Tetrandrine potently inhibits herpes simplex virus type-1-induced keratitis in BALB/c mice.

This study investigated the effect of tetrandrine (TDR) on experimental herpes simplex keratitis (HSK) in mice. BALB/c mice were divided as follows: Group 1, untreated; Group 2, acyclovir (ACV)-treated from day 0 postinfection; Group 3, ACV-treated from day 7; Group 4, TDR-treated from day 0; and Group 5, TDR-treated from day 7. All mice were infected in the right cornea with herpes simplex virus (HSV) type I. TDR 30 mg/kg and ACV 120 mg/kg were administered intraperitoneally daily. The mice were observed for 14 days postinfection. Clinical inflammatory reactions and ocular histopathology were analyzed. The herpes specific antibody response and the delayed type hypersensitivity (DTH) response were studied. Of the 22 untreated mice, 16 developed HSK (incidence, 72.7%). TDR given from day 7 reduced the HSK incidence to 8.5% (p < 0.01); the incidence of HSK was 45.4% in mice treated with TDR from day 0 (p > 0.05). Systemic ACV given from day 0 inhibited HSK development (p < 0.01); ACV given from day 7 resulted in an HSK incidence of 50% (p > 0.05). The specific anti-HSV antibody response in the serum of mice treated with TDR or ACV either from day 0 or day 7 was significantly less than that of untreated mice (p < 0.01 and p < 0.05, respectively), and TDR treatment suppressed DTH responses to HSV (p < 0.05). Systemic TDR administered after HSV inoculation of the cornea significantly modulates murine HSK development at least partly by modifying the host immune/inflammatory response to the virus.

[99mTc-MIBI myocardial perfusion imaging in patients with diabetes].

Seventy-four patients with diabetes were taken on both stress and rest 99m-Tc-MIBI SPECT imaging: 44 patients (59.5%) were found having myocardial ischemic disorders, including 28 cases of myocardial ischemia, 7 cases of myocardial infarction and 9 cases of both conditions. 113 segments were involved and 46% were located in the posterior and inferior walls, indicating that the right coronary artery is most susceptive to ischemia. Diabetic patients complicated hypertension and/or hyperlipidemia were at higher risk of either early occurrence of coronary disease. 99mTc-MIBI SPECT imaging is helpful in early diagnosis and therefore is significant in directing.

[Ventriculo-peritoneal shunts for thirty-two patients with traumatic hydrocephalus].

Thirty-two patients with traumatic hydrocephalus were treated by ventriculo-peritoneal shunts. Twenty-two patients were improved (70%). Among them, seventeen patients showed satisfactory results (53%): ventriculo-peritoneal shunts have good effects and few complications; this method is easy to manipulate for traumatic hydrocephalus. We discussed the important points in operations.

[A study on the mechanism of taurine postponing the aging process of human fetal brain neural cells].

The relationship between taurine and aging of brain neural cells was studied by using primary human fetal brain neural cells cultured in serum-free medium. The results indicated that the activity of superoxide dismutase (SOD) was significantly higher in taurine added group than the control group cultured for 6, 9 and 15 days (P < 0.05). With different taurine levels, the SOD activities of 50 mg/L and 200 mg/L taurine groups were significantly higher than the control group cultured for 6 days (P < 0.05). After 3-day culture, the activity of glutathione peroxidase (GSH-Px) in the taurine added group was significantly higher than the control group (P < 0.05). The membrane lipid fluidity of neural cells decreased with time in control group, but was stable in the group treated with taurine. The fluidity was more stable in the groups treated with 50, 200 and 400 mg/L groups cultured for 6 days, compared with the control group (P < 0.05 or 0.01). The present study showed that taurine is necessary for maintaining membrane lipid fluidity of neural cells in proper levels, and plays an important role in postponing the aging process of brain neural cells.

[A study on the relationship between bone mineral content and the tissue calcium level in rats].

