This study analyzed the feasibility of, and the strategy for, DNA ploidy analysis of cervical condyloma and intraepithelial neoplasia by a computerized digital imaging system. Paraffin-embedded tissue provided satisfactory single-cell preparations for DNA ploidy analysis after enzyme digestion and additional procedures. Negative endocervical curettings and normal squamous mucosa were used as internal diploid controls. With suitable controls, 21 (81%) of the 26 aneuploid lesions were identified as such in the single-cell preparations. The remaining five lesions (not recognized as aneuploid in the single-cell preparations) had ploidy levels between 2.08n and 2.30n and required DNA measurements on 12-microns sections. Criteria for these DNA measurements were defined: specimens intended for DNA ploidy analysis should contain abnormal epithelium of at least 3 mm to 4 mm in greatest dimension and should be accompanied by diploid controls, such as endocervical curettings or normal ectocervical squamous mucosa. With a combination of single-cell preparations and 12-microns tissue sections, it was possible to accurately determine the DNA ploidy patterns of the cervical lesion specimens obtained by punch biopsies. Available evidence suggests that ploidy analysis can provide useful diagnostic and prognostic information.

Filamentous phage Cf16 undergoes a unique neolysogenic infectious cycle in Xanthomonas campestris pv. citri and generates stable lysogens. In contrast, we have isolated a distinctive unstable lysogen, designated LW. This new lysogen segregated spontaneously into three nonparental types, each with a unique combination of colony morphology, phage-producing capacity, and phage genome content. In a given population of LW lysogens the segregation frequency of these types varied randomly with drastic fluctuations from experiment to experiment. Heat and UV treatment failed to increase the segregation frequency with a magnitude matching the sporadic intrinsic fluctuations. The characteristics of an LW lysogen were transmitted through the phage it released. Designated as Cf16-v1, this phage, which produced clear instead of turbid plaques as its progenitor Cf16, was exceedingly unstable in the host cell. Only 4% of the infected cells, rather than 95% as in the case of Cf16, retained the phage genome and completed the neolysogenization process. The prophage integration site on the host chromosome, however, appeared to be the same for both phages.
A beta-thalassemia gene from a patient with beta+-thalassemia in South China was cloned and sequenced. A four nucleotide (TCTT) deletion in codons 41 and 42 was found. This is the most common beta-thalassemia gene in South China and the first mutant identified on China mainland.
In order to initiate a program of prenatal diagnosis for the prevention of beta-thalassemia in China, we have begun systematic studies of the beta-thalassemia mutations among the Chinese. DNA polymorphisms in the beta-globin gene cluster were examined in 46 beta-thalassemia chromosomes. Six different haplotypes were observed. One beta-thalassemia gene associated with a new haplotype was cloned and sequenced. The mutation was a single base substitution (A----G) at position -29 within the highly conserved proximal promoter element (the "TATA" box). This mutation was not observed previously in the Chinese. The beta-thalassemia genes were further screened with oligonucleotide probes specific for all known mutations in the Chinese. Five mutations were identified and accounted for 35 beta-thalassemia alleles.

Sections of human placental villi tissue and carcinoma cell lines (SMMC-7721, (MGC-803 and Kato-111; were examined using the indirect immunoperoxidase histological technique to further investigate the localization of the tumor-associated placental protein (PP) in human placental tissue and the expression of PP on carcinoma cell lines. PP was prepared from placenta in the first trimester. Placental tissue sections exposed to PP antisera showed a marked staining of the trophoblast cells, especially in syncytiotrophoblast. The strongly stained area was localized to the cell membrane of syncytiotrophoblast. In contrast, the nucleus and the cytoplasm of syncytiotrophoblast were stained weakly. The specific PP antisera also stained the carcinoma cell lines. Among different carcinoma cell lines, the degree of staining of liver carcinoma cell line (SMMC-7721) was higher than that of gastric carcinoma cell lines (MGC-803 and Kato-111). In the carcinoma cell lines the presence of antigen similar to PP suggests that oncogene (one) occur in human trophoblast cells as well as mice trophoblast cells.
Polypeptides synthesized in the cytoplasm of eukaryotes are generally initiated with methionine, but N-terminal methionine is absent from most mature proteins. Many proteins are also N alpha-acetylated. The removal of N-terminal methionine and N alpha-acetylation are catalyzed by two enzymes during translation. The substrate preferences of the methionine aminopeptidase (EC 3.4.11.x) and N alpha-acetyltransferase (EC 2.3.1.x) have been partially inferred from the distribution of amino-terminal residues and/or mutations found for appropriate mature proteins, but with some contradictions. In this study, a synthetic gene corresponding to the mature amino acid sequence of the plant protein thaumatin, expressed in yeast as a nonexported protein, i.e., lacking a signal peptide, has been used to delineate the specificities of these enzymes with respect to the penultimate amino acid. Site-directed mutagenesis, employing synthetic oligonucleotides, was utilized to construct genes encoding each of the 20 amino acids following the initiation methionine codon, and each protein derivative was isolated and characterized with respect to its amino-terminal structure. All four possible N-terminal variants--those with and without methionine and those with and without N alpha-acetylation--were obtained. These results define the specificity of these enzymes in situ and suggest that the nature of the penultimate amino-terminal residue is the major determinant of their selectivity.
We have cloned and sequenced the predominant germline V kappa gene segment expressed by B cells of strain A origin that synthesize antibodies with specificity for Ars. In hybridomas synthesizing anti-Ars antibodies, this V kappa gene segment (V kappa IdCR) has been found exclusively associated with the J kappa 1 gene segment without exhibiting junctional sequence variation. Sequence comparisons of the germline V kappa IdCR gene with expressed derivatives reveals that the latter frequently contain somatically introduced amino acid replacements. Taken together with results of previous structural analyses, these results show that the predominant population of IdCR+ V regions elicited in the secondary immune response is encoded by one or two combinations of V gene segments, has little junctional diversity, and is extensively diversified by somatic mutation in both heavy and light chains.
In this prospective study of 31 patients, paired receptor levels from biopsy and mastectomy specimens were found to be significantly correlated on biochemical assay. Despite this fact, 6 of 20 tumors that were estrogen receptor-positive on biopsy, had a negative assay from the mastectomy specimen, indicating the need for a fresh tumor specimen. Immunocytochemical staining with monoclonal antibody H222 Spy, an antibody specific for the estrogen receptor, was evaluated in 34 biopsy and mastectomy specimens. With a sensitivity of 85 percent and specificity of 93 percent, this technique compares favorably with the standard biochemical assay.

