In vivo retinal gene expression in early diabetes.

PURPOSE: Studies have demonstrated a causal role for specific molecules in the pathogenesis of diabetic retinopathy. Among the implicated mediators are growth factors such as vascular endothelial growth factor (VEGF) as well as adhesion molecules and proliferation- and apoptosis-related genes. However, a coordinated large-scale investigation of gene expression in the diabetic retina has not yet been reported. Here the retinal gene expression profile of diabetic and nondiabetic animals using cDNA microarrays were analyzed and compared. METHODS: Long-Evans rats were made diabetic with streptozotocin. Retinal gene expression was analyzed over 3 weeks using high-density nylon filter-based cDNA arrays. Genes were sorted into clusters according to their temporal expression profiles. They were also grouped according to their potential pathophysiological significance. The in vivo gene expression profiles of selected genes were verified via RNase protection assay. RESULTS: The rat GeneFilter contains a total of 5147 genes, of which 1691 are known genes and 3456 are expressed sequence tags (ESTs). On day 3, the expression of 27 known genes was increased by more than twofold. On days 7 and 21, the corresponding numbers were 60 and 12, respectively. A transient upregulation (>2-fold) in expression was seen in 627 of 5147 total genes. A subset of 926 genes exhibited a modest (<2-fold) decrease in expression. No genes showed a greater than twofold decrease in expression. Overall, the identity of the genes that were upregulated suggests that the response of the retina to the diabetic challenge contains an inflammatory component. Moreover, most regulatory activity occurs during the first week of diabetes. CONCLUSIONS: The development of a rational therapy for diabetic retinopathy will be assisted by detailed knowledge regarding the molecular pathophysiology of the disease. Here, an expression profile of an underlying retinal inflammatory process in early diabetes was extracted. Beyond providing insight into the general nature of the response to a pathogenic challenge, gene expression profiling may also allow the efficient identification of potential drug targets and markers for monitoring the course of disease.

[Pathological observation of 16 cases with non-epithelial tumor and tumor-like lesions of the prostate].

OBJECTIVE: To investigate the diagnosis, differential diagnosis and prognosis of stromal tumors and tumor-like lesions of the prostate. METHODS: By means of HE, immunohistochemistry and electron microscopy, the clinical and histological data of 16 cases with stromal tumors and tumor-like lesions of the prostate were observed. Some of them were followed-up. RESULTS: Of the 16 cases, 6 presented stromal nodular hyperplasia, 3 were stromal sarcoma, 1 malignant phyllodes-type tumor, 3 leiomyosarcoma, 1 fibrosarcoma, and one non-Hodgkin's lymphoma. CONCLUSIONS: Nearly all the leiomyomas of the prostate diagnosed previously are, in fact, stromal nodular hyperplasia. The phyllodes-type tumors are often confused with stromal sarcomas in needle biopsy specimens and cause difficulty in differential diagnosis. Diagnostic criteria of leiomyosarcoma, fibrosarcoma, non-Hodgkin's lymphoma of the prostate are the same as their counterparts in the soft tissue. Non-epithelial malignant tumors in prostate are of poor prognosis.

[Allogeneic peripheral blood stem cell transplantation for 3 patients with beta-thalassemia major and 1 patient with congenital idiopathic haemolytic anemia].

OBJECTIVE: To examine the feasibility of allogeneic peripheral blood stem cell transplantation (Allo-PBSCT) for the hereditary haemolytic anemia (HHA). METHODS: Allo-PBSCTs were performed on 3 patients with beta-thalassemia major and 1 patient with congenital idiopathic haemolytic anemia by the HLA-identical siblings' donors or 1 locus-mismatch parent. The harvested stem cell contained nucleated cells (4.7-14.4) X 10(8)/kg recipient weight, CD(34) (+) CD(38)(-) cells(4.8-32.9) X 10(6)/kg and colony-forming-unit-granulocyte-macrophages (CFU-GM)(3.22-9.8) X 10(5)/kg. The conditioning regimen consisted of busulfan, cyclophosphamide, melphalan, thiotepa, fludarabine and antithymocyte globulin (ATG). RESULTS: All patients were engrafted and accomplished with the acute graft-versus-host disease (aGVHD). A patient had the controlled chronic GVHD. Three patients are survivals with ex-anemia; their Hb levels have been maintaining normality without transfusion. One died of the hepatic veno-occlusive disease. CONCLUSION: Allo-PBSCT is an effective and a new way to treat beta-thalassemia major and congenital idiopathic haemolytic anemia.

