Eighty patients with Gan Yang Shang Kang Zheng (GYSKZ) and the related Zheng were questioned with three psychological scales: Type A Behavior Inventory, STAI and SDS and plasma AVP (arginine ressopressin) levels were determined by RIA. The TAB, S-AI, T-AI scores and plasma AVP levels of the patients with GYSKZ increased consistently and the changes of all parameters in different diseases in the same Zheng were similar. The results suggest that in patients with GYSKZ, type A behavior is the dominant personality and anxiety is the chief emotional disorder. Patients with GYSKZ are in the state of high level of psychological stress. Patients with GYSKZ and GHSYZ have some common pathophysiological basis.

The state of basement membrane(BM) and dendritic cells(DC) were observed by immunohistochemical methods with the use of anti-laminin antibody, anti-type IV collagen antibody, anti-S-100 protein antibody in 68 cases of laryngeal carcinoma and 21 cases of non-cancer laryngeal tissues. The results showed that among 68 patients with laryngeal carcinoma 39 cases had discontinuous BM. A continuous, linear BM was found mostly in normal larynx, atypia hyperplasia and papilloma, only 3 cases had discontinuous BM. 43 positive DC were detected in 68 cases of laryngeal carcinoma. There were rarely positive DC in non-cancer laryngeal tissues. The poorer the differentiation of carcinoma, the more the discontinuous BM and the lesser the number of DC. The discontinuous BM and DC(-) were seen more in advanced stages of carcinoma. The five-year survival rate in patients with continuous BM was significantly higher than that in patients with discontinuous BM, the five-year survival rate in patients with DC positive was also higher than that in patients with DC negative; the survival rate was highest in patients with continuous BM and DC positive. Our results indicated that the detection of the state of BM and dendritic cells were of value to the prognostic analysis in patients with laryngeal carcinoma.
Pepper roots contain 0.39% of essential oil. By GC-MS method, 44 peaks and 22 compounds were obtained and identified. The major component was trans-caryophyllene. The trace metal content analysis showed that the heavy metals were under the safety level.
OBJECTIVE: To study the characteristics of plasma norepinephrine (NE) and epinephrine (E) contents alteration and vegetative nervous function disturbances in patients with Liver Syndrome of TCM. METHODS: Subjects were selected by combination of disease diagnosis and Syndrome Differentiation. Plasma NE and E contents were determined by high performance liquid chromatography (HPLC) and electrochemical detector. The vegetative nervous function were assessed by comprehensive analysis of sympathetic and parasympathetic hyperfunction symptoms, physiologic indexes and physical loading tests. RESULTS: Patients of Liver Excess Syndrome had an elevated plasma NE and E contents and with the vegetative nervous system disturbance mainly hypersympathetic. On the contrary, patients of Liver Deficiency Syndrome had an lowered plasma NE and E contents and with the vegetative nervous system disturbance mainly hyperparasympathetic. CONCLUSION: Plasma NE and E contents and vegetative nervous function disturbance can be taken as the auxiliary laboratory diagnostic indexes for Syndrome Differentiation of Liver Excess and Liver Deficiency.
The relationship between electronystagmogram(ENG) and intensity of vestibular stimulation was observed in 62 pilots. 31 pilots received low intensity vestibular stimulation and the other 31 pilots received higher intensity vestibular stimulation. Electronystagmogram were recorded and analysed. The results showed that there was no relationship between the ENG parameters and stimulation intensity, but both slow phase and fast phase velocities and amplitudes were lower under Coriolis acceleration stimulus, while all of them were higher under angular acceleration and angular stimulations. It was also found that the fast phase amplitude of ENG was related to motion sickness symptoms of the subjects.
Possible effects of vestibular stimulation autonomic nerve function in pilots were studied by observing changes of Heart Rate Variability (HRV). Time domain and frequency domain analyses demonstrated that the primarily averages, standard deviation of the HRV averages(SD), least or greatest value, deviation of greatest minus least value, interrelated averages, square root of the mean of the sum of squares of differences between adjacent R-R intervals (RMSSD), total power(TP), low frequency(LF), high frequency(HF), LF/TP, HF/TP, and LF/HF all had significant increase or decrease (P <0.05 - <0.01) and the degrees of changes were related to the intensity of the stimulations.
OBJECTIVE: To evaluate the degree of pulmonary hypertension in patients with congenital heart disease (CHD) by using radionuclide pulmonary perfusion imaging. METHOD: The pulmonary perfusion in 81 patients with left to right cardiac shunts was examined by scintigram with macroaggregates of (99m)Tc-labeled human serum albumin (PPS). PPS was analyzed for (1) lung regional radionuclide count (LRC), (2) right upper and lower lung count ratio (RULR), (3) kidney count ratio (KCR). The results were compared with those of cardiac catheterization exam. RESULT: There were correlation between mean pulmonary arterial pressure (mPAP), total pulmonary resistance (TPR), and PPS (KCR, RULR with mPAP gamma = 0.25, 0.35; LRC, KCR with TPR gamma = -0.34, 0.29). Five patients repeated PPS test after early operation. There were no changes in PPS. CONCLUSION: The degree of reversibility of pulmonary hypertension in patients with CHD could be quantitatively analysed by PPS.
