNA, mitochondrial cytochrome c oxidase subunit I (mCOI), and ribosomal internal transcribed spacer 1 (ITS1) fragments were obtained from the worms by PCR. The PCR products were cleaved by 5 four-base pair restriction enzyme combinations (Msp I, Hae III, Alu I, Cfo I, Rsa I), electrophoresed and analyzed with PAUP 3.1.1. The fragment patterns of 28S rDNA and ITS1 demonstrated that two worms were in identical systematic tree with bootstrap number 94 and 100, respectively. As for mCOI, bootstrap number was 74 in a different tree. Above results are indicative of recent common ancestry between S. erinacei and S. mansonoides.

Intimal thickening develops without humoral immunity in a mouse aortic allograft model of chronic vascular rejection.

BACKGROUND: The major threat to the long-term survival of cardiac allograft recipients is the development of diffuse intimal thickening in the allograft coronary arteries through mechanisms that are poorly understood. Although antidonor antibodies have been associated with the development of this condition, a causal relationship has not been established. METHODS AND RESULTS: To determine whether humoral immune responses are necessary for the development of graft vascular disease, we performed abdominal aortic allografts from normal donor mice into different immunodeficient recipient mice: those lacking all donor-specific immune responses (severe combined immunodeficient [SCID] mice and recombination activating gene-1 [RAG-1]-deficient mice) and those lacking humoral immune responses alone owing to a targeted deletion of the joining region (JH) gene segments for the immunoglobulin heavy chain. At 6 to 9 weeks after transplantation, aortic allografts in normal immunocompetent recipients showed concentric intimal thickening extending the full length of the graft (percent luminal reduction, [%LR], 31.2 +/- 9.1 [mean +/- SD] and 38.5 +/- 3.6 in different donor-recipient strain combinations). In contrast, syngeneic (histocompatible) aortic grafts showed a normal-appearing vessel wall (%LR, 1.6 +/- 0.7). In both SCID and RAG-1-deficient recipients, aortic allografts showed a virtual absence of neointimal formation (%LR, 3.7 +/- 2.1 and 3.8 +/- 1.6 in SCID and RAG-1-deficient recipients, respectively), indicating a critical etiological role for alloimmune responses in this model. Importantly, allografts in JH-deficient mice showed marked intimal thickening (%LR, 35.7 +/- 7.9), with an appearance histologically indistinguishable from that of normal immunocompetent recipients. CONCLUSIONS: Neointimal formation in graft vascular disease is critically dependent on alloimmune responses of the host. Humoral effector mechanisms, however, may not be required.

Redescription of Macroorchis spinulosus Ando, 1918 (Digenea: Nanophyetidae) encysted in the fresh water crayfish, Cambaroides similis.

Macroorchis spinulosus Ando, 1918 (Digenea: Nanophyetidae) is redescribed based on adult flukes recovered from experimental mice, rats and cats which were fed with metacercariae encysted in the muscle of the crayfish. Cambaroides similis. A total of 117 adult flukes were harvested from the small intestine of the experimental animals at 5-21 days post-infection. The worm recovery rate was 31-37%, not significantly different among the three kinds of animals. The metacercariae were round, 0.18 mm in average diameter, encysted with two layers of thick walls, and possessing a stylet on the oral sucker and Y-shaped excretory bladder. The adult flukes were oval, 0.66-0.71 mm long and 0.36-0.41 mm wide, and characterized by the presence of a stylet on the oral sucker, variable location of the ovary near the mid-portion of the body, bilobed seminal vesicle, and anterolateral location of two testes. These morphological features were a little different from the original description, hence, an emended version of M. spinulosus was given. M. spinulosus is a new intestinal trematode of animals and possibly man in Korea.

A nationwide survey of naturally produced oysters for infection with Gymnophalloides seoi metacercariae.

A nationwide survey was performed to know the geographical distribution of Gymnophalloides seoi (Digenea: Gymnophallidae) metacercariae in Korea, by examining the infection status of locally produced oysters, Crassostrea gigas. A total of 24 coastal areas (myons) of 14 guns ( = counties) in Kyonggi-do, Chollabuk-do, Chollanam-do, Kyongsangnam-do, Kyongsangbuk-do, or Kangwon-do, where natural oysters are produced but G. seoi has never been reported, and 13 areas (myons) of Shinan-gun, Chollanam-do, nearby the known endemic area, were surveyed. Oysters from nonendemic areas were free from G. seoi infection, except Byonsan-myon of Buan-gun, Chollabuk-do, where one of 50 oysters examined was infected with 15 metacercariae of G. seoi. In Shinan-gun, oysters from 10 areas including Aphae-myon ( = township) and Anjwa-myon were infected with the metacercariae, with the infection rate ranging from 1.7% to 100% by areas. The intensity of infection was the highest in Aphae-myon. 785.9 metacercariae per oyster. The results indicate that high prevalence of G. seoi is confined to Shinan-gun, but low grade prevalence is also present in adjacent areas such as Buan-gun, Chollabuk-do.

Intestinal parasitic infections in the residents of an emigration camp in Tijuana, Mexico.

We examined stool specimens of the residents in the emigration camp in Tijuana, Mexico for helminth eggs or protozoan cysts with formalin-ethyl acetate concentration method in February and July 1992. Out of 92 examined samples, number of positive was 49 (53.3%). While number of cumulative positive was 66 (71.7%). Cysts of Entamoeba coli (29.3%) Giardia lamblia (9.8%), Entamoeba histolytica (7.6%), and eggs of Taenia spp. (6.5) were most frequently observed. Filtered water supply and chemotherapy were required in this camp.