The role of sexual related Y gene detection in the diagnosis of patients with gonadal dysgenesis.

OBJECTIVE: To clarify the role of sexual related Y (SRY) gene detection in the diagnosis of gonadal dysgenesis. METHODS: Sixteen cases of gonadal dysgenesis were included in this study: 5 with androgen insensitivity syndrome, 1 with 17-alpha-hydroxylase deficiency, 4 with true hermaphrodite, 2 with 45, X/46, XY gonadal dysgenesis, 1 with 45, X gonadal dysgenesis, 1 with XY pure gonadal dysgenesis, 1 with testicular regression, and 1 XY female who gave birth to a normal baby. SRY gene was detected by using polymerase chain reaction (PCR) in blood and gonad samples and by direct sequencing of the SRY motif. RESULTS: Among the 16 cases, 15 were blood SRY positive, among which 13 (86.7%) showed the presence of testicular tissue, and 2 showed ovaries without testicular tissue. One SRY negative case showed the presence of testicular tissue. In 3 cases, SRY detection in gonadal tissue correlated with pathological findings but not with blood karyotype. The correlation between peripheral blood SRY and the pathology of the gonads was 81.25% and the correlation between the presence of peripheral blood Y chromosome and pathology of the gonads was 68.75%. Sequencing of the SRY motif in an XY female who gave birth to a normal baby showed no mutation. CONCLUSIONS: SRY detection is more sensitive and specific than blood karyotype in the prediction of the presence of testicular tissue. Peripheral blood karyotype does not necessarily reflect gonadal type. There may be testicular related factors other than the SRY gene.

[Umbilical cord blood transplantation (UCBT) in thalassemia children].

OBJECTIVE: To evaluate the efficacy of allo-UCBT in thalassemia children. METHODS: Five patients with beta-thalassemia major (genotypes were homozygote of beta41-42, beta654 and double heterozygosities of beta41-42/beta654, beta41-42/-28, beta654/-28, respectively) were treated by allo-UCBT. The median age was 3 year and ten month old (31/2 - 7 2/12). Four donor-recipient pairs were HLA-identical and one 1 locus mismatch. The patients received a median UCB nucleated cells (NC) of 5.5 x 10(7)/kg body weight [range (4.7 - 7.7) x 10(7)/kg] and CD(34)(+)CD(38)(-) cells 2.8 x 10(5)/kg [range (0.6 - 4.5) x 10(5)/kg] and CFU-GM 1.09 x 10(5)/kg [range (0.24 - 230.00) x 10(5)/kg]. The conditioning regimen consisted ofbusalphan 16 - 20 mg/kg, cyclophosphamide 180 - 200 mg/kg, horse antithymocyte globulin (ATG) 90 mg/kg or rabbit anti-lymphocyte globulin (ALG) 25 mg/kg and melphalan of 90 mg/m(2) was added to four cases. Cyclosporine A (CsA) alone was administered in 3 cases, and a combination of CsA and methotrexate in the other 2 cases. RESULTS: Four cases were engrafted, but one rejected and recurred thalassemia state at day 60 past UCBT. One case had autologous reconstitution. The reconstitutions were found in all patients with neutrophils 0.5 x 10(9)/L at day 19 (14 - 22), platelets > 20 x 10(9)/L at day 37 (21 - 63). Two cases developed grade I and II acute GVHD respectively. None developed chronic GVHD. Three patients survived disease-freely (survival with ex-thalassemia state). The median follow-up was 18 months, the probability of survival was 100%. CONCLUSION: Allo-UCBT could reconstitute hematopoiesis with low frequency and mild grade of GVHD. It is an effective treatment for thalassemia.