OBJECTIVE: To observe the changes of ocular anterior segment and the relation between the intraocular pressure (IOP) and these changes after the topical application of cycloplegia using ultrasound biomicroscope (UBM), especially to observe the changes of ciliary body thickness and ciliary body-lens distance. METHODS: The quantitative measurement of UBM and the measurement of IOP had been done in 48 normal eyes before and after the topical use of 2% homatropine solution. The results were statistically analyzed. RESULTS: In the parameter reflecting the change of the anterior chamber angle after using 2% homatropine, the trabecular iris angle decreased, the angle open distance 250 decreased, the iris thickness 1 increased, the anterior chamber became deeper and the iris-lens contact distance became shorter, and all these differences were statistically significant (P < 0.05). In the parameter reflecting the changes of ciliary body, the ciliary body thickness and the ciliary process thickness decreased, the scleral ciliary body angle increased, the iris-zonule distance decreased and the ciliary body-lens distance increased, and all these differences were significant statistically (P < 0.05). The IOP increased from 2.35 +/- 0.46 kPa to 2.43 +/- 0.47 kPa after the topical use of 2% homatropine, but the differences were not statistically significant (P > 0.05). CONCLUSIONS: After the topical application of cycloplegia, the anterior chamber becomes deeper, the anterior chamber angle becomes narrower, the ciliary body becomes thinner and moves backward and the ciliary body-lens distance increases. By using UBM the structure of ocular anterior segment in its moving state can be observed and these structures can be measured quantitatively. The ultrasound biomicroscopic imaging has its advantages in the morphological study of ocular anterior segment.
8 allele and group specificity at HLA-DRB1 locus were genotyped in 159 unrelated individuals of Liaoning Han, with polymerase chain reaction-sequence specific primers. The range of gene frequencies was from 0.02201-0.23899. Heterozagosity was 83%. 33 out of 36 theoretical genotypes were identified and the result conformed to Hardy-Weinberg equilibrium law. The present method can be used not only in paternity test, individual identification, but also in clinic graft matching between donor and recipient as well as the study of HLA.
Endogenous and exogenous opioids can influence and modulate neuronal and glial cell function via an opioid receptor mediated mechanism, leading to either protection or damage of the brain. Opiates such as morphine have been postulated to promote the progression of HIV-1 and the development of secondary opportunistic infections. Kappa opioid receptor ligands, on the other hand, may play a neuroprotective role. More studies are needed to delineate how opioids exert their effects on glial cells as well as neurons with the goal of developing new therapeutic approaches for neurodegenerative disease.
In light of recent reports linking K+ channel modulation with food intake and macronutrient preference, we investigated the effect of anorectic agent dexfenfluramine (d-FF), a 5-HT reuptake inhibitor and releasing agent, on the delayed rectifier K+ (DRK) channels in rat lingual taste cells using the patch-clamp technique in whole-cell configuration. In a concentration-dependent manner, d-FF caused a reduction of the DRK currents in taste cells with an IC50 of 30.5 microM. Other anorectics that promote 5-HT activity such as fenfluramine, sibutramine and m-chlorophenylpiperazine (a specific 5-HT2C receptor agonist) produced inhibition of DRK currents of a similar pattern with a respective IC50 of 69.0, 8.6 and 95.4 microM. The actions of all compounds had rapid onset and were readily reversible. The inhibitory effects were not secondary to their stimulation of 5-HT, because direct application of 5-HT up to 1 mM did not alter DRK current. In addition, d-FF-induced current reduction was not prevented by either the 5-HT synthesis inhibitor p-chlorophenylalanine or 5-HT receptor antagonist metergoline. d-FF was also tested in cardiac ventricular myocytes that are reportedly abundant in DRK channels and was found to depress the DRK currents concentration-dependently with an IC50 of 250.9 microM. These results indicate an important pharmacological role for d-FF as an inhibitor of the DRK channels. The common inhibitory effect on DRK channels in oral taste cells and cardiac cells by this class of compounds might contribute to the anorectic and some of the detrimental cardiovascular effect associated with long-term exposure.