The relationship of bone mineral density (BMD) and bone mineral content (BMC) of femur bone with the tissue and blood calcium contents was studied with 98 Wistar rats (15-week old). The results indicated that the body length, body weight, bone width, BMC, BMC/weight, BMD/weight and BMD/length in male were markedly different from those in female animals. The BMD of alive rats was significantly correlated with the total calcium intake, length, weight and calcium retention rate (P < 0.05). BMC was significantly related to sex, length, weight, liver calcium content and total calcium intake. Multiple regression analysis indicated that the animal body length, weight, total calcium intake and calcium content in heart were positively correlated with the BMD of alive animals. The animal body length and plasma calcium content were positively correlated with BMD of separated bones after the animals were sacrificed. The present results indicated that the BMC and BMD of alive rats could reflect the calcium status and bone mineral accumulation in the body because of the significant correlation with the growth rate, calcium retention rate, muscle and plasma calcium contents, but not with the separated bone BMD or BMC.

[Endovascular treatment of brain arteriovenous malformation].

Eighteen patients with arteriovenous malformations (AVMs) of the brain were treated with endovascular techniques between 1993 and 1996. The following angiograms showed complete obliteration of the nidus in 2 patients, 70%-90% reduction in nidus volume in 5, 50%-70% reduction in 3, and < 50% in the remaining 3. Two patients suffered from moderate neurologic deficits after embolization. One week after the treatment, 4 patients with superficial AVMs underwent surgery, and 2 with deep-situated AVMs received gamma-knife therapy. 14 patients were followed up, suffered from more frequent epilepsy, and 1 had intracranial haemorrhage 6 months after embolization. A few AVMs can be cured by endovascular embolization. In regard to some large AVMs or AVMs situated at important functional areas, embolization can facilitate surgery and radiotherapy afterwards.

[A clinical review of 700 cases of coronary artery bypass grafting].

To assess the changing trends in patients profiles, operative procedures, and the result of coronary artery bypass grafting (CABG) we reviewed the clinical data of 700 consecutive patients who had anisolated CABG (471, 67.3%), CABG combined with left ventricular aneurysm (170, 24.29%), CABG and valve procedures (48, 6.9%), CABG and ventricular septal defect repair (16, 2.3%) at the Fuwai Hospital. The patients were divided into group A (recent three years) and group B (before 1993). The incidence was significantly increased (P < 0.05) in diabetes, hypercholesterolemia, ventricular dysfunction, left main stem coronary artery disease and three vessles lesion. There was a high incidence of hypertension (41.3%), old myocardiac infarction (36.4%), combined left ventricular aneurysm (24.29%) and IABP needed (9.4%), however no significantl difference was notes between the two groups. Internal mammary artery was used in group A (25.6%) and group B (2.6%) (P < 0.005). The hospital mortality (group A, 2.7% and group B, 9.6%) and perioperative myocarial infarction (group A, 3.2% and group B, 9.0%) decreased significantly (P < 0.005). We conclude that despite a high incidence of hypertension, diabetes, hypercholoesterolemia, old myocardial infarction, ventricular dysfunction and diffuse coronary artery disease the patients tolerate CABG surgery well and obtain a good result.

[Color Doppler imaging diagnosis of ocular tumor].

OBJECTIVE: To study the characteristics of color Doppler imaging (CDI) of ocular tumors for the diagnosis and differential diagnosis of the tumors. METHODS: 92 cases with ocular tumor (105 eyes) were examined by CDI to observe the vasculature of ocular tumors, and the various parameters of blood vessels and blood flow in the lesions were measured. SAS software was used to analyze the results. RESULTS: Blood flow signal can be found in retinoblastoma, melanoma of choroid, angioma of choroid, angioma of optic nerve papilla, Coats disease, persistent hyperplastic primary vitreous (PHPV) and metastatic tumors of choroid, but we can not find any blood flow signal in choroid osteoma and melanocytoma of optic nerve papilla. The blood flow parameters of central retinal artery in choroidal melanoma and that of ophthalmic artery, central retinal artery and posterior ciliary artery in angioma of choroid are lower than normal (P < 0.05). In the comparison of the blood velocity in the posterior ciliary artery between choroidal melanoma and choroidal angioma, it was found that the former is much higher than the latter (P < 0.05). CONCLUSION: CDI is very important in the diagnosis and differential diagnosis of ocular tumors.

[The change of calcitonin gene-related peptide positive fibers in inflammatory pulps].