[Studies on chemical constituents from the flower of Citrus aurantium].

The chemical constituents from the flower of Citrus aurantium were studied. 11 compounds were isolated and identified including neohesperidin(I), synephrin(II), 5,8-epidioxyergosta-6,22-dien-3 beta-ol(III), adenosine(IV), asparagine(V), tyrosine(VI), valine(VII), isoleucine(VIII), alanine(IX),beta-sitosterol(X) and beta-daucosterol(XI).

[Inhibitory effect of antisense c-myb cDNA on the growth of asynchronous glioma cells in vivo].

OBJECTIVE: To examine the therapeutic effect of oncogene c-myb antisense oligodeoxynucleotide (AON) on rat C6 glioma in vivo. METHODS: C6 glioma cells were implanted into the subcutis of nude mice, the animals were randomly divided into three groups (12 mice respectively) when the tumor grew about 10 mm in size, and the mice with subcutaneous tumor foci were treated with sense oligodeoxynucleotide (SON), AON and normal saline. Three animals of each group were killed after treatment at 4, 8, 12 and 16 days respectively. The dynamic growth manifestations, features, histopathological changes and apoptosis of the glioma in each group were observed. RESULTS: AON suppressed the growth of C6 glioma cells, the suppression rates being 66.7%, 71.4%, 72% and 73% at 4, 8, 12, 16 days respectively (P < 0.05). The changes in weight were concurrent with the variations of tumor volume, the tumor weight of AON groups was significantly decreased (P < 0.05). The immunohistochemical assay showed the expression of c-myb proto-oncogenes was significantly decreased in AON groups (P < 0.05). The flow cytometry analysis found the apoptosis cell percentage of AON groups to be 13.4%, 27.1%, 46.1% and 48.4% at 4, 8, 12 and 16 days respectively, and this percentage of AON groups was about 3.5 times the apoptosis cell percentage of SON and control groups at the corresponding time. In addition, noticeable inflammatory infiltration and calcification were observed after the treatment with AON. CONCLUSION: The above findings indicate that c-myb gene might be associated with the suppression of carcinogenesis in brain glioma, and oncogene c-myb might be chosen as a target for antisense gene therapy of gliomas.

[Eight cases of rhabdomyosarcoma in head and neck].

OBJECTIVE: To improve the clinical diagnosis and treatment level of rhabdomyosarcoma in head and neck. METHOD: It was based on the documents that 8 cases of rhabdomyosarcoma in head and neck had been diagnosed by pathological diagnosis. RESULT: The 8 cases were treated by operation. Among 8 patients, 5 took radiotherapy after operation. 1 took chemotherapy after operation. We kept in touch with 7 patients, the survival rate of 5 years was 57.1% (4/7). CONCLUSION: We must pay attention to the painful or painless phyma in the otolaryngology and head and neck. It has the possibility of being rhabdomyosarcoma. We diagnose the rhabdomyosarcoma through biopsy and immuohistochemistry and with the references from the examination of CT, MR. In order to improve the survival rate of patients of rhabdomyosarcoma, we should use the comprehensive treatment, including operation, radiotherapy, and chemotherapy.

[Purification and characterization of 2-carbonyl reductase from marine bacteria Bacillus sp].

ANADPH-dependent 2-Oxoaldehyde reductase was isolated and purified from a marine bacteria Bacillus sp. The purification procedure involved ammonium sulfate fractionation and Q Sepharose FF, Hydroxyapatite, Sephadex G-100 column chromatographies. The specific activity of the purified enzyme was increased by 141.1 folds over crude extract and the recovery yield was 11.4%. 2-Oxoaldehyde compounds were found to be speciall good substrates. The optimum pH of the enzyme activity was 6.2-6.6. The Km coefficient for 3-deoxyglucosone was 2.5 mmol/L. The molecular weight of the enzyme was estimated to be 33 kD The enzyme activity is stable below 30 degrees C and pH 5.0-8.0. EDTA, beta-mercaptoethanol and dithiothreitol enhanced the enzyme activity. On the other hand, the enzyme activity was partially lost by idoacetic acid or N-ethylmaleimide.