BACKGROUND: The present study investigated the effects of drinking milk or water before viewing an optokinetic rotation drum on the severity of motion sickness and abnormal gastric myoelectric activity. METHODS: There were 27 subjects who drank 300 ml of milk or water, or drank nothing, in three separate sessions with 2-d intervals between every two sessions. Subjects in each session then viewed a vertically striped optokinetic rotation drum for 16 min. Symptoms of motion sickness and electrogastrograms (EGGs) were measured. RESULTS: Subjects reported no significant difference in symptoms of motion sickness among sessions of drinking milk, water, or nothing. EGG recordings indicated that subjects in sessions of drinking milk and water significantly increased normal gastric myoelectric activity at EGG 3 cycles per minute (cpm). Subjects in all three sessions increased abnormal EGG activity at 4-9 cpm during drum rotation period, however, the ratios of EGG 4-9 cpm of drum rotation and baseline periods were not significantly different among sessions. CONCLUSION: These results demonstrated that drinking milk or water before viewing an optokinetic rotation drum exhibited no effect on the severity of symptoms of motion sickness and abnormal gastric myoelectric activity.
PURPOSE: The present study investigated the relationship of the frequency of nystagmus to the severity of optokinetic rotation-induced motion sickness. METHODS AND RESULTS: Eighty-seven subjects viewed a vertically striped, rotating drum for 16 min. Subjects' electrooculograms, ratings of vection, and subjective symptoms of motion sickness (SSMS) were measured during the drum rotation period. Subjects' electrogastrograms (EGGs) were recorded throughout the baseline and drum rotation periods. The ratios of the spectral intensity of EGG 4-9 cycles per minute (cpm) between drum rotation and baseline periods were calculated. The results indicated that the frequency of nystagmus was positively correlated with the ratings of vection (r = 0.40, p < 0.0001), scores of SSMS (r = 0.61, p < 0.0001), and ratios of EGG 4-9 cpm spectral intensity between drum rotation and baseline periods (r = 0.54, p < 0.0001). CONCLUSION: These results demonstrated that more rapid eye movement indexed by higher frequency of optokinetic nystagmus are related to the development of symptoms of motion sickness and gastric tachyarrhythmia.
Nitric oxide (NO) plays an important role in host defense as well as cell injury within the CNS. In contrast to rodent species, human astrocytes are the major glial source of NO. Although interleukin (IL)-1 stimulates astrocyte inducible NO synthase (iNOS) expression, the mechanism is poorly defined. In the present study using primary human fetal astrocyte cultures, we found that IL-1 beta stimulated activation of nuclear factor kappa B (NF-kappa B) within 2 h, iNOS mRNA expression at 8 h, and maximal NO production by 5 days post-treatment. This IL-1-induced activation of astrocyte iNOS was suppressed by pyrrolidine dithiocarbamate, an inhibitor of NF-kappa B activation, suggesting involvement of a NF-kappa B mechanism.
Structure-activity relationships of a lead hydroxamic acid inhibitor of recombinant human stromelysin were systematically defined by taking advantage of a concise synthesis that allowed diverse functionality to be explored at each position in a template. An ex vivo rat model and an in vivo rabbit model of stromelysin-induced cartilage degradation were used to further optimize these analogs for oral activity and duration of action. The culmination of these modifications resulted in CGS 27023A, a potent, orally active stromelysin inhibitor that blocks the erosion of cartilage matrix.
The expression and activity of receptor tyrosine kinases (RTK) at the cell surface can be modulated by several different pathways including the proteolytic release of the extracellular domain as a soluble receptor. We investigated the regulation of tie receptor expression, an orphan RTK restricted to cells of hematopoietic and endothelial lineages, on primary human endothelial cells and a stably transfected Chinese hamster ovary (CHO) cell line. Tie was expressed in cells as a doublet of 135 and 125 kD; the 135-kD band represented mature cell surface receptor containing sialic acid and N-linked oligosaccharide residues, whereas the 125-kD band represented an intracellular pool of immature receptor. Phorbol 12-myristate 13-acetate (PMA) had dramatic effects on tie expression at the cell surface. Within 15 minutes of PMA treatment, the 135-kD band disappeared from the cell surface and was accompanied by the appearance of a 100-kD band in cell supernatants. The 100-kD band continued to accumulate in the media throughout the duration of PMA treatment during which mature tie receptor was undetectable on the cell surface by fluorescence-activated cell sorting (FACS) or in cell lysates by immunoblot analysis. Using specific antibodies, this 100-kD species was shown to be a soluble form of the tie receptor containing the extracellular domain. PMA-dependent release of soluble tie was mediated through the activation of protein kinase C (PKC); soluble tie was not released in the presence of PKC inhibitors, an inactive PMA analog, or following the downregulation of PKC through chronic PMA treatment. These results indicate that tie receptor expression on endothelial cells is regulated by the release of a soluble extracellular fragment following activation of PKC. Parallel pathways regulating c-kit, tumor necrosis factor (TNF), and colony-stimulating factor (CSF) receptor expression suggest that the release of extracellular receptor fragments represents an alternative mechanism through which cells modulate responses to growth factors and cytokines.