Deep cavities were prepared on rats' molars, and fresh carious dentin was enclosed in to establish animal pulpitis model. Immunohistochemical method was used to observe the distribution of calcitonin gene-related peptide (CGRP) positive nerve fibers in rats' normal dental pulps and inflammatory pulps at 4d, 8d, 14d respectively. CGRP positive fibers were found throughout the normal pulps, a large number of fibers penetrated into dentinal tubules. In 4d group, CGRP positive fibers became denser and the number of fibers penetrating into dentinal tubules also increased. In 8d group, a necrotic zone could be found in the coronal pulps, lots of fibers accumulated around the necrotic zone. In the radical pulps, CGRP positive fibers formed multiple arborizations. 14d postinflammation, all the pulps were necrotic, and the periapexs were richly innervated by CGRP positive fibers. The results suggesting that CGRP may play a role in the inflammation and repair of dental pulp.

[Mutiple factors analysis of the effects of excimer laser photorefractive keratectomy].

PURPOSE: To observe the effect of excimer laser photorefractive keratectomy (PRK) correcting myopia and astigmatism, and to study the related factors. METHOD: PRK was performed with the aim to correct myopia errors from 1.00 to 16.00 diopters on 529 myopia eyes (306 patients). All eyes were divided into two groups: Group I: -1.00-6.00 D(294 eyes); group II: -6.25-16.00 D(235 eyes). All the patients were followed up for 3 to 12 months. RESULTS: At postoperative 12 months the rates of uncorrected visual acuity more than or equal to 10/20 or 20/20 were 97.5%, 77.5% in group I, and 80.3%, 32.8% in group II respectively; 92.5% of eyes were within +/- 1.00 diopter of the desired emmetropia in group I, but 42.6% in group II; The influential factors of changes of postoperative diopters was the severity of corneal haze, methods of removing corneal epithelium, intraocular pressure, the central corneal thickness. CONCLUSION: The excimer laser PRK is regarded to be a very predictable, safe, stable, and effective method to correct myopia up to -6.00 D. Prior to operation, the plan would be disigned according to diopters, ages, corneal thickness intraocular pressure. During operation, the corneal epithelium bad better to be removed by laser.

[Observation on acupoint electric potential in pilots under vestibular stimulation].

To study the possibility of using acupoint electric potential as an index for evaluation of pilot's reaction to vestibular stimulation, electric potential was measured on both sides of left and right Neiguan points and Zusanli points in 80 pilots. Rotation chair was used as the method of stimulation. The results showed that changes of acupoint electric potentials were not significant (P >0.05) during 5 min uniform rotation. While the changes under head motion stimulation were significant (P<0.05 or 0.01) as compared with the pretest values. Relationship between symptoms and the corresponding acupoint potential changes remain to be studied further.

[Changes of electrogastrogram in pilots under vestibular stimulus].

The active electrogastrogram (EGG) of 81 pilots under vestibular stimulus was recorded and analysed by the method of running spectrum analysis. The subjects were randomly divided into group A and B. The results showed: that changes in dominant frequency instability coefficient (DFIC), dominant power instability coefficient(DPIC), tachygastria, bradygastria and normal slow wave components (NSW) of EGG occured under vestibular stimulus. These parameters of EGG may be used as indices to reflect the function of autonomic nervous system under vestibular stimulus. No difference of those parameters had been found between group A and B during the period of per-rotating, and the period of rotation.

[EGG and nausea under vestibular stimulus].

81 pilots were exposed to vestibular stimulus with Coriolis acceleration. They were divided into two groups, the first group consisted of 38 pilots with nausea symptoms, the second group 43 pilots without nausea symptom. Dominant frequency instability coefficient (DFIC), dominant power instability coefficient (DPIC), Tachygastria, Bradygastria and normal slow wave components (NSW) of the electrogastrogram (EGG) were analyzed. The results showed that Bradygastria and NSW of EGG were related to nausea symptom of pilots under Coriolis stimulation.

[Detection of human papillomavirus types 16, 18 DNA related sequences in bronchogenic carcinoma].

In order to study the relationship between human papillomavirus (HPV) and bronchogenic carcinoma, "high-risk" HPV 16, 18 DNA sequences were detected in samples from 50 lung cancer patients, 18 patients with benign pulmonary diseases and 4 fetal lung tissues by polymerase chain reaction (PCR) and dot-blot hybridization using biotin-labelled probes. The results showed that HPV 16, 18 DNA related sequences were found in 32% of lung cancer specimens, including 10 cases of HPV 16, 5 cases of HPV 18, 1 case of both types. 48.2% (13/27) of the squamous cell carcinomas were shown to be positive for HPV 16, 18 DNA; in addition, two adenocarcinomas and one small cell carcinoma were positive for HPV 16 DNA. No specimen from benign diseases and fetal lung tissue showed positive result. Most of HPV DNA positive cases had a history of heavy smoking. These results implicated that primary bronchogenic carcinomas may be related to HPV infection.