[Effect of manganese exposure on brain development in postnatal mice].

Effects of manganese (Mn) exposure on brain development in postnatal mice were studied. The results were as follows: (1) The gain of body weight, brain weight and the ratio of brain weight to body weight in the high dose Mn-exposed group was significantly decreased. (2) In the Morris Water Maze Test, the average latency discovering the hidden platform was obviously increased every day during the first 5 days in the high Mn-exposed group, but the latency probing the quadrant in which the hidden platform was located previously was reduced at the 6th day. (3) The protein content of brain tissue was significantly decreased in the high Mn-exposed mice, but the activity of AChE in brain was decreased. (4) The immunoreactivity of glial fibrillary acid protein (GFAP) and the average relative density of GFAP-positive products in hippocampus (area CA3) of both low and high Mn-exposed group, especially the high dose ones, was significantly higher than that of the control group.

[Amplifying variable region gene of light chain of monoclonal antibody against human retinoblastoma by PCR].

OBJECTIVE: To acquire the variable region gene of light chain of monoclonal antibody against human retinoblastoma. METHODS: Total RNA were extracted from hybridoma cells secreting specific monoclonal antibody(McAb) against human retinoblastoma(RB), then transcripted reversely into cDNA with random primers. The variable region of the light chain(VL) gene fragments were ampliflied using polymerase chain reaction(PCR) method. Agrose gel electrophoresis was confirmed. RESULTS: 1.5% agrose gel electrophoresis indicated that VL gene was about 340 base pairs. CONCLUSION: The light chain variable region gene of the McAb against human RB was amplified successfully, which lays a good basis for construction of a recombinant antibody.

[The measurement of multifocal electroretinography].

PURPOSE: Discussing the testing procedure of multifocal electroretinography(ERG). METHODS: The testing procedure of 131 multifocal ERG records was reviewed. RESULTS: Some variable factors existed in the measurement of multifocal ERG. These factors included the subject's co-operation, the mounting of electrode, the fixation of tested eye and adjustment of focus. The good records of multifocal ERG depend upon the skill of the operator. CONCLUSION: The testing procedure of multifocal ERG must be standardized so as to ensure the reliability and repeatability of the results.

Histopathological study of trabeculum after excimer laser trabeculectomy ab interno.

PURPOSE: To study the clinical manifestations and histopathologic changes of trabeculum after excimer laser trabeculectomy ab interno (ELT), and to investigate the mechanisms of ELT in reducing intraocular pressure. METHODS: ELT was performed on ten rabbit eyes and postoperative responses were documented. Corneoscleral tissue samples were harvested consecutively each week until the 5th postoperative week and these samples were examined under light and electrical microscopy. RESULTS: Mild stimulation signs were present postoperatively in nine of ten eyes, but no serious complications were experienced. Obvious inflammation was observed in one rabbit eye as a result of iris damage during the surgical manipulations. Local fractures on the trabecular meshwork and openings into Schlemm's canal were detected in all tissue samples under light microscope. Mitochondria were found to be turgescent and dilated like vacuoles and endoplasmic reticula were found to be dilated under electrical microscope in the early postoperative period. Later, all trabecular cells returned normal and no fibroblast cells were ever detected. CONCLUSIONS: Permanent openings through trabecular meshwork into the inner wall of Schlemm's canal can be created with ELT. The outflow resistance of aqueous humor can be reduced with these openings and intraocular pressure can be controlled thereafter.

The electroretinogram in patients with retinal vascular occlusion.

PURPOSE: To measure and analyze the electroretinogram (ERG) in patients with retinal vascular occlusion. METHOD: Fifty-eight cases (59 eyes) of retinal vascular occlusion and the fellow eyes of 47 cases were tested with Ganzfeld ERG. The scotopic and photopic ERG, and oscillatory potentials were tested according to the ERG standard of ISCEV. RESULTS: The abnormal rates of OPs and b wave were higher in retinal vascular occlusion. The abnormal ERG appeared mostly in CRVO among four types of retinal vascular occlusion. The comparisons of some amplitudes and latencies between the fellow eyes and the affected eyes showed statistically significant difference. CONCLUSION: The oscillatory potentials and b waves have important roles in evaluating the retinal function of patients with retinal vascular occlusions. There are some clinical significance for comparing ERG between the affected eye and the fellow eye.