It is reported that unilateral grafts of neonatal striatal tissue protect the recipient from the lethal aphagia and adipsia produced by bilateral intrastriatal injection of 10 nmol of kainic acid in rats. It is shown that neither adult striatum nor neonatal tissue from other sites have the same lifesaving effect and that the salutary effect of the graft is dependent upon graft survival. Grafts from a histoincompatible donor are apparently rejected, leading to the death of the recipient. Cyclosporine inhibits rejection thereby enabling recipient survival. It is postulated that the graft exerts a neurohumoral influence that protects the striatum from the toxic effect of kainate.
The earliest outgrowth of nerve fibers from identified spinal neurons labeled with horseradish peroxidase (HRP) was traced along surgically rearranged pathways in the central nervous system (CNS) of Xenopus embryos. Parts of the CNS were misaligned or inverted rostrocaudally by grafting a segment of labeled spinal cord in place of the same or different spinal cord segment of an unlabeled embryo or by joining two rostral half embryos (head-to-head) or two caudal half embryos (tail-to-tail), one half of which was derived from a labeled embryo in each combination. Donor embryos were labeled by injection of HRP into a selected blastomere at the 16- or 32-cell stage. Host embryos were unlabeled. Grafts from labeled donors to unlabeled host embryos were made at early neural tube stages before outgrowth of any nerve fibers had started (Jacobson and Huang, 1985). Routes taken by labeled nerve fibers growing into unlabeled CNS were observed at later stages, and the rates of nerve fiber elongation were calculated. Labeled nerve fibers were normal in appearance, and elongated without branching, at normal rates (22-71 micron/h). In head-to-head and tail-to-tail embryos and in embryos with inverted spinal cord grafts, nerve fibers continued elongating without branching in the direction opposite to normal in the CNS. Many fibers reached lengths that were far greater than normal. No reorientation of such maldirected nerve fibers was seen. These results indicate that nerve fiber elongation is not guided by axially polarized pathway cues or markers and that nerve fibers do not grow to predetermined lengths. However, neurites preferred to grow along stereotyped nerve fiber pathways even when forced to grow in the wrong direction or when confronted with nonneural tissue.
A Chinese beta +-thalassemia gene in a new haplotype was chosen for cloning and sequencing. The mutation identified was an A-G transition at position -29 in the TATA box of the beta-globin gene. This mutation has not been seen previously in Chinese but has been documented in American blacks on a different chromosomal background. This observation provides further evidence for independent origins of the same mutation in distinct ethnic groups.
Pituitary glands from neonatal donors were transplanted to the median eminence of hypophysectomized adult rats. Rats with transplants were then treated for 2 weeks with the immunosuppressive drug cyclosporine. For 5 weeks thereafter, blood was drawn at regular intervals for determination of serum thyroxine, prolactin, and luteinizing hormone. Cyclosporine-treated recipients of grafts with minor histocompatibility differences had normal levels of thyroxine and prolactin, whereas untreated animals did not. In addition, the treated animals responded to oophorectomy with a marked elevation in serum luteinizing hormone. This evidence indicates that cyclosporine enables successful transplantation across a minor histocompatibility barrier. It also suggests that these grafts interact with the hypothalamus. Transplantation across a major histocompatibility barrier was unsuccessful even in the presence of cyclosporine.
To make possible prenatal diagnosis of beta-thalassemia in China and Southeast Asia by direct detection of mutant beta-globin genes, we have determined the spectrum of mutations producing the disorder in this region of the world. Seventy-eight beta-thalassemia genes from Chinese and Southeast Asians were randomly obtained, and the relevant mutation was characterized in 76 (98%) of them. Seven different point mutations were found among the 78 genes studied. Of these seven beta-thalassemia alleles, two constitute 62%, and two others account for 29% of the total. Since only four alleles make up 91% of the mutant genes, prenatal diagnosis of beta-thalassemia in China and Southeast Asia should be feasible by simplified techniques for direct detection of point mutations.