[Analysis of single cells by capillary electrophoresis].

A review with 46 references is given on the application of capillary electrophoresis to the analysis of single cells. Injection modes, detectors and future prospects of this microseparation technique for single-cell analysis are described in detail.

[Evaluation of capillary chromatographic columns packed by electrokinetic packing method].

In this paper, a method for electrokinetic packing capillary columns is reported. A higher column effeciency was obtained by performing electrochromatography on electrokinetic packing columns. The highest column efficiency in number of theoretical plate per meter was more than 200000, corresponding to reduced plate height less than 2. The reproducibilities of the same column in different intervals and different columns prepared from the same or different batches were compared. The relative standard deviations of the number of theoretical plate and retention time were less than 10% and 8%, respectively. The results indicated that high column efficiency and good reproducibility can be obtained on these new capillary packed columns.

[Effects of different form calcium on growth and tissue calcium level in rats].

In order to solve the problem of calcium deficiency and to look for economic and efficient source of calcium, the effects of calcium carbonate, active calcium and calcium lactate on growth, development and tissue calcium level in rats were compared. Fifty-six 3-week old weaning rats were fed with calcium deficient diet (containing vitamin D 500 IU per kg diet) for 3 weeks, and then were divided into four groups randomly with 14 rats in each group, half male and half female. The diet of control group (A) was the basic diet, while the three experiment diets were supplemented with calcium carbonate (B), active calcium (C) and calcium lactate (D) (3000 mg calcium per kg diet), respectively. The experiment term was 12 weeks. The results showed that the body weight and length of calcium supplemented group were significantly higher than that of control group (P<0.05). Among the calcium supplemented groups, no significant differences were observed except the difference of body length between the group D and the group A in female. Calcium deficiency dramatically hindered the development with reduced dietary intake and decreased food consumption efficiency. The calcium levels in plasma, red blood cells and liver were significantly higher in the supplemented groups than that in the control group (P<0.05), however, there was no difference among the supplemented groups. No significant difference of calcium levels in muscle and heart was observed among all groups. Based on needs for reaching the RDA with additional 400 mg/d from present calcium status in Chinese population, the calcium carbonate is the most economic one and the ideal calcium source for supplementation.

Modulation of ATP-sensitive and large-conductance Ca++-activated K+ channels by Zeneca ZD6169 in guinea pig bladder smooth muscle cells.

ZD6169, the S-enantiomer of the racemic N-(4-benzoylphenyl)-3,3,3-trifluoro-2-hydroxy-2-methylpropionamide , was reported to possess a mechanoinhibitory effect on bladder detrusor smooth muscle through its ability to activate the KATP-sensitive K+ channel (KATP). In this study, the effects of ZD6169 and its racemic mixture on the whole-cell KATP and large-conductance CA+2-activated K+ (BKCa) currents in isolated smooth muscle cells from guinea pig bladder detrusor were examined by the patch-clamp technique. ZD6169 produced a multiple stimulatory and inhibitory effect on the KATP current. With a threshold effective concentration of 0.5 microM, it produced a glyburide-sensitive activation that reached a maximum between 5 and 10 microM. ZD6169 at concentrations greater than 20 microM markedly inhibited KATP channel, which resulted in a bell-shaped concentration-response relationship. Over a similar concentration range, ZD6169 caused a sizable stimulation of the BKCa current. All effects were readily reversible. Consistent with the data in whole-cell recordings, ZD6169 activated single BKCa channel in inside-out patches by increasing its open-state probability. Several lines of evidence showed that the opening of BKCa channel was Ca+2 independent. The activity profile of the racemic ZD6169 on KATP and BKCa channels had remarkable resemblance to that of ZD6169. Our results indicate that ZD6169 exerts diverse effects on multiple types of K+ channels. A combination of the absence of KATP channel activation and the presence of BKCa channel stimulation by ZD6169 at higher concentrations may be responsible, in part, for its reported weaker cardiovascular side effects than cromakalim.