[Correlation of JNK/SAPK activity and the production of ICAM-1 on renal mesangial cells in vitro].

OBJECTIVE: To observe the activity of c-Jun N-terminal kinase/stress-activated protein kinase (JNK/SAPK) and assess the correlation between JNK/SAPK activity and ICAM-1 expression on mesangial cells in mesangial proliferative glomerulonephritis(MSPGN). METHODS: Seven patients with MSPGN and 6 controls were selected for this study. JNK/SAPK activity was detected by immunoprecipitation and western blotting; RT-PCR was used to detect ICAM-1 mRNA expression, and flow cytometry was used to analyze cell surface ICAM-1 expression. RESULTS: Either in spontaneous condition or under the stimulus of TNF alpha, the activity of JNK/SAPK on mesangial cells in vitro in MSPGN was higher than that of the controls(P < 0.05); the activity of JNK/SAPK in MSPGN was found to have a positive correlation with the levels of ICAM-1 mRNA and protein. The levels of ICAM-1 mRNA and protein were increased by TNF alpha stimulation while the levels ICAM-1 mRNA and protein were decreased by DMAP inhibition (P < 0.01). CONCLUSION: JNK/SAPK may have been aberrantly activated in MSPGN, which may involve in the overexpression of ICAM-1 in MSPGN.

[Lymphocyte subpopulation, interleukin-2 and high affinity interleukin-2 receptor expression in primary nephrotic syndrome].

OBJECTIVE: To understand the cellular immune response during relapse and remission stage in primary nephrotic syndrome (PNS). METHODS: We applied the radioligand binding assay (RBA), the bioactivity measurement of IL-2, and monoclonal antibody sensitized red blood cell method to evaluate the expression of IL-2R, the production of IL-2 and T lymphocyte subsets of PBMC from 22 patients suffering from PNS, 17 patients with PNS in remission, and 25 normal subjects matched age and sex. RESULTS: CD3, CD4, CD8, IL-2, IL-2R in relapse were significantly lower than those in control (P < 0.05); all above except CD8 in remission patients were significantly higher than those in relapse, but lower than those in control (P < 0.05). CONCLUSION: This study suggested that the cell mediated immunity (CMI) during acute nephrotic phase decreased; the deficiency of CMI of PNS in remission improved a lot when compared with the acute phase, but still not recovered completely; the deficiency of CMI with PNS in remission might be one of the causes of easily recurring of PNS.

[Defect analysis in mercury cadmium telluride by Raman scattering].

Telluride precipitates in HgCdTe(MCT) and Raman scattering peaks of Hg vacancies are reported. We also analyze the relationship between the crystal surface perfection of MCT and its second-order scattering peaks. Intense bands at 125 and 145 cm-1 confirm them to be precipitates, and there is 2 cm-1 shift to high frequency in comparison with intense bands of normal trigonal Te. This shows the compressive stress exists around Te precipitates. The weak Raman peak at 108 cm-1 arise from Hg vacancies. This peak is only found in p-type MCT and MCT including p-n junction, and is not found in n-type MCT. The intensity of the second-order scattering peaks in MCT is very sensitive to its crystal surface perfect. To good surface perfect, these peaks are very distinct, and to poor surface perfect, they are not distinct. The intensity measurement of 2LO(T) mode in the second-order scattering peaks can provide useful information for surface analysis in crystal preparation process.

[Study on the detaining effect of hypocrelin a in the cancer cell using microspectrum].

Microspectra of mouse lymph cancer cells which one is cultured with HA and the other is cultured without HA have obtained in the range of 20,000-12,000 cm-1. The result shows tha two microspectra are quite different. This point out the cancer cell has changed because of HA. In the microspectrum of cancer cell cultured with HA, we have got the fluorescent peak of HA. This prompts HA has detained in the cancer cell. A new method which studies cells after being affected by drugs using microspectrum has also being developed.
